ABSTRACT

In vitro studies with organ (oviduct and trachea) and chicken embryo kidney cell cultures were attempted to assess the pathogenicity of locally isolated infectious bronchitis virus (IBV-P:120) initially isolated from the oviduct of young chicks. In oviduct cultures infected with IBV, ciliary movements decreased as early as 24 hours postinoculation (PI), and on the 6th day ciliary movements ceased completely. Cytopathic changes were also noticed. Immunofluorescent antigen was detected from 1 to 6 days PI, the maximum being on the 3rd day. The characteristic microscopic changes in the oviduct explants were reduced by 24 hours PI and had completely ceased on the 5th day. Cytopathic effect and immunofluorescent antigen were present from 1 to 8 days PI, being maximum on the 5th day. Histological changes marked by loss of cilia, rounding of the epithelial cells, degeneration, and sloughing were detected from 2 to 8 days PI. Low-embryo-passaged (EP-7) IBV did not produce cytopathic effect on the chicken embryo kidney cell cultures. On the contrary, high-embryo-passaged (EP-14) virus produced cytopathic effect at the third tissue-culture-passage level.

ABSTRACT

Canine venereal tumor cells were grown in monolayer and tumor tissue fragments were maintained in vitro for 56 days in medium-199. Attachment and cell replication were evident within 17 hours and monolayer was obtained by the tenth day. Growth at first comprised mainly round cells with abundance of cytoplasm and vesicular nuclei, and a few spindle-shaped cells. Later cells became more elongated and like fibroblasts. Cell degeneration started by the 17th day and most were degenerated by the 24th day. Necrosis and depletion of cell population were prominent in tumor explants during the first 2 weeks. Distinct cell multiplication was evident by the 21st day and tumor fragments were repopulated with cells resembling the original tumor by the 56th day. Two distinct morphological cell types were seen: small cells with vesicular, round to oval nuclei and acidophilic cytoplasm; and the large cells with large hyperchromatic nuclei and acidophilic cytoplasm.

ABSTRACT

PIP: Entoviruses and Mycoplasma bovigenitalium were isolated from 37 bovine abortions and characterized. Enteroviruses were isolated in 2 cases and Mycoplasma bovigenitalium in 1 case. Characteristics of the isolates of enterovirus were: resistance to ether and chloroform, lability at 56 degrees C for 30 minutes, resistance to pH 3.2 buffer, cationic stabilization in the presence of M MgC1(2) solution, and neutralization by La Placa group 2 antisera. Morphological, biochemical, serological, and epi-immunofluorescent methods were used in the identification of Mycoplasma bovigenitalium. The etiologic role of these agents in bovine abortions is discussed.^ieng

ABSTRACT

Studies were undertaken to determine the presence of lesions and immuno-fluorescent (IF) antigen of Marek's disease (MD) in the comb, wattles and skin overlaying the shank of MD affected birds. Lesions were more common in the comb than in the wattles and shanks, and were characterized by degeneration of epithelial lining cells and accumulation of lympho-reticular cells around the blood vessels of the dermis. Diffuse infiltration of these cells replacing varialbe areas of dermis were also seen in some cases. Immunofluorescent antigen was mostly confined to epidermal epithelium and was detected in comb from majority of cases, and the frequency of detection of IF antigen was similar to the presence of MD lesions in these tissues. This study indicates that the presence of lesions and viral antigen (If) of MD in these cutaneous appendages might play a significant role in epidemiology and diagnosis of this disease.

ABSTRACT

Granular vulvovaginitis (GVV) was experimentally produced in female kids by topical application of M. agalactiae on the scarified vulvar mucosa. Grossly visible GVV lesions were seen in 25 out of 30 infected kids, yet all were positive for microscopic lesions. Microscope lesions that appeared by 7 days post infection (DPI) were comprised of stromal oedema, lymphocytic infiltration into the lamina propria and perivascular accumulation of a few lymphocytes. The lesions observed between 28 and 49 DPI were comparable to those of spontaneous cases (severe). The changes seen in kids euthanized between 56 to 70 DPI were suggestive of the chronic stage of the disease. M. agalactiae was reisolated from all the infected kids from 7 to 70 days after inoculation. The pathology and pathogenesis of this condition have been described and discussed.
