ABSTRACT

The composition and diversity of the helminth component communities in eels Anguilla anguilla were determined in three separate localities in Italy: an Adriatic coastal lagoon, Comacchio and two freshwater localities, the River Po and the Lake Piediluco. Data from Comacchio lagoon were analysed over 15 years to determine whether community composition and diversity changed significantly overtime. The community was species rich (nine species, all marine except Proteocephalus macrocephalus) and was dominated by a suite of digeneans: Deropristis inflata, Helicometra fasciata, Lecithochirium musculus and Bucephalus anguillae. The community showed little change in composition over the period, but the relative abundance and dominance of the species did alter. By contrast, the component communities in the freshwater localities were species poor and the dominant species were freshwater acanthocephalans, Pomphorhyncus laevis in River Po and Acanthocephalus rhinensis in Lake Piediluco. The helminth community of Lake Piediluco with five species was richer than that of the River Po with only three species, but was poorer than that of Comacchio lagoons. Similarity indices between samples from Comacchio were high; between the lagoon and the freshwater localities and between the two freshwater localities, similarity indices were very low. Helminth component community structure in coastal lagoons was comparable across Europe. The helminth community in the River Po was similar to those in the River Tiber and other European rivers whilst that in Lake Piediluco was similar to that in other European lakes. Levels of the pathogenic Anguillicoloides crassus in swim bladders were consistently lower in prevalence and abundance in the coastal lagoons than in freshwater localities. This suggests that this parasite may have little impact on migrating eels if they are indeed primarily of marine origin and so it may be of little importance in the recent decline of eel populations throughout Europe.

ABSTRACT

Detailed information on plant penetration activities by pear psylla Cacopsylla pyri L. (Hemiptera Psyllidae) is essential to study phytoplasma transmission of "Candidatus Phytoplasma pyri" responsible of pear decline disease (PD) and to trace and evaluate resistant traits in new pear tree selections for advanced breeding programs. The electrical penetration graph technique or (full) EPG may relevantly contribute to this knowledge. C. pyri EPG waveforms were characterized on basis of amplitude, frequency, voltage level, and electrical origin. Additionally, stylet tracks and the putative location of stylet tips in the plant tissue were histologically related to EPG waveforms by light and transmission electron microscopy observations after stylectomy. More than one waveform occurred in the same tissue: PA, PB, PC1 and PC2 were all detected in the mesophyll, and PE1 and PE2 were both recorded in the phloem. Waveform PE1 was always preceded by transient waveform PD, as previously described in other psyllids. Interestingly, no brief intracellular punctures (potential drop waveforms) were observed during plant penetration, opposite of what is usually recorded in aphids and other Sternorrhyncha.

ABSTRACT

Karavasta is the widest and most important lagoon in Albania. This study aimed to assess the ecological quality status of the lagoon, acquire knowledge of a natural environment which might be exploited for aquaculture, and give management hints on the basis of anthropogenic impact and ecological conditions. A sampling campaign was carried out in 2008: at six stations, benthic fauna, water, and sediment parameters were considered. Statistical analyses were carried out through multivariate procedures (PCA, classification-clustering, SIMPER, RDA, DISTLM, PERMANOVA). Ecological quality was assessed through the AZTI Marine Biotic Index (AMBI), the multivariate AMBI (M-AMBI) and the Benthic Index based on Taxonomic Sufficiency (BITS). Sediment characteristics (percent organic matter, %OM; redox potential discontinuity layer depth, RPDL; particle size composition) and salinity represented contributory influences on lagoon communities. It was possible to distinguish and characterise a confined area, and benthic communities, from a marine-influenced area and its biota. The number of species was quite low when compared with other open Adriatic lagoons. The M-AMBI and BITS classifications gave quite similar results, which seemed consistent with the ecological conditions of the lagoon, that is a distinction in the ecological quality between the seaward and landward stations, with higher ecological quality (EcoQ) at the seaward stations. Given the pressures and the ecological condition of Karavasta, an intensification of aquaculture activities must be considered with caution, since the lagoon seems at significant risk of serious hypereutrophication. This situation is made worse by the limited water exchange with the marine environment due to the irregular dredging of the communication channels.

ABSTRACT

Habitat Suitability (HS) models have been extensively used by conservation planners to estimate the spatial distribution of threatened species and of species of commercial interest. In this work we compare three HS models for the estimation of commercial yield potential and the identification of suitable sites for Tapes philippinarum rearing in the Sacca di Goro lagoon (Italy) on the basis of six environmental factors. The habitat suitability index (HSI) is based on expert opinion while the habitat suitability conditional (HSC) is calibrated on observational data. The habitat suitability mixed (HSM) model is a two-part model combining expert knowledge and regression analysis: the first component of the model uses logistic regression to identify the areas in which clams are likely to be present; the second part applies the same parameter-specific suitability functions of the HSI model only in the areas previously identified as productive by the logistic component. The HS models were validated on an independent data set and estimates of potential yield of the Goro lagoon were compared. The effectiveness of the three approaches is then discussed in terms of predicted yield and identification of suitable sites for farming.

ABSTRACT

The number and distribution of the 18S-28S and 5S rRNA (rDNA) gene sequences were examined on mitotic chromosomes of six sturgeon species by two-colour in situ hybridization. Four of the six species, Huso huso, Acipenser stellatus, Acipenser sturio, and Acipenser ruthenus, with about 120 chromosomes, showed from six to eight 18S-28S rDNA signals, while 5S rDNA signals were on only one chromosome pair. The two species with 250-270 chromosomes, Acipenser baerii and Acipenser transmontanus, showed from 10 to 12 18S-28S sites and two chromosome pairs bearing 5S rDNA signals. In all examined species, the rather intense 5S rDNA signals apparently overlapped those of 18S-28S rDNA. These data support the diploid-tetraploid relationships between the two chromosome groups of sturgeons. The close association between the two rDNA families in species belonging to an ancestral fish order, such as Acipenseriformes, supports the hypothesis that the association represents a primitive condition.

ABSTRACT

This study was undertaken in order to investigate the possible interactions between nitric oxide and arachidonic acid (AA) in Venus verrucosa oocytes. We perifused isolated oocytes to determine the effect of the following substances on [3H]arachidonic acid release ([3H]AA): (1) A 23187, a calcium ionophore; (2) nitric oxide (NO) donors; (3) 1,1,1-trifluoromethyl-6,9,12,15 heicosatetraen-2-one (AACOCF(3)), a specific phospholipase A(2) (PLA(2)) inhibitor; (4) [5'-hydroxymethyl-2'-furyl]-1-benzyl indazole (YC-1) and 1H-[1,2,4]oxadiazolo[4,3-alpha]quinoxalin-1-one (ODQ), specific soluble guanylyl cyclase activator and inhibitor, respectively; (5) L-arginine, the substrate of nitric oxide synthase; (6) L-nitroarginine methyl esther (L-NAME), an inhibitor of nitric oxide synthase. Our results demonstrated that: (a) the calcium ionophore dose-dependently increased [3H]arachidonic acid release; (b) the NO donors sodium nitroprusside (SNP) and linsidomine (SIN-1) highly increased [3H]arachidonic acid output, while S-nitroso-N-acetylpenicillamine (SNAP) was without effect; (c) AACOCF(3) completely blocked the [3H]arachidonic acid release induced by SNP and SIN-1; (d) YC-1 increased [3H]arachidonic acid release, while ODQ completely counteracted SNP response; (e) [3H]arachidonic acid output was also increased by L-arginine; (f) a similar effect was, paradoxically, obtained in the presence of L-NAME. Furthermore, using RT-PCR we demonstrated in the same cells the presence of a nitric oxide synthase (NOS) mRNA, whose expression was not modulated by interleukin 1beta (IL-1beta). These results demonstrate the presence of a both calcium-dependent and NO-sensitive PLA(2) and of nitric oxide synthase in V. verrucosa oocytes. Our data also suggest a co-action of the two pathways in the control of reproduction in this bivalve.
