ABSTRACT

Were used twelve (12) adult anteaters (Myrmecophaga tridactyla), adults, 6 (six) males and 6 (six) females, weighing from 20 to 27.32kg from free life. The thoracic cavity was opened until visualization of the whole heart and lungs and later injection of the coronary vessels. The right coronary artery emerged through a single coronary ostium of the aorta, 50%, emitting the intermediate branch and the subsinuous interventricular branch, had a path directed to the subsurface interventricular groove. In the other 50%, the right coronary artery was not present, showing only its branches, intermediate branch and subsurface interventricular branch with emergence of the aorta. Left coronary artery presented, in 83.33%, origin from the aorta in single ostium, issuing the circumflex and interventricular paraconal branches. In 16.66%, the left coronary artery was not evidenced originating from the aorta, but its branches, circumflex and interventricular paraconal.(AU)


Foram utilizados 12 tamanduás-bandeira (Myrmecophaga tridactyla), adultos, sendo seis machos e seis fêmeas, pesando de 20 a 27,32kg, provenientes de vida livre. Foi realizada abertura da cavidade torácica até a visualização do conjunto coração e pulmões e, posteriormente, injeção dos vasos coronários. A artéria coronária direita emergia, 50% dela, por um único óstio coronário da aorta, emitindo o ramo intermédio e o ramo interventricular subsinuoso; tinha trajeto direcionado ao sulco interventricular subsinuoso. Nos outros 50%, a artéria coronária direita não estava presente, exibindo somente seus ramos, ramo intermédio e ramo interventricular subsinuoso com emergência da aorta. A artéria coronária esquerda apresentou, em 83,33%, origem a partir da aorta em óstio único, emitindo os ramos circunflexo e interventricular paraconal. Em 16,66%, a artéria coronária esquerda não foi evidenciada originando-se da aorta, mas, sim, de seus ramos, circunflexo e interventricular paraconal.(AU)

ABSTRACT

Osteosarcoma (OSA) is the most common malignant bone cancer in children and dogs. The therapeutic protocols adopted for dogs and humans are very similar, involving surgical options such as amputation. Besides surgical options, radiotherapy and chemotherapy also are adopted. However, hematologic, gastrointestinal and renal toxicity may occur because of chemotherapy treatments. Recent study clearly showed that mesenchymal stem cells (MSCs) combined with recombinant human bone morphogenetic protein (rhBMP-2) may be associated with decreases of the tumorigenic potential of canine OSA. The aim of this study was to analyse the efficacy of chemotherapy with carboplatin and rhBMP-2 with MSCs in a canine OSA in vivo model. Canine OSA cells were implanted in mice Balb-c/nude with MSCs, rhBMP-2 and carboplatin. Flow cytometry and PCR for markers involved in tumour suppression pathways were analysed. Results showed that the combination of MSCs and rhBMP-2 reduced tumour mass and infiltration of neoplastic cells in tissues more efficiently than carboplatin alone. Thus it was demonstrated that the use of rhBMP-2 and MSCs, in combination with conventional antineoplastic, may be an efficient treatment strategy.

ABSTRACT

O presente trabalho utilizou oito pulmões de tamanduá-bandeira (Myrmecophaga tridactyla), jovens e adultos, de ambos os sexos, provenientes do Setor de Atendimento Clínico Cirúrgico de Animais Selvagens (SACCAS), do Hospital Veterinário "Dr. Halim Atique"( Unirp, localizado na cidade de São José do Rio Preto, estado de São Paulo. Os animais foram encaminhados ao Laboratório de Anatomia Veterinária, onde se procedeu à separação dos órgãos, à lavagem em água corrente e à retirada do excesso de sangue, para serem fixados em formol a 10%. Após a fixação dos pulmões, estes foram dissecados para identificar os segmentos. Os animais apresentaram lobação pulmonar à direita em cranial, médio, caudal e acessório, sendo o lobo cranial dividido em partes cranial e caudal. A lobação pulmonar à esquerda apresentou cranial e caudal em 100% dos animais. Por sua vez, o lobo cranial, em 87,5%, dividiu-se em partes cranial e caudal, e em 12,5% não houve divisão. O pulmão direito apresentou maior tamanho do que o pulmão esquerdo. A árvore brônquica apresentou os brônquios principais direito e esquerdo, os quais se ramificam em brônquios lobares, que dão origem aos segmentos bronquiopulmonares. Em relação à segmentação bronquiopulmonar, foram observadas, à direita, de 14 (12,5%) a 18 segmentos (25%) e, à esquerda, de 10 (25%) a 30 segmentos (12,5%).(AU)


The study used eight lungs of giant anteaters (Myrmecophaga tridactyla), youth and adults of both sexes, from the Clinical Surgical Care Industry Wildlife (SACCAS), Veterinary Hospital "Dr. Halim Atique "- Unirp, located in the city of São José do Rio Preto, São Paulo. The animals were sent to the Veterinary Anatomy Laboratory, where organs were separated and washed under running waterto remove excess blood, then fixed in 10% formalin. After fixation, lungs were dissected to identify the segments. The animals have pulmonary lobations right cranial, middle, caudal and accessory; and the cranial lobe divided into cranial and caudal parts. Pulmonary lobation in the left cranial and caudal form were present in 100% of animals. The cranial lobe was evident in 87.5%, divided into cranial and caudal part, and 12.5% with no division. The right lung was shown to be larger than the left lung. Bronchial tree presented the main right and left bronchial tubes that branch into lobar bronchi that give rise to bronquiopulmonares segments. In relation to bronquiopulmonar targeting, 14 (12.5%) to 18 right segments (25%) and 10 (25%) to 30 (12.5%) left segments.(AU)

ABSTRACT

During a research visit for tissue collection at an abattoir located in Pelotas, Brazil, one female genital tract showed both enlarged oviducts. The reproductive tract was collected and analyzed. Occluded uterine tubes and an increase in the organ volume due to the large amount of fluid in the organ lumen were the macroscopic findings. Three samples, corresponding to isthmus, ampulla and infundibulum from each uterine tube and one sample from the endometrium were collected. Samples were fixed in Bouin's solution and processed in light microscopy. Microscopically a decrease in the number of folds and also an increase in the lumen of the organ were observed, mainly in the ampulla and infundibulum. The epithelial lining of the uterine tubes ranged from ciliated to simple squamous. Inflammatory cells were observed between the epithelial cells and in the lamina propria. Hydrosalpinx is difficult to diagnose and can be a cause of infertility in the mare.


Durante uma visita de pesquisa para coleta de tecido em um abatedouro localizado na cidade de Pelotas, Brasil, foi observado um trato genital com aumento do tamanho dos ovidutos. O trato reprodutivo foi coletado e analisado. Os achados macroscópicos observados foram tubas uterinas ocluídas e com aumento do volume do órgão devido à grande quantidade de líquido na luz do órgão. Três amostras, correspondendo a istmo, ampola e infundíbulo, e uma amostra do endométrio foram coletadas. As amostras foram fixadas em solução de Bouin e processadas para microscopia de luz. Microscopicamente foi observada uma diminuição no número de pregas e um aumento do lúmen do órgão, principalmente na ampola e no infundíbulo. O epitélio de revestimento das tubas uterinas variou de cilíndrico ciliado a pavimentoso simples. Células inflamatórias foram observadas entre as células epiteliais e na lâmina própria das tubas uterinas. A hidrossalpinge é difícil de ser diagnosticada e pode ser uma causa de infertilidade na égua.

WHAT IS KNOWN AND OBJECTIVE: Use of cisplatin can induce type I hypersensitivity reactions that may also be linked to the quality of the drug utilized. We observed cases of hypersensitivity that appeared to be associated with the brand of cisplatin used. The aim of this study was to compare two different brands of cisplatin in relation to type I hypersensitivity reactions. METHODS: Brand A was used in a tertiary care teaching hospital until 2012, and use of brand B started from January 2013, when the first hypersensitivity cases were observed. Patients were categorized based on symptom. Cisplatin of both brands was analysed by high-performance liquid chromatography (HPLC) and high-resolution electrospray ionization mass spectrometry (ESI-(+)-MS) and characterized according to US Pharmacopeia. RESULTS AND DISCUSSION: There were no cases of hypersensitivity associated with the use of cisplatin brand A, whereas four of 127 outpatients that used cisplatin brand B were affected. The two brands were in accordance with the US Pharmacopeia parameters, and there was no significant difference in the total platinum levels between the two brands when analysed by HPLC. However, high-resolution ESI-(+)-MS analyses show that brand B contains approximately 2.7 times more hydrolysed cisplatin than brand A. WHAT IS NEW AND CONCLUSION: The increase in the hydrolysed form of cisplatin found in brand B may be the cause of the hypersensitivity reaction observed in a subset of patients. We present the first study of the quality of drugs by high-resolution ESI-(+)-MS. Drug regulatory agencies and manufacturers should consider including measurement of hydrolysed cisplatin as a quality criterion for cisplatin formulations.

ABSTRACT

Benzaldehyde semicarbazone (BS) has presented positive results in several pharmacological models, including anticonvulsivant and anti-inflammatory models. The present study evaluated the preclinical toxicity (acute and subchronic), as well as the toxicokinetic and gastroprotective effects of BS against ethanol lesions. Oral doses of 300 and 2000mg/kg were used in the preclinical acute toxicity study; 100, 200, and 300mg/kg were used in both the subchronic toxicity evaluation and the gastric study; and 300mg/kg was used in the toxicokinetic study. No impact from the dose of 300mg/kg could be identified; while, one animal died at 2000mg/kg in the acute toxicity test. In the subchronic toxicity test, changes in the biochemical parameters of the liver, as well as in the histopatological evaluation, demonstrated that BS is a hepatotoxic drug. BS proved to be effective for moderate and severe gastric lesions. In the toxicokinetics study, BS presented a low concentration and rapid plasma disappearance. Several results also indicate that BS is likely to be mostly eliminated from the liver and may well undergo a first-pass effect after oral absorption. It was impossible to estimate the noobserved-adverse-effect-levels (NOAEL) and lowest-observed-adverse-effect-levels (LOAEL) due to the presence of hepatotoxicity in all tested doses.

ABSTRACT

Information on gene alterations associated to poorly differentiated (PDTC) and anaplastic thyroid carcinomas (ATC) is scarce. Using human cancer cell lines as a tool for gene discovery, we performed a cytogenetic and oligo-array analysis in five new cell lines derived from two PDTC and three ATC. In PDTC we evidenced, as important, the involvement of the MAPK/ERK kinase pathway, and downregulation of a group of suppressor genes that include E-cadherin. In ATC, downregulation of a specific group of oncosuppressor genes was also observed. Our ATC cell lines presented chromosomal markers of gene amplification, and we were able to identify for the first time the nature of the involved amplicon target genes. We found that the main molecular differences between the two cell line types were related to signal transduction pathways, cell adhesion and motility process. TaqMan experiments performed for five amplicon target genes and for two genes, which allowed a clear distinction between ATC and PDTC: CDH13 and PLAU corroborated array results, not only in the cell lines, but also in an additional set of primary 14 PDTC and three ATC. We suggest that our findings may represent new tools for the development of more effective therapies to the hitherto untreatable ATC.

The hydro-alcoholic extract of the aerial parts of Lithraea molleoides, given orally at a dose of 1000 mg/kg, showed significant anti-ulcerogenic activity on ulcer induced by indomethacin and absolute alcohol in rats.

ABSTRACT

The biochemical characterization of a new lectin (Hypnea cervicornis agglutinin or HCA) isolated from the Brazilian red alga H. cervicornis is reported. The haemagglutinating activity of the lectin was only inhibited by the glycoprotein porcine stomach mucin at a minimum inhibitory concentration of 19 microg x mL(-1). No haemagglutination inhibition was detected after the addition of simple sugars. The MALDI-TOF molecular masses of native and reduced and carbamidomethylated HCA were, respectively, 9196.6 Da and 9988.2 Da, indicating that the primary structure of the protein is crosslinked by 7 disulfide bonds. This unusual structural feature among lectins, along with its N-terminal sequence and amino-acid composition, clearly shows that HCA belongs to a protein family distinct from the isolectins Hypnin A1 and A2 isolated from the related Japanese alga Hypnea japonica. On the other hand, HCA displayed a high degree of similarity to the agglutinin from the Brazilian species Hypnea musciformis. Our data indicate the occurrence of structural diversity among lectins of closely related species living in distant ecosystems, i.e., the Pacific coast of Japan and the Atlantic coast of Brazil, and support the hypothesis that the lectin content (lectinome) might serve as a biomarker for taxonomical purposes.

ABSTRACT

Cordiaverbenacea D.C. (Boraginaceae) is found along the coastal regions of Brazil, from the Amazon basin to the southernmost state of Rio Grande do Sul. The aerial parts of this plant have been used in folk medicine since colonial times for their anti-rheumatic, anti-inflammatory, analgesic and healing properties. The objectives of this study were: the extraction of the essential oil of the aerial parts of this species and quantification of the yield thereof; the identification and quantification of the principal components of the essential oil; and the determination of its antimicrobial activity by plate diffusion method. The yield of essential oil was 0.19% (v/w) in relation to the fresh flowering aerial parts and 0.23% (v/w) of the leaves. The main constituents of the essential oil, identified by gas chromatography/mass spectrometry (GC/MS), were alpha-pinene (29.69%), trans-caryophyllene (25.27%) and aloaromadendrene (9.99%). The Gram-positive bacteria and yeast tested were sensitive to the essential oil of Cordiaverbenacea D.C., but most of the Gram-negative bacteria were resistant.

ABSTRACT

The placental vasculature of five hystricomorph rodents was examined by latex injection of the blood vessels, immunohistochemistry and scanning electron microscopy of vessel casts. The pattern of branching of the vessels is described at the level of fine structure. The placenta is divided into lobes separated by interlobular trophoblast. Fetal arteries course through the interlobular areas and give rise to capillaries from which blood drains into veins at the centre of the lobes. Maternal blood reaches the placenta through spiral arteries that pass around the perimeter of the subplacenta. They supply large maternal blood sinuses, lined by trophoblast, which run through the interlobular areas and into the centre of the lobes. Here they supply fine channels that run parallel to the fetal capillaries, so that maternal blood flows from the centre of the lobe to the periphery. This arrangement provides the morphological basis for countercurrent exchange. The maternal channels of the labyrinth drain into spaces formed by the latticework of the interlobular trophoblast and thence through venous lacunae to a basal venous lacunar ring. The subplacenta is supplied by a single fetal artery. The vessels within the subplacenta pursue a tortuous course with dilatations and constrictions as in an endocrine gland.

ABSTRACT

The genetic alterations that underlie the progression of follicular thyroid carcinoma towards anaplasia are still largely uncharacterised. We compared the Comparative Genomic Hybridization (CGH) profiles of 20 follicular (FTCs), 12 poorly differentiated (PDTCs) and seven anaplastic thyroid carcinomas (ATCs), in order to identify the chromosomal imbalances potentially associated with cancer progression. We found: (i) when considering that a 'direct' transformation of FTC towards anaplasia occurs, the defined significantly important alterations were the increase of gains at 3q (P<0.05) and 20q (P<0.01), and the increase of losses at 7q (P<0.05) and Xp (P<0.01); (ii) regarding poorly differentiated carcinomas as an intermediate independent entity in the anaplastic transformation of follicular cancers, evidenced as important alterations towards anaplasia, were the proportional decrease in copy sequences at 7p, 7q, 12q and 13q resulting from the significant decrease of DNA gains at 7p and 12q (P<0.05), and the significant increase of losses at 7q and 13q (P<0.05). These results unveil the chromosomal regions where genes of interest in thyroid anaplastic transformation are to be located, and demonstrate that different gene dosage copy sequence imbalances are associated to the 'direct' pathway of transformation of follicular into anaplastic cancers and to the progressive FTC --> PDTC --> ATC pathway.

Sementes de três espécies de Cordia L., da família Boraginaceae, coletadas em diferentes cidades do Estado de São Paulo foram analisadas quanto ao teor de lipídeos e composição de ácidos graxos, com ênfase na presença do ácido gama-linolênico, um ácido graxo de grande interesse terapêutico. Os lipídeos totais foram fracionados em classes (lipídeos neutros, fosfolipídeos e glicolipídeos) através de cromatografia em coluna aberta de gel de sílica e a composição de ácidos graxos foi determinada por cromatografia a gás. As concentrações de lipídeos totais apresentaram grande variação entre as amostras (25,84 a 62,81 por cento), sendo que a classe lipídica dominante foi de lipídeos neutros (média de 88,9 por cento dos lipídeos totais). Os ácidos insaturados representaram 75 por cento da composição total dos ácidos graxos, sendo o principal representante o ácido oléico. Verificou-se a presença do ácido gama-linolênico em todas as amostras analisadas, em concentrações que variaram de 0,63 a 2,54 por cento, valores superiores àqueles relatados pela literatura para outras espécies do gênero Cordia L.


Seeds of three Cordia L. species (Boraginaceae), collected in different cities of the State of São Paulo were analysed in relation to the lipid content and fatty acid composition, with emphasis on the presence of gamma-linolenic acid, a fatty acid with therapeutical interest. The total lipids were separated into classes (neutral lipids, phospholipids and glucolipids) by open column chromatography with silica gel and the fatty acid composition was determined by gas chromatography. Although the levels of total lipids varied considerably between samples (25.84 to 62.81 percent), the dominant class was that of neutral lipids (88.9 percent of the total lipids). The insaturated acids represented 75 percent of the total composition of fatty acids, with oleic acid being the main representative. The presence of gamma-linolenic acid was ascertained in all the samples analysed, in concentrations from 0.63 to 2.54 percent, higher values than those related in literature for other Cordia.
