ABSTRACT

The aim of the present study was to isolate and evaluate the diversity of rhizobial and endophytic bacterial strains from undisturbed native rainforests within an iron ore mining site of the Serra Norte de Carajás in the Eastern Brazilian Amazon region to assess their biotechnological utility in reclamation of areas. Experiments were conducted to capture strains from samples of the soil of these forests at the sites Arenito II, Noroeste II, and Sul IV using Macroptilium atropurpureum and Mimosa acutistipula var. ferrea as trap host plants. Only M. atropurpureum nodulated, and the different bacterial strains were isolated from its nodules. There was no difference in the number of nodules among the areas, but the Arenito II bacterial community was the most efficient, indicated by the aboveground biomass production and suitable shoot mass/root mass ratio. Fifty-two (52) bacterial isolates were obtained, distributed in five groups, including nodulating and endophytic bacteria: 32 from Arenito II, 12 from Noroeste II, and 8 from Sul IV. The nodulating Bradyrhizobium genus was common to the three areas, whereas Paraburkholderia was found only in Arenito II. The nodD1 gene was amplified in all the strains of both nodulating genera. Strains of the nodulating genus Methylobacterium were also isolated from the three areas; however, they did not nodulate the host of origin, and their nodD1 gene was not amplified. Endophytic strains were also isolated from the genera Paenibacillus, Pantoea, and Leifsonia in Arenito II, Leifsonia in Noroeste I, and Paenibacillus in Sul IV. The greater nodulation and rhizobial and endophytic bacterial diversity observed in Arenito II were probably due to the more suitable edaphic properties of the area. The isolated strains were incorporated in the collection of the Department of Soil Science of UFLA and will be investigated in relation to their symbiotic characteristics with native host plants, as well as their ability to perform other biological processes.

ABSTRACT

Pharmaceuticals and personal care products (PPCPs) constitute a group of chemicals of concern because of their potential toxicity when reaching aquatic environments. Wastewaters are one of the main pathways of introduction into the environment of the chemical compounds used in PPCPs because, in most cases, wastewater treatment facilities are not 100% efficient in their removal. This problem is accentuated in rural zones and isolated communities where conventional treatment systems are too expensive to build and operate. Waste-stabilization ponds and constructed wetlands (CWs) are natural wastewater treatment systems which are used to improve the quality of sewage from small communities because of their low cost and easy maintenance. There is growing interest in combining the two technologies to make a more robust system, taking into account their respective strengths and weaknesses. In this work, a combined macrophyte pond-CW system was evaluated for the presence at three sampling points (influent, pond effluent and CW effluent) of fifteen steroid hormones and six benzotriazole ultraviolet stabilizers (BUVSs). None of the targeted BUVS compounds were detected in either the influent or effluent, probably because of the particular characteristics of the population served by the wastewater system. In contrast, eight different steroid hormone compounds were detected at concentrations ranging from 17.3 to 247.7 ng·L-1 in influent samples and from 8.1 to 22.1 ng·L-1 in final effluent samples. The pond-CW system showed high elimination rates of steroid hormone residues with average removal efficiencies of over 77%. This efficacy was confirmed in the ecological risk assessment evaluation that was performed. Final effluents showed a low ecological risk associated with steroid hormones in contrast to the medium-high ecological risks found in the influent samples.

ABSTRACT

Objetivo: Estimar la prevalencia de uso de anticoncepción hormonal de solo gestágenos (AHSG) entre las mujeres demandantes de anticoncepción reversible en atención primaria (AP). Diseño: Estudio descriptivo transversal multicéntrico. Emplazamiento: AP de la Comunidad de Madrid. Participantes: Mujeres entre 16-50 años, usuarias de anticoncepción reversible, que hablen español y hubieran acudido en el último año a consulta de AP. Mediciones principales: Encuesta telefónica. Variable principal: método anticonceptivo utilizado: solo gestágenos (sí/no). Edad, paridad, país de origen, tipo de método anticonceptivo (MAC) utilizado, motivo de elección, duración, fuente de información, lugar de obtención, satisfacción con el método. Resultados: 417 mujeres. Edad: media 30,3 años (DE: 7,7). Españolas: 69%, estudios secundarios y universitarios: 82%, nulíparas: 57%. Tipo de MAC utilizado: AHSG 14%, hormonal combinado (AHC): 74%, DIU cobre: 2%, preservativo: 10%. La prevalencia de uso de AHSG fue del 13,9% (IC95%: 10,6-17,2). Entre los AHSG el método más utilizado fue el inyectable trimestral de acetato de medroxiprogesterona (4,6%), píldora oral de desogestrel (4,1%), DIU-LNG (3,9%), implante subdérmico de etonogestrel (1,9%). El médico de familia fue el prescriptor del MAC en el 71% de las mujeres. Satisfacción: alta (mediana 10 sobre 10). Utilizar AHSG se asoció con mayor edad, ser extranjera y motivos de prescripción: lactancia y tener contraindicación médica para AHC (p < 0,05). Conclusiones: La prevalencia de uso para la AHSG fue del 14%, la satisfacción fue muy alta para todos los MAC. El perfil de usuaria para la AHSG corresponde a mujer de mayor edad, extranjera y con condiciones como la lactancia o la contraindicación para otros MAC

ABSTRACT

Despite the ubiquity and importance of indigenous arbuscular mycorrhizal fungi (AMF) for plant ecosystems; functioning of indigenus mycorrhizal symbiosis (IMS) and related environmental factors at coastal Caribbean ecosystems remains still scarce. In order to determine functionality of IMS under contrasting land uses and wet seasons from Cuba, the influence of the water stress on some AMF functionality parameters from a semi-natural savannah (NS), a recovered savannah (RS) and an agro-ecosystem (AG) from the Managed Floristic Reserve San Ubaldo-Sabanalamar, Pinar del Rio, Cuba were assessed during two-years. Soil and root samples were collected in April and October, during the dry and wet seasons, respectively, in 2008 and 2010. Four plots in each ecosystem were selected, and five soil sub-samples were randomly collected, bulked, mixed homogeneously and used as the composite sample per plot. The host plant root biomass, arbuscular mycorrhizal colonization of the host plant, density of the intraradical and extraradical AMF mycelia, fungal endophyte biomass and AMF spore density were assessed. The host plant root biomass increased in the NS environment during the dry season, and approximately 12.85g root/dm³ dry soil was recorded. The colonization degree were significantly higher in all environments during the wet season of the second year, with means ranging from 79% to 89%. The extraradical mycelia were significantly more abundant in the dry season of the second year in all environments, with a maximum of 279mg/dm³ in the RS ecosystem. The density of AMF spores was highest in the dry season of the second year for the three studied ecosystems. The RS ecosystem hosted 5 670 spores/100g dry soil. In general, the influence of rainfall seasonality on the function of AMF was stronger than the influence of ecosystem management. The root biomass and extraradical mycelia were high in the dry seasons, suggesting strategies to increase the volume of soil for the mutual benefit of the symbionts. The increase in spore density during the dry seasons appears as an adaptation allowing AMF to survive period of water shortage. This study improves our understanding of the adaptative responses of arbuscular mycorrhizal symbiosis to seasonal variations in soil water availability. Rev. Biol. Trop. 63 (2): 341-356. En el presente estudio se evaluó, durante dos años, la influencia de los períodos seco y lluvioso sobre el funcionamiento de hongos formadores de micorrizas arbusculares (HFMA) simbiontes de plantas nativas de una sabana semi natural, una sabana recuperada de la actividad agrícola y un agroecosistema, ubicados en la Reserva Florística Manejada San Ubaldo-Sabanalamar, Pinar del Río, Cuba. Se recolectaron muestras de suelo en abril y octubre (período seco y húmedo de 2008 y 2010, respectivamente). Dentro de cada ecosistema se seleccionaron cuatro parcelas y se recolectaron cinco submuestras al azar, las que fueron homogeneizadas para formar una muestra compuesta por parcela. Se cuantificó la biomasa de raicillas, la colonización micorrízica de las plantas hospedadoras en el campo, el micelio extrarradical, el micelio endófito y la densidad de esporas. La biomasa de raicillas en la sabana semi natural incrementó durante los períodos secos, la cual alcanzó hasta 12.85 g/dm3 de suelo. En todos los ecosistemas, la mayor colonización micorrízica ocurrió en la época lluviosa del segundo año con valores que oscilaron entre 79 y 89 %. Las mayores biomasas de micelio externo fueron registradas en los tres ecosistemas durante el período seco del segundo año, con un valor máximo de 279 mg/dm3 de suelo en la sabana recuperada. La mayor densidad de esporas de HFMA fue determinada en el periodo seco del segundo año para los tres ecosistemas estudiados, con el valor más alto en la sabana recuperada con 5 670 esporas/100 g de suelo seco. De manera general, se evidenció un efecto de la estacionalidad de la lluvia más que del manejo de ecosistema sobre el funcionamiento de los HFMA. La mayor actividad colonizadora se evidenció en períodos húmedos, mientras que mayor biomasa de raicillas y micelio externo durante los períodos secos, lo que sugiere estrategias de incremento del volumen del suelo explorado por parte de los HFMA como de las plantas hospedadoras. La proliferación de esporas evidenció la formación de estructuras de resistencia de los HFMA ante condiciones adversas. Los resultados obtenidos pondrían en evidencia la plasticidad de la simbiosis micorrízica ante variaciones en la disponibilidad de agua.

ABSTRACT

Asteraceae family shows a great diversity in Cuba and presents threatened taxons in high number. Pinar del Río has 19 threatened species belonging to this family and three of them are characteristic of a sandy (quartzitic) soil at the Reserva Florística Manejada San Ubaldo-Sabanalamar (Floristic Reserve San Ubaldo-Sabanalamar). The arbuscular mycorrhizal status and colonization rate in Aster grisebachii, Erigeron bellidiastroides, Erigeron cuneifolius, Pectis juniperina and Sachsia polycephala (Asteraceae) from this Reserve was determined in October 2009 and April 2010, during hydrologic rainy and dry period, respectively. All species were mycotrophic in both periods with a high percentage of colonization rates, between 66 and 100% in rainy period and between 84 and 100% in dry period. Vesicles, arbuscules, spores, hyphal and arbuscular coils of arbuscular mycorrhizae were observed. Paris -type and Arum -type morphologies (both) were observed indistinctly in all the species, and Intermediate type was observed only in Aster grisebachii and Erigeron bellidiastroides, which could be a hint that these species are colonized by more than one fungal species, and therefore enhancing the relevance of fungal identity as a considerable character in the morphology association. Also, septate hyphae and microsclerotium of dark septate endophytes were identifyed inside the roots of all the species.


La familia Asteraceae muestra una gran diversidad en Cuba y presenta un elevado número de taxones amenazados. Pinar del Río cuenta con 19 especies amenazadas pertenecientes a esta familia, de las cuales tres son características de los suelos arenosos cuarcíticos de la Reserva Florística Manejada San Ubaldo-Sabanalamar. Se determinó el estatus micorrízico arbuscular y la tasa de colonización de Aster grisebachii, Erigeron bellidiastroides, Erigeron cuneifolius, Pectis juniperina y Sachsia polycephala (Asteraceae) presentes en esta Reserva, en octubre del 2009 y abril del 2010, durante los períodos hidrológicos húmedo y seco, respectivamente. Todas las especies estaban micorrizadas en ambos períodos con elevados porcentaje de colonización micorrízica, entre 66 y 100% en el período lluvioso y entre 84 y 100% en el período seco. Se identificaron vesículas, arbúsculos, esporas, enrollados hifales y arbusculares característicos de las micorrizas arbusculares. Los tipos morfológicos Paris y Arum (ambos) fueron observados indistintamente en todas las especies, y el tipo Intermedio solo se observó en Aster grisebachii y Erigeron bellidiastroides, lo cual sugiere la colonización de estas especies por más de una especie fúngica y con ello la importancia de la identidad fúngica en la determinación de la morfología de la asociación. Se identificaron además, hifas septadas y microesclerosios de endófitos oscuros septados en el interior de las raíces de todas las especies estudiadas.
