ABSTRACT

Many tropical species live close to their thermal limits within a narrow niche. Here, we investigate the ecophysiological limits of the tropical tree Podocarpus trinitensis, which is endemic to Trinidad and Tobago where most populations exist as isolated stands on hilltops. Five wild stands from a range of elevations were compared in the field with measurements of leaf temperature, canopy cover, stomatal conductance (gs), chlorophyll content and several chlorophyll fluorescence parameters. A parallel greenhouse experiment was used to acclimate seedlings to 'CONTROL' and 'HEAT' treatments (with mid-day air temperatures of 34.5 and 37 °C respectively), after which the above parameters were measured along with photosynthetic light and temperature response curves, leaf morphology and in vitro Fv/Fm thermostability. There was a positive association between improved physiological performance and elevation. In the high elevation sites, leaf temperatures were significantly lower while most of the physiological parameters were higher (gs, chlorophyll content, ɸ PSII, ETRmax and Isat90). In the greenhouse, HEAT and CONTROL plants were similar for most parameters, except leaf temperature (which was coupled with air temperature) and leaf mass per unit area (which was higher in HEAT plants). Temperature response curves showed an optimum temperature for photosynthesis of 30 ± 0.5 °C (TOpt) and in vitro Fv/Fm indicated a critical temperature of 47.4 ± 0.38 °C for HEAT and 48.2 ± 0.24 °C for CONTROL (T50), with no indication of heat acclimation. Podocarpus trinitensis was found to be shade tolerant. In the field, seedlings established under a close canopy (>95% canopy cover) and had a low light saturation point (LCP). In the greenhouse, where more light was available, seedlings retained a low light compensation point, light saturation point (LSP) and maximum photosynthetic rate (Amax). The results suggest that P. trinitensis is moderately heat tolerant with the higher elevation sites being more habitable, but stands are also able to survive near sea level under a closed canopy. The narrow niche, along with the 30 ± 0.5 °C optimum temperature for photosynthesis and the lack of thermal plasticity in critical temperature, suggests that P. trinitensis has little room to acclimate to temperatures higher than those currently experienced.

ABSTRACT

The islands of the Caribbean are considered to be a "biodiversity hotspot." Collectively, a high level of endemism for several plant groups has been reported for this region. Biodiversity conservation should, in part, be informed by taxonomy, population status, and distribution of flora. One taxonomic impediment to species inventory and management is correct identification as conventional morphology-based assessment is subject to several caveats. DNA barcoding can be a useful tool to quickly and accurately identify species and has the potential to prompt the discovery of new species. In this study, the ability of DNA barcoding to confirm the identities of 14 endangered endemic vascular plant species in Trinidad was assessed using three DNA barcodes (matK, rbcL, and rpoC1). Herbarium identifications were previously made for all species under study. matK, rbcL, and rpoC1 markers were successful in amplifying target regions for seven of the 14 species. rpoC1 sequences required extensive editing and were unusable. rbcL primers resulted in cleanest reads, however, matK appeared to be superior to rbcL based on a number of parameters assessed including level of DNA polymorphism in the sequences, genetic distance, reference library coverage based on BLASTN statistics, direct sequence comparisons within "best match" and "best close match" criteria, and finally, degree of clustering with moderate to strong bootstrap support (>60%) in neighbor-joining tree-based comparisons. The performance of both markers seemed to be species-specific based on the parameters examined. Overall, the Trinidad sequences were accurately identified to the genus level for all endemic plant species successfully amplified and sequenced using both matK and rbcL markers. DNA barcoding can contribute to taxonomic and biodiversity research and will complement efforts to select taxa for various molecular ecology and population genetics studies.

BACKGROUND: The purpose of the study is to determine the epidemiology of road traffic collisions (RTCs) in Trinidad and Tobago by characterizing RTCs in terms of number of collisions, fatalities, victim profiles, and locations for the purpose of informing accident prevention programs. Previous studies of RTCs in Trinidad and Tobago were primarily concerned with patterns of drivers use of seat belts, road collisions as a cause of mortality in young men, and the economic burden of road collisions. Attempts were made to model road fatalities, but limited epidemiological data meant that it was difficult to determine trends or develop models. METHODS: This study determined the epidemiology of RTCs in Trinidad and Tobago over the period 2000-2011 using data collected by the Trinidad and Tobago Road Traffic Branch of the Police Service and secondary data from the Central Statistical Office. Data were analyzed using Excel, SPSS, and R statistical packages. RESULTS: Fatalities were greater among men (80%) than among women (20%) and were highest on two major freeways in Trinidad [the Churchill-Roosevelt Highway and the Sir Solomon Hochoy Highway]. Most collisions occurred during the night among individuals between the ages of 15 and 44 years. Fatalities among drivers steadily increased over the study period and overtook fatalities among pedestrians, who were the group most affected in 2000. Most fatalities occurred at weekends. CONCLUSIONS: These patterns can inform (i) education programs and (ii) road and traffic control measures.
