ABSTRACT

Natural and anthropogenic environmental impacts can introduce contaminants into sensitive habitats, threatening ecosystems and human health. Consistent monitoring of coastal areas provides critical environmental assessment data. Sediments and Eastern Oyster (Crassostrea virginica) tissues were collected at fourteen South Carolina (SC) and four North Carolina (NC) sites as part of the National Oceanic and Atmospheric Administration's Mussel Watch environmental monitoring program. Cellular and molecular techniques were employed to measure C. virginica stress response, specifically, Lipid Peroxidation (LPx), Glutathione (GSH), and qPCR techniques. Gene specific primers targeted for detecting oxidative stress and cellular death were developed in C. virginica to gauge response to current environmental conditions using gill and hepatopancreas (HP) tissue. In order to validate gene specific markers as additional assessment tools, a 96 h zinc (Zn) laboratory exposure was performed. Cellular biomarker data revealed tissue specific responses. Hepatopancreas data showed C. virginica exhibited stress through the lipid peroxidation assay amongst sampling sites, however, response was managed through glutathione detoxification. Gill tissue data had significantly lower levels of cellular biomarker response compared to hepatopancreas. Molecular biomarkers targeting these cellular stress pathways through qPCR analysis show upregulation of Metallothionein in hepatopancreas and gill tissue with a concurrent > 2-fold upregulation in the detoxification marker Superoxide Dismutase (SOD) at three NC sites. SC sites displayed higher stress levels through LPx assays and down-regulation in GPx gene activity. Laboratory zinc exposure revealed no significance in cellular biomarker results, however, molecular data showed gills responding to zinc treatment through upregulation of Metallothionein, SOD and Cathepsin L, indicating an acute response in gills. Collectively, chemical, cellular and molecular methods clarify sentinel stress response of biological impacts and aid in evaluating environmental health in coastal ecosystems. This combined methodological approach provides a detailed analysis of environmental conditions and improves land-use management decisions.

ABSTRACT

The National Oceanic and Atmospheric Administration (NOAA), National Centers for Coastal Ocean Science (NCCOS) Mussel Watch Program (MWP), conducts basin-wide monitoring and place-based assessments using dreissenid mussels as bioindicators of chemical contamination in the Laurentian Great Lakes. Polycyclic aromatic hydrocarbons (PAHs) body burden results for the period 2009-2018 were combined into one dataset from multiple MWP studies allowing for a robust characterization of PAH contamination. Patterns in PAH data were identified using descriptive statistics and machine learning techniques. Relationships between total PAH concentration in dreissenid mussel tissue, impervious surface percentages, and PAH relative concentration were identified and used to build a predictive model for the Great Lakes Basin. Significant positive correlation was identified by the Spearman's rank correlation test between total PAH concentration and percent impervious surface. The findings support the paradigm that PAHs are primarily derived from land-based sources. Offshore and riverine locations had the lowest and highest median total PAH concentrations, respectively. PAH assemblages and ratios indicated that pyrogenic sources were more predominant than petrogenic sources and that PAHs at offshore sites exhibited relatively more weathering compared to inshore sites.

ABSTRACT

Along the littoral, a growing number of anthropogenic structures have caused substantial habitat destruction. Despite their detrimental impact, these constructions could play a role in the functioning of coastal ecosystems. The objective of this work was to assess the distribution of juvenile coastal fish along a seascape composed of various natural and artificial habitats in order to determine the potential role of coastal infrastructures as juvenile habitat. We surveyed juvenile populations on various infrastructures and natural sites along a 100 km shoreline of the French Mediterranean coast. Juvenile densities varied according to the level of artificialization of the sites. Densities were the highest on coastal defense structures, intermediate in natural sites and lowest in harbors. Focusing inside harbors revealed highly variable densities depending on the type of habitat, with densities on ripraps or jetties that were equivalent to those of natural sites. Our results underline the importance of anthropogenic structures as potential juvenile habitats, which is too often not considered in management plans.

ABSTRACT

Coastal nursery habitats are essential for the renewal of adult fish populations. We quantified the availability of a coastal nursery habitat (shallow heterogeneous rocky bottoms) and the spatial variability of its juvenile fish populations along 250km of the Catalan coastline (France and Spain). Nurseries were present in 27% of the coastline, but only 2% of them benefited from strict protection status. For nine taxa characteristic of this habitat, total juvenile densities varied significantly between nursery sites along the coastline, with the highest densities being found on the northern sites. Recruitment level (i.e. a proxy of nursery value) was not explained by protection level, but it was moderately and positively correlated with an anthropization index. Patterns of spatial variations were taxa-specific. Exceptional observations of four juveniles of the protected grouper Epinephelus marginatus were recorded. Our data on habitat availability and recruitment levels provides important informations which help to focus MPA management efforts.
