ABSTRACT

Phenylmethylsiloxanes, as modified products of dimethylsiloxanes, have been used in personal care products (PCPs) and household appliances, with indoor dust serving as one potential reservoir due to their particle-binding properties. This study measured six isomers of two phenylmethylsiloxanes (P3 and P4) in PCPs (99 %) intakes of phenylmethylsiloxanes for adults, while dust ingestion/adsorption (0.19 ng/d) may play important roles for toddlers/infants with little usage of phenylmethylsiloxanes-containing PCPs. Additionally, total daily intakes of PhMeSi(OH)2 (0.30-0.84 ng/d) via ingestion and dermal absorption of dust were higher than P3 (0.06-0.31 ng/d) and P4 (0.02-0.09 ng/d), suggesting exposure risk of degradation product of phenylmethylsiloxanes deserving attention.

ABSTRACT

Osteosarcoma (OS) is the most common malignant tumor of the bone tissue with the lowest survival rate among all pediatric cancers. OS cells grow vigorously under malnutrition; however, the mechanism by which they adapt to metabolic stress via metabolic reprogramming remains undefined. Here, we demonstrated that USP33, a member of the DUBs family, was significantly upregulated in the tissues of patients with OS compared to normal tissues. Moreover, high USP33 expression was significantly associated with poor survival. Functional assays suggested that USP33 promoted OS cell growth through the induction of aerobic glycolysis. Additionally, we confirmed that 6-phosphofructo-2-kinase/fructose-2, 6-biphosphatase 3 (PFKFB3) was critical for USP33-induced proliferation and aerobic glycolysis in OS cells, and the protein expression levels of PFKFB3 and USP33 were positively correlated in the OS tissues. Mechanistically, USP33 stabilized the expression of PFKFB3 by suppressing the ubiquitin mediated PFKFB3 degradation. Collectively, these findings reveal a mechanism by which OS cells survive in a dystrophic tumor microenvironment, with the USP33-PFKFB3 axis as a critical driver of aerobic glycolysis and OS proliferation. Furthermore, these findings reveal novel insights into the adaptation of cancer cells to metabolic stress in OS.

ABSTRACT

Background: Children with benign childhood epilepsy with centro-temporal spikes (BECT) have spikes, sharps, and composite waves on their electroencephalogram (EEG). It is necessary to detect spikes to diagnose BECT clinically. The template matching method can identify spikes effectively. However, due to the individual specificity, finding representative templates to detect spikes in actual applications is often challenging. Purpose: This paper proposes a spike detection method using functional brain networks based on phase locking value (FBN-PLV) and deep learning. Methods: To obtain high detection effect, this method uses a specific template matching method and the 'peak-to-peak' phenomenon of montages to obtain a set of candidate spikes. With the set of candidate spikes, functional brain networks (FBN) are constructed based on phase locking value (PLV) to extract the features of the network structure during spike discharge with phase synchronization. Finally, the time domain features of the candidate spikes and the structural features of the FBN-PLV are input into the artificial neural network (ANN) to identify the spikes. Results: Based on FBN-PLV and ANN, the EEG data sets of four BECT cases from the Children's Hospital, Zhejiang University School of Medicine are tested with the AC of 97.6%, SE of 98.3%, and SP 96.8%.
