ABSTRACT

Alzheimer's disease (AD) presents a significant challenge due to its prevalence and lack of cure, driving the quest for effective treatments. Anshen Bunao Syrup, a traditional Chinese medicine known for its neuroprotective properties, shows promise in addressing this need. However, understanding its precise mechanisms in AD remains elusive. This study aimed to investigate Anshen Bunao Syrup's therapeutic potential in AD treatment using a scopolamine-induced AD rat model. Assessments included novel-object recognition and Morris water maze tasks to evaluate spatial learning and memory, alongside Nissl staining and ELISA analyses for neuronal damage and biomarker levels. Results demonstrated that Anshen Bunao Syrup effectively mitigated cognitive dysfunction by inhibiting amyloid-ß and phosphorylation Tau aggregation, thereby reducing neuronal damage. Metabolomics profiling of rats cortex revealed alterations in key metabolites implicated in tryptophan and fatty acid metabolism pathways, suggesting a role in the therapeutic effects of Anshen Bunao Syrup. Additionally, ELISA and correlation analyses indicated attenuation of oxidative stress and immune response through metabolic remodeling. In conclusion, this study provides compelling evidence for the neuroprotective effects of Anshen Bunao Syrup in AD models, shedding light on its potential as a therapeutic agent for AD prevention and treatment.

ABSTRACT

Sucrose isomerase (SIase) catalyzes the hydrolysis and isomerization of sucrose into isomaltulose, a functional sugar extensively used in the food industry. However, the lack of safe and efficient heterologous expression systems for SIase has constrained its production and application. In this study, an engineered Bacillus subtilis strain for antibiotic-free SIase production was developed via a food-grade expression system. First, the B. subtilis strain TEA was modified through the CRISPR/Cas9 system, resulting in a mutant strain TEA4, which exhibited enhanced capabilities for recombinant protein expression. For efficient and safe production of SIase, different constitutive and inducible promoters were evaluated. The maltose-inducible promoter Poglv was found to have an extracellular SIase activity of 21.7 U mL-1 in engineered strain TEA4. Subsequent optimization of the culture medium further increased SIase activity to 26.4 U mL-1 during shake flask cultivation. Eventually, using the crude enzyme solution of the engineered strain in biotransformation reactions resulted in a high yield of isomaltulose under high concentrations sucrose, achieving a maximum yield of 83.1%. These findings demonstrated an engineered B. subtilis strain for antibiotic-free SIase production, paving the way for its scale-up industrial production and application.

ABSTRACT

Fusarium crown rot (FCR), primarily caused by Fusarium pseudograminearum, has emerged as a new threat to wheat production and quality in North China. Genetic enhancement of wheat resistance to FCR remains the most effective approach for disease control. In this study, we phenotyped 435 Chinese wheat cultivars through FCR inoculation at the seedling stage in a greenhouse. Our findings revealed that only approximately 10.8% of the wheat germplasms displayed moderate or high resistance to FCR. A genome-wide association study (GWAS) using high-density 660K SNP led to the discovery of a novel quantitative trait locus on the long arm of chromosome 3B, designated as Qfcr.hebau-3BL. A total of 12 significantly associated SNPs were closely clustered within a 1.05 Mb physical interval. SNP-based molecular markers were developed to facilitate the practical application of Qfcr.hebau-3BL. Among the five candidate FCR resistance genes within the Qfcr.hebau-3BL, we focused on TraesCS3B02G307700, which encodes a protein kinase, due to its expression pattern. Functional validation revealed two transcripts, TaSTK1.1 and TaSTK1.2, with opposing roles in plant resistance to fungal disease. These findings provide insights into the genetic basis of FCR resistance in wheat and offer valuable resources for breeding resistant varieties.

ABSTRACT

Systemic acquired resistance (SAR) is an inducible disease resistance phenomenon in plant species, providing plants with broad-spectrum resistance to secondary pathogen infections beyond the initial infection site. In Arabidopsis, SAR can be triggered by direct pathogen infection or treatment with the phytohormone salicylic acid (SA), as well as its analogues 2,6-dichloroisonicotinic acid (INA) and benzothiadiazole (BTH). The SA receptor non-expressor of pathogenesis-related protein gene 1 (NPR1) protein serves as a key regulator in controlling SAR signaling transduction. Similarly, in common wheat (Triticum aestivum), pathogen infection or treatment with the SA analogue BTH can induce broad-spectrum resistance to powdery mildew, leaf rust, Fusarium head blight, and other diseases. However, unlike SAR in the model plant Arabidopsis or rice, SAR-like responses in wheat exhibit unique features and regulatory pathways. The acquired resistance (AR) induced by the model pathogen Pseudomonas syringae pv. tomato strain DC3000 is regulated by NPR1, but its effects are limited to the adjacent region of the same leaf and not systemic. On the other hand, the systemic immunity (SI) triggered by Xanthomonas translucens pv. cerealis (Xtc) or Pseudomonas syringae pv. japonica (Psj) is not controlled by NPR1 or SA, but rather closely associated with jasmonate (JA), abscisic acid (ABA), and several transcription factors. Furthermore, the BTH-induced resistance (BIR) partially depends on NPR1 activation, leading to a broader and stronger plant defense response. This paper provides a systematic review of the research progress on SAR in wheat, emphasizes the key regulatory role of NPR1 in wheat SAR, and summarizes the potential of pathogenesis-related protein (PR) genes in genetically modifying wheat to enhance broad-spectrum disease resistance. This review lays an important foundation for further analyzing the molecular mechanism of SAR and genetically improving broad-spectrum disease resistance in wheat.

ABSTRACT

Maltotetraose amylase, which catalyzes the hydrolysis of amylaceous polysaccharides into maltooligosaccharides with maltotetraose as the main product, is extensively used in the food industry. However, the lack of efficient expression system for maltotetraose amylase has hampered its production and application. In this study, high-level production of a maltotetraose amylase mutant (referred to as Pp-Mta∆CBM) from Pseudomonas saccharophila was achieved in Pichia pastoris X-33. First, the gene of maltotetraose amylase with the carbohydrate-binding module (CBM) removed was codon-optimized and cloned into the pPICZαA vector, followed by transformation into P. pastoris X-33 for expression. Using the promoter PAOX1 and signal peptide α-factor, high-level production of Pp-Mta∆CBM with minimal extracellular impurity proteins was achieved, resulting in an extracellular activity of 367.9 U/mL after 7 days of cultivation in shake flasks. Next, the expressed Pp-Mta∆CBM was purified and characterized. This recombinant enzyme was glycosylated and has maximum activity at 55 ℃ and pH 7.0. Its Km for soluble starch was 4.1 g/L, and its kcat was 3237.6 s-1. Finally, the Pp-Mta∆CBM was found to produce a maximum maltotetraose yield of 57.1% in the presence of 200 g/L of substrate. The findings presented in this study demonstrate the efficient production of Pp-Mta∆CBM in P. pastoris, providing a new expression system for maltotetraose amylase and laying the foundation for its scale-up production and industrial application.

ABSTRACT

Household dust contains a wide variety of semi-volatile organic compounds (SVOCs) that may pose health risks. We developed a method integrating non-targeted analysis (NTA) and targeted analysis (TA) to identify SVOCs in indoor dust. Based on a combined use of gas and liquid chromatography with high-resolution mass spectrometry, an automated, time-efficient NTA workflow was developed, and high accuracy was observed. A total of 128 compounds were identified at confidence level 1 or 2 in NIST standard reference material dust (SRM 2585). Among them, 113 compounds had not been reported previously, and this suggested the value of NTA in characterizing contaminants in dust. Additionally, TA was done to avoid the loss of trace compounds. By integrating data obtained from the NTA and TA approaches, SVOCs in SRM 2585 were prioritized based on exposure and chemical toxicity. Six of the top 20 compounds have never been reported in SRM 2585, including melamine, dinonyl phthalate, oxybenzone, diheptyl phthalate, drometrizole, and 2-phenylphenol. Additionally, significant influences of analytical instruments and sample preparation on NTA results were observed. Overall, the developed method provided a powerful tool for identifying SVOCs in indoor dust, which is necessary to obtain a more complete understanding of chemical exposures and risks in indoor environments.

ABSTRACT

Climate-change-induced extreme weather events increase heat-related mortality and health risks for urbanites, which may also affect urbanites' expressed happiness (EH) and well-being. However, the links among EH, climate, and socioeconomic factors remain unclear. Here we collected ∼6 million geotagged tweets from 44 Chinese prefecture-level cities based on Sina Weibo and performed a quadratic regression model to explore the relationships between summer heat and EH. A three-stage analysis was developed to examine spatiotemporal heterogeneity and identify factors contributing to disparities in urbanites' EH. Results show that all cities exhibited a similar hump-shaped relationship, with an overall optimal temperature (OT) of 22.8 °C. The estimated OT varied geographically, with 25.3, 23.8, and 20.0 °C from north to south. Moreover, a 1 standard deviation increase in heatwave intensity was associated with a 0.813 (95% CI: 0.177, 1.449) standard deviation decrease in EH. Notably, within the geographic scope of this study, it was observed that urbanites in northern China and economically underdeveloped cities faced significantly lower heat risks during the summer heat. This research provides insight for future studies and practical applications concerning extreme weather events, urbanites' mental health, and sustainable urban development goal.

ABSTRACT

Introduction: In recent years, research has used psycholinguistic features in public discourse, networking behaviors on social media and profile information to train models for depression detection. However, the most widely adopted approach for the extraction of psycholinguistic features is to use the Linguistic Inquiry Word Count (LIWC) dictionary and various affective lexicons. Other features related to cultural factors and suicide risk have not been explored. Moreover, the use of social networking behavioral features and profile features would limit the generalizability of the model. Therefore, our study aimed at building a prediction model of depression for text-only social media data through a wider range of possible linguistic features related to depression, and illuminate the relationship between linguistic expression and depression. Methods: We collected 789 users' depression scores as well as their past posts on Weibo, and extracted a total of 117 lexical features via Simplified Chinese Linguistic Inquiry Word Count, Chinese Suicide Dictionary, Chinese Version of Moral Foundations Dictionary, Chinese Version of Moral Motivation Dictionary, and Chinese Individualism/Collectivism Dictionary. Results: Results showed that all the dictionaries contributed to the prediction. The best performing model occurred with linear regression, with the Pearson correlation coefficient between predicted values and self-reported values was 0.33, the R-squared was 0.10, and the split-half reliability was 0.75. Discussion: This study did not only develop a predictive model applicable to text-only social media data, but also demonstrated the importance taking cultural psychological factors and suicide related expressions into consideration in the calculation of word frequency. Our research provided a more comprehensive understanding of how lexicons related to cultural psychology and suicide risk were associated with depression, and could contribute to the recognition of depression.

ABSTRACT

Systemic acquired resistance (SAR) is a broad-spectrum plant defense phenomena controlled by the salicylic acid receptor NPR1. Key regulators of the SAR signaling pathway showed great potentials to improve crop resistance to various diseases. In our previous investigation, a barley transcription factor gene HvWRKY6 was identified as downstream of NPR1 during SAR. However, the broad-spectrum resistance features and molecular mechanisms of HvWRKY6 remain to be explored. In this study, a transgenic wheat line exogenously expressing HvWRKY6 showed improved resistance to leaf rust, Fusarium crown rot (FCR), and sharp eyespot. The model pathogen Pseudomonas syringae pv. tomato DC3000 was employed to induce the SAR response in wheat plants' leaf region adjacent to the infiltration area. Transcriptome sequencing revealed activation of broad-spectrum defense responses by expressing HvWRKY6 in a pathogen-independent manner. Based on the differentially expressed genes in plant hormone signal transduction, we speculated that the enhanced resistance in HvWRKY6-OE wheat transgenic line was associated with activation of the salicylic acid pathway and suppression of the abscisic acid and jasmonic acid pathways. These findings suggest that the transgenic line HvWRKY6-OE might be applied for the genetic improvement of wheat to several fungal diseases; the underlying resistance mechanism was clarified.

ABSTRACT

High levels of trust between employees and leaders moderate the relationship between organizational management practices. A collaborative environment encourages employees to have more Psychological Empowerment, which in turn leads to better performance. Based on Intrinsic Work Motivation and Self-Evaluation, this paper uses Perceived Leader Trust as an independent variable, Employee Work Performance as a dependent variable, and introduces Psychological Empowerment to explore the internal mechanism of perceived trust. This paper proposes a total of 28 hypotheses, and 25 hypotheses have been verified. The specific research conclusions are as follows: (1) Perceived Leader Trust has a positive impact on Employee Work Performance. (2) Perceived Leader Trust positively affects employees' Psychological Empowerment. Perceived Leader Dependence has a significant impact on all dimensions of Psychological Empowerment, but the relationship between Perceived Information Disclosure and Work Meaning is not significant. (3) Psychological Empowerment is positively correlated with Employee Work Performance, in which the four dimensions of Psychological Empowerment are significantly related to Employee Task Performance, while Work Meaning and Autonomy are not significantly related to Employee Relationship Performance. (4) Psychological Empowerment, as the overall perception of employees, plays a partial mediating role between Perceived Leader Trust and Employee Work Performance. This paper verifies the role of Psychological Empowerment between Perceived Leader Trust and Employee Work Performance, and explores the internal mechanism of Perceived Leader Trust from the perspective of employees' Intrinsic Work Motivation, which promotes the development of organizational management practices.

ABSTRACT

The voluntary frontier settlement hypothesis holds that frontier movements can promote the formation of individualism in the frontier area. The Chuangguandong Movement is one of China's voluntary frontier movements that potentially had a positive impact on the formation of high individualism in the northeastern provinces. Previous studies used independent/interdependent measures of self-construal scale, symbolic self-inflation, nepotism tasks, and percentage of most common names, to examine the differences in the independence between Heilongjiang and Shandong residents, which may be related to the Chuangguandong Movement. However, these studies were limited by certain factors such as sample size and objectivity of materials acquisition. In this study, we obtained Sina Weibo big data for period 2010-2020 to overcome the limitation of previous work. Using text feature extraction and keyword word frequency calculation methods based on the individualism/collectivism dictionary, we found that the level of individualism in Northeast China was higher than that in Shandong Province, which was consistent with previous research. Through the discussion of the four representative theoretical frameworks of individualism, the voluntary frontier settlement theory was considered as a potential explanation for the high degree of individualism in Northeast China.

Cross-cultural studies from a global perspective contend that China and India are both collectivistic cultures. However, it remains unclear whether and why China and India differ in their collectivism. This study examines whether the cultural heritage concerning argumentation explains why Chinese people are more collectivistic than Indians. Convenient samples were taken from online surveys (N China = 398, N India = 418), and 186 participants from the United States were included in the contrast group. In multiple methods conducted here, the Chinese respondents scored higher in holistic thought, compatriotism, nepotism, familism, and self-interdependence than the Indian respondents, while scoring lower in assertiveness and argumentativeness. Although China and India were more collectivistic than the United States, these findings support the hypothesis that Chinese people are more collectivistic than Indians. The study extended our knowledge of individualism-collectivism beyond east-west comparison.

The global COVID-19 pandemic may significantly affect the experiences of death and bereavement. This study aimed to learn from recent outbreaks of infectious diseases and further understand their impacts on bereavement. We obtained psychological status scores for 32 individuals bereaved due to COVID-19 and 127 individuals bereaved due to non-COVID-19 causes using the online ecological recognition (OER) approach. Next, a sentiment analysis and independent sample t-test were performed to examine the differences between these two groups. The results indicated that the individuals bereaved due to COVID-19 were more insecure and more preoccupied with the grief of the moment than those bereaved due to non-COVID-19 reasons, while the latter group had higher depression scores than the former group. This study can guide policy-makers and clinical practitioners to provide more targeted and sustainable post-bereavement support for both bereaved groups during the COVID-19 period.

In the world of social media, people are free to choose names based on their preferences, which may potentially reflect certain levels of uniqueness. In this study, we have attempted to explore the possibility of applying the ecological theory of individualism/collectivism in the context of social media. We, thus, examined provincial variations in the uniqueness of nicknames among more than 13 million Sina Weibo users. Initially, the nickname uniqueness indicator was set at the provincial level. It was found that the uniqueness of nicknames was the highest in provinces with temperate climates, for example Guangdong, and the lowest in provinces with demanding climate, such as Ningxia. Regression analysis results partially supported that inhabitants in provinces with temperate climate were more likely to use unique nicknames on social media compared to those from harsh climate. This finding is significant in terms of ecology.

The pathogen-prevalence hypothesis postulates that collectivism would be strengthened in the long term in tandem with recurrent attacks of infectious diseases. However, it is unclear whether a one-time pathogen epidemic would elevate collectivism. The outbreak of COVID-19 and the widespread prevalence of online social networks have provided researchers an opportunity to explore this issue. This study sampled and analyzed the posts of 126,165 active users on Weibo, a leading Chinese online social network. It used independent-sample t-tests to examine whether COVID-19 had an impact on Chinese collectivistic value-related behaviors by comparing the usage frequency of personal pronouns, group-related words, and relationship-related words before and after the outbreak. Overall, most collectivist words exhibited a significant upward trend after the outbreak. In turn, this tendency pointed to a rising sense of collectivism (versus individualism). Hence, this study confirmed the pathogen-prevalence hypothesis in real settings, finding that an outbreak of an infectious disease such as COVID-19 could exert an impact on collectivism and may deliver a theoretical basis for psychological protection against the threat of COVID-19. However, further evaluation is required to ascertain whether this trend is universal or culture-specific.

In the current paper, two different studies were designed to investigate body image dissatisfaction and perception of the attractive female body in Chinese and Croatian women and men using the correlational and experimental study research approach. Study 1 comprised 266 Chinese (160 women; 106 men) and 261 Croatian participants (161 women; 100 men). Women from both countries were asked to complete the measures of the Contour Rating Scale, SATAQ - 4, Body Area Scale and demographic data, while men were asked to complete the measures of the Contour Rating Scale and demographic data via online surveys. The obtained results indicated that thin internalization predicted body image dissatisfaction in both samples. Besides, while both samples scored relatively high on thin-ideal internalization, Chinese women, who were also in general less satisfied with their body image, had higher scores. Chinese women also scored higher on musculature internalization and felt more pressure from family, friends, and media to conform to standard beauty norms compared to Croatian women. The samples also differed in their perceptions of preferable body size, where Chinese women had a more negative perception of their actual body size. The obtained results furthermore revealed that Chinese men preferred thinner women compared to Croatian men. Moreover, both samples misjudged what their female counterparts found attractive, thinking that women wanted to have even thinner bodies than they actually reported. Similarly, women from both cultures revealed a preference for thinner figures than the ones selected as the most attractive by the opposite sex. Finally, in Study 2, experimental manipulation of thin-ideal was used to examine whether thin priming affected body image dissatisfaction. The obtained results revealed that when primed with thin-ideal women from both countries felt lower satisfaction with their body size. The observed effect was stronger for Chinese participants. Limitations of the current study are discussed in the conclusion.

In this study, we examined the Chinese Han and Mongolian, two ethnic groups that belong to the same national and geographic regions but vary in their degrees of social interdependence and independence. We assumed that the Mongolian, who have traditionally been known as a herding community, exhibit a greater independent social orientation compared to the Han Chinese, who have traditionally been known as an agrarian community. Through three different studies we used the explicit measurement of self-construal, implicit cultural task of self - inflation and the practice of name-giving (i.e., baby names as a cultural product) to test our hypothesis. The obtained results revealed that compared to Han Chinese, Mongolian scored higher on independent - self subscale, had greater levels of self-symbolic inflation and were less likely to give common names to their babies. These findings suggest that Mongolians are more independent than Han Chinese. In view of that, the present study contributes to a better understanding of the complexity of Chinese culture in terms of interdependence and independence, and provides further support for historical sustenance theory.

Voluntary frontier settlement leads to independent agency. Since this type of research has not yet been implemented in ongoing voluntary settlement frontiers, we conducted several cultural tasks to investigate Shenzhen, known as China's ongoing "South Frontier," which is composed mostly of people that have emigrated from other Chinese provinces within the past 30 years. We hypothesized that residents of Shenzhen are more independent than those in other regions of Mainland China. As predicted, residents of Shenzhen scored higher than China inland residents in self-reported independent beliefs and scored lower in nepotism. The results indicate that, even in a short-term ongoing frontier, voluntary settlement is associated with independent agency.

ABSTRACT

Unmerited authorship is a practice common to many countries around the world, but are there systematic cultural differences in the practice? We tested whether scientists from collectivistic countries are more likely to add unmerited coauthors than scientists from individualistic countries. We analyzed archival data from top scientific journals (Study 1) and found that national collectivism predicted the number of authors, which might suggest more unmerited authors. Next, we found that collectivistic scientists were more likely to add unmerited coauthors than individualistic scientists, both between cultures (Studies 2-3) and within cultures (Study 4). Finally, we found that priming people with collectivistic self-construal primes made them more likely to endorse questionable authorship attitudes (Study 5). These findings show that culture collectivism is related to unmerited authorship.

Understanding urbanization and evaluating its impact are vital for formulating global sustainable development. The results obtained from evaluating the impact of urbanization, however, depend on the kind of measurement used. With the goal of increasing our understanding of the impact of urbanization, we developed direct and indirect subjective indicators to measure how people assess their living situation. The survey revealed that the projected endorsements and perceived social ambiance of people toward living in different types of settlements did not improve along with the urbanization level in China. The assessment scores from the city dwellers were not significantly different from those from the country areas and, more surprisingly, both were significantly higher than the assessment scores of the town dwellers, which we had expected to fall between the assessment scores of the country and city dwellers. Instead their scores were the lowest. We dubbed this V-shaped relationship the "town dislocation effect." When searching for a potential explanation for this effect, we found additional town dislocation effects in social support, loss aversion, and receptivity toward genetically modified food. Further analysis showed that only social support mediated the relationship between the three tiers of settlements (cities, country areas, and towns) and the subjective indicator. The projected endorsements yielded significant subjective assessments that could enhance our understanding of Chinese urbanization. Towns posed specific problems that require special attention.
