ABSTRACT

The Psittaciformes are among the most popular pets due to their intelligence, ability, and ease of maintenance in small environments. However, the absence of adequate environmental stimuli generated by confinement can predispose these animals to characteristic stress conditions, leaving them susceptible to the triggering of various diseases, among which those of bacterial origin stand out. The objective of this study was to carry out a survey of enterobacteria and evaluate the antimicrobial sensitivity profile of bacteria isolated from parrots from a pet shop in the city of Fortaleza, Ceará. Ninety-six samples were collected from four pet shops (which were classified as A, B, C and D), eight samples of local swabs from budgerigar (Melopsittacus undulatus), were collected from each establishment eight from cockatiels (Nymphicus hollandicus) and eight from lovebirds (Agapornis sp.). Isolation of enterobacteria is under the methodology used by Lopes et al. (2015) with modifications. The method used to study bacterial resistance was the Kirby-Bauer method, following the standards stipulated by the Clinical and Laboratory Standards Institute. Sixty-eight enterobacteria strains from ten different species, Proteus mirabilis, Citrobacter diversus, Pantoea agglomerans, Escherichia coli, Providencia stuartii, Hafnia alvei, Proteus vulgaris, Serratia liquefaciens, Enterobacter sakasakii and Citrobacter amalonaticus, were isolated. P. agglomerans was the bacterium with the highest frequency of isolates from pet shop parrots, making up 23.5% of the isolates; the second-most isolated strain was P. mirabilis with 17.7%. In this study, 79% of the isolated strains were resistant to at least one class of antimicrobials tested. Tetracycline proved to be the most resistant antimicrobial (44%), followed by polymyxin B (38%) and nalidixic acid (25%). Among the 68 strains, 19% did not show resistance to any of the classes of antimicrobials tested. The condition of multidrug resistance - resistance to ≥3 classes of antimicrobials - was observed in 18% of the isolated strains.(AU)


Os psittaciformes estão entre os animais de estimação mais populares devido sua inteligência, habilidade, além da facilidade de manutenção da espécie em pequenos ambientes. Contudo, a ausência de estímulos ambientais adequados gerados pelo confinamento, podem predispor esses animais a quadros característicos de estresse, deixando-os susceptíveis ao desencadeamento de várias doenças dentre elas se destacam as de origem bacteriana. O objetivo desse trabalho foi realizar uma pesquisa de enterobactérias e avaliar o perfil de sensibilidade antimicrobiana de bactérias isoladas de psitacídeos de pet shop da cidade de Fortaleza, Ceará. Foram coletadas 96 amostras de quatro pet shops (os quais foram classificados em A, B, C e D), sendo coletados de cada estabelecimento oito amostras de suabes clocais oriundos de periquitos australianos (Melopsittacus undulatus), oito de calopsitas (Nymphicus hollandicus) e oito de agapornis (Agapornis sp.). O isolamento de enterobactérias está de acordo com a metodologia utilizada por Lopes et al. (2015) com modificações. O método utilizado para o estudo de resistência bacteriana foi o de Kirby-Bauer, seguindo os padrões estipulados pela Clinical and Laboratory Standards Institute. Foi isolado um total de 68 cepas de enterobactérias, de dez espécies diferentes, Proteus mirabilis, Citrobacter diversus, Pantoea agglomerans, Escherichia coli, Providencia stuartii, Hafnia alvei, Proteus vulgaris, Serratia liquefaciens, Enterobacter sakasakii e Citrobacter amalonaticus. Pantoea agglomerans foi a bactéria com maior percentagem de frequência dos isolados de psitacídeos de pet shop, perfazendo um total de 23,5% dos isolados, a segunda cepa mais isolada foi Proteus mirabilis com 17,7%. Neste estudo 79% das cepas isoladas foram resistentes a pelo menos uma classe de antimicrobianos testados, tetraciclina demonstrou ser o antimicrobiano com maior resistência (44%), seguido da polimixina B (38%) e do ácido nalidíxico (25%). Dentre as 68 cepas isoladas, 19% não apresentaram resistência a qualquer uma das classes de antimicrobianos testadas. A condição de multirresistência, ou seja, resistência a ≥3 classes de antimicrobianos foi observado em 18% das cepas isoladas.(AU)

Currently, this endemic species is only found in three areas in Ceará state, in Brazil. Cloacal swab samples were collected from 27 individuals and environmental swabs (drag swabs) from five nests used by these birds. Twenty-seven strains from nine species of Enterobacteriaceae were recovered from cloacal swabs, and the most prevalent bacteria strains were Hafnia alvei (22%) and Pantoea agglomerans (22%). From environmental nest samples, seven strains from three bacterial species were isolated, being the P. agglomerans the most frequent species (100%). Twenty-two of the 27 isolates (81.4%) exhibited antibiotic resistance, varying from one to eight of the 12 antimicrobials commonly used. Resistance to amoxicillin was the most prevalent (70.4%), followed by azithromycin (22.2%) and ceftriaxone (18.5%). None of the strains were resistant to gentamicin, tobramycin, ciprofloxacin or tetracycline. The H. alvei was the main species presenting multidrug resistance, including resistance against meropenem, which is an important finding. These results could provide interesting information on the health of these endangered wild grey-breasted parakeets. They could also indicate that the obtained isolates are part of a group of bacteria that are typical components of the enteric microbiota of birds, which present elevated rates of resistance to amoxicillin.(AU)


O periquito-de-cara-suja (Pyrrhura griseipectus) é uma espécie de psitacídeo considerado pela IUCN como ameaçado de extinção, resultado do comércio ilegal e do desmatamento. Atualmente, essa espécie endêmica é encontrada apenas em três áreas no estado do Ceará, Brasil. O objetivo deste estudo foi investigar a frequência e a diversidade de Enterobacteriaceae em periquitos de peito cinza adultos selvagens e determinar sua suscetibilidade a agentes antimicrobianos. Amostras de suabes cloacais foram coletadas de 27 indivíduos e de suabes ambientais (suabes de arrasto) de cinco ninhos utilizados por essas aves. Vinte e sete cepas de nove espécies de Enterobacteriaceae foram isoladas a partir de suabes cloacais, sendo as cepas bacterianas mais prevalentes Hafnia alvei (22%) e Pantoea agglomerans (22%). Das amostras ambientais de ninhos foram isoladas sete linhagens de três espécies bacterianas, sendo P. agglomerans a espécie mais frequente (100%). Vinte e dois dos 27 isolados (81,4%) exibiram resistência a antibióticos, variando de um a oito dos 12 antimicrobianos comumente usados. A resistência a amoxicilina foi a mais prevalente (70,4%), seguida por azitromicina (22,2%) e ceftriaxona (18,5%). Nenhuma das cepas era resistente à gentamicina, tobramicina, ciprofloxacina ou tetraciclina. H. alvei foi a principal espécie que apresentou resistência a múltiplas drogas e que também esteve associada a um outro achado relevante desta pesquisa, que foi a detecção de um caso de resistência ao meropenem. ABSTRACT

This study aimed to verify the presence of members from the Enterobacteriaceae family and determine antimicrobial susceptibility profiles of the isolates in canaries bred in northeastern Brazil; in addition, the presence of diarrheagenic Escherichia coli (DEC) and avian pathogenic Escherichia coli (APEC) was also verified in these birds. Samples were collected during an exhibition organized by the Brazilian Ornithological Federation in July 2015 in Fortaleza, Brazil. A total of 88 fecal samples were collected and submitted to pre-enrichment step using buffered peptone water, followed by enrichment with the following broths: brain-heart infusion, Rappaport-Vassiliadis, and Selenite-Cystine. Subsequently, aliquots were streaked on MacConkey, brilliant green and salmonella-shigella agar plates. Colonies were selected according to morphological characteristics and submitted to biochemical identification and antimicrobial susceptibility tests with disk-diffusion technique. E. coli strains were evaluated for the presence of eight DEC genes and five APEC genes through conventional polymerase chain reaction (PCR) screening. The most frequent species observed were Pantoea agglomerans (25%), Serratia liquefaciens (12.5%), and Enterobacter aerogenes (9.1%). A single rough strain of Salmonella enterica subsp. enterica was identified in one sample (1.1%). High resistance rates to amoxicillin (78.7%) and ampicillin (75.4%) were identified. Polymyxin B (9.8%), gentamycin (6.6%), and enrofloxacin (6.6%) were the most efficient antibiotics. The total number of multidrug-resistant strains (isolates resistant to more than three antimicrobial classes) was 23 (37.7%). Four E. coli strains were tested for the virulence genes, and two were positive for APEC virulence genes: one strain was positive for iutA and the other for hlyF. In conclusion, canaries in northeastern Brazil participating in exhibitions may present Salmonella spp., Escherichia coli and other enterobacteria in the intestinal microbiota with antimicrobial resistance. These results indicate that, although the E. coli strains recovered from canaries in this study have some virulence genes, they still do not fulfill all the requirements to be considered APEC.(AU)


O objetivo deste trabalho foi verificar a presença de enterobactérias e determinar o perfil de sensibilidade aos antimicrobianos dos isolados oriundos de canários belgas criados em cativeiro do Nordeste do Brasil, adicionalmente verificou-se a presença de Escherichia coli diarreiogênicas (DEC) e E. coli patogênica aviária (APEC) nesses animais. A colheita das amostras ocorreu durante uma exposição de canários belgas organizada pela Federação Ornitológica do Brasil (FOB), em julho de 2015, na cidade de Fortaleza, Ceará, Brasil. Um total de 88 amostras de fezes foram coletadas e submetidas a pré-enriquecimento utilizando água peptonada, caldo de enriquecimento Brain Heart Infusion, Rappaport-Vassiliadis e Selenito-Cistina. Fez-se triagem em placas de ágar MacConkey, Verde Brilhante e ágar Salmonella Shigella. As colônias foram selecionadas e submetidas à identificação bioquímica e susceptibilidade antimicrobiana. Estirpes de Escherichia coli foram avaliadas quanto a presença de 8 genes de virulência de DEC e cinco de APEC por reação em cadeia da polimerase convencional (PCR). As enterobactérias encontradas com maior frequência foram Pantoea agglomerans (25%), Serratia liquefaciens (12,5%) e Enterobacter aerogenes (9,1%). Uma única estirpe de Salmonella enterica subsp. enterica (rugosa) esteve presente em um dos isolados (1,1%). Altos percentuais de resistência foram encontrados para dois antibióticos: amoxicilina (78,7%) e ampicilina (75,4%). Polimixina B (9,8%), gentamicina (6,8%) e enrofloxacina (6,5%) foram os antibióticos com melhor eficiência. O total de estirpes multirresistentes (a mais de três classes de antimicrobianos) foi de 23 (37,7%). Das quatro estirpes de E. coli isoladas, duas foram positivas para os genes de APEC, sendo uma estipe para o gene iss e outra para os genes iutA e hlyF. Portanto, canários belgas criados em cativeiro no Brasil que participam de exposições podem apresentar Salmonella spp., Escherichia coli e outras enterobactérias em sua microbiota intestinal com resistência antimicrobiana. Estes resultados indicam que as estirpes de E. coli isoladas de canário belga no presente estudo apresentam alguns, mas não todos, genes de virulência para serem caracterizadas como E. coli patogênica para aves (APEC).(AU)

ABSTRACT

Fanconi anemia is a rare, cancer-prone disease with mutations in 22 genes. The primary defect results in altered DNA repair mechanisms that fuel a severe proinflammatory condition in the bone marrow, leading to cellular depletion of the hematopoietic system and eventually to bone marrow failure. During the past three decades, a plethora of dysfunctions have been highlighted in the Fanconi anemia phenotype, but recent research allows us to glimpse an even more complex scenario where defective lipid metabolism could have important consequences in hematopoietic stem cell differentiation.

ABSTRACT

This study aimed to isolate Escherichia coli and Salmonella enterica from captured feral pigeons in Fortaleza, Brazil, and, in addition to evaluate the antimicrobial susceptibility profiles and diagnose diarrheagenic E. coli strains. Pigeons were captured in four public locations in Fortaleza with three techniques. Individual cloacal swab samples were collected and submitted to bacterial isolation, biochemical identification and antimicrobial susceptibility test. Disk diffusion technique was used with twelve antibiotics. E. coli strains were submitted to DNA extraction followed by PCR to diagnose five diarrheagenic pathotypes. A total of 124 birds were captured. One bird was positive for Salmonella enterica (0.81%) and 121 (97.58%) were positive for E. coli. Among these, 110 isolates were submitted to antimicrobial susceptibility test and 28.18% (31/110) presented resistance to at least one antibiotic. Resistance to azithromycin was the most frequent (21.82%), followed by tetracycline (10.91%) and sulfamethoxazole with trimethoprim (8.9%). Multidrug resistance, calculated as a resistance to at least 3 antimicrobial classes, was identified in 3.64% (4/110) of strains. The maximum number of antimicrobial classes to which one strain was resistant was seven. Results demonstrated nine different resistance profiles and the most frequent was tetracycline and sulfamethoxazole with trimethoprim (4 strains), followed by chloramphenicol, azithromycin, tetracycline and sulfamethoxazole with trimethoprim (3 strains). Amoxicillin with clavulanic acid and tobramycin presented lowest levels of antimicrobial resistance, to which none of the tested strains were resistant. A single strain was positive for the eltB gene, which is a diagnostic tool to identify the Enterotoxigenic E. coli (ETEC) pathotype. None of the other investigated genes (stx1, stx2, estA, eaeA, ipaH, aatA and aaiC) were identified. The single isolate of S. enterica was a rough strain of Salmonella enterica subsp. enterica, but serotype identification was not possible. However, this isolate presented resistance to amoxicillin, amoxicillin with clavulanic acid, tetracycline and sulfamethoxazole with trimethoprim. Therefore, captured feral pigeons of Fortaleza presented a low prevalence of S. enterica and diarrheagenic E. coli. Considering the investigated pathogens, our results suggest a good health status and a low public health risk. However, important antimicrobial resistance profiles were identified.(AU)


O objetivo deste estudo foi isolar cepas de Escherichia coli e Salmonella enterica de pombos urbanos capturados em Fortaleza, Brasil, e avaliar os perfis de resistência antimicrobiana dos isolados, bem como diagnosticar patotipos diarreiogênicos de E. coli. Pombos foram capturados em quatro locais públicos de Fortaleza utilizando três técnicas. Amostras individuais de suabes cloacais foram coletadas e submetidas a isolamento bacteriano, seguido de identificação bioquímica e teste de susceptibilidade a antimicrobianos. A técnica de disco difusão foi utilizada para avaliar resistência antimicrobiana a doze antibióticos. Cepas de E. coli foram submetidas à extração de DNA seguido de PCR para o diagnóstico de cinco patotipos diarreiogênicos. Um total de 124 aves foram capturadas, a partir das quais em uma houve isolamento de Salmonella enterica (0,81%) e em 121 (97,58%) houve isolamento de E. coli. Destas, 110 isolados foram submetidos a teste de suscetibilidade a antimicrobianos e 28,18% (31/110) apresentaram resistência a pelo menos um antibiótico. Resistência a azitromicina foi a mais frequente (21,82%), seguida por tetraciclina (10,91%) e sulfametoxazol com trimetoprim (8,9%). Resistência a múltiplas drogas foi identificada em 3,64% (4/110) dos isolados e o número máximo de antibióticos aos quais uma única cepa foi resistente foi sete. Resultados demonstraram nove diferentes perfis de resistência e o mais frequente foi tetraciclina e sulfametoxazol com trimetoprim (4 cepas), seguido por cloranfenicol, azitromicina, tetraciclina e sulfametoxazol com trimetoprim (3 cepas). Amoxicilina com ácido clavulânico e tobramicina foram os antibióticos com menor resistência antimicrobiana, aos quais nenhuma cepa apresentou resistência. Uma única cepa foi positiva para o gene eltB que é usado para diagnóstico do patotipo E. coli enterotoxigênica (ETEC), enquanto que os demais genes investigados (stx1, stx2, estA, eaeA, ipaH, aatA e aaiC) não foram identificados. A única cepa de S. enterica isolada foi identificada como uma cepa rugosa de Salmonella enterica subsp. enterica e, portanto, a identificação do sorotipo não foi possível. Entretanto, este isolado apresentou resistência a amoxicilina, amoxicilina com ácido clavulânico, tetraciclina e sulfametoxazol com trimetoprim. Portanto, pombos urbanos capturados em Fortaleza apresentaram baixa prevalência de cepas de S. enterica e E. coli diarreiogênicas. Considerando os patógenos investigados, os resultados encontrados sugerem um bom status sanitário destas aves e um baixo risco à saúde pública. Entretanto, importantes perfis de resistência antimicrobiana foram identificados.(AU)

ABSTRACT

Insulations for rocket motors such as Flexible Thermal Protections (FTPs) and Rigid Thermal Protection (RTPs) act as thermal barriers against the hyperthermal environment from the solid propellant combustion. FTPs present dual function: to extend RTPs performance, and to attenuate the propellant contraction. FTPs used in the Brazilian Space Program have asbestos in their composition since the 70´s; however, they are hazardous for human health. In this context, a mixture of Expanded Perlite (PExp) and Cork Powder (CP) was investigated as a replacement for asbestos. Results showed reduction about 21 % in density and an increase of 13 % in the ablative properties. The low mechanical properties not interfere in this type of FTP due to your function of attenuate the propellant contraction.

ABSTRACT

Accelerometers are becoming ever more important sensors in animal-attached technology, providing data that allow determination of body posture and movement and thereby helping to elucidate behaviour in animals that are difficult to observe. We sought to validate the identification of sea turtle behaviours from accelerometer signals by deploying tags on the carapace of a juvenile loggerhead (Caretta caretta), an adult hawksbill (Eretmochelys imbricata) and an adult green turtle (Chelonia mydas) at Aquarium La Rochelle, France. We recorded tri-axial acceleration at 50 Hz for each species for a full day while two fixed cameras recorded their behaviours. We identified behaviours from the acceleration data using two different supervised learning algorithms, Random Forest and Classification And Regression Tree (CART), treating the data from the adult animals as separate from the juvenile data. We achieved a global accuracy of 81.30% for the adult hawksbill and green turtle CART model and 71.63% for the juvenile loggerhead, identifying 10 and 12 different behaviours, respectively. Equivalent figures were 86.96% for the adult hawksbill and green turtle Random Forest model and 79.49% for the juvenile loggerhead, for the same behaviours. The use of Random Forest combined with CART algorithms allowed us to understand the decision rules implicated in behaviour discrimination, and thus remove or group together some 'confused' or under--represented behaviours in order to get the most accurate models. This study is the first to validate accelerometer data to identify turtle behaviours and the approach can now be tested on other captive sea turtle species.

ABSTRACT

Diarrheagenic Escherichia coli (DEC) are considered one of the major causes of human diarrhea in developing countries. Some studies have pointed wild birds as important reservoirs for these pathogens. However, scarce species from the Psittaciformes order have been investigated. This study aimed to evaluate the presence of DEC strains in Psittaciformes from illegal wildlife trade. A total of 78 E. coli strains isolated from cloacal swab samples of 167 Psittaciformes in the Ceará State, Brazil, were evaluated regarding the presence of the following DEC virulence genes by polymerase chain reaction (PCR): eaeA and bfpA genes (Enteropathogenic E. coli - EPEC); stx1 and stx2 (Shiga toxin-producing E. coli - STEC); estA and eltB (Enterotoxigenic E. coli - ETEC); ipaH (Enteroinvasive E. coli - EIEC); aatA and aaiC (Enteroaggregative E. coli - EAEC). Positive strains for eaeA and bfpA genes were considered typical EPEC, while strain positive exclusively for the eaeA gene were classified as atypical EPEC. The eaeA gene was identified in 20 E. coli strains and bfpA in 22 isolates. In addition, 11 and 9 belonged to tEPEC and aEPEC, respectively. No strain was positive for stx1 or stx2. A total of 47 (60.3%) strains and a total of 136 birds (81.4%) were negative for the remaining DEC pathotypes investigated. In conclusion, psittacine from illegal wildlife trade in Ceará State, Brazil, presented a relevant prevalence of typical and atypical EPEC, potentially playing a role as reservoirs of DEC strains in the environment. Thus, proper control measures must be adopted to block the spread of these pathogens.(AU)


Escherichia coli diarreiogênicas (DEC) são consideradas uma das causas mais importantes de diarreia em países em desenvolvimento. Alguns estudos têm apontado aves silvestres como importantes reservatórios destes patógenos, entretanto, poucas espécies da ordem Psittaciformes têm sido investigada. O objetivo deste estudo foi analisar a presença de cepas de E. coli diarreiogênicas em Psittaciformes do tráfico de animais silvestres. Um total de 78 amostras de E. coli isoladas de suabes cloacais provenientes de 167 de Psittaciformes do Ceará, Brasil, foram avaliadas quanto a presença dos seguintes genes de virulência DEC por meio de reação em cadeia de polimerase (PCR): eaeA e bfpA (E. coli Enteropatogênica - EPEC); stx1 e stx2 (E. coli produtora de Shiga - STEC); estA e eltB (E. coli Enterotoxigênica - ETEC); ipaH (E. coli Enteroinvasiva - EIEC); aatA e aaiC (E. coli Enteroagregativa - EAEC). As cepas positivas para os genes eaeA e bfpA foram consideradas EPEC típicas, enquanto que as positivas exclusivamente para o gene eaeA foram classificadas como EPEC atípicas. O gene eaeA foi identificado em 20 cepas de E. coli e o gene bfpA em 22 dos isolados. Adicionalmente, 11 e 9 cepas foram classificadas como EPEC típicas e atípicas, respectivamente. Nenhuma cepa foi positiva para os genes stx1 e stx2. Um total de 47 cepas (60,3%) e um total de 136 aves (81,4%) foram negativas para os demais patotipos DEC pesquisados. Em conclusão, psitacídeos provenientes do tráfico de aves silvestres do estado do Ceará, Brasil, apresentaram relevante prevalência de EPEC típicas e atípicas, potencialmente participando como reservatórios de cepas DEC no ambiente. Portanto, medidas de controle devem ser adotadas para inibir a disseminação destes patógenos.(AU)

ABSTRACT

The Enterobacteriaceae family contains potentially zoonotic bacteria, and their presence in canaries is often reported, though the current status of these in bird flocks is unknown. Therefore, this study aimed to identify the most common genera of enterobacteria from canaries (Serinus canaria) and their antimicrobial resistance profiles. From February to June of 2013, a total of 387 cloacal swab samples from eight domiciliary breeding locations of Fortaleza city, Brazil, were collected and 58 necropsies were performed in canaries, which belonged to the Laboratory of Ornithological Studies. The samples were submitted to microbiological procedure using buffered peptone water and MacConkey agar. Colonies were selected according to their morphological characteristics on selective agar and submitted for biochemical identification and antimicrobial susceptibility. A total of 61 isolates were obtained, of which 42 were from cloacal swabs and 19 from necropsies. The most isolated bacteria was Escherichia coli with twenty five strains, followed by fourteen Klebsiellaspp., twelve Enterobacterspp., seven Pantoea agglomerans, two Serratiaspp. and one Proteus mirabilis. The antimicrobial to which the strains presented most resistance was sulfonamides with 55.7%, followed by ampicillin with 54.1% and tetracycline with 39.3%. The total of multidrug-resistant bacteria (MDR) was 34 (55.7%). In conclusion, canaries harbor members of the Enterobacteriaceae family and common strains present a high antimicrobial resistance rate, with a high frequency of MDR bacteria.


A família Enterobacteriaceae possui bactérias com potencial zoonótico e a presença destas bactérias em canários é relatada na literatura, porém a realidade dos plantéis de criadores de canários é desconhecida. Portanto, este trabalho teve como objetivo isolar enterobactérias de canários belga (Serinus canarius) com o intuito de conhecer os gêneros mais comuns nestas aves e suas respectivas resistências a antimicrobianos. De fevereiro a junho de 2013 foram coletadas 387 amostras de swabs cloacais de canários de oito propriedades da cidade de Fortaleza, Brasil e de 58 necropsias de aves do acervo próprio do Laboratório de Estudos Ornitológicos. As amostras foram submetidas a isolamento microbiológico utilizando-se água peptonada e ágar MacConkey. As colônias foram selecionadas de acordo com suas características morfológicas nas placas, submetidas à tipificação bioquímica para identificação e ao teste de sensibilidade a antimicrobianos. Foram isoladas 61 cepas, sendo 42 de suabes cloacais e 19 de necropsias. A bactéria mais isolada foi Escherichia coli com vinte e cinco cepas, seguida por catorze Klebsiella spp., doze Enterobacter spp., sete Pantoea agglomerans, duas Serratiaspp. e uma cepa de Proteus mirabilis. As cepas apresentaram maior resistência a sulfonamidas com 55,7%, seguidas por ampicilina com 54,1% e tetraciclina com 39,3%. Além disso, o total de cepas resistentes a múltiplas drogas (RMD) foi 34 (55,7%). Portanto, conclui-se que os canários albergam enterobactérias e que as cepas apresentam alto índice de resistência a antimicrobianos, com alta frequência de cepas RMD.

OBJECTIVE: To measure VEGF/PlGF heterodimer levels in preterm infants born to mothers with preeclampsia. METHODS: Neonates with birth weight <2000 g and gestational age ≤34 weeks were divided into two groups: born to mothers with Preeclampsia (PE) and controls. Neonates transferred from outside after the 72nd hour of life, death before blood collection, major congenital malformations or inborn errors of metabolism, and mothers with multiple pregnancies, STORCH complex infections, HIV or autoimmune conditions were excluded. Blood was collected within 72 h of birth and again at 28 days. VEGF/PlGF heterodimer levels were measured by ELISA. RESULTS: We included 73 neonates (24 born to mothers with PE and 49 without PE). Mean gestational age was 30.32 ± 2.88 weeks and mean birth weight was 1288.62 ± 462.22 g. Median VEGF/PlGF levels were significantly higher in infants born to mothers with PE. VEGF/PlGF levels were inversely proportional to birth weight. There were no between-group differences in blood samples collected at age 28 days. CONCLUSION: Higher VEGF/PlGF levels were higher in neonates exposed to PE, and there was a significant negative correlation between birth weight and VEGF/PlGF levels. Further studies to elucidate the role of this substance in the fetal and neonatal period are needed.

ABSTRACT

INTRODUÇÃO: Existem poucos estudos contemporâneos avaliando o implante de stent direto em pacientes com infarto agudo do miocárdio com supradesnivelamento do segmento ST (IAM). Método: Estudo de coorte prospectivo com pacientes consecutivamente atendidos com IAM em um centro de referêncioa em cardiologia entre dezembro de 2009 e janeiro de 2010. Dados clínicos e laboratoriais, características angiográficas e evolução hospitalar foram avaliados. Foram excluídos pacientes com mais de 12 horas de evolução, com menos de 18 anos de idade ou por recusa em participar do estudo. Os pacientes submetidos a implante de stent direto foram comparados àqueles tratados com pré-dilatação. Os dados foram armazenados em banco de dados dedicado e analisados com SPSS 17.0. Resultados: No período de estudo, 98 pacientes foram submetidos a angioplastia primária, dos quais em 33 (34,7 por cento) foi implantado stent direto e em 65 (65,3 por cento) foi realizada pré-dilatação. A média de idade dos 98 pacientes foi de 58,6 +- 10,6 anos e 63 por cento eram do sexo masculino. Os procedimentos com stent direto foram menos frequentes em lesões calcificadas (9 por cento vs. 30 por cento; P + 0,02) e mais frequentes naqueles com TIMI 3 pré-procedimento (41 por cento vs. 18 por cento; P=0,06), sendo a tromboaspiração...


BACKGROUND: There are few contemporary studies assessing direct stenting in patients with acute ST-segment elevation myocardial infarction (AMI). METHOD: Prospective cohort study in consecutive patients with AMI seen at a reference cardiology center from December 2009 to January 2010. Clinical and laboratory data, angiographic characteristics and hospital outcomes were evaluated. Patients with over 12 hours of symptom onset, with less than 18 years of age and those who denied participating in the study were excluded. Patients undergoing direct stenting were compared to those treated with stent with balloon pre-dilatation. Data were entered in a dedicated database and analyzed by SPSS 17.0. RESULTS: During the study period, 98 patients were submitted to primary angioplasty, of which 33 (34.7%) received direct stenting and 65 (65.3%) underwent stent with balloon pre-dilatation. Mean age was 58.6 ± 10.6 years and 63% were men. Direct stenting was less frequent in calcified lesions (9% vs. 30%; P = 0.02) and more frequent in those with TIMI 3 before the procedure (41% vs. 18%; P = 0.06) and thrombus aspiration was more frequently used in these cases (41% vs. 14%; P = 0.003). There was no statistically significant difference for the clinical outcome in both groups of patients. CONCLUSION: Direct stenting was more frequently used in patients with normal flow prior the procedure and in combination with thrombus aspiration and less used in calcified lesions. It was not associated to significant differences in clinical outcome rates.

ABSTRACT

INTRODUÇÃO: O implante de stents coronários é atualmente a técnica mais utilizada para revascularização miocárdica. A análise de registros com grande número de pacientes é importante para avaliação da efetividade de um tratamento na prática clínica do mundo real. MÉTODO: Os perfis clínico e angiográfico, a incidência de complicações e os desfechos tardios foram analisados em uma coorte de pacientes tratados com intervenções coronárias percutâneas (ICPs) com implante de stent no período de abril de 1996 a dezembro de 2007. Os dados foram coletados prospectivamente em um banco de dados informatizado e analisados com programa SPSS 11.0. RESULTADOS: No período do estudo, 5.284 pacientes foram submetidos a 6.254 intervenções coronárias percutâneas com implante de stents. A média de idade foi de 60,59 ± 10,63 anos e 73% eram do sexo masculino. A maioria foi tratada por angina instável (50%) e 14% por infarto agudo do miocárdio (IAM) em evolução. Sucesso clínico foi obtido em 95,8% dos pacientes, complicações vasculares ocorreram em 3,6% e trombose de stent foi observada em 0,5%. Seguimento clínico de um ano foi obtido em uma amostra aleatória de 1.610 pacientes, sendo observadas as seguintes taxas de eventos: óbito cardiovascular, 3,7%; IAM, 2,9%; e revascularização miocárdica, 15,7%. Os preditores de eventos cardiovasculares maiores foram o diâmetro de referência do vaso [razão de chance (OR): 0,69; intervalo de confiança (IC) 95%: 0,51-0,94; P = 0,018] e comprometimento multiarterial (OR: 1,38; IC 95%: 1,001-1,90; P = 0,049). CONCLUSÃO: Os resultados demonstram a efetividade das ICPs em grande população de pacientes não selecionados do mundo real.


BACKGROUND: Coronary stenting is currently the most widely used technique for myocardial revascularization. The analysis of registries with large numbers of patients is important to evaluate treatment efficacy in the real world clinical practice. METHOD: The clinical and angiographic profile, incidence of complications and late outcomes in a cohort of patients treated with percutaneous coronary interventions (PCI) with coronary stent implantation from April 1996 to December 2007 were analyzed. Data was prospectively collected using a dedicated database, and analyzed by SPSS 11.0. RESULTS: In the study period, 5,284 patients underwent 6,254 stent implantation procedures. Mean age was 60.59 ± 10.63 years, and 73% were male. Most patients were treated for unstable angina (50%) and 14% for acute myocardial infarction (AMI). Clinical success was achieved in 95.8 % of procedures, with vascular complications occurring in 3.6% and stent thrombosis in 0.5% of the patients. One-year clinical follow-up was obtained in a random sample of 1,610 patients, with the following event rates: cardiovascular death = 3.7%, AMI = 2.9%, CABG = 15.7%. The predictors of major cardiovascular events were the reference diameter of the vessel [odds ratio (OR): 0.69, 95% confidence interval (CI): 0.51-0.94; P = 0.018] and multivessel disease (OR: 1.38; 95% CI: 1,001-1,90; P = 0.049). CONCLUSION: These results show the efficacy of percutaneous coronary interventions in a large population of unselected patients of the real world clinical practice.

ABSTRACT

Pelizaeus-Merzbacher disease (PMD) is an X-linked myelination disorder most frequently caused by duplication of a genomic segment of variable length containing the PLP1 gene. We studied five PMD male patients affected by the classic PMD form carrying a PLP1 gene duplication. On the basis of clinical and neuroradiological features, two of the five patients appeared to be the most severely affected. In order to establish a possible genotype-phenotype correlation, the extent of the duplication was determined in each patient and in the respective mother by quantifying the copy number of genomic markers surrounding the PLP1 gene by a real-time PCR-based approach. Duplications, ranging in size from 167-195 to 580-700 kb, were in the same genomic interval of the majority of the reported duplications. The extent of the duplicated genomic segments does not correlate with the clinical severity. Interestingly enough, each duplication had one of the two breakpoints in or near to low copy repeats (LCRs), supporting recent evidence concerning a possible role of LCRs in the generation of the duplications in PMD.

ABSTRACT

Mucolipidosis type II (ML II) is a fatal, autosomal recessive, lysosomal storage disorder characterized by severe clinical and radiologic features. ML II results from mutations in alpha and beta subunits, encoded by the GlcNAc-1-phosphotransferase gene (GNPTAB). Most of the 40 different GNPTAB mutations reported so far are insertions and deletions predicting diverse types of aberrant proteins. Alu mobile elements have however never been involved in these events up to now. The Italian ML II patient of our study showed an Alu retrotrasposition in GNPTAB exon 5. The Alu insertion mutation (NM_024312.3:c.555_556insHSU14569) generated a transcript with a skipping of the target exon 5 and a frameshift p.S122fs, causing a premature translation termination codon at position 123. This insertion mutation was found in compound heterozygosity with the frameshift p.S887KfsX33, resulting from a new mono-nucleotide duplication (c.2659dupA) that occurred in GNPTAB exon 13. A possible involvement of cis-splicing elements having an exonic location, such as exon enhancers (ESEs), is discussed as mechanism that led to the production of the aberrant mRNA splicing.

ABSTRACT

Gaucher disease (GD) is a lysosomal storage disorder with a wide spectrum of phenotypic presentations. We report the case histories of two adult brothers with GD who developed both parkinsonism and psychiatric symptoms. Direct sequencing and real-time polymerase chain reaction were used to establish that the patients were homozygous for mutation L444P. While parkinsonism has been described previously in GD, these patients had atypical features, including a complicated mood disorder. The comorbidity of GD and a mood disorder is a new finding, as psychiatric manifestations of GD have been described rarely. The etiology of the mental illness could be related to the processes contributing to the development of parkinsonism.

ABSTRACT

Niemann-Pick disease (NPD) results from the deficiency of lysosomal acid sphingomyelinase (SMPD1). To date, out of more than 70-disease associated alleles only a few of them have a significant frequency in various ethnic groups. In contrast, the remainder of the mutations are rare or private. In this paper we report the molecular characterization of an Italian series consisting of twenty-five NPD patients with the severe neurodegenerative A phenotype. Mutation detection identified a total of nineteen different mutations, including 14 novel mutations and five previously reported lesions. The known p.P189fs and the novel p.T542fs were the most frequent mutations accounting for 34% and 18% of the alleles, respectively. Screening the alleles for the three common polymorphisms revealed the variant c.1516G>A (exon 6) and the repeat in exon 1, but not the variant c.965C>T (exon 2). In absence of frequent mutations, the prognostic value of genotyping is limited. However, new genotype/phenotype correlations were observed for this disorder that could in the future facilitate genetic counseling and guide selection of patients for therapy.

ABSTRACT

The major challenge of this paper comprises an attempt to compare the wealth of folk-information based on "traditional knowledge" with the efficacy of a chemo-biological background based on scientific knowledge. Surprisingly, correlations between plant uses (as food and medicine) and evolutionary status according to morphology (as assigned by Sporne indices) and metabolism (as assigned by diversity of phytochemicals) obey strikingly uniform systematic and evolutionary trends. The resulting patterns suggest a dynamic chemical mechanism for the bioactivity in plants regulated by the antagonistic gallate/caffeate pair. This finding is an important step toward the construction of a coherent chemo-biological language by a dynamic holistic quantitative methodology, one of the most potent prospects for understanding the functioning of nature.

ABSTRACT

Hibernating adults of Oebalus poecilus (Dallas) were collected from May to December of 2000, in a bamboo thicket located in Eldorado do Sul County (30º 02’ S e 51º 23’ W), RS. The fungi isolation was done from dead insects, which presented a white micelial growth, in PDA culture medium. The pathogenicity of the isolate was tested in laboratory conditions using three concentrations of conidium suspension (5 x 105; 5 x 107; 1.25 x 109 conidia/ml). The fungus was identified as Beauveria bassiana (Bals.) Vuill. and was deposited in a culture collection of entomopathogenic fungi in Embrapa Sojawith access number Bb353. The isolate was pathogenic in all concentrations, being the highest mortality (84.4%) recorded in 1.25 x 109 conídios/ml concentration. This is the first record of this fungus on O. poecillus, in natural conditions.


Adultos hibernantes de Oebalus poecilus (Dallas) foram coletados de maio a dezembro de 2000, em um taquaral situado no município de Eldorado do Sul (30º 02’ S e 51º 23’ W), RS. A partir deum inseto morto que apresentava crescimento micelial branco sobre o corpo, realizou-se o isolamento do fungo em meio de cultura BDA. Em laboratório testou-se a patogenicidade do isolado em três concentrações de suspensões de conídios (5 x 105; 5 x 107; 1,25 x 109 conídios/ml). O fungo foi identificado como Beauveria bassiana (Bals.) Vuill., e armazenado na coleção de culturas de fungos entomopatogênicos da Embrapa Soja com o número de acesso Bb353. O isolado mostrou-se patogênico em todas as concentrações utilizadas, sendo o maior percentual de mortalidade (84,4%) registrado na concentração mais elevada de conídios. Este foi o primeiro registro, em condições naturais, de B. bassiana sobre O. poecilus.
