ABSTRACT

As excellent biocontrol agents and plant growth promoters, Trichoderma species are agriculturally important. Trichoderma spp. cultures can be produced using solid-state or submerged cultivation, the latter being much less labor intensive and easier to control and automate. The aim of the study was to investigate the ability to increase the shelf-life of T. asperellum cells by optimizing cultivation media and upscaling the submerged cultivation process. Four different cultivation media were used with or without the addition of Tween 80 and stored with or without incorporation into peat, and viability, expressed as CFU/g, was assessed during one year of storage in an industrial warehouse. The addition of Tween 80 had a positive effect on the biomass yield. The culture medium played a major role in the ability of the mycelium to produce spores, which in turn influenced the amount of CFU. This effect was less pronounced when the biomass was mixed with peat prior to storage. A procedure that increases the number of CFU in a peat-based product formulation is recommended, namely, incubation of the mixture at 30 °C for 10 days prior to storage at 15 °C over an extended period of time.

ABSTRACT

T. asperellum MSCL 309 was used in the study. T. asperellum was grown in the stirred bioreactor under submerged cultivation. The resulting biomass was filtered to obtain a thick biomass. The viability and antifungal activity of T. asperellum biomass samples were determined simultaneously by studying the colonization of the malt extract agar medium surface and its competitiveness with the plant pathogenic fungus Fusarium graminearum using in vitro dual culture experiments. Treatment with starch, alone or in combination with copper (II) sulphate and/or hydrochloric acid did not significantly affect fungal viability compared to control. An important factor in maintaining viability was the addition of hydrochloric acid, which significantly increased the storage life of biomass. In all post-treatments, F. graminearum was overgrown with T. asperellum in seven days, and accordingly, the larger the area occupied by T. asperellum, the smaller the area of F. graminearum colonization. Viability and antifungal activity of T. asperellum persisted throughout the experiment, at least for eight weeks. All the post-treatment methods we studied improved the viability and antifungal activity of Trichoderma, at least in terms of the area of the colonized surface. For the development of long-term viable and active T. asperellum preparations, we recommend the use of acidification of T. asperellum biomass obtained by submerged fermentation.

ABSTRACT

Recently, sewage sludge (SS) disposal has become one of the greatest global challenges. In this study, we aimed to evaluate the effect of faba bean straw (Straw-B), wheat straw (Straw-W), and wood-chip pellets (WCP) amended to SS, as well as bioaugmentation (BA), on the physicochemical characteristics and structure of the microbial community of the treated SS. Sixteen days of incubation of SS-containing mixtures revealed the highest efficiency of Straw-W(BA) in terms of SS stabilisation, i.e., the highest and most stable respiration intensity, the lowest ammonia emission, and the highest stimulation effect on the cress seedling growth. Shotgun sequencing data analysis showed that Proteobacteria dominated in the raw SS with 60.17% reads, which consisted of 16.40%, 29.18%, and 12.33% of Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria, respectively. All treated samples were characterised by an increased abundance of Firmicutes (32.70-53.84%). A remarkable increase in virus abundance (0.34% reads) was detected in the treated SS, which was incubated without C amendment and bioaugmentation. The addition of C sources to the SS changed some physicochemical characteristics of the mixture. All of these findings provide novel insights toward a mechanistic understanding of the fate of the human sewage microbiome in wastewater and other environments.

ABSTRACT

AIM: Folic acid supplementation before and during pregnancy decreases rates of neural tube defects. However, many women fail to adhere to folic acid supplementation recommendations. This study explored factors associated with women's intention to take folic acid before and during pregnancy, using the Theory of Planned Behaviour, with an emphasis on differences between Israeli-born women and immigrant women from the Former Soviet Union. METHODS: In this cross-sectional study, 100 Israeli born-women and 100 women from the Former Soviet Union of childbearing age completed a questionnaire, based on the Theory of Planned Behaviour. RESULTS: The findings indicated a significant difference in the rates at which Israeli-born and FSU-born women took folic acid before and during pregnancy, as well as a significant difference in their intention to take folic acid supplementation in future pregnancies. The theoretical model explained 88.7% of variance in women's intention to take folic acid, with the most influential variable being behavioural attitudes towards taking folic acid. CONCLUSION: The study emphasizes the need to increase knowledge and change attitudes and beliefs about folic acid supplementation among women and their significant others, as well as the importance of cultural, language, and economic barriers when treating Former Soviet Union immigrant populations.
