ABSTRACT

The aim of this research was to determine the effect of gut health parameters on the flock's final weight of broilers and to calculate an accurate equation to estimate this weight with information available at 7, 14, and 21 days, in field conditions. Gut health parameters (gizzard erosion, coccidiosis, feed passage, and redness, gut tone, consistency of content, and presence of mucus for each part of the small intestine [duodenum, jejunum, and ileum], and color, consistency, and presence of gas for caeca content) were evaluated at 7 and 14 days. Other parameters evaluated for impact on flock final weight were body weight and mortality, both at 7, 14, and 21 days; stocking density; litter reuse; and downtime period. Structural equation model evaluation of the data showed that stocking density and litter reuse did not affect (P > 0.05) flock final weight, while downtime period, body weight (14 and 21 days), and mortality (14 and 21 days) directly affected (P ≤ 0.05) the flock final weight. Gut health parameters did not directly affect the flock's final weight; however, they affected body weight and mortality at 14 days, thus showing an indirect effect on the flock's final weight. It was also possible to determine two accurate equations to estimate the flock's final weight using information available at both 14 (R2 = 0.56) and 21 (R2 = 0.77) days.

Variance and performance of two sampling plans for aflatoxins quantification in maize were evaluated. Eight lots of maize were sampled using two plans: manual, using sampling spear for kernels; and automatic, using a continuous flow to collect milled maize. Total variance and sampling, preparation, and analysis variance were determined and compared between plans through multifactor analysis of variance. Four theoretical distribution models were used to compare aflatoxins quantification distributions in eight maize lots. The acceptance and rejection probabilities for a lot under certain aflatoxin concentration were determined using variance and the information on the selected distribution model to build the operational characteristic curves (OC). Sampling and total variance were lower at the automatic plan. The OC curve from the automatic plan reduced both consumer and producer risks in comparison to the manual plan. The automatic plan is more efficient than the manual one because it expresses more accurately the real aflatoxin contamination in maize.

ABSTRACT

The objective of this study was to determine the effects of three doses of fumonisin B1 (0, 100, and 200mg/kg of feed) on biological variables (relative weight of liver [RWL], total plasma protein [TPP], albumin [Alb], calcium [Ca], phosphorus [P], uric acid [UA], alanine aminotransferase [ALT], aspartate aminotransferase [AST], gamma glutamyltransferase [GGT], alkaline phosphatase [AP], total cholesterol [Chol], triglycerides [Tri], sphinganine-to-sphingosine ratio [SA:SO], and C-reactive protein [CRP]), morphological evaluation of the small intestine (villus height [VH], crypt depth [CD], and villus-to-crypt ratio [V:C]), histological evaluation, and on performance (body weight [BW], feed intake [FI], and feed conversion rate [FCR]) of broiler chickens. Significant effects of FB were observed on BW and FI (reduced), on RWL, TPP, Ca, ALT, AST, GGT, Chol, and Tri (increased) at both 14 and 28 days evaluations. In addition, significant increase was observed on FCR, Alb, P, SA:SO, and CRP and significant reduction in UA, VH, and V:C only at the 28 days evaluation. Significant histological lesions were observed on liver and kidney of FB inoculated broilers at 14 and 28 days. Those results show that FB has a significant effect on biological and histological variables and on performance of broiler chickens.


O objetivo deste trabalho foi determinar os efeitos de três doses de fumonisina B1 (0, 100 e 200 mg/kg de ração) sobre variáveis biológicas (peso relativo de fígado [RWL], proteínas plasmáticas totais [TPP], albumina [Alb], cálcio [Ca], fósforo [P], ácido úrico [UA], alanina aminotransferase [ALT], aspartato aminotransferase [AST], gama glutamiltransferase [GGT] fosfatase alcalina [AP], colesterol total [Chol], triglicerídeos [Tri], relação esfinganina:esfingosina [SA:SO] e proteína C-reativa [CRP]), avaliação morfológica do intestino delgado (altura de vilosidades [VH], profundidade de criptas [CD] e relação entre vilosidade e cripta [V:C]), avaliações histológicas e no desempenho (peso corporal [BW], consumo de ração [FI] e conversão alimentar [FCR]) de frangos de corte. Efeitos significativos da FB foram observados sobre BW e FI (diminuição), sobre RWL, TPP, Ca, ALT, AST, GGT, Chol e Tri (aumento) aos 14 e 28 dias. Além disso, houve aumento significativo nos parâmetros FCR, Alb, P, SA:SO e CRP e redução nos parâmetros UA, VH e V:C somente aos 28 dias. Lesões histológicas significativas foram observadas no fígado e rins das aves intoxicadas, tanto aos 14 quanto aos 28 dias. Estes resultados indicam que a FB tem um efeito significativo sobre variáveis biológicas e de desempenho de frangos de corte.

ABSTRACT

O objetivo deste trabalho foi avaliar a eficiência de dois planos de amostragem para análise de fumonisinas (B1+B2) em 11 lotes de milho. Empregaram-se o plano de amostragem manual em grãos inteiros, através do calador graneleiro e o plano de Amostragem Automático em Fluxo Contínuo em grãos moídos (AAFC). Para a comparação dos dois planos de amostragem, foram utilizadas duas medidas de dispersão: a variância e o coeficiente de variação. Em cada plano de amostragem, foram determinadas as variâncias associadas às três etapas do procedimento de detecção das fumonisinas (amostragem, preparação da amostra e análise) e os coeficientes de variação total. A correlação entre as médias de concentrações de fumonisinas e as variâncias de cada etapa do procedimento de detecção foi avaliada para cada plano amostral através da análise de regressão. Na AAFC de milho moído para análise de fumonisinas, obtiveram-se resultados inferiores (P<0,01) de variância na etapa de amostragem (0,0226mg kg-1)², assim como no coeficiente de variação total (6,37%), quando comparados com a variância da amostragem manual (0,0685mg kg-1)² e coeficiente de variação total do plano de amostragem manual em grãos inteiros (8,94%), apresentando, portanto, maior eficiência na detecção de fumonisinas em milho.


The aim of this research was to evaluate the efficiency of two sampling plans for fumonisins (B1+B2) analysis in 11 lots of maize. For sample collection the two systems used were a manual system in whole grains, using sampling spear, and an automatic system in milled grains, using the continuous flow sampling system (CFSS). For comparison purpose, two dispersion measurements were used: variance and coefficient of variation. For each sampling plan the variance associated to the three steps of fumonisins quantification (sampling, sample preparation, and analysis), and the total coefficient of variation were determined. The correlation between the average fumonisins concentration and the variances for each phase of the detection procedure was evaluated for each sampling plan using analysis of regression. At the automatic sampling plan in milled grains for fumonisins analysis in maize both sampling variance (0.0226mg kg-1)² and total coefficient of variation (6.37%) were lower (P<0.01) than in the manual sampling plan in whole grains whit (0.0685mg kg-1)² of variance and 8.94% of coefficient of variation, showing so better efficiency for fumonisin detection on maize.

ABSTRACT

CONTEXT: Taraxacum officinale Weber (Asteraceae), known as dandelion, is used for medicinal purposes due to its choleretic, diuretic, antitumor, antioxidant, antiinflammatory, and hepatoprotective properties. OBJECTIVE: We sought to investigate the protective activity of T. officinale fruit extract against sodium nitroprusside (SNP)-induced decreased cellular viability and increased lipid peroxidation in the cortex, hippocampus, and striatum of rats in vitro. To explain the mechanism of the extract's antioxidant activity, its putative scavenger activities against NO, DPPH·, OH·, and H(2)O(2) were determined. METHODS: Slices of cortex, hippocampus, and striatum were treated with 50 µM SNP and T. officinale fruit ethanolic extract (1-20 µg/mL) to determine cellular viability by MTT reduction assay. Lipid peroxidation was measure in cortical, hippocampal and striatal slices incubates with SNP (5 µM) and T. officinale fruit extract (1-20 µg/mL). We also determined the scavenger activities of T. officinale fruit extract against NO·, DPPH·, OH·, and H(2)O(2), as well as its iron chelating capacity. RESULTS: The extract (1, 5, 10, and 20 µg/mL) protected against SNP-induced decreases in cellular viability and increases in lipid peroxidation in the cortex, hippocampus, and striatum of rats. The extract had scavenger activity against DPPH· and NO· at low concentrations and was able to protect against H(2)O(2) and Fe(2+)-induced deoxyribose oxidation. CONCLUSION: T. officinale fruit extract has antioxidant activity and protects brain slices against SNP-induced cellular death. Possible mechanisms of action include its scavenger activities against reactive oxygen species (ROS) and reactive nitrogen species (RNS), which are attributed to the presence of phenolic compounds in the extract.

ABSTRACT

Although the production of patulin in apple fruits is mainly by Penicillium expansum, there is no information on the ability of heat resistant moulds that may survive pasteurization to produce this mycotoxin in juice packages during storage and distribution. In this study, the production of patulin by Byssochlamys spp (Byssochlamys nivea FRR 4421, B. nivea ATCC 24008 and Byssochlamys fulva IOC 4518) in cloudy and clarified apple juices packaged in laminated paperboard packages or in polyethylene terephthalate bottles (PET) and stored at both 21 degrees C and 30 degrees C, was investigated. The three Byssochlamys strains were able to produce patulin in both cloudy and clarified apple juices. Overall, the lower the storage temperature, the lower the patulin levels and mycelium dry weight in the apple juices (p<0.05). The greatest variations in pH and degrees Brix were observed in the juices from which the greatest mycelium dry weights were recovered. The maximum levels of patulin recovered from the juices were ca. 150 microg/kg at 21 degrees C and 220 microg/kg at 30 degrees C. HPLC-UV, HPCL-DAD and mass spectrometry analyses confirmed the ability of B. fulva IOC 4518 to produce patulin. Due to the heat resistance of B. nivea and B. fulva and their ability to produce patulin either in PET bottles or in laminated paperboard packages, the control of contamination and the incidence of these fungi should be a matter of concern for food safety. Control measures taken by juice industries must also focus on controlling the ascospores of heat resistant moulds.

ABSTRACT

O objetivo desta pesquisa foi estudar os efeitos das aflatoxinas sobre o desempenho e plumagem de frangos de corte. Foram utilizados 240 pintos machos, alimentados de 1 a 42 dias de idade com racão ad libitum sem aflatoxinas (grupo controle) e contaminada com 3mg de aflatoxinas/kg de racão (grupo tratado). As aves foram analisadas duas vezes ao dia em relacão ao seu desenvolvimento. O consumo de racão e peso dos animais foram avaliados ao final de cada fase de criacão (21, 35 e 42 dias). Ao final do experimento, realizaram-se as avaliacões patológicas, a pesagem das penas e a avaliacão dos seus aminoácidos. As aves do grupo tratado apresentaram sinais caracteristicos de intoxicacão por aflatoxinas, como desuniformidade da estatura, reducão no consumo de racão e ganho de peso e alteracões microscópicas e macroscópicas em órgãos e tecidos. Observou-se que o coracão e o fígado das aves intoxicadas apresentaram um incremento significativo no peso médio relativo, a massa de penas foi reduzida significativamente (33,8 por cento) e não houve alteracões na concentracão e na composicão dos aminoácidos, apesar do baixo desempenho dos animais intoxicados.
