ABSTRACT

BACKGROUND: Recently, a simple and rapid commercial assay (BD MGIT TBc ID) has been developed using a monoclonal antibody anti-MPT64 for the differentiation of Mycobacterium tuberculosis complex from other mycobacteria by immunochromatography. METHODS: We evaluate in this work the clinical usefulness of the test for the identification of 51 strains of M. tuberculosis complex and 24 strains of other mycobacteria belonging to 14 different species, compared with the method of hybridization with DNA probes. RESULTS: Immunochromatographic method performance was excellent, with sensitivity, specificity, positive and negative predictive values of 98, 100, 96.1, and 98.7%, respectively. CONCLUSIONS: These results indicate that immunochromatographic assay can be safely used for rapid identification of M. tuberculosis complex in combination with culture in liquid media. The test is extremely simple, provides results in just 15 minutes, requires no complex equipment or specialized personnel and may be a good alternative to molecular methods, especially in small laboratories.

Introducción: Recientemente, se ha desarrollado un sencillo y rápido método comercial (BD MGIT TBc ID) que emplea un anticuerpo monoclonal anti-MPT64 para la diferenciación del complejo Mycobacterium tuberculosis de otras micobacterias por inmunocromatografía. Métodos: Evaluamos en este trabajo la utilidad clínica del método para la identificación de 51 cepas del complejo M. tuberculosis y 24 cepas de otras micobacterias pertenecientes a 14 especies diferentes, comparándolo con el método de hibridación con sondas de ADN. Resultados: El resultado del método inmunocromatográfico fue excelente, con una sensibilidad, especificidad y valores predicitivos positivo y negativo de 98, 100, 96,1 y 98,7%, respectivamente. Conclusiones: Estos resultados indican que el método inmunocromatográfico puede ser usado con seguridad para la identificación rápida del complejo M. tuberculosis en combinación con el cultivo en medios líquidos. El método es extremadamente sencillo, ofrece resultados en sólo 15 minutos, no requiere equipamiento complejo ni personal especializado y puede ser una buena alternativa a los métodos moleculares, especialmente en pequeños laboratorios(AU)
