ABSTRACT

Avaliou-se a correlação entre estruturas fetais e extrafetais com a predição do dia antes do parto (DAP) em raças de cães miniaturas. Para isso, realizou-se um experimento, utilizando-se 12 cadelas, com peso corporal entre 3,0kg e 5,0kg, sendo seis da raça Chihuahua, duas da raça Shih-Tzu, duas da raça Spitz Alemão e duas da raça Yorkshire. Foram mensurados, por meio da ultrassonografia, diâmetro biparietal (DBP), diâmetro torácico (DTX), diâmetro abdominal (DAB), comprimento craniocaudal (CCC), diâmetro da cavidade coriônica interna (DCI) e espessura da placenta (EP), a partir do 15º dia após a última monta. Foram estudadas as correlações simples e a significância dos coeficientes de regressão linear simples e o coeficiente de determinação (R), com nível de significância estabelecido em P<0,05. Entre os parâmetros avaliados, DBP, DTX, DAB e CCC foram os mais correlacionados com tempo gestacional, podendo ser utilizados para prever dia antes do parto em cadelas de raças miniaturas.(AU)


The correlation between fetal and extra-fetal structures with the pre-delivery prediction (DAP) in miniature dog breeds was evaluated. For this, an experiment was carried out using 12 bitches, with body weight between 3.0kg and 5.0kg, being 6 Chihuahua, 2 Shih-Tzu, 2 German Spitz and 2 Yorkshire breed. The Biparietal Diameter (BD), Thoracic Diameter (TD), Abdominal Diameter (AD), Crown-rump length (CRL), Internal Chorionic Cavity Diameter (ICD) and Placenta Thickness (PT) were measured by ultrasonography from the 15th day after the last mating. The simple correlations and significance of simple linear regression coefficients and the coefficient of determination (R) were studied, with a significance level of P<0.05. BD, T, AD and CRL were the most correlated with gestational time, and can be used to predict day before delivery in miniature breed bitches.(AU)

ABSTRACT

A utilização da gema de ovo dificulta a padronização de meios diluidores e apresenta riscos biológicos. Assim, este estudo avaliou diferentes concentrações da lipoproteína de baixa densidade (LDL), em substituição à gema de ovo, para a confecção de diluentes para criopreservação espermática em ovinos. Foram utilizados um diluidor controle (CTR= 20% de gema de ovo) e cinco tratamentos, substituindo-se a gema pelas diferentes proporções de LDL (T1=6%; T2=8%; T3=12%; T4=16%; T5=20%), todos à base de TRIS-glicerol. Para o estudo, utilizaram-se dois ejaculados, de seis reprodutores da raça Santa Inês. Sessenta dias após a criopreservação, as amostras foram descongeladas e avaliadas subjetivamente quanto à motilidade total (MT, %) e progressiva (MP, %), ao vigor (1-5) e à integridade funcional (choque hisposmótico com água destilada, %) e estrutural (corante supravital eosina, %) das membranas espermáticas. As avaliações de vigor e funcionalidade de membrana não diferiram (P>0,05) entre os grupos. Entretanto, os grupos T4 (P<0,01) e T5 (P<0,05) foram superiores ao CTR para os parâmetros MT, MP e integridade estrutural de membrana, o que confirma que as LDLs podem ser alternativas eficientes para substituição da gema de ovo em diluidores para criopreservação de sêmen ovino.(AU)


The use of egg yolk makes it difficult to standardize extenders and presents biological hazards. Thus, this study evaluated different concentrations of low-density lipoprotein (LDL) to replace yolk extenders for production of sperm for cryopreservation in ovine. A control extender was used (CTR= 20% yolk) and five treatments, replacing the yolk by different ratios of LDL (T1= 6%; T2= 8%, T3= 12%; T4= 16%; T5= 20%) all based on TRIS-glycerol. For the study, two ejaculates from six Santa Ines breeding were used. Sixty days after cryopreservation, the samples were thawed and evaluated for total motility (MT, %) and progressive motility (MP, %), vigor (1-5) and the functional integrity (hyposmotic shock with distilled water, %) and structural (supravital dye eosin, %) of the sperm membranes. The evaluations of strength and membrane functionality didn't differ (P> 0.05) between groups. However, T4 (P< 0.01) and T5 (P< 0.05) groups were superior to the CTR for the MT, MP, and membrane structural integrity parameters, which confirms that LDLs can be efficient alternatives for yolk replacement in extenders for cryopreservation of ovine semen.(AU)

ABSTRACT

The attitudes toward people with epilepsy are influenced by the degree of knowledge of the condition. Teachers usually do not receive any formal instruction on epilepsy during their training. This study was done to access three hundred teachers' knowledge and attitudes toward epilepsy by answering a questionnaire in order to quantify their knowledge, attitude and practice toward epilepsy. Almost all the teachers had heard about epilepsy yet could not demonstrate discrimination among the students. Some teachers still thought that epilepsy was contagious. A few others either objected to having epileptic children in their classes or did not think that an epileptic child could achieve higher studies. The knowledge about the clinical characteristics and initial procedures to attend a person during a seizure was unsatisfactory. We conclude that schools should offer some kind of knowledge or assistance in health services and physicians must ensure that these teachers have sufficient knowledge of epilepsy. Also general public education campaigns should be encouraged in this field.

ABSTRACT

Este trabalho objetivou relacionar o diâmetro da vesícula embrionária/fetal (DVE) e comprimento craniocaudal do embriäo/feto (CCC) do 15§ ao 45§ dia de gestaçäo com a idade de gestaçäo em ovelhas. Utilizaram-se 19 ovelhas adultas da raça Bergamácia, com cio sincronizado e cobertas por um mesmo reprodutor. A avaliaçäo ultra-sonográfica foi realizada diariamente do 15§ ao 30§ dia após a cobriçäo e a cada cinco dias do 30§ ao 45§ dia, utilizando-se aparelho com transdutor linear de 5,0 e 7,5MHZ. O crescimento diário médio do DVE e CCC foi de 1,40mm e 1,33mm, respectivamente. A calibragem inversa obtida para DVE foi: Dias = -11 + 50(0,1 + 0,04log DVE sobrescrito) 1/2(sobrescrito), com R2(sobrescrito) = 0,85 e para CCC foi: Dias = 13,63 + 12,5(-0,71 + 0,l6CCC)1/2(sobrescrito), com R 2 sobrescrito = 0,93. Tanto o DVE como o CCC obtidos por ultra-sonografia säo medidas pelas quais se pode prever a idade de gestaçäo em ovelhas da raça Bergamácia

ABSTRACT

The relationship between polymorphisms in the interleukin-1 receptor antagonist (IL-1ra) gene and microbial vaginal colonization was examined in 88 asymptomatic women of reproductive age. Alleles of the intron 2 region of the IL-1ra gene were identified by polymerase chain reaction (PCR). PCR was also used to detect Ureaplasma urealyticum, Mycoplasma hominis, and Candida albicans; bacterial vaginosis (BV) was identified by clinical criteria. Among the 31 women with vaginal U. urealyticum, only 3 (9.7%) were homozygous for allele 2 of the IL-1ra gene; 21 (36.8%) of the 57 women who were negative for this organism were positive for allele 2 (P=.006). Only 7 women were positive for M. hominis; none were allele 2 homozygotes as opposed to 24 (29.6%) of the 81 women negative for M. hominis. There was no relation between C. albicans or BV and any IL-1ra allele. Reduced susceptibility to vaginal colonization with mycoplasmas may be associated with homozygosity of the 2 allele of the IL-1ra gene.

ABSTRACT

OBJECTIVE: Predisposing factors influencing recurrences of bacterial vaginosis (BV) or vaginitis from Candida remain unidentified for most women. As a component of studies to determine host susceptibility factors to genital tract infections in women, we measured expression of the 60-kDa and 70-kDa heat shock proteins (hsp60 and hsp70, respectively) in the circulation of women with or without a history of recurrent BV or candidal vaginitis and with or without a current lower genital tract infection. Heat shock protein expression is associated with a down-regulation of pro-inflammatory immune responses that would inhibit microbial infection. METHOD: The investigators measured hsp60 and hsp70, antibodies to these proteins, the pro-inflammatory cytokine tumor necrosis factor-alpha (TNF-alpha), and the anti-inflammatory cytokine interleukin-10 (IL-10) in sera by ELISA. The study population consisted of 100 women who attended a gynecology clinic in Campinas, Brazil. Of those, 55 had a history of recurrent vulvovaginitis (RV), while 45 were controls with no such history. Only women who were asymptomatic for at least 1 month were studied. RESULTS: Although all were asymptomatic, clinical and microbiological examination revealed that five of the women with a history of RV and two controls had a current candidal vaginal infection; 16 RV patients and 12 controls had BV; and six RV patients had both BV and candidiasis. Twenty-eight RV patients and 31 controls had no clinical or microbiological detectable vaginal infection. Among the RV patients, hsp60 and hsp70 were more prevalent in those with current BV (40.9% and 50.0%, respectively) or a candidal infection (45.5% and 54.5%) than in women with no current infection (21.4% and 17.9%). In the women with no history of RV, BV was not associated with a high prevalence of hsp60 (8.3%) or hsp70 (8.3%). Interleukin-10 and TNF were not more prevalent in vaginitis patients or controls with a current candidal infection or BV than in uninfected subjects. CONCLUSION: The high prevalence of circulating hsp60 and hsp70 in women with a history of RV and current BV or vaginal candidiasis, but not in women with no history of RV, suggests that differences in heat shock protein induction may be related to susceptibility to recurrent vaginal infections.

ABSTRACT

The expression of the 60-kD and 70-kD heat shock proteins (hsp60 and hsp70) in the vaginas of 43 asymptomatic women of reproductive age with or without a history of recurrent vulvovaginitis (RVV) were compared. Vaginal wash samples were obtained and assayed by enzyme-linked immunosorbent assay (ELISA) for human hsp60 and hsp70. Heat shock protein 70 was not detected in any of the 19 women with no history of RVV, and hsp60 was present in only one woman in this group. In contrast, in the RVV group, 11 (45.8%) were hsp60-positive and eight (33.3%) were hsp70-positive. The presence of either heat shock protein in the vagina was associated with an elevated vaginal pH (>4.5). Bacterial vaginosis or Candida was identified in some of the asymptomatic subjects; their occurrence was significantly higher in women with vaginal hsp70 than in women with no heat shock proteins. Oral contraceptives were used by 35.7% of subjects who were negative for vaginal heat shock proteins, as opposed to only 12.5% of women who were positive for hsp70 and 8.3% who were positive for hsp60. Expression of heat shock proteins in the vagina may indicate an altered vaginal environment and a susceptibility to vulvovaginal symptoms.

ABSTRACT

OBJECTIVES: The cause of recurrent vulvovaginitis remains unexplained in most cases. Heat shock protein synthesis is induced under conditions of stress; its presence in vaginal samples from women who were between episodes of recurrent vulvovaginitis thus might reflect a persistent perturbation in the local milieu. STUDY DESIGN: We undertook an evaluation by means of enzyme-linked immunosorbent assay of 60-kd heat shock protein and inducible 70-kd heat shock protein expressions in vaginal wash samples from 24 symptom-free women with a history of recurrent vulvovaginitis and 19 matched control subjects. The samples were also tested for Candida albicans, Chlamydia trachomatis, Ureaplasma urealyticum, Mycoplasma hominis, and human papillomavirus by polymerase chain reaction; for bacterial vaginosis by clinical and microbiologic evaluation; and for interleukin 10, interleukin 1, interleukin 8, RANTES, and eotaxin by enzyme-linked immunosorbent assay. RESULTS: The presence of 60-kd heat shock protein was detected in 11 women with recurrent vulvovaginitis (45.8%) and 1 control subject (5.3%, P =.005). Similarly, 70-kd heat shock protein was present in 8 patients with recurrent vulvovaginitis (33.3%) and no control subjects (P =.005). The presence of 60-kd heat shock protein and the presence of 70-kd heat shock protein were correlated with each other (P =.02), as were both 60-kd heat shock protein (P =.006) and 70-kd heat shock protein (P =.01) correlated with IL-10. There was no relation between the presence of 60-kd heat shock protein or 70-kd heat shock protein and detection of IL-1, IL-8, or any microorganism. CONCLUSION: The expression of heat shock proteins and IL-10 in the vaginas of women with a history of recurrent vulvovaginitis but not in the vaginas of control subjects suggests the existence of differences in the vaginal milieu between the 2 groups, even when both are without vaginal symptoms.

A simple method for the collection, preservation, shipment, and testing of minute amounts of dried monoclonal antibodies for typing Neisseria meningitidis B is described. The monoclonal antibodies collected on filter paper were extracted in PBS and evaluated by Dot-blot employing whole cells of N. meningitidis B as antigen. The dried filter paper with monoclonal antibodies could be stored at room temperature for as long as 30 days without detectable changes in antibody response when used for typing outer membrane antigens of N. meningitidis B.
