ABSTRACT

Environmental and management pressures are considered a threat for preserving plant communities worldwide. Identification of long-term impacts of changing management practices on plant community composition must, therefore, be a major priority to ensure improvement in conservation value. Land abandonment/wilding is one topical area where there is little available information on long-term impacts. To address this, here, we describe plant species compositional change over relatively long-time scales in a range of four British upland communities (high-level grasslands, intermediate grasslands, blanket bogs and high-level bogs) and its relationships with a series of life-history and plant trait variables. Our aims were to determine whether the business-as-usual sheep grazing practice was maintaining these communities, and if grazing was to be removed as part of abandonment/wilding strategies, would there be a conservation benefit. We used a series of long-term experimental grazing-exclosure studies at twelve sites (>40 years) on the Moor House National Nature Reserve in northern England. Each site consisted of paired plots; i.e. sheep-grazed versus ungrazed. Our results showed that there was relatively little change in dominant plant species with most change occurring within sub-dominant species. Similarly, different temporal responses in plant species richness were detected between the experiments (richness was reduced in three experiments, only one increased). In any case, the vegetation temporal trajectories were moving in similar direction in grazed and ungrazed plots at most sites. Interestingly, blanket bog experiments showed a clear compositional convergence for both grazed and ungrazed plots, although, the between-site differences related to a combination of elevation and past burning testament. Finally, the bryophytes (especially liverworts) and lichens were the groups that contributed most to the reductions in species richness. Moreover, implementation of a no-stock grazing strategy under a land abandonment/wilding approach will not bring about much plant change in the short-term in the plant communities studied here.

ABSTRACT

Only a few studies have considered the N budget of peat soils and this in turn has limited the ability of studies to consider the impact of changes in climate and atmospheric deposition upon the N budget of a peat soil. This study considered the total N budget of an upland peat-covered catchment over the period 1993 to 2009. The study has shown: i) Over the period of study the total N atmospheric deposition declined from 3.5 to 0.7 tonnes N/km2/yr. ii) The total fluvial export of N at soil source varied from 0.41 to 1.85 tonnes N/km2/yr with the fluvial flux being greater than the atmospheric input in 3 years of the study, implying significant internal processing. iii) Measuring the C:N ratio of organic matter pools in the ecosystem shows that gross primary productivity and litter decomposition represent outputs of N from the soil while DOC production and humification represent inputs of N. iv) Overall, the total N budget of the peat ecosystem varies from − 1.0 to + 2.5 tonnes N/km2/yr, i.e. in some years the ecosystem is a net source of N. The time series of the total N budget suggests that the ecosystem is responding to the occurrence of severe droughts with a long-term decline in N storage that could be interpreted as a response to long-term high N deposition rates, even if those rates have now diminished.

ABSTRACT

Persistence of HIV DNA presents a major barrier to the complete control of HIV infection under current therapies. Most studies suggest that cells with latently integrated HIV decay very slowly under therapy. However, it is much more difficult to study the turnover and persistence of HIV DNA during active infection. We have developed an "escape clock" approach for measuring the turnover of HIV DNA in resting CD4+ T cells. This approach studies the replacement of wild-type (WT) SIV DNA present in early infection by CTL escape mutant (EM) strains during later infection. Using a strain-specific real time PCR assay, we quantified the relative amounts of WT and EM strains in plasma SIV RNA and cellular SIV DNA. Thus we can track the formation and turnover of SIV DNA in sorted resting CD4+ T cells. We studied serial plasma and PBMC samples from 20 SIV-infected Mane-A*10 positive pigtail macaques that have a signature Gag CTL escape mutation. In animals with low viral load, WT virus laid down early in infection is extremely stable, and the decay of this WT species is very slow, consistent with findings in subjects on anti-retroviral medications. However, during active, high level infection, most SIV DNA in resting cells was turning over rapidly, suggesting a large pool of short-lived DNA produced by recent infection events. Our results suggest that, in order to reduce the formation of a stable population of SIV DNA, it will be important either to intervene very early or intervene during active replication.
