ABSTRACT

Studies have indicated that glyphosate induces endocrine disruption and may adversely affect the male reproductive system. However, evidence of its effects on ovarian function is poorly understood so far, making further studies necessary on the mechanisms of the glyphosate toxicity in the female reproductive system. The aim of this work was to evaluate the effect of a subacute exposure (28 days) to the glyphosate-based formulation Roundup® (1.05, 10.5 and 105 µg/kg b.w. of glyphosate) on steroidogenesis, oxidative stress, systems involved in cell redox control and histopathological parameters in rat ovaries. Hence we quantify plasma estradiol and progesterone by chemiluminescence; non-protein thiol levels, TBARS, superoxide dismutase and catalase activity by spectrophotometry; gene expression of steroidogenic enzymes and redox systems by real-time PCR; and ovarian follicles by optical microscopy. Our results demonstrated that oral exposure increased progesterone levels and the mRNA expression of 3ß-hydroxysteroid dehydrogenase. Histopathological analysis revealed a decrease in the number of primary follicles and an increase in the number of corpus luteum in rats exposed to Roundup®. An imbalance of the oxidative status was also evidenced by decreasing the catalase activity at all groups exposed to the herbicide. Increased lipid peroxidation and gene expression of glutarredoxin and decreased of glutathione reductase were also observed. Our results indicate that Roundup® causes endocrine disruption of hormones related to female fertility and reproduction and changes the oxidative status by altering antioxidant activity, inducing lipid peroxidation, as well as changing the gene expression of the glutathione-glutarredoxin system in rat ovaries.

ABSTRACT

The corn grains from the extremities of corncob are known to have a spherical shape and the grains from the center of corncob have higher length and lower thickness. It is understood that these differences in grain dimensions can affect post-harvest processes and the properties of the grains. Therefore, the objective of this study was to evaluate the effects of drying temperature (60, 80, and 100 °C) of corn from the center and extremities of corncob on drying parameters, protein and starch properties, and carotenoid profile. At 60 °C, the drying rate and effective moisture diffusivity of corn from the center and extremities of the corncob did not differ. However, at 80 and 100 °C these parameters were higher in the corn from the center. Corncob part and separation did not significantly affect corn pasting properties. However, they caused significant changes in the properties of the grain proteins, mainly the reduction of the solubility and inactivation of the lipase enzyme, and the reduction of the lutein and ß-carotene contents in the grains from the center and separated after drying. The results of this research show the need to evaluate the effects of these drying conditions during grain storage. In addition, the implementation of an industrial separation step and/or the development of new corn cultivars with more homogeneous grains should be studied.

ABSTRACT

Senecio spp. is one of the most frequent plant-related poisonings in cattle. Its ingestion generates the disease seneciosis, characterized by hepatic damages. Liver biopsies and serum markers dosage are tools used in diagnosis; however, many breeding cattle are undiagnosed. MicroRNAs are non-coding RNA, stable in biological fluids. Their difference in expression levels may indicate the presence of the poisoning. We analyzed the miRNA profiling to identify potential diagnostic biomarkers for Senecio brasiliensis poisoning. The expression of miR-21, miR-885, miR-122, miR-181b, miR-30a, miR-378, and let-7 f were evaluated in the serum of exposed cattle. At least one histological change was found in liver and lower quantity of albumin and high AST and ALP were also detected. MiRNAs miR-30a, miR-378, miR-21, miR-885, and miR-122 presented significantly higher expression in intoxicated animals than in healthy animals. Furthermore, miR-122, miR-885, and, especially, miR-21 signatures demonstrated high sensitivity and specificity, with potential application for detecting poisoning.

ABSTRACT

Our aim with this study was to evaluate the consumption, performance, quantitative characteristics of carcasses, biochemical profile, plasma levels of ghrelin and leptin, expression of the receptor for ghrelin (GHS-R1a) in the hypothalamus and duodenum, and the number of goblet cells in the duodenum of calves subjected to milk volume restriction and supplemented with 2-hydroxy-4-(methylthio)butanoic acid (HMTBa). We used 21 Holstein mixed-breed calves, aged between 3 and 15 d with an average weight of 36.8 kg, and housed in pens with troughs for hay, concentrate, and water. The study included two consecutive experimental periods (first period [P1] and second period [P2]) of 21 d each, with 7 d of adaptation to the diet and facilities. The calves were distributed in a completely randomized design in three treatments with seven repetitions. 1) Control: 6 liters of milk/d during P1 and 6 liters of milk/day during P2; 2) RES (milk restriction): 3 liters of milk/day during P1 and 6 liters of milk/day during P2; and 3) RES + HMTBa: 3 liters of milk/day during P1 and 6 liters of milk/day during P2 + 3.3 g of HMTBa/day in both periods. HMTBa was supplied in milk, and the amount of concentrated ration and hay provided and leftovers were recorded daily to estimate dry matter (DM) and crude protein consumption. Mean daily weight gain (DWG), final weight (FW), and feed conversion (FC) were obtained at the beginning and at the end of each 21-d period. Plasma concentrations of ghrelin and leptin, triglycerides, total protein, urea, lactate, creatinine, alkaline phosphatase, and cholesterol were measured for P1 and P2 at the end of each 21-d period. At the end of P2, animals were slaughtered; sections of the duodenum were collected to evaluate the expression of GHS-R1a and quantity of goblet cells; hypothalamus was used to evaluate the expression of GHS-R1a; rumen was used to evaluate the thickness of epithelium and keratin and the density, height, and width of ruminal papillae. In P1, total DM consumption, FW, DWG, glucose, and triglycerides were lower in the RES and RES + HMTBa groups (P < 0.001). In P2, there was an improvement in the FC of the RES + HMTBa group (compared with Control and RES groups) and a lower urea concentration in the RES group (compared with Control and RES + HMTBa groups) (P < 0.001). No differences were observed among groups regarding hormonal concentrations, histological parameters, and GHS-R1a expression in the duodenum and hypothalamus. Therefore, milk restriction combined with HMTBa supplementation promoted greater compensatory gain by a mechanism independent of changes in GHS-R1a expression and hormone levels of ghrelin and leptin.

ABSTRACT: This study aimed to assess liver damage and interferon-stimulated gene 15 (ISG15) blood expression as a consequence of embryonic signaling on maternal recognition of pregnancy in beef cattle presenting natural ingestion of Senecio spp. Epidemiological aspects, as the presence of the plant, associated to gamma glutamyl transferase (GGT) activity can be used as Senecio spp. poisoning diagnosis. Maternal recognition of pregnancy period occurs when the embryo secretes interferon tau (IFNT) to signal its presence to the mother and eventually extend corpus luteum (CL) lifespan. In our study, liver damage was determined by concentration serum GGT, cytological and histopathological examinations. Reproductive status was evaluated by concentration of progesterone, CL diameter and ISG15 mRNA expression on Day 19 following fixed-time artificial insemination (FTAI). Cows were categorized into two groups based on concentration of GGT: Group 1 (GGT 30U/L) and 2 (GGT>31U/L). No difference on body condition scores was observed. All the cows presented liver damage based on cytology and histopathological exams. Cows from the Group 1 had higher pregnancy rate, presenting larger CL diameter and greater concentration of progesterone. Interestingly, ISG15 mRNA expression had no difference between Groups 1 and 2, even presenting difference in pregnancy status. These findings suggest embryonic loss beyond Day 19. It suggests late embryonic mortality may be associated to liver insufficiency. In conclusion, liver injury and/or concentration of GGT does not alter ISG15 expression on blood neutrophils, however cows presenting lower concentration of GGT ( 30U/L) had increased pregnancy status. Therefore, the concentration of GGT allow us to screen liver status and foresee a successful pregnancy in beef cattle.


RESUMO: O objetivo deste estudo foi avaliar a lesão hepática e a expressão sanguínea do gene estimulado por interferon 15 (ISG15) durante a sinalização embrionária, no reconhecimento materno da gestação, em bovinos de corte apresentando ingestão natural de Senecio spp. Fatores epidemiológicos, como a presença da planta, associados à atividade da gama glutamil transferase (GGT) podem ser utilizados como diagnóstico da intoxicação por Senecio spp. O reconhecimento materno da gestação ocorre quando o embrião secreta interferon tau (IFNT) para sinalizar sua presença à mãe. Em nosso estudo, a lesão hepática foi determinada pela concentração sérica de GGT, pelos exames citológicos e histopatológicos. O estado reprodutivo foi avaliado pela concentração de progesterona, diâmetro de corpo lúteo (CL) e expressão de mRNA ISG15 no Dia 19 após a inseminação artificial em tempo fixo (IATF). As vacas foram separadas em dois grupos com base na concentração de GGT sanguíneo: Grupo 1 (GGT 30U/L) e Grupo 2 (GGT>31U/L). Não foi observada nenhuma diferença no escore de condição corporal entre os grupos. Na citologia e nos exames histopatológicos todas as vacas apresentaram lesão hepática. As vacas do Grupo 1 apresentaram maior taxa de prenhez, maior diâmetro do CL e maior concentração de progesterona. Diferente do esperado, a expressão do mRNA ISG15 não foi diferente entre os Grupos 1 e 2, mesmo apresentando diferença na taxa de prenhez. Esses achados sugerem perda embrionária após o Ddia 19. Isso demonstra que a mortalidade embrionária tardia pode estar associada à insuficiência hepática. Dessa forma, conclui-se que a lesão hepática e/ou concentração de GGT não altera a expressão de ISG15 nos neutrófilos sanguíneos, porém vacas com menor concentração de GGT ( 30U/L) apresentaram maiores taxas de prenhez. Assim, a concentração de GGT nos permite avaliar a saúde hepática e prever uma gestação bem-sucedida em bovinos de corte.

The objective of this study was to evaluate the effects of drying temperature (30, 50, 70, and 90 °C) and genotype (yellow floury corn, white floury corn, and yellow flint corn) on the morphology and technological and thermal properties of corn starch. The white and yellow genotypes with floury endosperm (soft) had spherical starch granules, while the granules of the yellow flint genotype were polyhedral. White floury corn showed higher extraction yield, higher resistance during granules' swelling before the physical collapse, as it was not affected by the increase in drying temperature, and at 90 °C it presented higher breakdown and energy necessary for gelatinization. There were peak viscosity and gelatinization enthalpy reduction on starch from yellow floury corn and yellow flint corn according to the increase in drying temperature. Drying temperatures below 50 °C are indicated for all genotypes studied. White floury corn is indicated due to the higher extraction yield presented. Yellow floury corn is indicated for application in chilled and frozen products due to lower starch retrogradation. The yellow flint corn is indicated for application as a thickening agent due to higher gel hardness.

Synthetic preservatives are widely present in processed foods, but most of them have carcinogenic potential, requiring the development of new natural alternatives such as fruit extracts, for microbial control. The objective of the study was to evaluate the chemical characterization, antioxidant, and antimicrobial activity of the sugar apple pulp (Annona squamosa L.). Physicochemical characteristics were evaluated, an extract was prepared, and its antioxidant activity by DPPH method and antimicrobial by disk diffusion. Minimal inhibitory concentration and minimum bactericidal concentration against strains of Salmonella typhimurium, Escherichia coli, Listeria monocytogenes, and Staphylococcus aureus were evaluated. The physicochemical analysis revealed that sugar apple pulp had 75.0% moisture, 3.0% ash, 4.0% protein, 0.2% lipids, 3.3% fibers, and 14.5% carbohydrates. The antioxidant activity of the extract by the DPPH method was 20.6%. The pulp extract from the sugar apple had inhibition zone for Staphylococcus aureus, satisfactory inhibitory effect against Staphylococcus aureus, Escherichia coli, Listeria monocytogenes, and Salmonella Typhimurium, but did not present a bactericidal effect. Sugar apple pulp presents adequate levels of nutrients and potential for food application due to its microbiological activity and antioxidant properties.


Los conservantes sintéticos están ampliamente presentes en los alimentos procesados, pero la mayoría tienen potencial carcinogénico, lo que requiere el desarrollo de nuevas alternativas naturales para el control microbiano, como los extractos de frutas. El objetivo del estudio fue evaluar la caracterización química, la actividad antioxidante y antimicrobiana de la pulpa de manzana de azúcar (Annona squamosa L.). Se evaluaron las características fisicoquímicas, y se evaluó su actividad antioxidante mediante el método DPPH y antimicrobiano por difusión en disco, concentración inhibitoria mínima y concentración bactericida mínima contra cepas de Salmonella Typhimurium, Escherichia coli, Listeria monocytogenes y Staphylococcus aureus. El análisis fisicoquímico reveló que la pulpa de manzana de azúcar tiene 75.0% de humedad, 3.0% de cenizas, 4.0% de proteínas, 0.2% de lípidos, 3.3% de fibras y 14.5% de carbohidratos. La actividad antioxidante del extracto por el método DPPH fue del 20.6%. El extracto de pulpa de la manzana de azúcar tenía zona de inhibición para Staphylococcus aureus, efecto inhibidor satisfactorio contra Staphylococcus aureus, Escherichia coli, Listeria monocytogenes y Salmonella Typhimurium, pero no presenta efecto bactericida. La pulpa de manzana de azúcar presenta niveles adecuados de nutrientes y potencial para la aplicación de alimentos debido a su actividad microbiológica y propiedades antioxidantes.

ABSTRACT

ABSTRACT Objective Explore scientific publications on the use of gene therapy in dentistry. Methods A bibliographic survey was carried out with articles published in the last eleven years, available on online platforms that had the highest correlation to the proposed theme. Results Since the 1960s, researchers have attempted to establish guidelines for treatments using gene therapy; however, it was only in the mid-1990s that their use in science was authorized. Gene therapy in dentistry has the ability to alter and improve genetically, through stem cells in dental tissues associated with recombinant viral vectors, therapeutic protocols against diseases that do notrespond to conventional treatments. Conclusion The perspectives of dentistry concerning this resource have been positive, mainly in the reestablishment and regeneration of tissues in pulp pathologies, periodontopathies, bone disorders, orofacial pain among others. It is known that this form of therapy still requires more research, however, in the near future, it may be a safe first option treatment in hospital and outpatient settings.


RESUMO Objetivo Explorar publicações cientificas acerca do uso da terapia gênica na Odontologia. Métodos Foi realizado um levantamento bibliográfico com artigos dos últimos dez anos disponíveis em plataformas online que continham maior correlação com o temaproposto. Resultados Desde os anos 60 pesquisadores tentam estabelecer diretrizes para tratamentos com a terapia gênica, no entanto, somente em meados dos anos 90 o seu uso na ciência foi autorizado. A terapia gênica em odontologia tem a capacidade de alterar e melhorar geneticamente, através de células-tronco em tecidos dentais e sua associação a vetores viras recombinados, protocolos terapêuticos contra enfermidades que não tem resposta aos tratamentos convencionais. Conclusão A perspectiva da odontologia diante deste recurso tem sido positiva, principalmente no reestabelecimento e regeneração de tecidos em patologias pulpares, periodopatias, distúrbios ósseos, dores orofaciais entre outros. Sabe-se que esta terapia ainda requer muitas pesquisas, entretanto em um futuro próximo poderá ser um tratamento de primeira escolha e de forma segura nos ambientes hospitalares e ambulatoriais.

The conventional prolonged parboiling process results in high operation cost and grain darkening, which may limit consumption. Moreover, residue generation by rice industries is another challenge. The objective of this study was to evaluate the use of microwave irradiation during soaking and gelatinization stages of parboiling rice. Processing time, colorimetric profile, broken and nongelatinized grains, sucrose and glucose content, free 5-hydroxymethyl-2-furfural, and residual phosphorus were evaluated. As the soaking and gelatinization times during microwave treatments increased, the colorimetric parameters increased; however, the values were lower than those with the conventional process. Regardless of soaking time, a decrease in broken and nongelatinized grains was obtained by using the lowest steaming time (5 min). Additionally, lower residual phosphorus content was found in soaking water (10 and 20 min) when using microwave irradiation. Under favorable conditions, a reduction in the levels of broken and nongelatinized grains, residual phosphorus, and color changes was observed, indicating that microwave irradiation may be more beneficial than conventional parboiling. PRACTICAL APPLICATION: Parboiling requires a high volume of water and soaking time, which leads to high costs, underutilization of infrastructures, and high residue in the water after processing. The rapid parboiling process involves the use of microwaves during the soaking and gelatinization stages. The main advantages of the microwave parboiling process include reduced processing time, ranging from 83% to 95%, higher gelatinization, greater yield, reduced darkening, and reduced residual phosphorus in the effluents by 60%. This report can aid industries in streamlining their processes, thereby providing a high-quality, lower cost, and environmentally safe product.

ABSTRACT Celtis iguanaea (Jacq.) Sarg., Cannabaceae, is popularly used in the treatment of diabetes mellitus. However, chemical and pharmacological investigations are lacking. In this study, we investigated the effects of the hydroalcoholic extract from C. iguanaea on markers of cardiovascular diseases and the glucose metabolism in cholesterol-fed rats. Therefore, hypercholesterolemic rats (1% cholesterol) were orally treated with C. iguanaea extract (C-150, CI-300, or CI-600 mg/kg) or simvastatin (4 mg/kg) (n = 6) once a day (30 days) with a hypercholesterolemic diet. A control group (C) was given saline. C. iguanaea extract showed significant decreases in serum levels of total cholesterol, LDL-cholesterol, HMG-CoA-reductase, interleukin-1 and 6, TNF-α and IFN-γ when compared to group C (p < 0.001). Hypoglycemic effects were observed along with a decrease of the activity of sucrase (CI-600), maltase (CI-150, CI-300), and an increase in muscle glycogen levels (CI-300). Antioxidant effects were observed in plasma by the decrease of TBARS and increase of nonprotein thiols levels (CI-600). The histopathological analysis showed a significant decrease in the liver fat area for C. iguanaea extract compared to group C (p < 0.001). Our results suggest that the biological effects of C. iguanaea extract could be related to the flavonoids that possibly exert antioxidant, enzymatic inhibitory, and insulin-mimetic effects.

ABSTRACT

Jatropha curcas cake, a by-product of biodiesel production, is rich in protein and has potential to be used in livestock feed; however, the presence of antinutritional factors and phorbol esters limits its use. Thus, this study investigated toxicological and reproductive effects in male Wistar rats after subchronic exposure to J. curcas cake subjected to detoxification procedures. Rats were divided into seven groups (n = 10) and treated for 60 days. The control group received commercial feed, while experimental groups received a diet containing 5% J. curcas cake nonhydrolyzed or hydrolyzed with 5 M NaOH. The cakes were unwashed or washed with ethanol or water and were autoclaved at 121°C for 30 minutes. Alkaline hydrolysis combined with ethanol washing decreased the phorbol ester concentration in the cake by 98%. Histopathological findings included diffuse degeneration of the liver and edema around the pulmonary vessels in the nonhydrolyzed groups. In addition, nontreated females mated with males of nonhydrolyzed unwashed group showed a decreased number of live fetuses and an increased placental weight. There were no signs of toxicity in rats given hydrolyzed cakes washed and unwashed, indicating that alkaline hydrolysis associated with heat treatment is an efficient method for detoxification of the J. curcas cake.

ABSTRACT

Systemic hypertension is known to be a common consequence of chronic renal disease, which is frequently diagnosed in dogs with visceral leishmaniasis. Although many veterinary investigations have looked at the renal injury caused by Leishmania spp., the role played by this complication in the development of arterial hypertension documented in some animals with visceral leishmaniasis is not completely understood. In this study, 18 adult dogs with naturally-occurring visceral leishmaniasis and varying clinical signs underwent an indirect blood pressure measurement. Also, sera and spot urine were used for laboratory tests. The median systolic blood pressure was 135.2mmHg (95% confidence interval: 128.5-147.7), median mean arterial pressure was 105.8mmHg (98.3-110.4), and median diastolic arterial pressure was 88.5mmHg (77.8-92.5). No differences existed between asymptomatic and symptomatic animals regarding arterial pressure, and no correlations were documented between blood pressure and serum creatinine, blood urea, urine protein-to-creatinine ratio, urine specific gravity, and the fractional excretion of sodium and potassium. Although an association between hypertension and the identification of inflammation on histopathology could not be demonstrated in hypertensive animals, the assessment of kidney samples from 12 dogs indicated mild inflammation with a lymphoplasmacytic infiltrate (6/12), moderate inflammation with multifocal lymphoplasmacytic and histiocytic infiltrates (3/12), and multifocal degeneration and protein casts (2/12). Anti-Leishmania spp. immunohistochemistry assays stained the renal epithelium in 2/12 of the animals. Even though mild systemic hypertension was documented in a small subset of animals, no relationship between the severity of clinical signs and hypertension could be anticipated.(AU)


A hipertensão sistêmica é reconhecida como uma consequência comum da doença renal crônica, cujo diagnóstico é frequente em cães com leishmaniose visceral. Embora muitas pesquisas veterinárias tenham investigado a lesão renal causada pela Leishmania spp., o papel dessa complicação no desenvolvimento da hipertensão arterial documentada em alguns animais com leishmaniose visceral ainda não é completamente compreendido. Neste estudo, 18 cães adultos com diagnóstico de leishmaniose visceral e sinais clínicos variáveis foram submetidos à avaliação indireta da pressão arterial. Além disso, foram coletados soro e urina para análises laboratoriais. As medianas das pressões arteriais sistólica, média e diastólica foram 135,2mmHg (intervalo de confiança de 95%: 128,5-147,7), 105,8mmHg (98,3-110,4) e 88,5mmHg (77,8-92,5), respectivamente. Não foram constatadas diferenças entre os cães assintomáticos e sintomáticos em relação à pressão arterial, assim como não houve correlação entre a pressão arterial e a creatinina e uréia séricas, relação proteína-creatinina urinária, densidade urinária e excreção fracionada de sódio e potássio. Embora não tenha sido evidenciada associação entre hipertensão arterial e inflamação do tecido renal à histopatologia, a avaliação das amostras oriundas de 12 cães indicou inflamação leve, com infiltrado linfoplasmocitário (6/12), inflamação moderada com infiltrados linfoplasmocitário e histiocítico multifocais (3/12), além de degeneração multifocal e cilindros protéicos (2/12). Ensaios imunoistoquímicos anti-Leishmania spp. marcaram o epitélio renal em 2/12 animais. Apesar de hipertensão leve ter sido documentada em uma pequena parcela dos cães estudados, não se evidenciou relação entre a severidade dos sinais clínicos e o desenvolvimento de hipertensão arterial.(AU)

ABSTRACT

Congenital heart diseases are not commonly diagnosed in wild animals. It is not surprising that few reports exist in the literature, so that prevalence of these anomalies is unknown in wild species. We report a case of a ventricular septal defect documented in a free-ranging crab-eating fox (Cerdocyon thous). This animal presented with rapid, labored breathing, and on physical examination, pulmonary crackles and a holosystolic murmur were auscultated. The echocardiogram with Doppler showed discontinuity of the dorsal section of the ventricular septum, which allowed a turbulent systolic flow to move from the left to the right ventricle. The postmortem examination confirmed the absence of a dorsal connection between the septum and the atrioventricular junction, and pronounced left ventricular myocardial dilation was observed on histopathology. To the authors' knowledge, this is the first report of a perimembranous ventricular septal defect in a crab-eating fox.

Samanea tubulosa is a plant used for medicinal and feeding purposes. However, ingestion of S. tubulosa pods has been associated with bovine abortion. Thus, the aim of this work was to investigate the effects of diet containing 5% of S. tubulosa pod meal on male and female Wistar rats. Diet was administered to male rats (n = 10) for 60 days before mating. Female rats (n = 10) received the treatment for 30 days, during cohabitation and from gestational day (GD) 0 to GD20. Treated animals were mated with untreated rats. In male rats, plant consumption caused decreased food consumption and 20% fertility index reduction. Litters from treated males presented lower body weight and crownrump length. Female rats treated with the plant increased water and food intake and body weight. Decreases in fertility, fecundity and gestation indices and increase of placenta weight and mean number of corpora lutea were found. Thus, owing to the possible general and reproductive toxic effects, long-term consumption of S. tubulosa is not recommended for phytotherapic or food purposes...


Samanea tubulosa é uma planta utilizada na fitoterapia e na alimentação animal. Entretanto, a ingestão de vagens de S. tubulosa tem sido associada à ocorrência de abortos em bovinos. Assim, o objetivo do trabalho foi investigar os efeitos da dieta contendo 5% de vagens de S. tubulosa em ratos machos e fêmeas Wistar. A dieta foi administrada para ratos machos (n = 10) por 60 dias antes do acasalamento. Ratos fêmeas (n = 10) receberam o tratamento por 30 dias, durante a coabitação e do dia gestacional (GD) 0 ao GD 20. As fêmeas tratadas foram acasaladas com ratos não tratados. Em machos, o consumo da planta causou diminuição no consumo de ração e redução de 20% no índice de fertilidade. A prole de machos tratados apresentou menor ganho de peso e comprimento cabeça cauda. Fêmeas tratadas com a planta apresentaram aumento do consumo de ração e água e do peso corporal. Ainda, foram observadas diminuição na fertilidade, fecundidade e no índice de gestação e aumento do peso da placenta e no número médio de corpos lúteos. Desse modo, em decorrência aos possíveis efeitos tóxicos sistêmicos e reprodutivos, o consumo prolongado de S. tubulosa não é recomendado para fins fitoterápicos ou alimentar...

Since little information is available regarding cellular antigen mapping and the involvement of non-neuronal cells in the pathogenesis of bovine herpesvirus type 5 (BHV-5) infection, it were determined the BHV-5 distribution, the astrocytic reactivity, the involvement of lymphocytes and the presence of matrix metalloproteinase (MMP)-9 in the brain of rabbits experimentally infected with BHV-5. Twelve New Zealand rabbits that were seronegative for BHV-5 were used for virus inoculation, and five rabbits were used as mock-infected controls. The rabbits were kept in separate areas and were inoculated intranasally with 500 µl of virus suspension (EVI 88 Brazilian isolate) into each nostril (virus titer, 10(7.5) TCID50). Control rabbits were inoculated with the same volume of minimum essential medium. Five days before virus inoculation, the rabbits were submitted to daily administration of dexamethasone. After virus inoculation, the rabbits were monitored clinically on a daily basis. Seven rabbits showed respiratory symptoms and four animals exhibited neurological symptoms. Tissue sections were collected for histological examination and immunohistochemistry to examine BHV-5 antigens, astrocytes, T and B lymphocytes and MMP-9. By means of immunohistochemical and PCR methods, BHV-5 was detected in the entire brain of the animals which presented with neurological symptoms, especially in the trigeminal ganglion and cerebral cortices. Furthermore, BHV-5 antigens were detected in neurons and/or other non-neural cells. In addition to the neurons, most infiltrating CD3 T lymphocytes observed in these areas were positive for MMP-9 and also for BHV-5 antigen. These infected cells might contribute to the spread of the virus to the rabbit brain along the trigeminal ganglia and olfactory nerve pathways.

ABSTRACT

O aumento na produção de biodiesel vem acompanhado do incremento na geração de subprodutos, dentre os quais a glicerina bruta e o glicerol, que vem sendo objeto de inúmeros trabalhos em que se destaca seu uso como fonte energética para alimentação de animais de produção. Entretanto, sua utilização tem sido observada com atenção devido à presença de contaminantes como o metanol, utilizado no processo de separação dos ácidos graxos do glicerol. O metanol é um álcool e tem sido responsável por intoxicações, principalmente em seres humanos e primatas, caracterizadas por acidose metabólica e problemas oculares, apresentando menor toxidade a outras espécies animais. Os efeitos tóxicos da intoxicação por metanol são atribuídos principalmente ao ácido fórmico, um metabólito do processo de degradação do metanol no fígado. Objetivou-se com esse estudo verificar a possível ação tóxica da glicerina bruta e do metanol por análises histopatológicas do sistema nervoso central (SNC), fígado, rúmen, intestino delgado e rim de 24 bovinos confinados por 84 dias, consumindo rações contendo até 24% de glicerina bruta na matéria seca (MS),a qual apresentava 80,35% de glicerol, e <0,01% de metanol. À análise histopatológica, não foram observadas alterações patológicas em amostras provenientes de vacas e novilhos de raças de aptidão leiteira com até 24% de inclusão de glicerol com base na MS, da dieta. O ganho de peso total e a conversão alimentar não foram afetados pela inclusão da glicerol. Conclui-se que a glicerina bruta pode ser incluída até 24% da MS total nas dietas de novilhos e vacas de aptidão leiteira sem comprometer o desempenho dos animais e apresentar toxicidade.


The increase in biodiesel production has led to higher production of crude glycerin, thus, numerous studies have highlighted the use of crude glycerin and glycerol as energy source to be used for animal feed production. However, its use has been closely watched because of the presence of contaminants such as methanol used in the process of separating fatty acids from glycerol. Methanol is an alcohol and has been responsible for intoxication in humans and primates, characterized by metabolic acidosis and eye problems, with less toxicity to other species. The toxic effects of methanol poisoning are attributed mainly to formic acid, a metabolite of the degradation process of methanol in the liver. The objective of this study was to verify the possible toxic action of crude glycerin and methanol by pathological examinations of the central nervous system (CNS), liver, rumen, small intestine and kidney from 24 feedlot cattle for 84 days, feed containing up to 24% of crude glycerin in dry matter (DM) presenting in its composition 80.35% glycerol, and <0.01% methanol. There were no pathological changes observed in dairy cows and steers up to 24% of glycerol inclusion based on the diet DM. The total weight gain and feed conversion were not affected by inclusion of glycerol. We conclude that crude glycerin can be included up to 24% of total DM in the diets of steers and dairy cows without compromising animal performance or display toxicity.

ABSTRACT

Laryngeal cancer is the second most frequent head and neck cancer in the Brazilian male population. For treatment, radiotherapy combined with chemotherapy is now used in substitution for total laryngectomy, becoming the standard treatment for advanced larynx cancer cases, with the aim of organ preservation. However, this method needs assessment of the side effects caused to normal tissue and organ functionality after treatment and the relation of these clinical factors to the individual characteristics of patients. Thus, the clinical characteristics of 229 patients with laryngeal cancer treated with radiotherapy were evaluated by medical records analysis in relation to normal tissue radiosensibility. Significant relations between smoking (p = 0.018) and combined chemoradiotherapy assistance (p = 0.03) were identified with high frequency of treatment suspension cases. The application of combined chemoradiotherapy also resulted in a higher incidence of oral mucositis (p = 0.04), xerostomia (p = 0.001) and treatment side effects to GIT (p = 0.04). Advanced clinical staging was associated with worse prognosis (p = 0.002) and a higher occurrence of treatment failure (p < 0.001). Radiotherapy was also less effective depending on the primary tumor location (p = 0.001)

A compactação dos pré-estômagos é um transtorno digestório pouco frequente. E pode ter origem primária ou secundária. Osautores descrevem o primeiro relato de compactação ruminoabomasal causado pela ingestão de caule de bananeira (Musasp.) em bovinos. Os casos ocorreram em duas vacas, mestiças de aptidão leiteira, com idades de 5 e 4 anos respectivamente,oriundas de uma propriedade rural no município de Araguaína, Estado do Tocantins (TO). O histórico, os achados clínicos, osresultados dos exames hematológicos e a análise do fluido ruminal permitiram o diagnóstico, que foram confirmados por necropsiae laparotomia exploratória. O uso de caule de bananeira (Musa sp.) como única fonte de volumoso favoreceu a ocorrência dacompactação ruminoabomasal. No entanto, essa fonte de alimento pode ser considerada uma boa opção para a alimentação debovinos, desde que seja fornecido em proporções adequadas na dieta.

Buchenavia tomentosa Eichler is a common plant in Brazilian cerrado. Fruits of this plant are employed in human feeding and folk medicine. Cattle producers affirm that consumption of the fruits cause abortion in cows, and even death. Considering that the plant may be consumed by pregnant women and animals, the present study was undertaken to evaluate the possible toxic effects of the ingestion of B. tomentosa fruit (10% added to the diet), from the first to the twenty-first days of gestation, on reproductive parameters and on physical and neurobehavioral development of rats offspring. An increase in food consumption at pregnancy days 11 and 17, and weight increase at day 17 of pregnancy were observed. Besides that, we verified an increase in weight of male offspring on post natal day 1. Other parameters were not affected by plant consumption. These results indicate that the consumption of B. tomentosa at 10% during pregnancy cause slight toxicological effects. The changes verified in the present study indicate toxic action of the fruit possibly induced by flavonoids with hormonal action; however, further studies must be accomplished to corroborate this hypothesis.


Buchenavia tomentosa Eichler é uma planta típica do cerrado brasileiro. Os frutos desta planta são empregados na alimentação humana e medicina popular. Criadores de bovinos afirmam que o consumo desta planta produz aborto em vacas bem como a morte destes animais. Uma vez que a planta pode ser consumida pelo homem e animais, em idade fértil e inclusive gestantes, o presente estudo avaliou os possíveis efeitos tóxicos da ingestão de dieta com 10% de frutos de B. tomentosa do primeiro ao vigésimo primeiro dia de gestação sobre os parâmetros reprodutivos e sobre o desenvolvimento físico e neurocomportamental das ninhadas de ratos. Foram observadas elevações no consumo de alimentos nos dias 11 e 17 de gestação e no peso ao dia 17 de gestação. Aumento do peso dos filhotes machos foi verificado no primeiro dia após o nascimento. Outros parâmetros não foram afetados pelo consumo da planta. Estes resultados indicam que a ingestão, durante a gestação, de uma dieta acrescida com 10% de B. tomentosa causa efeitos tóxicos leves. As alterações verificadas no presente estudo indicam ação tóxica do fruto possivelmente induzidos por flavonoides de ação hormonal; no entanto, estudos detalhados devem ser conduzidos para garantir esta hipótese.
