ABSTRACT

The Brazilian Cerrado biome consists of a great variety of endemic species with several bioactive compounds, and Anadenanthera peregrina (L.) Speg is a promising species. In this study, we aimed to perform phytochemical characterization and evaluate the antioxidant and antibacterial activities against Staphylococcus aureus and Escherichia coli of the hydroethanolic extract of A. peregrina stem bark. The barks were collected in the Botanical Garden of Goiânia, Brazil. The hydroethanolic extract was obtained by percolation and subjected to physicochemical screening, total phenolic content estimation, high-performance liquid chromatography (HPLC) fingerprinting, and antioxidant (IC50 values were calculated for the 2,2-diphenyl-1-picrylhydrazyl assay - DPPH) and antibacterial activity determination. The pH of the extract was 5.21 and density was 0.956 g/cm3. The phytochemical screening indicated the presence of cardiac glycosides, organic acids, reducing sugars, hemolytic saponins, phenols, coumarins, condensed tannins, flavonoids, catechins, depsides, and depsidones derived from benzoquinones. The extract showed intense hemolytic activity. The total phenolic content was 6.40 g GAE 100 g-¹. The HPLC fingerprinting analysis revealed the presence of gallic acid, catechin, and epicatechin. We confirmed the antioxidant activity of the extract. Furthermore, the extract did not inhibit the growth of E. coli colonies at any volume tested, but there were halos around S. aureus colonies at all three volumes tested. These results contribute to a better understanding of the chemical composition of A. peregrina stem bark and further support the medicinal applications of this species.

ABSTRACT

Sepsis induces a severe systemic inflammatory response that may result in multiple organ dysfunction and death. Studies using a protein derived from natural Hevea brasiliensis (rubber tree) latex, denominated Hev b 13, have demonstrated important anti-inflammatory effects, but no data have been published regarding its effects on sepsis. The aim of this study was to investigate the effects of Hev b 13 on the inflammatory response and lung lesions of septal rats. Male Wistar rats were submitted to cecal ligation and puncture (CLP), randomized into groups and treated with subcutaneously administered doses of 0.5/2.0/3.0 mg/Kg of Hev b 13. Next, animals were subdivided into three different points in time (1, 6 and 24 hours after treatments) for collection of blood samples and euthanasia accompanied by organ removal. Total and differential leukocyte counts, cytokine dosage and histological assessment were analyzed. Treatment with Hev b 13 resulted in a significant decline in total and differential leukocytes as well as suppression of TNF-α and IL-6 production, associated with the increase in IL-10 and IL-4 in plasma and lung tissue. Moreover, it reduced morphological and pathological changes found in the lungs, including neutrophil infiltration, edema and alveolar thickening. The present study concluded that Hev b 13 exerts anti-inflammatory effects and attenuates lung lesions in septal rats, showing potential for clinical application.(AU)

O objetivo deste estudo foi investigar os efeitos da Hev b 13 na resposta inflamatória e na lesão pulmonar de ratos com sepse. Ratos machos da linhagem Wistar foram submetidos a ligação e perfuração do ceco (LPC), randomizados em grupos e tratados com as doses 0,5/2,0/3,0 mg/Kg de Hev b 13 subcutâneo. Após subdividiu-se os animais em três pontos diferentes de tempo (1, 6 e 24 horas após os tratamentos) para coleta de amostras sanguíneas e eutanásia com remoção dos órgãos. Contagem total e diferencial de leucócitos, dosagem de citocinas e avaliação histológica foram analisadas. O tratamento com a Hev b 13 resultou em diminuição significativa de leucócitos totais e diferenciais bem como suprimiu a produção de TNF-α e IL-6, associado ao aumento de IL-10 e IL-4 no plasma e tecido pulmonar. Além disso, reduziu as alterações morfológicas e patológicas encontradas nos pulmões, incluindo infiltrado de neutrófilos, edema e espessamento alveolar. Este estudo concluiu que a Hev b 13 tem efeitos anti-inflamatórios e atenua lesões pulmonares em ratos com sepse, apresentando potencialidades para aplicabilidade clínica.(AU)

Abstract Predatory mites that belong to the Phytoseiidae family are one of the main natural enemies of phytophagous mites, thus allowing for their use as a biological control. Phytoseiulus macropilis (Banks, 1904) (Acari: Phytoseiidae) is among the main species of predatory mites used for this purpose. Tetranychus urticae Koch, 1836 (Acari: Tetranychidae) is considered to be one of the most important species of mite pests and has been described as attacking over 1,100 species of plants in 140 families with economic value. The objective of the present study was to investigate, in the laboratory, the reproductive parameters of the predatory mite P. macropilis when fed T. urticae. Experiments were conducted under laboratory conditions at 25 ± 2 °C of temperature, 70 ± 10% RH and 14 hours of photophase. In addition, biological aspects were evaluated and a fertility life table was established. The results of these experiments demonstrated that the longevity of adult female was 27.5 days and adult male was 29.0 days. The population was estimated to increase approximately 27 times (Ro) in mean generation time (T), which was 17.7 days. Lastly, the mite population grew 1.2 times/day (λ) and doubled every 3.7 days (TD).


Resumo Os ácaros predadores pertencentes à família Phytoseiidae constituem um dos principais inimigos naturais de ácaros fitófagos, o que possibilita o seu uso em controle biológico. Entre as principais espécies destaca-se Phytoseiulus macropilis (Banks, 1904) (Acari: Phytoseiidae). O ácaro Tetranychus urticae Koch, 1836 (Acari: Tetranychidae) é considerado uma das espécies de ácaros-praga mais importantes, atacando mais de 1.100 espécies de plantas em 140 famílias de valor econômico. Este trabalho foi realizado com o objetivo de estudar, em laboratório, os parâmetros reprodutivos do ácaro predador P. macropilis, quando alimentado com T. urticae. Os estudos foram conduzidos em laboratório à temperatura de 25 ± 2 °C, 70 ± 10% de UR e 14 horas de fotofase e foram avaliados os aspectos biológicos e confeccionada a tabela de vida de fertilidade. Foi constatada uma longevidade de 27,5 dias para fêmeas adultas e 29,0 dias para machos. A população foi estimada em aumentar aproximadamente 27 vezes (Ro) no período médio de duração de geração (T) que foi de 17,7 dias. A população do ácaro cresceu 1,2vezes/dia (λ) e dobrou a cada 3,7 dias (TD).

ABSTRACT

RESUMO O Barbatimão (Stryphnodendron adstringens) planta medicinal encontrada no bioma Cerrado apresenta propriedades físico-químicas que lhe garante importantes atividades farmacológicas tais como: anti-inflamatória, analgésica e uma atividade protetora da mucosa gástrica. A casca do tronco é a principal matéria-prima usada para o desenvolvimento de produtos medicinais. Neste estudo, o objetivo foi investigar a influência da solução aquosa da casca do barbatimão no processo de formação de vasos sanguíneos na membrana corioalontoide de ovo embrionado de galinha. Foram utilizadas 30g da casca triturada em um litro de água. Este processo permitiu a obtenção da Solução Aquosa de Barbatimão - SAB em uma concentração de 30mg/mL. A atividade angiogênica da solução aquosa do barbatimão foi avaliada mediante realização de testes laboratoriais “in vivo”, utilizando como modelo experimental a membrana do ovo embrionado de galinha (MCA). Utilizou-se como controle indutor o Regederm®, o qual apresenta atividade angiogênica conhecida. Os resultados demonstraram que a SAB apresentou um percentual de vascularização na MCA de (50.4%) não tendo diferença (p>0,05) aos valores detectados no controle indutor (52,9%). Com os resultados obtidos, percebe-se que o barbatimão apresenta atividade angiogênica no modelo experimental utilizado.


ABSTRACT The Barbatimão (Stryphnodendron adstringens) medicinal plant found in the Cerrado biome has physicochemical properties which guarantee important pharmacological activities such as anti-inflammatic, analgesic and protective activities of gastric mucosa. The bark of the trunk is the main raw material used for the development of medicinal products. In this study, the objective was to investigate the influence of the aqueous solution of barbatimão bark in the formation of blood vessels in the membrane of embryonated chicken egg corioalontoid. 30g of shredded bark was used in one liter of water. This process enabled the obtention of aqueous Barbatimão - BSA at a concentration of 30mg / ml. The angiogenic activity of the aqueous solution of barbatimão was assessed by laboratory testing “in vivo”, using the chorioallantoic membrane of embryonated chicken egg (MCA) as an experimental model. TheRegederm® controlinductor was used, which exhibits known angiogenic activity. The results showed that the percentage of BSA showed a vascularization of the MCA (50.4%) there was no difference (p> 0.05) in the values detected in the control inductor (52.9%). With the obtained results, it is clear that barbatimão shows angiogenic activity under the experimental model used.

To assess the pro-angiogenic activity of Euphorbia tirucalli, commonly known as "avelós" plant, we performed a series of tests by applying an aqueous E. tirucalli latex solution (10 mg/mL) to the chorioallantoic membranes (CAMs) of 80 fertilized chicken eggs incubated in a temperature- and humidity-controlled automatic incubator. The results indicated that the aqueous latex solution increased vascular network formation compared to that with the negative control (p < 0.05) and the inhibitor control (p < 0.05). This suggests that under the experimental conditions tested, the aqueous latex solution induced an inflammatory response leading to neoangiogenesis.

Com o objetivo de analisar a atividade angiogênica apresentada pela Euphorbia tirucalli, popularmente conhecida comumente como “avelós”, foram realizados ensaios utilizando solução aquosa de látex na concentração de 10 mg/ml, aplicada em membrana corioalantóide (MCA) de 80 ovos férteis de galinha, incubado em estufa automática com temperatura e umidade controladas. Os resultados apontaram que a ação da solução aquosa provocou aumento da percentagem da rede vascular formada em relação aos controles negativo (p<0,05) e inibidor (p<0,05), indicando que nas condições deste experimento, foi responsável pela ativação da resposta inflamatória e crescimento de novos vasos sanguíneos.(AU)

The present study aimed to assess the cytotoxic, genotoxic, and antigenotoxic activities of sucupira oil (Pterodon emarginatus), which is commonly used as an anti-rheumatic, analgesic, antimicrobial, anticercariae, and anti-inflammatory. We used the mouse bone marrow micronucleus test as an experimental model. The experimental groups, which consisted of 5 animals, was administered sucupira oil (100 mg/kg body weight) intraperitoneally and evaluated 24 h after the treatment. The negative control group was treated with sterile distilled water, and the positive control group received an intraperitoneal dose of 4 mg/kg mitomycin C, a dose that corresponds to 80% of its median lethal dose. Cytotoxicity was determined by the polychromatic to normochromatic erythrocytes ratio (PCE/NCE). Sucupira oil had no significant effects (P > 0.05) on the frequency of micronucleated polychromatic erythrocytes as compared to the negative control group. However, the difference was significant (P < 0.005) as compared to the positive control group. The PCE/NCE (100 mg/kg oil and 4 mg/kg mitomycin) ratio did not differ between the experimental group and the positive control group, but it differed significantly when compared to the negative control group (P < 0.05). Thus, these findings suggested that the P. emarginatus oil showed no cytotoxic, mutagenic, or antimutagenic activities at a dose of 100 mg/kg.

Pest attacks such as that of the spider mite, Tetranychus urticae Koch, 1836 (Acari: Tetranychidae), is one of the problems faced by farmers, especially in the greenhouse, due to the difficulty of its control with the use of chemicals because of the development of fast resistance making it hard to control it. The objective of this work was to study the life history of the predatory mite N. californicus as a contribution to its mass laboratory rearing, having castor bean plant [Ricinus communis L. (Euphorbiaceae)] pollen as food, for its subsequent use as a natural enemy of T. urticae on a cultivation of greenhouse rosebushes. The studies were carried out in the laboratory, at 25 ± 2°C of temperature, 70 ± 10% RH and a 14 hour photophase. The biological aspects and the fertility life table were appraised. Longevity of 32.9 days was verified for adult females and 40.4 days for males. The intrinsic rate of increase (rm) was 0.2 and the mean generation time (T) was 17.2 days. The population doubled every 4.1 days. The results obtained were similar to those in which the predatory mite N. californicus fed on T. urticae.

O ácaro predador, Neoseiulus californicus (McGregor, 1954) (Acari: Phytoseiidae) é um dos principais inimigos naturais de ácaros tetraniquídeos em vários países, promovendo um eficiente controle em várias culturas alimentícias e ornamentais. O ataque de pragas como o ácaro-rajado, Tetranychus urticae Koch, 1836 (Acari: Tetranychidae), é um dos problemas enfrentados pelos produtores, principalmente em casa de vegetação, pela dificuldade de seu controle com o uso de agroquímicos devido ao rápido desenvolvimento de resistência o que dificulta seu controle. Foi objetivo deste trabalho estudar a história de vida do ácaro predador N. californicus como contribuição à sua criação massal em laboratório tendo como alimento pólen de mamoneira [Ricinus communis L. (Euphorbiaceae)] para posterior uso como inimigo natural de T. urticae em cultivo de roseiras (Rosa spp.) em casa de vegetação. Os estudos foram conduzidos em laboratório à temperatura de 25 ± 2°C, 70 ± 10% de UR e 14 horas de fotofase. Foram avaliados os aspectos biológicos e confeccionada a tabela de vida de fertilidade. Foi constatada uma longevidade de 32,9 dias para fêmeas adultas e 40,4 dias para machos. A estimativa da capacidade inata de crescimento da população (rm) foi 0,2 dias e a duração média de geração (T) foi 17,2 dias. A população dobrou a cada 4,1 dias. Os resultados obtidos foram semelhantes àqueles nos quais o predador N. californicus foi alimentado com T. urticae.(AU)

This study evaluated the predatory capacity of Euseius alatus (DeLeon) as a biological control agent of the pest mite Oligonychus ilicis (McGregor) on coffee leaves under laboratory conditions, using arenas containing 25 O. ilicis per coffee (Coffea arabica) leaf to one specimen of each stage of the predator mite. The functional response and oviposition rate of adult females of E. alatus were evaluated on coffee leaf arenas and offered from 1 to 125 immature stages of O. ilicis per arena. The number of preys killed and the number of eggs laid by the predator were evaluated every 24 h during 8 days. The preys consumed were daily replaced. Male and female adults of E. alatus were the most efficient in killing all developmental stages of O. ilicis. Larvae and nymphs of O. ilicis were the most consumed by all stages of the predatory mite. The functional response and oviposition rates of E. alatus increased as the prey density increased, with a positive and highly significant correlation. Regression analysis suggested a type II functional response, with a maximum predation of 22 O. ilicis/arena and a maximum oviposition rate of 1.7 eggs/day at a density of 70 O. ilicis/arena.

Synadenium umbellatum Pax, popularly known as "cola-nota", is a medicinal plant that grows in tropical regions. The latex of this plant is used against various diseases, such as diabetes mellitus, leprosy, tripanosomiasis, leukemia, and several malignant tumors. The mutagenic, antimutagenic, and cytotoxic effects of the latex of this plant were investigated by measuring the frequency of micronuclei in mice bone marrow cells. To evaluate mutagenicity, the animals were treated with four doses of latex (10, 30, 50, and 100 mg/kg body weight). To study the antimutagenic activity, the animals were simultaneously treated with latex and mitomycin C (4 mg/kg). The cytotoxicity was evaluated by polychromatic and normochromatic erythrocytes ratio. Our results showed a significant increase of frequency of micronucleated polychromatic erythrocytes (MNPCE) compared to the negative control group (p < 0.05). Concerning antimutagenicity, the doses of 10 and 30 mg/kg co-administered with mitomycin C showed significant decrease in MNPCE frequency compared to the positive control group (p < 0.05). However, no significant reduction in MNPCE frequency (p > 0.05) was detected at the doses of 50 and 100 mg/kg. Under our experimental conditions, the results obtained indicate strong mutagenic and cytotoxic activity of S. umbellatum latex except the dose of 10 mg/kg and moderate antimutagenic effect at lower doses.
