ABSTRACT

Soybean is an important food and oil crop widely cultivated globally. However, water deficit can seriously affect the yield and quality of soybeans. In order to ensure the stability and increase of soybean yield and improve agricultural water use efficiency (WUE), research on improving drought tolerance and the efficiency of water utilization of soybeans under drought stress has become particularly important. This study utilized the drought-tolerant variety Heinong 44 (HN44) and the drought-sensitive variety Suinong 14 (SN14) to analyze physiological responses and transcriptome changes during the gradual water deficit at the early seed-filling stage. The results indicated that under drought conditions, HN44 had smaller stomata, higher stomatal density, and lower stomatal conductance (Gs) and transpiration rate as compared to SN14. Additionally, HN44 had a higher abscisic acid (ABA) content and faster changes in stomatal morphology and Gs to maintain a dynamic balance between net photosynthetic rate (Pn) and Gs. Additionally, drought-tolerant variety HN44 had high instantaneous WUE under water deficit. Further, HN44 retained a high level of superoxide dismutase (SOD) activity and proline content, mitigating malondialdehyde (MDA) accumulation and drought-induced damage. Comprehensive analysis of transcriptome data revealed that HN44 had fewer differentially expressed genes (DEGs) under light drought stress, reacting insensitivity to water deficit. At the initial stage of drought stress, both varieties had a large number of upregulated DEGs to cope with the drought stress. Under severe drought stress, HN44 had fewer downregulated genes enriched in the photosynthesis pathway than SN14, while it had more upregulated genes enriched in the ABA-mediated signaling and glutathione metabolism pathways than SN14. During gradual water deficit, HN44 demonstrated better drought-tolerant physiological characteristics and water use efficiency than SN14 through key DEGs such as GmbZIP4, LOC100810474, and LOC100819313 in the major pathways. Key transcription factors were screened and identified, providing further clarity on the molecular regulatory pathways responsible for the physiological differences in drought tolerance among these varieties. This study deepened the understanding of the drought resistance mechanisms in soybeans, providing valuable references for drought-resistant soybean breeding.

ABSTRACT

Job resources can buffer the deleterious effect of adverse work environments. Extant studies on the interaction pattern between job resources and adverse environments were confined to the diathesis stress model. This traditional perspective has received the challenge from the differential susceptibility model and the vantage sensitivity model. Additionally, stress reactivity may be one of the important job resources at the personal biological level, but its moderating role was short of empirical research. This study aimed to examine how stress reactivity interacts with work environments in predicting job burnouts among 341 Chinese hospital female nurses. This study selected job control and job support representative of supportive environments and psychological demands representative of an adverse environment and the cortisol content in 1-cm hair segment as a biomarker to assess individual's stress reactivity in 1 month. The nurses self-reported their work environments and job burnouts and provided 1-cm hair segments closest to the scalp. Hair cortisol content was measured with high-performance liquid chromatography-tandem mass spectrometry. The interaction pattern was examined with multiple linear regressions and the analysis of region of significance (RoS). The regression revealed that the interaction of hair cortisol content with job control could positively predict professional efficiency among nurses, with psychological demands could negatively predict emotional exhaustion, and with coworker support could negatively predict professional efficiency. The RoS analysis revealed that nurses with high cortisol levels had not only significantly higher professional efficiency than those with low cortisol levels in high job control but also significantly lower professional efficiency in low job control. Nurses with high cortisol levels had significantly higher emotional exhaustion than those with low cortisol levels in low psychological demands. Nurses with low cortisol levels had not only significantly higher professional efficiency than those with high cortisol levels in high coworker support but also significantly lower professional efficiency in low coworker support. The interaction patterns of stress reactivity with both job control and coworker support were consistent with the differential susceptibility model, but the interaction between stress reactivity and psychological demands supported the vantage sensitivity model.

Extant studies mostly focused on the buffering role of social and external organizational resources and personal mental resources. However, there is no research exploring the moderating role of personal physiological resources (e.g., stress reactivity). The present study is aimed at examining the interactive effect of emotional labor and stress reactivity on job burnout. The present study utilized cortisol content in a 1 cm hair segment as the biomarker of total stress reactivity in one month. The participants were 229 female hospital nurses randomly recruited from city hospitals, China. They self-reported their emotional labor strategies and job burnout syndromes and provided 1 cm hair segments closest to the scalp two weeks later after the survey. Hair cortisol content was determined with high-performance liquid chromatography-tandem mass spectrometry. The results revealed that hair cortisol can moderate the associations of surface acting with emotional exhaustion and personal burnout; of deep acting with emotional exhaustion, depersonalization, and personal burnout; and of expression of naturally felt emotions with professional inefficacy. In particular, nurses with high cortisol levels not only showed higher emotional exhaustion than those with low cortisol levels under high surface acting but also showed lower emotional exhaustion under low surface acting. A similar situation was true for nurses' emotional exhaustion and depersonalization in the context of deep acting. Nurses with low hair cortisol levels not only showed higher professional inefficacy than those with high hair cortisol levels under low expression of naturally felt emotions but also showed lower professional inefficacy under high expression of naturally felt emotions. Additionally, nurses with high hair cortisol levels showed lower personal burnout than those with low hair cortisol levels under low surface acting or high deep acting. In summary, the interaction pattern between stress reactivity and emotional labor was varied with the nature of emotional labor strategy and job burnout.

ABSTRACT

OBJECTIVES: The purpose of this study was to explore whether two types of emotional labor, surface acting and deep acting, are related to hair cortisol concentration among kindergarten teachers. METHODS: Surface acting and deep acting over the last month were measured with the Chinese version of the emotional labor scale in 43 kindergarten teachers. Hair samples with 1 cm in length were cut from their posterior vertex region to represent cortisol excretion over one month. Cortisol concentrations were analyzed with high-performance liquid chromatography-tandem mass spectrometry. RESULTS: Positive association of emotion labor with hair cortisol concentration was significant for surface acting (r = 0.34, p < 0.05) and not significant for deep acting (r = 0.14, p > 0.05). CONCLUSIONS: More surface acting showed to be associated stronger with stress responses or higher HPA axis activity.

ABSTRACT

OBJECTIVES: The purpose of this study was to explore whether work strain (i.e., job demands and job control) and subjective need for recovery (NFR) after work are related to measured concentration of cumulative cortisol. METHODS: Participants were 43 teachers recruited from kindergarten. They self-reported their NFR, job demands and job control over the last month. NFR was measured with the NFR scale. Job demands and control were assessed with the Chinese version of the Job Content Questionnaire. Hair cortisol was used to represent cumulative cortisol excretion. Hair cortisol concentration (HCC) was measured with high-performance liquid chromatography-tandem mass spectrometry. RESULTS: No significant correlations were found between job demands or job control and HCC. NFR was significantly and inversely correlated with HCC (r = -0.41, p = 0.006). The inverse association between NFR and HCC remained significant when age and job demands and job control were controlled for (p = 0.02). CONCLUSIONS: The activity of the hypothalamic-pituitary-adrenal axis declines with the accumulation of NFR after working time.

ABSTRACT

OBJECTIVE: The present study aims to investigate the relationship between effort-reward imbalance and hair cortisol concentration among teachers to examine whether hair cortisol can be a biomarker of chronic work stress. METHODS: Hair samples were collected from 39 female teachers from three kindergartens. Cortisol was extracted from the hair samples with methanol, and cortisol concentrations were measured with high performance liquid chromatography-tandem mass spectrometry. Work stress was measured using the effort-reward imbalance scale. RESULTS: The ratio of effort to reward showed significantly positive association with hair cortisol concentration. CONCLUSION: The cortisol concentration in the system increases with the effort-reward imbalance. Measurement of hair cortisol can become a useful biomarker of chronic work stress.

ABSTRACT

OBJECTIVES: To investigate the clinical effect and therapeutic mechanism of Nanmiqing capsule made of rheum palmatum, leech, astragalus memberanaceus on patients with chronic prostatitis(CP). METHODS: Seventy-six CP cases were treated with Nanmiqing, while 32 CP cases were treated with Qianliekang as a control. The changes of EPS were observed pre- and post-treatment. The rat model of CP got by Xiaozhiling inducing were treated with Nanmiqing and Qianliekang respectively. The concentration of endothelin, TXB2, 6-keto-PGF1 alpha and SOD, IgG, IgA in plasma were measured pre- and post-treatment, meanwhile, pathological changes of prostate tissues were observed. RESULTS: The total effective rate was 89.47% in treatment group, which was significantly higher than 71.88% in the control group (P < 0.01). Experimental study for CP rats showed that the Nanmiqing was more effective medicine than Qianliekang (P < 0.01). CONCLUSIONS: Nanmiqing was an effective medicine for CP. The mechanism of clearing heat and resolving toxin, activating blood and removing stasis and reinforcing Qi in chinese medicine could be the explanation of the useful treatment including three therapentic ways.
