ABSTRACT

Implementation of marker-assisted selection (MAS) in modern beekeeping would improve sustainability, especially in breeding programs aiming for resilience against the parasitic mite Varroa destructor. Selecting honey bee colonies for natural resistance traits, such as brood-intrinsic suppression of varroa mite reproduction, reduces the use of chemical acaricides while respecting local adaptation. In 2019, eight genomic variants associated with varroa non-reproduction in drone brood were discovered in a single colony from the Amsterdam Water Dune population in the Netherlands. Recently, a new study tested the applicability of these eight genetic variants for the same phenotype on a population-wide scale in Flanders, Belgium. As the properties of some variants varied between the two studies, one hypothesized that the difference in genetic ancestry of the sampled colonies may underly these contribution shifts. In order to frame this, we determined the allele frequencies of the eight genetic variants in more than 360 Apis mellifera colonies across the European continent and found that variant type allele frequencies of these variants are primarily related to the A. mellifera subspecies or phylogenetic honey bee lineage. Our results confirm that population-specific genetic markers should always be evaluated in a new population prior to using them in MAS programs.

ABSTRACT

Uncertainty of measurement values (MU) is crucial to their reliable geochemical interpretation. MU can be estimated using the Duplicate Method, which requires the taking of a small proportion of duplicated samples, and can be applied at any spatial scale. The distance between the duplicated samples is selected to reflect the effect of analyte heterogeneity on the measurement result (i.e. estimated concentration) within each sampling target, at the particular scale of investigation. Three published case studies, at different spatial scales, are used to explain how the Duplicate Method can be applied to estimate MU. They also illustrate how MU can be used to improve geochemical interpretation and validate measurement procedures (that include sampling) by judging their fitness for purpose. At the kilometre scale, measurements from the GEMAS survey of agricultural soils across Europe are used to estimate their MU for the first time. The MU for 53 elements range from an uncertainty factor of 1.01 to over 10. The MU contributes more that 20% to the total variance for 8 of the 53 elements, showing that the measurement procedure was not fit for purpose in those cases. At the micron scale, measurements of oxygen isotopes in candidate quartz reference materials had MU that was dominated by its sampling component, caused by sometimes unacceptable heterogeneity. A third case study of Pb in soils at 12 UK sites showed that the Duplicate Method can also be used to quantify the heterogeneity (as factor 1.03 to 2.4), and that it can indicate different possible sources of an element.

ABSTRACT: Lang, HM, Duffourc, MM, Bazyler, CD, Ramsey, MW, and Gentles, JA. The relationship between cell-free DNA and resistance training volume load. J Strength Cond Res 38(6): 1008-1012, 2024-The primary purpose of this study was to assess the sensitivity of cell-free DNA (cf-DNA) to different resistance training volume loads. The secondary purpose was to examine the relationship between change in cf-DNA and relative strength. Researchers hypothesized that (a) cf-DNA concentrations would increase with increasing volume load and (b) increases in relative strength would result in predicted decreases to %Δ of cf-DNA. Thirty subjects were recruited for this study, 15 men and 15 women. Blood was collected through venous draws into 4-ml vacutainers at 3 time points: immediately before (T1), after 3 sets (T2), and after 6 sets (T3) of the back squat exercise. A critical alpha of 0.05 was set for inferential statistics. A repeated-measures ANOVA showed that cf-DNA increased significantly from T1 (407.72 ± 320.83) to T2 (1,244.6 ± 875.83) ( p < 0.01) and T1 (407.72 ± 320.83) to T3 (1,331.15 ± 1,141.66) ( p < 0.01), whereas no difference was found from T2 to T3 ( p = 1.00). The linear regression model used to examine the predictive capabilities relative strength had on cf-DNA %Δ from T1 to T3 was found to be significant ( p = 0.04; R2 = 0.15). The results of this study demonstrate the short response of cf-DNA in relation to variations in resistance training volume load. Results also demonstrated the positive relationship between relative strength and cf-DNA %Δ. The current study builds on the body of research that cf-DNA provides insight regarding the level of immune response after exercise training.

ABSTRACT

A series of liquid crystalline porphyrins was synthesized, purified, and characterized. Differential scanning calorimetry (DSC) and hot-stage polarized optical microscopy (HS-POM) revealed that the porphyrins in the series with shorter alkyl arm lengths exhibit kinetic cold crystallization, wherein the molecules spontaneously organize into large, disc-like structures that remain stable upon cooling. Using DSC, the kinetic and thermodynamic parameters related to these materials were determined. Analysis of non-isothermal crystallization revealed the presence of multiple nucleation and growth processes related to cold crystallization.

ABSTRACT

The adsorption of heptacene (7 A) on Cu(110) and Cu(110)-(2 × 1)-O was studied with scanning tunneling microscopy, photoemission orbital tomography and density functional calculations to reveal the influence of surface passivation on the molecular geometry and electronic states. We found that the charge transfer into the 7 A molecules on Cu(110) is completely suppressed for the oxygen-modified Cu surface. The molecules are aligned along the Cu-O rows and uncharged. They are tilted due to the geometry enforced by the substrate and the ability to maximize intermolecular π-π overlap, which leads to strong π-band dispersion. The HOMO-LUMO gap of these decoupled molecules is significantly larger than that reported on weakly interacting metal surfaces. Finally, the Cu-O stripe phase was used as a template for nanostructured molecular growth and to assess possible confinement effects.

ABSTRACT

The COVID-19 pandemic has posed unprecedented challenges to global healthcare systems, highlighting the need for accurate and timely risk prediction models that can prioritize patient care and allocate resources effectively. This study presents DeepCOVID-Fuse, a deep learning fusion model that predicts risk levels in patients with confirmed COVID-19 by combining chest radiographs (CXRs) and clinical variables. The study collected initial CXRs, clinical variables, and outcomes (i.e., mortality, intubation, hospital length of stay, Intensive care units (ICU) admission) from February to April 2020, with risk levels determined by the outcomes. The fusion model was trained on 1657 patients (Age: 58.30 ± 17.74; Female: 807) and validated on 428 patients (56.41 ± 17.03; 190) from the local healthcare system and tested on 439 patients (56.51 ± 17.78; 205) from a different holdout hospital. The performance of well-trained fusion models on full or partial modalities was compared using DeLong and McNemar tests. Results show that DeepCOVID-Fuse significantly (p < 0.05) outperformed models trained only on CXRs or clinical variables, with an accuracy of 0.658 and an area under the receiver operating characteristic curve (AUC) of 0.842. The fusion model achieves good outcome predictions even when only one of the modalities is used in testing, demonstrating its ability to learn better feature representations across different modalities during training.

ABSTRACT

Solid tumors contain hypoxic regions that contribute to anticancer therapy resistance. Thus, mitigating tumor hypoxia may enhance the efficacy of radiation therapy which is commonly utilized for patients with prostate cancer. Increasing perfusion pressure in the prostate with head-up tilt (HUT) may augment prostate tumor perfusion and decrease hypoxia. The purpose of this study was to determine if an increase in the vascular hydrostatic gradient via 70° HUT increases tumor perfusion and decreases tumor hypoxia in a preclinical orthotopic model of prostate cancer. Male Copenhagen rats (n = 17) were orthotopically injected with Dunning R-3327 (AT-1) prostate adenocarcinoma cells to induce prostate tumors. After tumors were established, prostate tumor perfusion and hypoxia were measured in rats during level (0°) and 70° HUT positions. To compare the magnitude of the hydrostatic column to that present in humans, ultrasound was used to measure the heart to prostate distance in male human subjects to estimate the prostate vascular hydrostatic pressure with the upright posture. In young rats, no differences were detected in prostate tumor perfusion or prostate tumor hypoxia with 70° HUT versus the level position. However, from the retrospective study, young rats increased prostate vascular resistance to HUT, whereas aged rats lacked this response. Tumor vessels co-opted from existing functional vasculature in young rats may be sufficient to negate increases in perfusion pressure with HUT seen in aged rats. Additionally, in humans, the estimated hydrostatic column at the level of the prostate is five times greater than that of the rat. Therefore, 70° HUT may elicit increases in prostate/prostate tumor blood flow in humans that is not seen in rats.

ABSTRACT

We have experimentally determined the adsorption structure, charge state, and metalation state of porphin, the fundamental building block of porphyrins, on ultrathin Ag(001)-supported MgO(001) films by scanning tunneling microscopy and photoemission spectroscopy, supported by calculations based on density functional theory. By tuning the substrate work function to values below and above the critical work function for charging, we succeeded in the preparation of 2H-P monolayers which contain negatively charged and uncharged molecules. It is shown that the porphin molecules self-metalate at room temperature, forming the corresponding Mg-porphin, irrespective of their charge state. This is in contrast to self-metalation of tetraphenyl porphyrin (TPP), which occurs on planar MgO(001) only if the molecules are negatively charged. The different reactivity is explained by the reduced molecule-substrate distance of the planar porphin molecule compared to the bulkier TPP. The results of this study shed light on the mechanism of porphyrin self-metalation on oxides and highlight the role of the adsorption geometry on the chemical reactivity.

ABSTRACT

Polycyclic aromatic compounds with fused benzene rings offer an extraordinary versatility as next-generation organic semiconducting materials for nanoelectronics and optoelectronics due to their tunable characteristics, including charge-carrier mobility and optical absorption. Nonplanarity can be an additional parameter to customize their electronic and optical properties without changing the aromatic core. In this work, we report a combined experimental and theoretical study in which we directly observe large, geometry-induced modifications in the frontier orbitals of a prototypical dye molecule when adsorbed on an atomically thin dielectric interlayer on a metallic substrate. Experimentally, we employ angle-resolved photoemission experiments, interpreted in the framework of the photoemission orbital tomography technique. We demonstrate its sensitivity to detect geometrical bends in adsorbed molecules and highlight the role of the photon energy used in experiment for detecting such geometrical distortions. Theoretically, we conduct density functional calculations to determine the geometric and electronic structure of the adsorbed molecule and simulate the photoemission angular distribution patterns. While we found an overall good agreement between experimental and theoretical data, our results also unveil limitations in current van der Waals corrected density functional approaches for such organic/dielectric interfaces. Hence, photoemission orbital tomography provides a vital experimental benchmark for such systems. By comparison with the state of the same molecule on a metallic substrate, we also offer an explanation why the adsorption on the dielectric induces such large bends in the molecule.

ABSTRACT

When a molecule interacts chemically with a metal surface, the orbitals of the molecule hybridise with metal states to form the new eigenstates of the coupled system. Spatial overlap and energy matching are determining parameters of the hybridisation. However, since every molecular orbital does not only have a characteristic spatial shape, but also a specific momentum distribution, one may additionally expect a momentum matching condition; after all, each hybridising wave function of the metal has a defined wave vector, too. Here, we report photoemission orbital tomography measurements of hybrid orbitals that emerge from molecular orbitals at a molecule-on-metal interface. We find that in the hybrid orbitals only those partial waves of the original orbital survive which match the metal band structure. Moreover, we find that the conversion of the metal's surface state into a hybrid interface state is also governed by momentum matching constraints. Our experiments demonstrate the possibility to measure hybridisation momentum-selectively, thereby enabling deep insights into the complicated interplay of bulk states, surface states, and molecular orbitals in the formation of the electronic interface structure at molecule-on-metal hybrid interfaces.

ABSTRACT

Tracing the modifications of molecules in surface chemical reactions benefits from the possibility to image their orbitals. While delocalized frontier orbitals with π character are imaged routinely with photoemission orbital tomography, they are not always sensitive to local chemical modifications, particularly the making and breaking of bonds at the molecular periphery. For such bonds, σ orbitals would be far more revealing. Here, we show that these orbitals can indeed be imaged in a remarkably broad energy range and that the plane wave approximation, an important ingredient of photoemission orbital tomography, is also well fulfilled for these orbitals. This makes photoemission orbital tomography a unique tool for the detailed analysis of surface chemical reactions. We demonstrate this by identifying the reaction product of a dehalogenation and cyclodehydrogenation reaction.

ABSTRACT

OBJECTIVE: To understand African Americans' perceptions, barriers, and facilitators to recruitment, enrollment, adoption, maintenance, and retention in a nutrition and physical activity promotion program. DESIGN: Four focus groups were conducted. SETTING: Two community settings located in Jackson and Hattiesburg, Mississippi. PARTICIPANTS: Participants (n = 28) were aged 18-50 years. MAIN OUTCOME MEASURE(S): Barriers and facilitators associated with healthy eating, physical activity, achieving a healthy weight, and participation in a health behavior change program. ANALYSIS: A conventional thematic content analysis approach includes data familiarization, initial code generation, initial theme generation, themes review, and team review for finalization of themes. RESULTS: Major themes related to health behaviors and participation in a behavior change program were identified by participants, including time constraints, costs, social support, consistency and self-efficacy, motivation for longevity and disease prevention, physical appearance, fear of injury/pain, social norms/stigma associated with outdoor physical activity, body criticism from family members, and having empathic and validating program staff support. CONCLUSIONS AND IMPLICATIONS: Several social determinants of health were identified as essential considerations for promoting healthy nutrition and physical activity behaviors among African American adult Mississippians. Cultural and spiritual implications were also identified. Study insights inform policy approaches for designing culturally appropriate health behavior change programs in the Deep South.

ABSTRACT

The ability to navigate scientific obstacles is widely recognized as a hallmark of a scientific disposition and is one predictor of science, technology, engineering, and mathematics persistence for early-career scientists. However, the development of this competency in undergraduate research has been largely underexplored. This study addresses this gap by examining introductory students' emotional and behavioral responses to research-related challenges and failures that occur in two sequential research-based courses. We describe commonly reported emotions, coping responses, and perceived outcomes and examine relationships between these themes, student demographics, and course enrollment. Students commonly experience frustration, confusion, and disappointment when coping with challenges and failures. Yet the predominance of students report coping responses likely to be adaptive in academic contexts despite experiencing negative emotions. Being enrolled in the second course of a research-based course sequence was related to several shifts in response to challenges during data collection, including less reporting of confusion and fewer reports of learning to be cautious from students. Overall, students in both the first and second courses reported many positive outcomes indicating improvements in their ability to cope with challenge and failure. We assert that educators can improve research-based educational courses by scaffolding students' research trials, failures, and iterations to support students' perseverance.

ABSTRACT

Hexacene, composed of six linearly fused benzene rings, is an organic semiconductor material with superior electronic properties. The fundamental understanding of the electronic and chemical properties is prerequisite to any possible application in devices. We investigate the orientation and interface properties of highly ordered hexacene monolayers on Ag(110) and Cu(110) with X-ray photoemission spectroscopy (XPS), photoemission orbital tomography (POT), X-ray absorption spectroscopy (XAS), low-energy electron diffraction (LEED), scanning tunneling microscopy (STM), and density functional theory (DFT). We find pronounced differences in the structural arrangement of the molecules and the electronic properties at the metal/organic interfaces for the two substrates. While on Cu(110) the molecules adsorb with their long molecular axis parallel to the high symmetry substrate direction, on Ag(110), hexacene adsorbs in an azimuthally slightly rotated geometry with respect to the metal rows of the substrate. In both cases, molecular planes are oriented parallel to the substrate. A pronounced charge transfer from both substrates to different molecular states affects the effective charge of different C atoms of the molecule. Through analysis of experimental and theoretical data, we found out that on Ag(110) the LUMO of the molecule is occupied through charge transfer from the metal, whereas on Cu(110) even the LUMO+1 receives a charge. Interface dipoles are determined to a large extent by the push-back effect, which are also found to differ significantly between 6A/Ag(110) and 6A/Cu(110).

Charge-transfer processes at molecule-metal interfaces play a key role in tuning the charge injection properties in organic-based devices and thus, ultimately, the device performance. Here, the metal's work function and the adsorbate's electron affinity are the key factors that govern the electron transfer at the organic/metal interface. In our combined experimental and theoretical work, we demonstrate that the adsorbate's orientation may also be decisive for the charge transfer. By thermal cycloreversion of diheptacene isomers, we manage to produce highly oriented monolayers of the rodlike, electron-acceptor molecule heptacene on a Cu(110) surface with molecules oriented either along or perpendicular to the close-packed metal rows. This is confirmed by scanning tunneling microscopy (STM) images as well as by angle-resolved ultraviolet photoemission spectroscopy (ARUPS). By utilizing photoemission tomography momentum maps, we show that the lowest unoccupied molecular orbital (LUMO) is fully occupied and also, the LUMO + 1 gets significantly filled when heptacene is oriented along the Cu rows. Conversely, for perpendicularly aligned heptacene, the molecular energy levels are shifted significantly toward the Fermi energy, preventing charge transfer to the LUMO + 1. These findings are fully confirmed by our density functional calculations and demonstrate the possibility to tune the charge transfer and level alignment at organic-metal interfaces through the adjustable molecular alignment.

ABSTRACT

PURPOSE: Little is known about racial/ethnic minority substance use and disorder-related disparities among sexual minorities as compared to their heterosexual counterparts. This study aimed to understand the associations between sexual minority status and substance use and disorders among U.S. adults stratified by race/ethnicity. Methods: We analyzed data from a sample of U.S. adults (n = 35,981) surveyed during the 2012-2013 National Epidemiological Survey on Alcohol and Related Conditions-III. Respondents were categorized into four sexual minority statuses: heterosexual, gay/lesbian, bisexual, and conflicting. Respondents' substance (tobacco, alcohol, and marijuana) use and disorders were also assessed using standard measures. Weighted multivariable logistic regressions were used to examine the relationships between sexual minority status and substance use and disorders, stratified by race/ethnicity, adjusting for socio-demographic characteristics. Results: Sexual minority adults experienced a higher prevalence of substance use and disorders than heterosexuals. For example, bisexuals were more likely than heterosexuals to use marijuana (AOR = 3.45, 95% CI = 2.64-4.50) and have tobacco use disorders (AOR = 2.58, 95% CI = 2.02-3.28). These associations were stronger among racial/ethnic minorities. For instance, bisexual non-Hispanic Blacks were more than twice as likely (AOR = 3.17, 95% CI = 2.16-4.65) to be current tobacco users than their heterosexual counterparts; while this association was weaker for bisexual non-Hispanic Whites (AOR = 1.99, 95% CI = 1.47-2.71). Conclusion: Sexual minority adults, especially those who are also racial/ethnic minorities, experience a significantly higher burden of substance use and disorders than heterosexuals. Efforts to screen and treat substance use and disorders among this particular population are critically needed to improve their health outcomes and reduce health disparities.

OBJECTIVES: On the basis of the data from school-aged children, there is consistent evidence that there is a prolonged course of auditory development for perceiving speech embedded in competing background sounds. Furthermore, age-related differences are prolonged and pronounced for a two-talker speech masker compared to a speech-shaped noise masker. However, little is known about the course of development during the toddler and preschool years because it is difficult to collect reliable behavioral data from this age range. The goal of this study was to extend our lower age limit to include toddlers and preschoolers to characterize the developmental trajectory for masked speech detection thresholds across childhood. DESIGN: Participants were 2- to 15-year-old children (n = 67) and adults (n = 17), all with normal hearing. Thresholds (71%) were measured for detecting a two-syllable word embedded in one of two maskers: speech-shaped noise or two-talker speech. The masker was presented at 55 dB SPL throughout testing. Stimuli were presented to the left ear via a lightweight headphone. Data were collected using an observer-based testing method in which the participant's behavior was judged by an experimenter using a two-interval, two-alternative testing paradigm. The participant's response to the stimulus was shaped by training him/her to perform a conditioned play-based response to the sound. For children, receptive vocabulary and working memory were measured. Data were fitted with a linear regression model to establish the course of development for each masker condition. Appropriateness of the test method was also evaluated by determining if there were age-related differences in training data, inter-rater reliability, or slope or upper asymptote estimates from pooled psychometric functions across different age groups. RESULTS: Child and adult speech detection thresholds were poorer in the two-talker masker than in the speech-shaped noise masker, but different developmental trajectories were seen for the two masker conditions. For the speech-shaped noise masker, threshold improved by about 5 dB across the age span tested, with adult-like performance being reached around 10 years of age. For the two-talker masker condition, thresholds improved by about 7 dB between 2.5 and 15 years. However, the linear fit for this condition failed to achieve adult-like performance because of limited data from teenagers. No significant age-related differences were seen in training data, probe hit rate, or inter-rater reliability. Furthermore, slope and upper asymptote estimates from pooled psychometric functions were similar across different child age groups. CONCLUSIONS: Different developmental patterns were seen across the two maskers, with more pronounced child-adult differences and prolonged immaturity during childhood for the two-talker masker relative to the speech-shaped noise masker. Our data do not support the idea that there is rapid improvement of masked speech detection thresholds between 2.5 and 5 years of age. This study also highlights that our observer-based method can be used to collect reliable behavioral data from toddlers and preschoolers-a time period where we know little about auditory development.

J Strength Cond Res 35(5): 1217-1222, 2021-The purpose of this study was to determine if successive heavy sets of back squats can augment the concentric velocity of a lighter down set performed by strength-trained men. Twelve trained men with experience in the back squat volunteered to perform a 5 repetition maximum (5RM) along with 2 separate squat sessions consisting of 3 sets of 5 repetitions with 85% of their 5RM. One condition involved performing a "down set" (DS) after the 3 working sets at 85% of 5RM equivalent to 60% of the working-set load that was also performed during the warm-up. A "No down set" condition involved performing an additional warm-up set before the working sets with 60% of the working-set load instead of the down set to determine if velocity was augmented because of postactivation potentiation in the DS condition. In both conditions, 3 minutes of rest was applied between all sets. A paired sample t-test was used to compare the mean concentric velocities (MCVs) of the working sets of both conditions, and a repeated measures analysis of variance was used to assess differences in MCVs between sets performed at 60% of the working-set load. Cohen's d effect sizes were reported for all comparisons, and the critical alpha was set at p ≤ 0.05. No significant differences were observed in the working-set MCVs in both conditions (p = 0.412, d = 0.246) or between MCVs in the down set and equivalent warm-up set load in the DS condition (p = 0.270, d = 0.002).Although performing a down set may still be efficacious for developing power across a broad spectrum of loads, the results of this study suggest successive heavy sets of back squats do not acutely augment down set concentric velocity in strength-trained men.
