ABSTRACT

Long transportation times remain a challenge to the satisfactory diagnosis of Campylobacter fetus subsp. venerealis (Cfv). Here we demonstrated that samples of frozen bovine preputial mucus maintained at -20 °C for 10 days can be used as an alternative source for molecular diagnosis of Cfv. This approach will improve the analysis of this bacterium.

ABSTRACT

This paper reports the abortion of a male Aberdeen Angus bovine by a vaccine strain of Bacillus anthracis, describing the pathological and microbiological findings and the genome sequence. Necropsy findings included multifocal areas of hemorrhage in different organs. Histologically, various organs showed hemorrhage, fibrin exudation, necrosis associated with countless bacillary bacterial clumps and severe neutrophilic inflammatory infiltrate. In the microbiological examination, numerous rough, nonhemolytic, gray and dry colonies with irregular edges were isolated from liver, lung and abomasum content samples. Gram staining revealed square-ended Gram-positive rods arranged in chains. B. anthracis identification was confirmed by detection of the molecular chromosomal marker Ba813. The genomes from the isolated B. anthracis (named SPV842_15) and from the isolated vaccinal strain (Brazilian vaccinal strain), which was recovered from a commercial vaccine used in the pregnant cow, were sequenced. Genomic comparisons displayed a high level of nucleotide identity in the comparisons between B. anthracis SPV842_15 and the B. anthracis Brazilian vaccinal strain (98,2%). Furthermore, in both strains, only the plasmid pX01 sequence was detected. Although, vaccination against anthrax is characterized by an elevated protective profile and very low residual virulence, immunization with Sterne strains can cause abortion in cattle, presumably by the plasmid pX01 toxins in rare or special situations.(AU)


Este trabalho relata um aborto de um bovino, macho, Aberdeen Angus, por uma cepa vacinal de Bacillus anthracis, descreve os achados patológicos, microbiológicos e o sequenciamento do genoma. Os achados de necropsia incluíram áreas multifocais de hemorragias em diferentes órgãos. Histologicamente, órgãos afetados apresentaram hemorragia, exsudação de fibrina, necrose associada a miríades bacterianas bacilares e intenso infiltrado inflamatório neutrofílico. No exame microbiológico, foram isoladas numerosas colônias rugosas, não hemolíticas, cinzas e secas, com bordas irregulares a partir de amostras de fígado, pulmão e conteúdo do abomaso. A coloração de Gram revelou bastonetes Gram-positivos dispostos em cadeias. A identificação do B. anthracis foi confirmada pela detecção do marcador cromossômico molecular Ba813. Os genomas do isolado B. anthracis (SPV842_15) e do isolado vacinal (cepa vacinal brasileira), recuperado de uma vacina comercial utilizada na vaca prenhe, foram sequenciados. Comparações genômicas mostraram um elevado nível de identidade de nucleotídeos entre B. anthracis SPV842_15 e cepa vacinal brasileira (98,2%). Além disso, em ambas as estirpes foi detectada apenas a sequência do plasmídeo pX01. Embora a vacinação contra o antraz seja caracterizada por um perfil protetor elevado e uma virulência residual muito baixa, a imunização com estirpes de Sterne pode causar aborto em bovinos, presumivelmente pelas toxinas do plasmídeo pX01 em situações raras ou específicas.(AU)

A colângio-hepatite é considerada uma causa frequente de insuficiência hepática em gatos e é classificada em neutrofílica, linfocítica e esclerosante. Os objetivos deste estudo foram determinar a frequência de colângio-hepatite em gatos diagnosticados na Região Metropolitana de Porto Alegre, descrever seus aspectos anatomopatológicos e estabelecer uma associação com as infecções por Escherichia coli, vírus da imunodeficiência felina (FIV) e vírus da leucemia felina (FeLV). No período de janeiro de 2000 a julho de 2016 o Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul realizou 1915 necropsias de gatos, destes, 32 foram diagnosticados com colângio-hepatite, representando 1,7% dos casos. Destes, a colângio-hepatite linfocítica (CHL) foi diagnosticada em 68,7% (22/32), a neutrofílica (CHN) em 21,9% (7/32) e a esclerosante (CHE) com 9,4% (3/32). A idade variou de quatro meses a 16 anos, com a mediana de seis anos, acometendo predominantemente gatos sem raça definida. Somente na CHN observou-se predisposição por machos, verificado em 85,7% (6/7) dos casos. Enterite e pancreatite foram identificadas concomitantemente com a colângio-hepatite em 56,2% (18/32) dos casos, cada, e a formação de tríade foi identificada em 46,9% (15/32) dos gatos. Através da imuno-histoquímica, 68,2% (15/22) dos gatos com CHL, foram positivos para FIV, 40,9% (9/22) para FeLV e 31,8% (7/22) marcação para ambos os retrovírus. Na CHN, 85,7% (6/7) positivos para FIV, 57,1% (4/7) para FeLV e 42,8% (3/7) imunorreação para os dois retrovírus. Na CHE, 100% (3/3) dos casos apresentaram marcação para FeLV, 33,3% (1/3) para FIV e 33,3% (1/3) para ambos. Imunomarcação para E. coli foi observada em 27,3% (6/22) dos casos da CHL, 28,6% (2/7) da CHN e em 33,3% (1/3) da CHE. The aims of this study was to determine the frequency of cholangiohepatitis in cats diagnosed in the metropolitan region of Porto Alegre, to describe the anatomopathological aspects and to establish an association with Escherichia coli, feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV). From January 2000 to July 2016, the Department of Veterinary Pathology of the Universidade Federal do Rio Grande do Sul performed 1915 cat necropsies, of which 32 were diagnosed with cholangiohepatitis, representing 1.7% of the cases. Of these, lymphocytic cholangiohepatitis (LCH) was diagnosed in 68.7% (22/32), neutrophilic (NCH) in 21.9% (7/v) and sclerosing cholangiohepatitis (SCH) with 9.4% (3/32) of the cases. In general, age ranged from four months to 16 years, with the median age of six years, and predominantly affected cats with mixed breed. Only in NCH demonstrated male predisposition, verified in 85.7% (6/7) of this cases. In NCH, 85.7% (6/7) was positive for FIV, 57.1% (4/7) for FeLV, and 42.8% (3/7) immunoreactions for the two retroviruses. ABSTRACT

Descrevem-se os aspectos epidemiológicos, clínicos, patológicos e imuno-histoquímicos de um surto de parvovirose cardíaca em filhotes de cães. O surto ocorreu em um canil localizado na cidade de Parnamirim, Rio Grande do Norte, região Nordeste do Brasil. De uma ninhada de nove filhotes, um foi natimorto e seis morreram entre 35-57 dias de idade após apresentarem sinais clínicos cardiorrespiratórios com evolução de 10 minutos a três dias. Dos seis filhotes que morreram, dois foram encaminhados para necropsia. No exame macroscópico, ambos os animais apresentaram discreta efusão pericárdica, coração marcadamente globoso, difusa palidez nas superfícies epicárdica e miocárdica e dilatação da cavidade ventricular esquerda. Nos pulmões, observaram-se áreas multifocais avermelhadas na superfície pleural e ao corte fluía líquido espumoso e levemente avermelhado. O fígado estava difusamente aumentado de tamanho, com acentuação do padrão lobular e com áreas pálidas entremeadas por áreas escuras que, ao corte, se aprofundavam ao parênquima. Microscopicamente observou-se miocardite linfohistiocítica, necrosante, associada a fibrose intersticial e corpúsculos de inclusões virais basofílicos intranucleares em cardiomiócitos. Nos pulmões observou-se pneumonia intersticial e edema, e no fígado notou-se degeneração e necrose centrolobular a mediozonal associada à congestão e hemorragia. O diagnóstico foi confirmado por imuno-histoquímica. A forma miocárdica da parvovirose canina pode ocorrer ocasionalmente em filhotes de cadelas que não foram efetivamente vacinadas. Essa forma clínica da doença caracteriza-se por alterações cardiorrespiratórias e morte hiperaguda ou aguda dos animais afetados.(AU)


We describe the epidemiological, clinical, pathological and immunohistochemical aspects of parvoviral myocarditis outbreak in puppies. The outbreak occurred in a kennel located in Parnamirim, Rio Grande do Norte, Northeastern of Brazil. In a litter of nine pups, one was stillbirth and six died between 35-57 days of age after cardiopulmonary clinical signs with evolution of 10 minutes to three days. Of the six puppies that died, two were sent for necropsy. On gross examination, both animals had discreet pericardial effusion, markedly distended heart, diffuse pallor in epicardial and myocardial surfaces and dilation of the left ventricular cavity. The lungs were observed multifocal reddish areas in the pleural surface and at cutting flowed foamed and slightly red liquid. The liver was diffusely increased in size, with lobular standard accentuation and pale areas interspersed with dark areas wich deepened in the parenchyma. Microscopically observed linfohistiocítica myocarditis, necrotizing, associated with interstitial fibrosis and basophilic intranuclear viral inclusions corpuscles in cardiomyocytes. In the lungs there were edema and interstitial pneumonia and in the liver was noted centrilobular to mediozonal degeneration and necrosis associated with congestion and hemorrhage. The diagnosis was confirmed by immunohistochemistry. The parvoviral myocarditis can occasionally occur in puppies of bitches that have not been effectively vaccinated. This clinical form of the disease characterized by cardiorespiratory changes and hyperacute or acute death of the affected animals.(AU)

ABSTRACT

Enterotoxigenic Escherichia coli (ETEC) causes diarrhea in pigs, referred to as colibacillosis. The aim of this study was to optimize multiplex polymerase chain reaction (PCR) and immunohistochemistry (IHC) analyses of paraffin-embedded material to detect pathogenic E. coli strains causing colibacillosis in pigs. Multiplex PCR was optimized for fimbriae (F18, F4, F6, F5, and F41) and toxins (types A and B heat-stable toxins [STaP and STb], heat-labile toxin [LT], and type 2 Shiga toxin [STx2e]), and IHC was optimized for an anti-E. coli polyclonal antibody. Samples (132) from pigs received between 2006 and 2014 with clinical and histopathological diagnoses of colibacillosis were analyzed. E. coli was detected by IHC in 78.7%, and at least one virulence factor gene was detected in 71.2%. Pathogenic strains of ETEC with at least one fimbria and one toxin were detected in 40% of the samples in multiplex PCR. The most frequent virulence types were F18-STaP (7.5%), F18-STaP-STb (5.7%), and F4-STaP (3.8%). A statistically significant association was noted between virulence factors F4, F18, STaP, and STb and positive immunostaining results. Colibacillosis diagnosis through multiplex PCR and IHC of paraffin-embedded tissues is a practical approach, as samples can be fixed and stored for long periods before analysis.

The lungs were observed multifocal reddish areas in the pleural surface and at cutting flowed foamed and slightly red liquid. The liver was diffusely increased in size, with lobular standard accentuation and pale areas interspersed with dark areas wich deepened in the parenchyma. Microscopically observed linfohistiocítica myocarditis, necrotizing, associated with interstitial fibrosis and basophilic intranuclear viral inclusions corpuscles in cardiomyocytes. In the lungs there were edema and interstitial pneumonia and in the liver was noted centrilobular to mediozonal degeneration and necrosis associated with congestion and hemorrhage. The diagnosis was confirmed by immunohistochemistry. The parvoviral myocarditis can occasionally occur in puppies of bitches that have not been effectively vaccinated. This clinical form of the disease characterized by cardiorespiratory changes and hyperacute or acute death of the affected animals.


RESUMO: Descrevem-se os aspectos epidemiológicos, clínicos, patológicos e imuno-histoquímicos de um surto de parvovirose cardíaca em filhotes de cães. O surto ocorreu em um canil localizado na cidade de Parnamirim, Rio Grande do Norte, região Nordeste do Brasil. De uma ninhada de nove filhotes, um foi natimorto e seis morreram entre 35-57 dias de idade após apresentarem sinais clínicos cardiorrespiratórios com evolução de 10 minutos a três dias. Dos seis filhotes que morreram, dois foram encaminhados para necropsia. No exame macroscópico, ambos os animais apresentaram discreta efusão pericárdica, coração marcadamente globoso, difusa palidez nas superfícies epicárdica e miocárdica e dilatação da cavidade ventricular esquerda. Nos pulmões, observaram-se áreas multifocais avermelhadas na superfície pleural e ao corte fluía líquido espumoso e levemente avermelhado. O fígado estava difusamente aumentado de tamanho, com acentuação do padrão lobular e com áreas pálidas entremeadas por áreas escuras que, ao corte, se aprofundavam ao parênquima. Microscopicamente observou-se miocardite linfohistiocítica, necrosante, associada a fibrose intersticial e corpúsculos de inclusões virais basofílicos intranucleares em cardiomiócitos. Nos pulmões observou-se pneumonia intersticial e edema, e no fígado notou-se degeneração e necrose centrolobular a mediozonal associada à congestão e hemorragia. O diagnóstico foi confirmado por imuno-histoquímica. A forma miocárdica da parvovirose canina pode ocorrer ocasionalmente em filhotes de cadelas que não foram efetivamente vacinadas. Essa forma clínica da doença caracteriza-se por alterações cardiorrespiratórias e morte hiperaguda ou aguda dos animais afetados.

ABSTRACT: Cholangiohepatitis is considered a frequent cause of liver failure in cats, and are classified as neutrophilic, lymphocytic and sclerosing. The aims of this study was to determine the frequency of cholangiohepatitis in cats diagnosed in the metropolitan region of Porto Alegre, to describe the anatomopathological aspects and to establish an association with Escherichia coli, feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV). From January 2000 to July 2016, the Department of Veterinary Pathology of the Universidade Federal do Rio Grande do Sul performed 1915 cat necropsies, of which 32 were diagnosed with cholangiohepatitis, representing 1.7% of the cases. Of these, lymphocytic cholangiohepatitis (LCH) was diagnosed in 68.7% (22/32), neutrophilic (NCH) in 21.9% (7/v) and sclerosing cholangiohepatitis (SCH) with 9.4% (3/32) of the cases. In general, age ranged from four months to 16 years, with the median age of six years, and predominantly affected cats with mixed breed. Only in NCH demonstrated male predisposition, verified in 85.7% (6/7) of this cases. Enteritis and pancreatitis were identified concomitantly with cholangiohepatitis in 56.2% (18/32) cases, each, and triad formation was identified in 46.9% (15/32) of cats. In the immunohistochemistry, we observed that 68.2% (15/22) of the cats with LCH were positive for FIV, 40.9% (9/22) for FeLV, and 31.8% (7/22) positive for both retroviruses. In NCH, 85.7% (6/7) was positive for FIV, 57.1% (4/7) for FeLV, and 42.8% (3/7) immunoreactions for the two retroviruses. In SCH, 100% (3/3) of the cases presented FeLV marking, 33.3% (1/3) for FIV and 33.3% (1/3) for both. Immunostaining for E. coli was observed in 27.3% (6/22) of LCH, 28.6% (2/7) of NCH, and 33.3% (1/3) of SCH. E. coli, Enterococcus sp. and Klebsiella pneumoniae were the most frequently microorganisms isolated in the bacteriological examination. The visualization of E. coli by the immunohistochemistry in the hepatobiliary system of cats diagnosed with cholangiohepatitis suggests that the disease developed secondary to ascending bacterial infection.


RESUMO: A colângio-hepatite é considerada uma causa frequente de insuficiência hepática em gatos e é classificada em neutrofílica, linfocítica e esclerosante. Os objetivos deste estudo foram determinar a frequência de colângio-hepatite em gatos diagnosticados na Região Metropolitana de Porto Alegre, descrever seus aspectos anatomopatológicos e estabelecer uma associação com as infecções por Escherichia coli, vírus da imunodeficiência felina (FIV) e vírus da leucemia felina (FeLV). No período de janeiro de 2000 a julho de 2016 o Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul realizou 1915 necropsias de gatos, destes, 32 foram diagnosticados com colângio-hepatite, representando 1,7% dos casos. Destes, a colângio-hepatite linfocítica (CHL) foi diagnosticada em 68,7% (22/32), a neutrofílica (CHN) em 21,9% (7/32) e a esclerosante (CHE) com 9,4% (3/32). A idade variou de quatro meses a 16 anos, com a mediana de seis anos, acometendo predominantemente gatos sem raça definida. Somente na CHN observou-se predisposição por machos, verificado em 85,7% (6/7) dos casos. Enterite e pancreatite foram identificadas concomitantemente com a colângio-hepatite em 56,2% (18/32) dos casos, cada, e a formação de tríade foi identificada em 46,9% (15/32) dos gatos. Através da imuno-histoquímica, 68,2% (15/22) dos gatos com CHL, foram positivos para FIV, 40,9% (9/22) para FeLV e 31,8% (7/22) marcação para ambos os retrovírus. Na CHN, 85,7% (6/7) positivos para FIV, 57,1% (4/7) para FeLV e 42,8% (3/7) imunorreação para os dois retrovírus. Na CHE, 100% (3/3) dos casos apresentaram marcação para FeLV, 33,3% (1/3) para FIV e 33,3% (1/3) para ambos. Imunomarcação para E. coli foi observada em 27,3% (6/22) dos casos da CHL, 28,6% (2/7) da CHN e em 33,3% (1/3) da CHE. E. coli, Enterococcus sp. e Klebsiella pneumoniae foram os micro-organismos mais frequentes isolados no exame bacteriológico. A visualização da E. coli, através da IHQ no sistema hepatobiliar de gatos diagnosticados com colângio-hepatite associados à inflamação, sugere que a doença se desenvolveu secundariamente à infecção bacteriana ascendente.

Este trabalho descreve os achados anatomopatológicos de doenças fúngicas em gatos no período de janeiro de 2005 a dezembro de 2015 pelo SPV-UFRGS. Foram revisados 1.274 protocolos de necropsia e encontrou-se 17 casos de micoses, correspondendo a 1,33% dos diagnósticos. Quanto às amostras provenientes de biopsias, foram verificados 2.615 protocolos e detectou-se 59 casos de doenças fúngicas, totalizando 2,25% dos diagnósticos, sendo revisados seus achados macroscópicos, microscópicos, colorações especiais e cultivo micológico. O principal diagnóstico encontrado foi esporotricose com 34 casos de biópsia e cinco de necropsia. Criptococose obteve seis diagnósticos na necropsia e 13 na biópsia. O pseudomicetoma dermatofítico possui cinco casos de biópsia e dois de necropsia. Histoplasmose obteve um diagnóstico na necropsia e dois na biópsia. Aspergilose teve dois casos diagnosticados na necropsia. Candidíase e feo-hifomicose obtiveram um caso cada. Em quatro ocasiões, não se pode identificar o agente envolvido na lesão e foram classificados conforme a alteração morfológica em: dermatite fúngica, enterite fúngica, rinite micótica e dermatite e osteomielite fúngica. Através deste trabalho, pode-se identificar que a esporotricose foi o diagnóstico de doença fúngica mais frequente em felinos, seguido da criptococose e pseudomicetoma dermatofítico.(AU)

In four occasions, it was not possible to identify the agent involved and the diagnosis was made regarding the morphological changes: fungal dermatitis, fungal enteritis, mycotic rhinitis, and fungal dermatitis and osteomyelitis. In this study, sporotrichosis could be identified as the most frequent diagnosis of fungal disease in cats, followed by cryptococcosis and dermatophytic pseudomycetoma.(AU)

ABSTRACT

A case of ulcerative dermatitis caused by feline herpesvirus type 1 (FeHV-1) in an adult male domestic shorthair cat is reported. The cat was rescued from the streets and presented with ulcerative lesions at the nasal planum and tongue in addition to a history of occasional sneezing. Thirty days after of the first clinical evaluation, the cat died as a result of acute myeloid leukemia. During necropsy, ulcerative lesions were found on the superior lip, the skin of the nasal planum, and at the periorbital region. Ulcerations were also noted on the tongue and hard palate. Histological examination revealed extensive epidermal necrosis, which involved the subjacent dermis and adnexal structures; the inflammatory infiltrate consisted of neutrophils, mast cells, and lymphocytes. Amphophilic intranuclear inclusion bodies were occasionally observed in intact epithelial cells. In the immunohistochemical evaluation, positive intracytoplasmic immunolabeling was detected in the sebaceous and follicular epithelial cells as well as in the bronchiolar epithelial cells. Samples of lymphoid tissue tested positive for the presence of feline leukemia virus and feline immunodeficiency virus by immunohistochemistry. Pulmonary tissue fragments were immunolabeled for feline calicivirus. Samples obtained from a cutaneous lesion were subjected to virus isolation in a cellular culture, which revealed the cytopathic effects characteristic of herpesvirus. FeHV-1 was detected in the samples by polymerase chain reaction.(AU)


Descreve-se um caso de dermatite ulcerativa causada por herpesvírus felino tipo 1 (FeHV-1), em um gato adulto, macho, sem raça definida. O gato foi resgatado da rua e apresentava uma lesão ulcerativa no plano nasal e língua, além de espirros esporádicos. Trinta dias após o primeiro atendimento, o gato morreu por leucemia mieloide aguda. Na necropsia, o lábio superior e a pele do plano nasal e periorbital apresentaram extensa lesão ulcerativa, além de ulcerações na língua e no palato duro. Histologicamente havia extensa necrose da epiderme, estendendo-se à derme subjacente e estruturas anexas, associada ao infiltrado inflamatório, constituído por neutrófilos, mastócitos e linfócitos. Observaram-se ainda, ocasionalmente, em células epiteliais intactas, corpúsculos de inclusão intranucleares anfofílicos. Na avaliação imuno-histoquímica anti-FeHV-1 observou-se imunomarcação positiva intracitoplasmática nas células epiteliais e nas células epiteliais bronquiolares. Amostras de tecido linfoide apresentaram imunomarcação para vírus da leucemia felina, vírus da imunodeficiência felina, além de marcação para calicivírus em fragmentos pulmonares. Fragmentos da lesão cutânea foram submetidos a isolamento viral em cultivo celular, onde foi observado efeito citopático característico de herpesvírus e a amostra foi positiva na PCR para FeHV-1.(AU)

ABSTRACT

The objective of the study was to report on a fatal case of feline toxoplasmosis with coinfection with the feline leukemia virus (FeLV). A domestic cat (Felis silvestris catus) presented intense dyspnea and died three days later. In the necropsy, the lungs were firm, without collapse and with many white areas; moderate lymphadenomegaly and splenomegaly were also observed. The histopathological examination showed severe necrotic interstitial bronchopneumonia and mild necrotic hepatitis, associated with intralesional cysts and tachyzoites of Toxoplasma gondii that were positive by anti-T. gondii immunohistochemical (IHC) evaluation. The bone marrow showed chronic myeloid leukemia and the neoplastic cells were positive by anti-FeLV IHC evaluation. DNA extracted from lungs was positive for T. gondii by PCR targeting REP-529. T. gondii was characterized by PCR-RFLP and by the microsatellites technique. ToxoDB-PCR-RFLP #10, i.e. the archetypal type I, was identified. Microsatellite analysis showed that the strain was a variant of type I with two atypical alleles. This was the first time that a T. gondii clonal type I genotype was correlated with a case of acute toxoplasmosis in a host in Brazil.

O objetivo deste estudo foi relatar um caso de toxoplasmose felina fatal com coinfecção com o vírus da leucemia felina (FeLV). Um gato doméstico (Felis silvestris catus) apresentou intensa dispneia e morreu três dias depois. Na necropsia, observaram-se pulmões firmes, não colabados e com múltiplas áreas brancas, além de linfoadenomegalia e esplenomegalia moderadas. No exame histopatológico, evidenciaram-se broncopneumonia intersticial necrótica acentuada e hepatite necrótica discreta associada a cistos e taquizoítas de T. gondii intralesionais positivos na imuno-histoquímica (IHC) anti-T. gondii. Evidenciou-se ainda, na medula óssea, leucemia mieloide crônica com IHC anti-FeLV positiva nas células neoplásicas. O DNA extraído dos pulmões foi positivo para T. gondii por meio da PCR-REP-529. T. gondii foi caracterizado por PCR-RFLP e pela técnica de microssatélites. Foi identificado o genótipo ToxoDB-PCR-RFLP #10, i.e., o arquétipo tipo I. A análise por microssatélites mostrou que a cepa era uma variante do tipo I, com dois alelos atípicos. Esta é a primeira vez que T. gondii clonal tipo I foi relacionado com um caso agudo de toxoplasmosis em um hospedeiro no Brasil.(AU)

ABSTRACT

Abstract The objective of the study was to report on a fatal case of feline toxoplasmosis with coinfection with the feline leukemia virus (FeLV). A domestic cat (Felis silvestris catus) presented intense dyspnea and died three days later. In the necropsy, the lungs were firm, without collapse and with many white areas; moderate lymphadenomegaly and splenomegaly were also observed. The histopathological examination showed severe necrotic interstitial bronchopneumonia and mild necrotic hepatitis, associated with intralesional cysts and tachyzoites of Toxoplasma gondii that were positive by anti-T. gondii immunohistochemical (IHC) evaluation. The bone marrow showed chronic myeloid leukemia and the neoplastic cells were positive by anti-FeLV IHC evaluation. DNA extracted from lungs was positive for T. gondii by PCR targeting REP-529. T. gondii was characterized by PCR-RFLP and by the microsatellites technique. ToxoDB-PCR-RFLP #10, i.e. the archetypal type I, was identified. Microsatellite analysis showed that the strain was a variant of type I with two atypical alleles. This was the first time that a T. gondii clonal type I genotype was correlated with a case of acute toxoplasmosis in a host in Brazil.


Resumo O objetivo deste estudo foi relatar um caso de toxoplasmose felina fatal com coinfecção com o vírus da leucemia felina (FeLV). Um gato doméstico (Felis silvestris catus) apresentou intensa dispneia e morreu três dias depois. Na necropsia, observaram-se pulmões firmes, não colabados e com múltiplas áreas brancas, além de linfoadenomegalia e esplenomegalia moderadas. No exame histopatológico, evidenciaram-se broncopneumonia intersticial necrótica acentuada e hepatite necrótica discreta associada a cistos e taquizoítas de T. gondii intralesionais positivos na imuno-histoquímica (IHC) anti-T. gondii. Evidenciou-se ainda, na medula óssea, leucemia mieloide crônica com IHC anti-FeLV positiva nas células neoplásicas. O DNA extraído dos pulmões foi positivo para T. gondii por meio da PCR-REP-529. T. gondii foi caracterizado por PCR-RFLP e pela técnica de microssatélites. Foi identificado o genótipo ToxoDB-PCR-RFLP #10, i.e., o arquétipo tipo I. A análise por microssatélites mostrou que a cepa era uma variante do tipo I, com dois alelos atípicos. Esta é a primeira vez que T. gondii clonal tipo I foi relacionado com um caso agudo de toxoplasmosis em um hospedeiro no Brasil.

ABSTRACT

This study discusses the causes of bird deaths recorded in a small aviary dedicated to the breeding of a few exotic, wild bird species. Findings from 28 birds were examined over a period of five years. About 40% of the deaths occurred in the first two weeks after hatching in the two most numerous species in the flock and such losses were mainly a result of starvation caused by inadequate nutritional management. Additionally, 28% of the cases affected recently introduced birds. Despite frequent treatment with anthelmintics; a total of 21% of the deaths in the flock could be attributed to parasitic diseases, most of them in recently acquired birds. Only three of the deaths could be associated with advanced age, all of the further cases were also attributable to management fails. Our results indicated that propagation of these and other species requires an improvement of the management of the newly hatched and newcomer birds to considerably enhance the flocks performance. Such information may be useful in conservation initiatives and may justify, at least in part, captivity of these animals.(AU)


Este estudo discute as causas de morte de aves registradas em um pequeno criadouro de umas poucas espécies de aves silvestres exóticas. As mortes de 28 aves foram examinadas em um período de cinco anos. Cerca de 40% dos casos ocorreram na primeira semana de vida de filhotes das duas espécies mais numerosas no plantel e essas perdas foram consequentes à desnutrição por falhas no manejo nutricional. Outros 28% de casos ocorreram em aves, recentemente introduzidas, ou em fase de adaptação, no criadouro. Ainda que as aves do plantel sejam frequentemente dosificadas com anti-helmínticos, 21% das mortes no plantel foram secundárias a doenças parasitárias; a maioria afetou aves recentemente adquiridas. Exceto por três mortes que foram associadas com desordens atribuídas à idade avançada, todas as demais foram associadas com inadequações no manejo das aves. Esses resultados sugerem que, em sistemas dessa natureza, as aves recém-nascidas e as recém-introduzidas são categorias críticas que demandam concentração de esforços para o atendimento sistemático de suas necessidades. Tais dados podem ser aplicáveis em iniciativas de conservação de outras espécies e podem justificar, ainda que parcialmente, a manutenção desses animais, em cativeiro.(AU)
