ABSTRACT

ABSTRACT Purpose: This study aimed to determine whether early-stage intraocular pressure can be modulated using a thermal face mask. Methods: In this prospective clinical study, healthy participants were randomized on a 1:1:1 allocation ratio to three mask groups: hypothermic (G1), normothermic (G2), and hyperthermic (G3). After randomization, 108 eyes from 108 participants were submitted to clinical evaluations, including measurement of initial intraocular pressure (T1). The thermal mask was then applied for 10 minutes, followed by a second evaluation of intraocular pressure (T2) and assessment of any side effects. Results: The hypothermic group (G1) showed a significant reduction in mean intraocular pressure between T1 (16.97 ± 2.59 mmHg) and T2 (14.97 ± 2.44 mmHg) (p<0.001). G2 showed no significant pressure difference between T1 (16.50 ± 2.55 mmHg) and T2 (17.00 ± 2.29 mmHg) (p=0.054). G3 showed a significant increase in pressure from T1 (16.53 ± 2.69 mmHg) to T2 (18.58 ± 2.95 mmHg) (p<0.001). At T1, there was no difference between the three study groups (p=0.823), but at T2, the mean values of G3 were significantly higher than those of G1 and G2 (p<0.00). Conclusion: Temperature was shown to significantly modify intraocular pressure. Thermal masks allow the application of temperature in a controlled, reproducible manner. Further studies are needed to assess the duration of these effects and whether they are reproducible in patients with pathologies that affect intraocular pressure.

ABSTRACT

Thyroid tissue in teleosts is located mainly in the pharyngeal region, usually reaching other adjacent anatomical locations. Herein, a nodular lesion located in the left operculum of a Senegal seabream (Diplodus bellottii) was surgically excised and sent for microscopical evaluation. Microscopically, the lesion presented irregular borders and consisted in columnar epithelial cells arranged in a tubulopapillary pattern, surrounding a central lumen filled with acellular, acidophilic and homogeneous, material ('colloid'). To determine the lesion's histogenesis, immunohistochemistry was performed employing antibodies for AE1/AE3, CK7, thyroglobulin and vimentin. The neoplastic cells presented low mitotic index and positive immunolabelling for CK7 and thyroglobulin. Therefore, a diagnosis of ectopic thyroid adenoma was made. Herein, the successful employment of antibodies classically used in mammals for accurate diagnosis of thyroid disorders is described. Proliferation of thyroid tissue in fish may reflect environmental and physiological imbalances, making the study and correct diagnosis of these tumours in this species important.

ABSTRACT

Polydimethylsiloxane (PDMS) is an elastomer that has received primary attention from researchers due to its excellent physical, chemical, and thermal properties, together with biocompatibility and high flexibility properties. Another material that has been receiving attention is beeswax because it is a natural raw material, extremely ductile, and biodegradable, with peculiar hydrophobic properties. These materials are applied in hydrophobic coatings, clear films for foods, and films with controllable transparency. However, there is no study with a wide range of mechanical, optical, and wettability tests, and with various proportions of beeswax reported to date. Thus, we report an experimental study of these properties of pure PDMS with the addition of beeswax and manufactured in a multifunctional vacuum chamber. In this study, we report in a tensile test a 37% increase in deformation of a sample containing 1% beeswax (BW1%) when compared to pure PDMS (BW0%). The Shore A hardness test revealed a 27% increase in the BW8% sample compared to BW0%. In the optical test, the samples were subjected to a temperature of 80 °C and the BW1% sample increased 30% in transmittance when compared to room temperature making it as transparent as BW0% in the visible region. The thermogravimetric analysis showed thermal stability of the BW8% composite up to a temperature of 200 °C. The dynamic mechanical analysis test revealed a 100% increase in the storage modulus of the BW8% composite. Finally, in the wettability test, the composite BW8% presented a contact angle with water of 145°. As a result of this wide range of tests, it is possible to increase the hydrophobic properties of PDMS with beeswax and the composite has great potential for application in smart devices, food and medicines packaging films, and films with controllable transparency, water-repellent surfaces, and anti-corrosive coatings.

ABSTRACT

Chemical desiccation is widely used in agriculture to anticipate harvest and mitigate the effects of adverse environmental conditions. It is applied to both grains and seeds. Although this practice is widely used, there are still significant gaps in understanding the effects of different herbicide application times on seed quality and plant physiological responses. The objective of this study was to evaluate the effects of different herbicide application times on cowpea, focusing on seed quality, physiological responses, and biochemical composition, including chlorophylls, carotenoids, sugars, and proline, under nocturnal desiccation. In the first experiment, eight herbicides and two mixtures were applied at night: diquat, flumioxazin, diquat + flumioxazin, glufosinate ammonium, saflufenacil, carfentrazone, diquat + carfentrazone, atrazine, and glyphosate. All of the tested herbicides caused a reduction in normal seedling formation, with the diquat + carfentrazone combination resulting in 100% abnormal seedlings. A significant decrease in chlorophyll levels (chlorophyll a: 63.5%, chlorophyll b: 50.2%) was observed using diquat, which indicates damage to photosynthetic processes, while the carotenoid content increased. Total soluble sugars and proline were also negatively impacted, reflecting physiological stress and metabolic changes in seedlings. In the second experiment, three application times were tested with diquat, diquat + flumioxazin, and diquat + carfentrazone. Nocturnal application showed the most significant reduction in chlorophyll levels and increased carotenoid levels. Application at noon and late afternoon also significantly changed the soluble sugar and proline levels. These results indicate that the herbicide application time directly influences the seeds' physiological quality.

We adapted the percentage intensity approach to monitor accelerations and decelerations allowing players' individualisation. Forty-two players were monitored during four microcycles via global navigation satellite system devices. Raw velocity and time data were collected to calculate acceleration and deceleration magnitudes according to specific starting speed intervals, and the efforts intensities were established as very low (<25% of the maximal effort), low (25-50%), moderate (50-75%) and high (>75%). Linear regressions and Pearson correlation (r) analysed the relationship between maximal efforts and starting speeds; additionally, mean paired differences compared efforts magnitudes between subsequent starting speed intervals. Most very low intensity accelerations (86%) and decelerations (79%) started from <5 km.h-1. Correlation between maximal efforts and starting speeds were r = -0.97 (p < .001) for acceleration, and r = -0.94 (p < .01) for deceleration. Maximal acceleration decreased as starting speed increases (very large effect sizes), but deceleration is less starting speed dependent (unclear to large effect sizes). This adaptation allows practitioners to individualise accelerations and decelerations classification during real-life scenarios, leading to a more precise training prescription. The very low intensity interval could be excluded to consider only relevant efforts. Maximal acceleration should be collected for each starting speed interval because accelerations are starting speed dependents.

BACKGROUND: Monitoring training load and competition load is crucial for evaluating and improving athlete performance. This study proposes an applied approach to characterize and classify the training task specificity in relation to competition in a top-level rink hockey team, considering external and internal load from training tasks and competition. HYPOTHESIS: Training tasks and game demands have significant dose-response differences, and exercises can be classified successfully based on their physiological and biomechanical demands. STUDY DESIGN: Cross-sectional study. LEVEL OF EVIDENCE: Level 5. METHODS: Ten elite-level male rink hockey players participated in this study. Players were monitored on 6 different task categories during 8 training sessions and 2 official games. A linear mixed model with random intercepts was used to compare training tasks and competition load, accounting for individual repeated measures. A 2-step cluster analysis was performed to classify the training tasks and games based on physiological and biomechanical load, employing log-likelihood as the distance measure and Schwartz's Bayesian criterion. RESULTS: Average heartrate , maximum heartrate, and high-speed skating (18.1-30 km/h) were the best physiological load predictors, while the most effective biomechanical load predictors were impacts [8-10] g(n), decelerations [-10 to -3]m/s²(n), and accelerations [3-10]m/s²(n). Different physiological and biomechanical responses were verified between training tasks and match demands. A 4-quadrant efforts assessment for each task category revealed that training tasks used by the team in the analysis presented lower biomechanical and physiological load demands than competition. CONCLUSION: Training tasks failed to adequately replicate the specific demands of competition, especially regarding high mechanical stress, such as the absence of high-intensity impacts and decelerations. CLINICAL RELEVANCE: This method of classification of training tasks may allow coaches to understand further the specificity and contribution of each task to competition demands, consequently improving the capacity of load management and the preparedness and readiness of players for competition.

BACKGROUND: Usher's syndrome type II (USH2) is a rare genetic disorder encompassing hearing loss, vision impairment, and apparent intact vestibular function. Recent research suggests a potential involvement of the otolith vestibular receptors in USH2. AIMS/OBJECTIVES: Evaluate otolith dynamic function in USH2. MATERIAL AND METHODS: Twenty-two USH2 (median age 53.9 ± 2.99) and age-matched controls underwent a complete battery vestibular testing including air conducted cervical and ocular vestibular evoked myogenic potentials (c-VEMPs and o-VEMPs). Vestibular function tests were correlated with Activities Balance Scale (ABC) and Dizziness Handicap Inventory (DHI) scores. RESULTS: Fourteen USH2 reported previous vertigo (vs none control). Among 88 ears, c-VEMPs were absent in 15 USH2 cases and 4 controls (p = 0.034), while o-VEMPs were absent in 22 USH2 cases and 12 controls (p = 0.129). There were significant differences between USH2 vs controls in right ear o-VEMP N1 latencies (median 11.60/10.40, p < 0.010), N1-P1 amplitudes (median 5.15/10.10, p < 0.003) and in o-VEMP N1-P1 asymmetry ratio (median 24.78/40.50, p < 0.014). USH2 showed a strong correlation between o-VEMP amplitude and DHI score (p = 0.003, ρ = 0.769). No association was found between vertigo and VEMPs subgroups. CONCLUSIONS AND SIGNIFICANCE: Our findings suggest the presence of otolith dysfunction in USH2, which is independent from subjectively reported dizziness. Incorporating vestibular testing into USH2 evaluation and monitoring could enhance characterization of this multisensory disease.

OBJECTIVE: This study analyzed, using an umbrella review, existing systematic reviews on medications to prevent and control postoperative endodontic pain to guide professionals in choosing the most effective drug. MATERIALS AND METHODS: An electronic search in the PubMed (MEDLINE), LILACS, SciELO, EMBASE, Scopus, Web of Science, Cochrane Reviews, and Data Archiving and Networked Services (DANS) databases retrieved 17 systematic reviews. The study included only systematic reviews of clinical trials with or without meta-analyses evaluating effectiveness of medications in reducing pain after non-surgical endodontic treatment. RESULTS: The evidence showed that steroidal and non-steroidal anti-inflammatory drugs and opioids effectively controlled pain within six to 24 h. CONCLUSIONS: Dexamethasone, prednisolone, paracetamol, and mainly ibuprofen provided higher postoperative pain relief. The quality of evidence of the reviews ranged from very low to high, and the risk of bias from low to high, suggesting the need for well-designed clinical trials to provide confirmatory evidence. CLINICAL RELEVANCE: This review emphasizes the efficacy of developing protocols for pain control after endodontic therapy.

ABSTRACT

Implantable active dense CMOS neural probes unlock the possibility of spatiotemporally resolving the activity of hundreds of single neurons in multiple brain circuits to investigate brain dynamics. Mapping neural dynamics in brain circuits with anatomical structures spanning several millimeters, however, remains challenging. Here, we demonstrate the first CMOS neural probe for mapping intracortical neural dynamics (both LFPs and spikes) in awake, behaving mice from an area >4 mm2. By taking advantage of the modularity of our SiNAPS technology, we realized an eight shanks probe with 1024 electrode channels arranged on each shank in regular arrays with an electrode pitch <30 µm. Low-noise recordings from all electrodes at 20 kHz/channel demonstrate a field of view spanning the 2D lattice of the entire mice hippocampal circuit, together with cortical and thalamic regions. This arrangement allows combining large population unit recording across distributed networks with precise intra- and interlaminar/nuclear mapping of the oscillatory dynamics.

ABSTRACT

This case series reports eight eyes with keratoconus treated with laser implantation of one or two segments of progressive thickness corneal intrastromal ring (PT-ICRS). In this case series, it was evident that the insertion of PT-ICRS induces more pronounced corneal flattening at the thickest point, causing a reduction in distortion (coma) and lower astigmatism, resulting in a remarkable improvement in vision. Compared to the implementation of traditional intrastromal rings, the PT-ICRS variant showed superior results despite the small sample size. However, the same degree of asymmetry enhancement was not observed in cases in which a 330° PT-ICRS was implanted, despite the improvement in visual results when replacing a 320° traditional ring with a 330° PT-ICRS. These conclusions are limited as this is a case series with few cases.

ABSTRACT

This study aimed to provide a comprehensive picture of the life expectancy of dogs in Portugal, focusing on the impact of diverse factors including breed, sex, size, and skull shape. The final dataset, gathering data from the national registry database, consisted of 278,116 dogs with confirmed deaths. The mean lifespan at birth for all the dogs was around 8.91 years, with the female dogs tended to have a similar lifespan to male dogs. The analysis of life expectancy at birth for the 20 most common non-Portuguese breeds and 10 Portuguese breeds revealed that Yorkshire Terriers had the highest life expectancy (10.89 years) and French Bulldogs the lowest (6.27 years). Size and cephalic index were found to be influential factors, with large brachycephalic breeds exhibiting shorter life expectancies and smaller, mesocephalic breeds experiencing longer lifespans. Additionally, the cephalic index had a more substantial impact on life expectancy compared to body size. These findings enhance the understanding of the factors influencing canine longevity and aid in developing strategies to improve the health and lifespan of companion dogs.

Chemical desiccation in the preharvest of grains and seeds is commonly used in production fields. Using herbicides for this purpose is a viable alternative to reduce beans' exposure to adverse crop conditions. Our objectives were to evaluate (1) the efficacy of herbicides for accelerated defoliation of cowpea, (2) the impact of herbicide application on antioxidant enzyme activity and protein and amino acid contents in seeds, and (3) the effects of different herbicide application schedules on the physiological aspects of seeds. In the first experiment, in addition to the control treatment (without herbicides), seven herbicides and two mixtures were applied at night: diquat, flumioxazin, diquat + flumioxazin, glufosinate ammonium, saflufenacil, carfentrazone, diquat + carfentrazone, atrazine, and glyphosate. Diquat and its mixtures showed greater efficacy in anticipating the harvest. Flumioxazin and diquat alone reduced amino acid content by 61.72 and 51.44%, respectively. The same trend was observed for total soluble proteins. The activity of antioxidant enzymes (CAT, POD, PPO) increased, indicating oxidative stress caused by diquat and flumioxazin. In the second experiment, we tested three application times (6 a.m., 12 p.m., 6 p.m.) with diquat, diquat + flumioxazin, and diquat + carfentrazone. The lowest damage to chlorophyll a was at 6 a.m.; other times reduced photosynthetic pigments and increased carotenoid content. Total soluble sugars decreased by 27.74% with nocturnal application of diquat + flumioxazin. Our data indicate that herbicide use for desiccation affects seed quality. These findings highlight the need for selecting appropriate herbicides and application times. Future research should explore long-term impacts on crop yield and quality.

ABSTRACT

3-Tetrazolyl-ß-carbolines were prepared by the Pictet-Spengler approach using a tryptophan analogue as building block, in which the carboxylic acid was replaced by the bioisosteric tetrazole group. Knowing that ß-carbolines are often associated with psychopharmacological effects, the study of the 3-tetrazolyl-ß-carbolines as potential neuroprotective agents against Parkinson's disease was investigated. The evaluation of neuroprotective effects against 1-methyl-4-phenylpyridin-1-ium (MPP+)-induced cytotoxicity allowed to identify compounds with relevant neuroprotective activity. One derivative, 3-(1-benzyl-1H-tetrazol-5-yl)-1-(p-dimethylaminophenyl)-ß-carboline, stood out for its low cytotoxicity and excellent performance, preventing cell death induced by this neurotoxin. The most promising compounds were also evaluated for their neuroprotective properties against iron (III)-induced cytotoxicity. However, only one 3-tetrazolyl-ß-carboline derivative slightly reduced iron-induced cytotoxicity. Overall, the neuroprotective properties of 3-tetrazolyl-ß-carbolines have been demonstrated and this finding may contribute to the development of new therapies for Parkinson's disease.

ABSTRACT

INTRODUCTION: ChatGPT has attracted a lot of interest worldwide for its versatility in a range of natural language tasks, including in the education and evaluation industry. It can automate time- and labor-intensive tasks with clear economic and efficiency gains. METHODS: This study evaluated the potential of ChatGPT to automate psychometric analysis of test questions from the 2020 Portuguese National Residency Selection Exam (PNA). ChatGPT was queried 100 times with the 150 MCQ from the exam. Using ChatGPT's responses, difficulty indices were calculated for each question based on the proportion of correct answers. The predicted difficulty levels were compared to the actual difficulty levels of the 2020 exam MCQ's using methods from classical test theory. RESULTS: ChatGPT's predicted item difficulty indices positively correlated with the actual item difficulties (r (148) = -0.372, p < .001), suggesting a general consistency between the real and the predicted values. There was also a moderate significant negative correlation between the difficulty index predicted by ChatGPT and the number of challenges (r (148) = -0.302, p < .001), highlighting ChatGPT's potential for identifying less problematic questions. CONCLUSION: These findings unveiled ChatGPT's potential as a tool for assessment development, proving its capability to predict the psychometric characteristics of high-stakes test items in automated item calibration without pre-testing in real-life scenarios.

ABSTRACT

Joint hypermobility syndromes, particularly chronic pain associated with this condition, including Hypermobile Ehlers-Danlos Syndrome (hEDS) and Hypermobility Spectrum Disorders (HSD), present diagnostic challenges due to their multifactorial origins and remain poorly understood from biomechanical and genomic-molecular perspectives. Recent diagnostic guidelines have differentiated hEDS, HSD, and benign joint hypermobility, providing a more objective diagnostic framework. However, incorrect diagnoses and underdiagnoses persist, leading to prolonged journeys for affected individuals. Musculoskeletal manifestations, chronic pain, dysautonomia, and gastrointestinal symptoms illustrate the multifactorial impact of these conditions, affecting both the physical and emotional well-being of affected individuals. Infrared thermography (IRT) emerges as a promising tool for joint assessment, especially in detecting inflammatory processes. Thermal distribution patterns offer valuable insights into joint dysfunctions, although the direct correlation between pain and inflammation remains challenging. The prevalence of neuropathies among hypermobile individuals accentuates the discordance between pain perception and thermographic findings, further complicating diagnosis and management. Despite its potential, the clinical integration of IRT faces challenges, with conflicting evidence hindering its adoption. However, studies demonstrate objective temperature disparities between healthy and diseased joints, especially under dynamic thermography, suggesting its potential utility in clinical practice. Future research focused on refining diagnostic criteria and elucidating the underlying mechanisms of hypermobility syndromes will be essential to improve diagnostic accuracy and enhance patient care in this complex and multidimensional context.

This study compared the most common absolute sprint threshold (> 25.2 km/h) with relative and individualized thresholds (> 70%, > 75%, > 80%, > 85% and > 90% of peak match speed). Twenty elite soccer players, competing in the first division of the Portuguese League, were monitored using GNSS equipment during thirty-four official matches. Peak match speed was retrieved as the individual maximal speed reached during the full season. Distances were registered when speed overcame the absolute and the relative thresholds. Mean ± SD of peak speeds and distances covered were calculated, and Pearson correlation (r) and mean paired differences were performed to analyze relationships and differences between thresholds. The peak match speed was 32.9 ± 1.4 km/h. Correlations between distances covered using the absolute and relative thresholds varied from very strong (> 70%: r = 0.84, p < .001; > 75%: r = 0.89, p < .001; and > 80%: r = 0.88, p < .001), strong (> 85%: r = 0.79, p < .001), to moderate (> 90%: r = 0.59, p < .001). Overall, the > 75% (ES: 0.23 [95% CI: 0.16, 0.31]) and the > 90% (ES: -1.65 [95%CI: -1.85, -1.48]) relative thresholds presented the smallest and largest differences, respectively, with the absolute threshold. Differences were also found when considering the playing positions. While the distances covered by central midfielders were similar between the absolute and > 80% thresholds (-0.03 [-0.16, 0.10]), fullbacks covered largely more distance -1.88 [-2.42 -1.50]) in the absolute threshold than in the > 80% threshold. The distances covered by players varied based on the selected threshold, affecting the distances covered by different playing positions. Being the highest speed threshold within displacements thresholds, the absolute sprint threshold showed greater similarity to lower rather than higher relative thresholds.

ABSTRACT

OBJECTIVES: To explore postural disability in Usher Syndrome (USH) patients using temporal posturographic analysis to better elucidate sensory compensation strategies of deafblind patients for posture control and correlate the Activities-specific Balance Confidence (ABC) scale with posturographic variables. DESIGN: Thirty-four genetically confirmed USH patients (11 USH1, 21 USH2, 2 USH 4) from the Otolaryngology Outpatient Clinic and 35 controls were prospectively studied using both classical and wavelet temporal analysis of center of pressure (CoP) under different visual conditions on static and dynamic platforms. The functional impact of balance was assessed with the ABC scale. Classical data in the spatial domain, Sensorial Organization Test, and frequency analysis of the CoP were analyzed. RESULTS: On unstable surfaces, USH1 had greater CoP surface area with eyes open (38.51 ± 68.67) and closed (28.14 ± 31.64) versus controls (3.31 ± 4.60), p < 0.001 and (7.37 ± 7.91), p < 0.001, respectively. On an unstable platform, USH consistently showed increased postural sway, with elevated angular velocity versus controls with eyes open (USH1 [44.94 ± 62.54]; USH2 [55.64 ± 38.61]; controls [13.4 ± 8.57]) (p = 0.003; p < 0.001) and closed (USH1 [60.36 ± 49.85], USH2 [57.62 ± 42.36]; controls [27.31 ± 19.79]) (p = 0.002; p = 0.042). USH visual impairment appears to be the primary factor influencing postural deficits, with a statistically significant difference observed in the visual Sensorial Organization Test ratio for USH1 (80.73 ± 40.07, p = 0.04) and a highly significant difference for USH2 (75.48 ± 31.67, p < 0.001) versus controls (100). In contrast, vestibular (p = 0.08) and somatosensory (p = 0.537) factors did not reach statistical significance. USH exhibited lower visual dependence than controls (30.31 ± 30.08) (USH1 [6 ± 11.46], p = 0.004; USH2 [8 ± 14.15], p = 0.005). The postural instability index, that corresponds to the ratio of spectral power index and canceling time, differentiated USH from controls on unstable surface with eyes open USH1 (3.33 ± 1.85) p < 0.001; USH2 (3.87 ± 1.05) p < 0.002; controls (1.91 ± 0.85) and closed USH1 (3.91 ± 1.65) p = 0.005; USH2 (3.92 ± 1.05) p = 0.045; controls (2.74 ± 1.27), but not USH1 from USH2. The canceling time in the anteroposterior direction in lower zone distinguished USH subtypes on stable surface with optokinetic USH1 (0.88 ± 1.03), USH2 (0.29 ± 0.23), p = 0.026 and on unstable surface with eyes open USH1 (0.56 ± 1.26), USH2 (0.072 ± 0.09), p = 0.036. ABC scale could distinguish between USH patients and controls, but not between USH subtypes and it correlated with CoP surface area on unstable surface with eyes open only in USH1(ρ = 0.714, p = 0.047). CONCLUSIONS: USH patients, particularly USH1, exhibited poorer balance control than controls on unstable platform with eyes open and appeared to rely more on proprioceptive information while suppressing visual input. USH2 seems to use different multisensory balance strategies that do not align as well with the ABC scale. The advanced analysis provided insights into sensory compensation strategies in USH subtypes.
