ABSTRACT

Concentrations of total mercury, total selenium, and cadmium ([THg], [TSe], [Cd]) were determined in hair of California sea lion (Zalophus californianus) pups from four islands of the Gulf of California and the Baja California Pacific coast (NG, CG, NP, and CP) to identify geographical differences and the effect of Se against Hg toxicity (TSe:THg molar ratio). THg displayed a strong north-south trend for both ecoregions, while TSe presented a significantly high concentration only for CG. TSe:THg molar ratios decreased when [THg] increased, with the lowest ratios presenting in NG pups, in which [THg] exceeded toxicological thresholds of concern. [Cd] presented similar values at all study sites except CG, which presented the lowest level. The present study shows that proximity to urbanized coastal areas has a strong influence on [THg] in pups, while [TSe] and [Cd] are probably more related to the physiological requirements of the species, and environmental conditions.

ABSTRACT

The effect of two different anionic membranes on manganese deposition was studied in a two-compartment electrochemical reactor with a titanium cathode and a dimensionally stable RuO2|Ti anode. Chronopotentiometry, ICP-OES, SEM, XRD and elemental mapping were used to understand the changes in concentration and characteristics of the metallic deposition at different current densities with the anionic membranes AMI 7001s and Neosepta AMX. The results demonstrate that AMI reduces more manganese than AMX below -100 A m-2, generating more metallic deposition but also more low-solubility manganous by-products, whereas both membranes exhibited similar behaviours above -100 A m-2 reaching the maximum current efficiency (63%) at -200 A m-2. It was also observed that the membranes have a significant effect on sulphate consumption since they are anions.

ABSTRACT

We study the evolution of the probability density of ensembles of iterates of the logistic map that advance toward and finally remain at attractors of representative dynamical regimes. We consider the mirror families of superstable attractors along the period-doubling cascade and of chaotic-band attractors along the inverse band-splitting cascade. We examine also their common aperiodic accumulation point. The iteration time progress of the densities of trajectories is determined via the action of the Frobenius-Perron (FP) operator. As a difference with the study of individual orbits, the analysis of ensembles of positions offers a viewpoint from which the nonlinear dynamical features of this iconic model can be better characterized in statistical-mechanical terms. The scaling of densities along the considered families of attractors conforms to a renormalization-group (RG) structure, while their entropies are seen to attain extrema at the fixed points of the RG flows. Additionally, this entropy as a function of the map control parameter displays the characteristic features of an equation of state of a thermal system undergoing a second-order phase transition. We discuss our results.

ABSTRACT

To evaluate different methods for the determination of postmortem myofibrillar fragmentation, three protocols were compared: fragmentation index method (FI) after drying the filtrate residue at room temperature for 10 min (FIroom) or at 105 °C for 12 h (FIoven); and myofibrillar fragmentation index (MFI) by the absorbance method. Beef Longissimus thoracis steaks (n = 34) were randomized into two treatments (frozen/thawed and unfrozen) and aged for 0, 7, 14 or 21 days. An interaction (P < 0.05) between method and treatment, method and aging and treatment and aging was found. FIroom was the best to detect a difference in fragmentation at all aging days. In addition, FIroom and FIoven both detected the tenderization process that occurs by freezing while the MFI method did not, yet all three methods were similarly related to tenderness as measured by shear force. It is suggested that FIroom is the most effective method (time, cost) to indicate myofibrillar fragmentation, under the conditions imposed herein.

ABSTRACT

Cancer therapies that target a single protein or pathway may be limited by their specificity, thus missing key players that control cellular proliferation and contributing to the failure of the treatment. We propose that approaches to cancer therapy that hit multiple targets would limit the chances of escape. To this end, we have developed a bicistronic adenoviral vector encoding both the CDKN2A and p53 tumor suppressor genes. The bicistronic vector, AdCDKN2A-I-p53, supports the translation of both gene products from a single transcript, assuring that all transduced cells will express both proteins. We show that combined, but not single, gene transfer results in markedly reduced proliferation and increased cell death correlated with reduced levels of phosphorylated pRB, induction of CDKN1A and caspase 3 activity, yet avoiding the induction of senescence. Using isogenic cell lines, we show that these effects were not impeded by the presence of mutant p53. In a mouse model of in situ gene therapy, a single intratumoral treatment with the bicistronic vector conferred markedly inhibited tumor progression while the treatment with either CDKN2A or p53 alone only partially controlled tumor growth. Histologic analysis revealed widespread transduction, yet reduced proliferation and increased cell death was associated only with the simultaneous transfer of CDKN2A and p53. We propose that restoration of two of the most frequently altered genes in human cancer, mediated by AdCDKN2A-I-p53, is beneficial since multiple targets are reached, thus increasing the efficacy of the treatment.

ABSTRACT

AIM: The purpose of the present study was to evaluate the accuracy of computed tomography colonography (CTC) in the preoperative localization and TN staging of colon cancer. CTC can be an effective technique for preoperative evaluation of colon cancer and could facilitate the selection of high-risk patients who may benefit from neoadjuvant chemotherapy. METHOD: This was a prospective observational study conducted at a single tertiary-care centre. It involved 217 patients (225 tumours) who had colon cancer and underwent preoperative CTC and elective colectomy. The radiologist determined the TNM stage using postprocessing software with multiplanar images and virtual colonoscopy. The following criteria were analysed for every colon tumour: location, size and signs of direct colon wall invasion. The histopathological findings of the surgical colectomy specimens served as the reference standard for local staging. RESULTS: CTC detected all tumours and achieved an exact location in 208 cases (92.4%). CTC findings changed the surgical plan in 31 patients (14.3%) following colonoscopy. The accuracy in differentiating T3/T4 vs T1/T2 tumours was 87.1%, with a sensitivity and specificity of 88.5% and 84.1%, respectively (kappa = 0.71). For high-risk tumours (T3 ≥ 5 mm and T4), CTC showed an accuracy, sensitivity and specificity of 82.7%, 86% and 80%, respectively (kappa = 0.65). The accuracy of N-stage evaluation was 69.3%, the sensitivity 74% and the specificity 67.1% (kappa = 0.37). CONCLUSION: CTC provides accurate information for the assessment of tumour localization and T staging, allowing better surgical planning and also allows the selection of locally advanced tumours that may benefit from new treatments such as neoadjuvant chemotherapy.

ABSTRACT

Coagulase-negative staphylococci (CNS) are important pathogens causing nosocomial infections worldwide with increasing resistance to antimicrobials. The aim of this study was to characterize resistance aspects of CNS isolated from patients with bloodstream infections acquired in hospitals in Belo Horizonte, MG, Brazil. Staphylococcus strains were characterized using repetitive sequence-based polymerase chain reaction (rep-PCR) fingerprinting with (GTG)5 primer. Phenotypic resistance was analyzed using AST-P5085 card (bioMérieuxVitek®). PCR was used to detect mecA, vanA, blaZ, ermA/B/C, aac-aphD, and SCC-mec. For statistical analyses, we used hierarchical cluster, chi-square test (χ2), and correspondence. Several clusters were formed within the same species using (GTG)5 primer, and strains showed resistance to the following antimicrobials: benzylpenicillin (100%); oxacillin (93.1%); gentamicin (36.3%); ciprofloxacin (63.7%); moxifloxacin (32.7%); norfloxacin (81.0%); erythromycin (86.2%); clindamycin (75.8%); linezolid, teicoplanin and vancomycin (1.7%); tigecycline (0%); fusidic acid (10.35%); rifampicin (13.7%); and trimethoprim/sulfamethoxazole (46.5%). Regarding genotypic analyses, 40%, 0%, 78%, 42%, 100%, 24%, and 30% were positive for mecA, vanA, blaZ, ermA, ermB, ermC, and aac-aphD, respectively. Regarding staphylococcal cassette mec (SCCmec) type, 3.4% presented type I; 5.0% type II; 27.1% type III; 20.3% type IIIA; and 32.2% type IIIB. Six clusters were formed and frequency distributions of resistant strains to oxacillin, gentamicin, ciprofloxacin, moxifloxacin, norfloxacin, erythromycin, clindamycin, linezolid, teicoplanin, vancomycin, fusidic acid, rifampicin, and trimethoprim/sulfamethoxazole, and mecA, blaZ, ermC, aac-aphD, and SCCmec type differed (p < 0.001). In conclusion, the strains investigated in this study were multidrug resistant and carried multiple antibiotic resistance genes.

ABSTRACT

Semitendinosus (ST) muscle steaks were grouped according to three locations (proximal, middle, and distal end), grilled to endpoint temperature of 71C or cooked for 20, 30, 40, 50, or 60 s in a microwave oven (Mw). The location did not affect (p > .05) the cooking loss (CL) or shear force (SF) values. The CL increased (p < .05) with longer microwave cooking times, but only the Mw40 samples did not differ (p > .05) from the grill samples. None of the microwaves' SF values were different (p > .05) from the grill values, with treatments Mw30 to Mw50 showing moderate repeatability (R = 0.51-0.60) and Mw30 and Mw60 showing higher correlations (r > .71) with grill values. Cooking beef strips with a microwave is a potential method for tenderness evaluation, but requires additional study to evaluate and optimize this application in different muscles and for comparison to sensorial data. PRACTICAL APPLICATIONS: The work was intended to evaluate the possibility of using a microwave oven for cooking meat to be used in objective measurement protocols for meat tenderness and to optimize the conditions for this purpose. The use of a standardized microwave procedure allows a dramatic reduction in analysis time and may reduce error variance due to nonuniform cooking procedures.

ABSTRACT

Retronychia is a recently described cause of ingrowth of the nail plate on the ventral surface of the proximal nail fold. Clinical features are repeated episodes of proximal paronychia, nail plate thickening, and occasionally granulation tissue emergence. The usual treatments for paronychia such as antibiotics and antifungals are ineffective in these cases. Avulsion of the nail plate is the treatment of choice for these patients, but effective treatment is usually delayed owing to inadequate diagnosis. Herein, we describe a 28-year-old woman with a case of retronychia. She was treated for two months with oral and topical antifungal and antibiotics by her general practitioner. After proper diagnosis and avulsion of the nail she presented a normal and non-painful growth of the affected nail.

ABSTRACT

The objective of this study was to determine the effects of freezing prior to aging on the meat tenderness of young Nellore and Aberdeen Angus bulls. Samples of the longissimus thoracis muscle were submitted to two treatments: conventional aging and freezing (-20°C for 40 days) followed by thawing and aging periods. The meats were evaluated after 0, 7, 14 and 21 aging days (1°C). Freezing increased (P<0.05) purge, cooking loss and total exudate loss throughout aging. Nellore meats had greater total exudate loss and shorter sarcomere lengths (P<0.05). Freezing increased proteolysis during aging in the meats of both breeds, but reduced shear force was found (P<0.05) only in Aberdeen Angus meats and only at time zero. These results suggest that the meat tenderizing process by freezing prior to aging may contribute to meat tenderness in the first weeks of aging, but it is dependent on the animal breed.

ABSTRACT

This study was conducted to investigate the effect of core sampling on Warner-Bratzler shear force evaluations of beef and pork loins (Longissimus thoracis et lumborum muscles) and to determine the relationship between them. Steaks of 2.54 cm from beef and pork loins were cooked and five round cross-section cores and five square cross-section cores of each steak were taken for shear force evaluation. Core sampling influenced both beef and pork shear force values with higher (P<0.05) average values and standard deviations for square cross-section cores. There was a strong and linear relationship (P<0.01) between round and square cross-section cores for beef (R(2)=0.78), pork (R(2)=0.70) and for beef+pork (R(2)=0.82) samples. These results indicate that it is feasible to use square cross-section cores in Warner-Bratzler shear force protocol as an alternative and potential method to standardize sampling for shear force measurements.

ABSTRACT

NaTaO(3) perovskite-like materials were synthesized using sodium acetate and tantalum ethoxide as precursors in an ultrasonic bath at room temperature. The pristine sample was thermally treated at 600 °C and characterized using XRD, N(2) physisorption, DRS, SEM and TEM techniques. The structural characterization by X-ray powder diffraction revealed that the crystallization of the NaTaO(3) phase prepared at 600 °C showed agglomerates sizes in the micrometric scale, as confirmed by scanning electron microscopy (SEM). On the other hand, well-defined NaTaO(3) particles in the nanometric scale were determined using TEM. It was found that, for the treated sample, the band gap and BET area was 3.8 eV and 9.5m(2) g(-1), respectively. The annealed perovskite, deposited onto ITO glass, presented an important variation in the open circuit potential transient during UV light irradiation in neutral solution, compared with its counterpart prepared by solid-state method. These intrinsic properties, given by the preparation route, might be appropriate for increase its photocatalytic activity.

ABSTRACT

We examine the connectivity fluctuations across networks obtained when the horizontal visibility (HV) algorithm is used on trajectories generated by nonlinear circle maps at the quasiperiodic transition to chaos. The resultant HV graph is highly anomalous as the degrees fluctuate at all scales with amplitude that increases with the size of the network. We determine families of Pesin-like identities between entropy growth rates and generalized graph-theoretical Lyapunov exponents. An irrational winding number with pure periodic continued fraction characterizes each family. We illustrate our results for the so-called golden, silver, and bronze numbers.

ABSTRACT

We show that the key transport states, insulating and conducting, of large regular networks of scatterers can be described generically by negative and zero Lyapunov exponents, respectively, of Möbius maps that relate the scattering matrix of systems with successive sizes. The conductive phase is represented by weakly chaotic attractors that have been linked with anomalous transport and ergodicity breaking. Our conclusions, verified for serial as well as parallel stub and ring structures, reveal that mesoscopic behavior results from a drastic reduction of degrees of freedom.

ABSTRACT

Prolonged endurance exercise over several days induces increase in extracellular water (ECW). We aimed to investigate an association between the increase in ECW and the change in aldosterone and vasopressin in a multistage ultraendurance triathlon, the 'World Challenge Deca Iron Triathlon' with 10 Ironman triathlons within 10 days. Before and after each Ironman, body mass, ECW, urinary [Na(+)], urinary [K(+)], urinary specific gravity, urinary osmolality and aldosterone and vasopressin in plasma were measured. The 11 finishers completed the total distance of 38 km swimming, 1800 km cycling and 422 km running within 145.5 (18.8) hours and 25 (22) minutes. ECW increased by 0.9 (1.1) L from 14.6 (1.5) L prerace to 15.5 (1.9) L postrace (P < 0.0001). Aldosterone increased from 70.8 (104.5) pg/mL to 102.6 (104.6) pg/mL (P = 0.033); vasopressin remained unchanged. The increase in ECW was related neither to postrace aldosterone nor to postrace vasopressin. In conclusion, ECW and aldosterone increased after this multistage ultraendurance triathlon, but vasopressin did not. The increase in ECW and the increase in aldosterone were not associated.

BACKGROUND: Structural equation models (SEM) are used to model multiple traits and the casual links among them. The number of different causal structures that can be used to fit a SEM is typically very large, even when only a few traits are studied. In recent applications of SEM in quantitative genetics mixed model settings, causal structures were pre-selected based on prior beliefs alone. Alternatively, there are algorithms that search for structures that are compatible with the joint distribution of the data. However, such a search cannot be performed directly on the joint distribution of the phenotypes since causal relationships are possibly masked by genetic covariances. In this context, the application of the Inductive Causation (IC) algorithm to the joint distribution of phenotypes conditional to unobservable genetic effects has been proposed. METHODS: Here, we applied this approach to five traits in European quail: birth weight (BW), weight at 35 days of age (W35), age at first egg (AFE), average egg weight from 77 to 110 days of age (AEW), and number of eggs laid in the same period (NE). We have focused the discussion on the challenges and difficulties resulting from applying this method to field data. Statistical decisions regarding partial correlations were based on different Highest Posterior Density (HPD) interval contents and models based on the selected causal structures were compared using the Deviance Information Criterion (DIC). In addition, we used temporal information to perform additional edge orienting, overriding the algorithm output when necessary. RESULTS: As a result, the final causal structure consisted of two separated substructures: BW→AEW and W35→AFE→NE, where an arrow represents a direct effect. Comparison between a SEM with the selected structure and a Multiple Trait Animal Model using DIC indicated that the SEM is more plausible. CONCLUSIONS: Coupling prior knowledge with the output provided by the IC algorithm allowed further learning regarding phenotypic causal structures when compared to standard mixed effects SEM applications.

ABSTRACT

Multiple endocrine neoplasia (MEN) syndromes are characterized by the association of various endocrine neoplasias. Prophylactic thyroidectomy is the treatment of choice for patients with RET gene mutations. The age at which patients undergo prophylactic thyroidectomy may vary depending on the position of the RET gene codon. In cases of MEN 2B, when the mutation is carried in codons 883, 918 or 922, prophylactic thyroidectomy is performed prior to 6 months of age, due to the increased aggressiveness of these heterozygosities, which are capable of determining the onset of medullary cancer during the first months of life. We present two heterozygous twin patients with MEN 2B syndrome who were born 32 weeks premature, and who underwent prophylactic thyroidectomy at 7 months of age. The patients were carriers of the mutation at codon 918. We suggested the early surgery at 7 months as, due to their prematurity, the patients were required to gain weight to improve their condition prior to surgery. The two patients had medullary thyroid carcinoma without lymph node involvement. In conclusion, for a truly prophylactic thyroidectomy, such patients should undergo surgery within the first month of life, particularly if these patients are carriers of the mutation in codons 883, 918 or 922.

ABSTRACT

We exhibit a remarkable equivalence between the dynamics of an intermittent nonlinear map and the electronic transport properties (obtained via the scattering matrix) of a crystal defined on a double Cayley tree. This strict analogy reveals in detail the nature of the mobility edge normally studied near (not at) the metal-insulator transition in electronic systems. We provide an analytical expression for the conductance as a function of the system size that at the transition obeys a q-exponential form. This manifests as power-law decay or few and far between large spike oscillations according to different kinds of boundary conditions.

ABSTRACT

We demonstrate that the dynamics toward and within the Feigenbaum attractor combine to form a q -deformed statistical-mechanical construction. The rate at which ensemble trajectories converge to the attractor (and to the repellor) is described by a q entropy obtained from a partition function generated by summing distances between neighboring positions of the attractor. The values of the q indices involved are given by the unimodal map universal constants, while the thermodynamic structure is closely related to that formerly developed for multifractals. As an essential component in our demonstration we expose, in great detail, the features of the dynamics of trajectories that either evolve toward the Feigenbaum attractor or are captured by its matching repellor. The dynamical properties of the family of periodic superstable cycles in unimodal maps are seen to be key ingredients for the comprehension of the discrete scale invariance features present at the period-doubling transition to chaos. Elements in our analysis are the following. (i) The preimages of the attractor and repellor of each of the supercycles appear entrenched into a fractal hierarchical structure of increasing complexity as period doubling develops. (ii) The limiting form of this rank structure results in an infinite number of families of well-defined phase-space gaps in the positions of the Feigenbaum attractor or of its repellor. (iii) The gaps in each of these families can be ordered with decreasing width in accordance with power laws and are seen to appear sequentially in the dynamics generated by uniform distributions of initial conditions. (iv) The power law with log-periodic modulation associated with the rate of approach of trajectories toward the attractor (and to the repellor) is explained in terms of the progression of gap formation. (v) The relationship between the law of rate of convergence to the attractor and the inexhaustible hierarchy feature of the preimage structure is elucidated. (vi) A "mean field" evaluation of the atypical partition function, a thermodynamic interpretation of the time evolution process, and a crossover to ordinary exponential statistics are given. We make clear the dynamical origin of the anomalous thermodynamic framework existing at the Feigenbaum attractor.

ABSTRACT

The adsorption of nitrogen species, in neutral electrolyte solutions, onto boron-doped diamond (BDD) electrode surfaces from dissolved NO2, NO, and N2O gases was induced at 0 V/SCE. Modified BDD electrode surfaces showed a different electrochemical response toward the hydrogen evolution reaction than did a nonmodified electrode surface in electrolyte base solution. The formation of molecular hydrogen and nitrogen gaseous species was confirmed by the online differential electrochemical mass spectrometry (DEMS) technique. Among the three nitrogen oxides gases, NO2 substantially modifies the electrolyte via hydrolysis leading to the formation of NO3- and its adsorption on the BDD electrode surface. The BDD/(NO3-) interface was the only N2O and N2 species generating system.
