ABSTRACT

The understanding of excitonic transitions associated with polymeric aggregates is fundamental, as such transitions have implications on coherence lengths, coherence numbers and inter- and intra-chain binding parameters. In this context, the investigation of efficient solvents and other ways to control polymer aggregate formation is key for their consolidation as materials for new technologies. In this manuscript, we use Poly(3-hexothiophene) (P3HT) as a probe to investigate the significance of amylene (C5H10) and its association with methanol (MeOH) in both pure and C5H10-stabilized chloroform (CHCl3)-based polymeric solutions. Using the intensity ratio between the first and second vibronic transitions of the P3HT H-aggregates formed, values for their exciton bandwidths and interchain interactions are obtained and correlated with the presence of C5H10 and MeOH as agents determining the CHCl3 quality.

ABSTRACT

The electromagnetic interference (EMI) around the devices for ionizing radiation experiments is a serious concern. In order to reduce EMI and enhance the signal-to-noise ratio, a new type of analog fiber optic link (FOL) has been developed for fast pulse signal transmission under complicated electromagnetic fields. Electrical signals are converted to optical signals through an electro-optical conversion process using the amplitude modulation mechanism. After long-distance transmission, optical signals can also be restored via photoelectric conversion. Each link has a transmitter unit connected via a fiber optic cable to the receiver unit. The transmitter unit consists of a distributed feedback laser, a driver circuit made of amplifiers, and an automatic temperature control module, while the receiver unit consists of a PIN detector, a transimpedance amplifier, a main amplifier, and a third order active low-pass filter. A prototype of this link has been fabricated and tested. The test results show that the transmission bandwidth of the analog signals is DC ∼ 155 MHz, the transmission delay is a typical 21 ns, the linear dynamic range is greater than 56 dB (692 times larger), the output noise is less than 4.5 mV, and the transmission distance is up to several kilometers, which can meet the requirements of ionizing radiation experiments. The FOL demonstrates great performance in transmission for scientific research with high quality in terms of transmission bandwidth, linearity, security, and stability.

ABSTRACT

Polarimetry is generally used to determine the polarization state of light beams in various research fields, such as biomedicine, astronomy, and materials science. In particular, the rotating quarter-wave plate polarimeter is an inexpensive and versatile option used in several single-wavelength applications to determine the four Stokes parameters. Extending this technique to broadband spectroscopic measurements is of great scientific interest since the information on light polarization is highly sensitive to anisotropic phenomena. However, the need for achromatic polarizing elements, especially quarter-wave plates, requires special attention in their modeling. In this study, we implemented a rotating retarder spectropolarimeter for broadband measurements using a commercially available quasi-achromatic biplate retarder over the visible range. Here, we present a comprehensive approach for troubleshooting this type of spectropolarimeter through the observation of artifacts stemming from the standard single-plate retarder model. Then, we derive a more suitable model for a quasi-achromatic retarder consisting of a biplate junction. This new biplate model requires knowledge of the intrinsic dispersive properties of the biplate, namely the equivalent retardance, fast axis tilt, and rotatory angle. Hence, in this study, we also show a self-consistent methodology to determine these biplate properties using the same polarimeter apparatus so that accurate Stokes parameters can be determined independently. Finally, the comparison of data generated with the standard single-plate and new biplate models shows a significant improvement in the measurement precision of the investigated polarization states, which confirms that remodeling the retarder for reliable spectropolarimetry is necessary.

ABSTRACT

Obtained natural sands can present different particle size distributions (PSD), although they have the same mineralogical origin. These differences directly influence the physical and mechanical behavior of mortars and, therefore, the performance of mortar and ceramic renderings. Standardizing the particle size of sands based on pre-established requirements in normative standards (NBR 7214 or ASTM C778) is one way to minimize these effects. However, these standards do not consider the optimization of the granular skeleton through the analysis of bulk density and PSD, which may be insufficient to obtain satisfactory results. Therefore, this paper analyzes the effects of using different particle size ranges on the physical and mechanical behavior of cement and hydrated lime mortars. The properties of consistency index, bulk density, air content, capillary water absorption, water absorption by immersion, flexural strength, compressive strength, and dynamic modulus of elasticity were evaluated. For this purpose, standardized sands of the same mineralogical origin were made with different particle size ranges, being: (i) standardized sand constituted by 25% of coarse and fine fractions (S25-control), (ii) standardized sand constituted by 30% of coarse fraction and 20% of fine fraction (S30-20), and (iii) standardized sand composed by 40% of coarse fraction, and 10% of fine fraction (S40-10), respectively. The results indicated that variations in the particle size composition of the standardized sands are necessary to obtain mixtures with higher compactness and, therefore, mortars with better physical and mechanical performance. Thus, the dosage of the particle size fractions of standardized sand should consider the optimization of the granular skeleton, being the unit mass and the granulometric composition as important parameters to meet this premise.

ABSTRACT

To facilitate biomedical studies of disease mechanisms, a high-quality interactome that connects functionally related genes is needed to help investigators formulate pathway hypotheses and to interpret the biological logic of a phenotype at the biological process level. Interactions in the updated version of the human interactome resource (HIR V2) were inferred from 36 mathematical characterizations of six types of data that suggest functional associations between genes. This update of the HIR consists of 88 069 pairs of genes (23.2% functional interactions of HIR V2 are in common with the previous version of HIR), representing functional associations that are of strengths similar to those between well-studied protein interactions. Among these functional interactions, 57% may represent protein interactions, which are expected to cover 32% of the true human protein interactome. The gene set linkage analysis (GSLA) tool is developed based on the high-quality HIR V2 to identify the potential functional impacts of the observed transcriptomic changes, helping to elucidate their biological significance and complementing the currently widely used enrichment-based gene set interpretation tools. A case study shows that the annotations reported by the HIR V2/GSLA system are more comprehensive and concise compared to those obtained by the widely used gene set annotation tools such as PANTHER and DAVID. ABSTRACT

In this work, the natural and modified carnauba powder from the addition of bentonite was evaluated for the adsorption of Cu(II) ions in synthetic solution. The results showed that the carnauba powder treated with bentonite (CPTB) showed a better percentage of removal of Cu(II) ions when compared to natural carnauba powder (NCP). The best results for both adsorbents were obtained with pH 5. The adsorption kinetics was governed by the pseudo-second-order model for both bioadsorbents studied. While the isothermal behavior was governed by the Langmuir model and showed that the adsorption capacity of the CPTB for Cu(II) was 21.98 mg·g-1. The interaction of the metal and CPTB was also investigated by means of thermodynamic parameters showing that the adsorption process is not spontaneous, although the values of ΔG° decrease with the increase in temperature from 20 to 40 °C and endothermic causing an increase in the degree of disorder at the solid/liquid interface. The results showed that the CPTB is a material with potential adsorbent for the removal of copper ions.

ABSTRACT

AIM: To determine the cut-off value of the area ratio under the curve (ARUC) for predicting symptoms of stroke. MATERIALS AND METHODS: Contrast-enhanced ultrasound was used to analyse intraplaque neovascularization (IPN). The correlations between the ARUC and risk factors of stroke were examined. A receiver operating characteristic curve was used to determine the cut-off value of the ARUC. RESULTS: Using a quantitative analysis method for IPN, the ARUC was significantly higher in the symptomatic group than in the asymptomatic group (p=0.017). The ARUC was positively associated with the homocysteine level (r=0.429, p=0.002) and high-sensitivity C-reactive protein level (r=0.424, p=0.003). Regression analysis showed that the ARUC was a risk factor for symptoms of stroke. The receiver operating characteristic curve showed that the cut-off value for symptoms was 0.24; the sensitivity was 77%, and the specificity was 70%; the positive predictive value was 68%, and the negative predictive value was 78%. CONCLUSION: IPN was a risk factor for the occurrence of the clinical symptoms of stroke. Patients with an ARUC of >0.24 had a higher risk of stroke.

ABSTRACT

AIM: To investigate the role of utilizing size-based thresholds of stiffness gradient in diagnosing solid breast lesions and optimizing original Breast Imaging-Reporting And Data System (BI-RADS) classifications. MATERIALS AND METHODS: Two-hundred and twenty-seven consecutive women underwent shear-wave elastography (SWE) before ultrasound-guided biopsy, and 234 solid breast lesions categorized as BI-RADS 3-5 were analysed. Receiver operating characteristic curve analysis was performed based on histopathology. Diagnostic performance among SWE, BI-RADS, and their combination were compared. RESULTS: The stiffness gradient correlated with the standard deviation of elasticity (SD, r=0.90), and with Tozaki's pattern classification (r=0.64). The area under the receiver operating characteristic curves (AUC) for stiffness gradient (0.939) outperformed SD (0.897) or colour pattern (0.852). Due to significant association with lesion size (r=0.394, p<0.001), stiffness gradient's size-based thresholds (lesions >15 mm: 82.5 kPa; lesions ≤15 mm: 51.1 kPa) were established to reclassify BI-RADS 3-4b lesions. Upgrading category 3 lesions (over the corresponding cut-off value, 3 to 4a) and downgrading categories 4a-4b lesions (less than or equal to the corresponding cut-off value, 4b to 4a, 4a to 3), yielded significant improvement in specificity (90.28% versus 77.78%, p<0.001) and AUC (0.948 versus 0.926, p=0.035) than BI-RADS alone. No significant loss emerged in the sensitivity (88.89% versus 91.11%, p=0.500). CONCLUSION: Stiffness gradient exhibited better discriminatory ability than SD or four-colour pattern classification in determining solid breast lesions and applying its size-specific thresholds to categorize BI-RADS 3-4b lesions could improve diagnostic performance.

ABSTRACT

OBJECTIVE: ICOS/ICOSL plays a crucial part in various disease-mediated immune responses. However, the exact role of ICOS/ICOSL in type 2 diabetes mellitus (T2DM) development remains unexplored. This study aims to investigate the role of ICOS/ICOSL in the pathogenesis of T2DM. MATERIALS AND METHODS: Human peripheral blood T-lymphocytes (CD3) and umbilical vein endothelial cells (HUVECs) were treated with high-glucose (HG) or advanced glycation end products (AGEs). A portion of CD3 cells was co-cultured with HUVECs and treated with different mediums or anti-ICOS mAbs. The ICOS/ICOSL and caspase-3 protein expression was measured by Western blotting. ELISA (enzyme-linked immunosorbent assay), MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide), and NOx production assays were respectively used to detect cytokines level, cell viability and the production of NOx. RESULTS: HG and AGEs significantly upregulated ICOS/ICOSL expressions in T cells and HUVECs. T cell contact with HUVECs secreted more IFN-γ, IL-4, and IL-10 compared to non-contact cells, while cytokines from IL-6-, IL-1ß-, and CM- (the conditioned medium) treated cells did not differ from the control. A significant increase of IL-8 and IL-6 was found in HUVECs under both contact and non-contact conditions vs. control cells. Similar results were also observed in the comparison between CM1- (T cell condition medium) or CM2- (co-culture condition medium) treated cells and control cells. However, CM1 and CM2 treatment significantly inhibited cell viability and increased caspase-3 and NOx production; blocking ICOS/ICOSL remarkably decreased cytokines secretion, enhanced cell viability and reduced caspase-3 and NOx production. CONCLUSIONS: HG and AGEs cause T cell inflammatory response and vascular endothelial dysfunction by upregulating ICOS/ICOSL, which may be one of the possible mechanisms of cardiovascular complications development in T2DM patients.

OBJECTIVE: Diabetic nephropathy (DN) has become the major complication of diabetes. The progression of the disease impedes the efficacy of DN treatment. Therefore, the strategies to inhibit or reverse kidney damage in DN patients are of critical importance. We aim to investigate the effect of sitagliptin on DN within rat model and analyze the associated metabolism and expression of iNOS/GLP-1 receptor. MATERIALS AND METHODS: Diabetic model was generated by using Sprague-Dawley (SD) rats, which received an intra-peritoneal injection of 30 mg/kg streptozotocin. Rats were then treated with saline or 15 mg/(kg.d) sitagliptin by gavage. After 12 weeks, fasting blood glucose and insulin resistance were measured. Rat glucose and lipid metabolism were evaluated by high triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C). Western blot was used to measure expression of glucose-6-phosphatase (G6Pase), phosphoenolpyruvate carboxykinase (PEPCK), fatty acid synthase (FAS) and acetyl-CoA carboxylase (ACC) related with glucose-lipid metabolism, and expression of iNOS and GLP-1 expression in kidney tissues. RESULTS: After 12 weeks of feeding, the levels of blood glucose and lipid in sitagliptin group were significantly decreased compared to those in control group (p<0.05), whilst insulin sensitivity was enhanced (p<0.05). Western blot showed that sitagliptin downregulated the expressions of glucose-lipid metabolism proteins such as G6Pase, PEPCK, ACC and FAS in rat livers, inhibited iNOS expression in kidneys and elevated GLP-1 receptor activity (p<0.05). CONCLUSIONS: Sitagliptin effectively stabilizes blood glucose and lipid levels in DN rats, significantly improves glucose-lipid metabolism and protects kidney and vascular endothelial cells during DN pathogenesis through inhibiting iNOS expression and elevating GLP-1 receptor activity.

OBJECTIVE: To investigate the effect of nuclear factor-kappaB (NF-κB) on the myocardin-mediated differentiation of hysteromyoma cells. MATERIALS AND METHODS: Expression levels of myocardin in hysteromyoma cells from patients with hysteromyoma were detected. Normal uterine smooth muscle cells were used as control group. Overexpression of myocardin in hysteromyoma cells was achieved through lentivirus infection. Changes in expression levels of uterine smooth muscle cell maker p21, p57, Cyclin D1, PCNA, SM22α, and αSMA were detected. Hysteromyoma cells with lentivirus infection were stimulated by lipopolysaccharide (LPS), and changes in expression levels of myocardin were detected. RESULTS: Compared with normal uterine smooth muscle cells, the expression level of myocardin in hysteromyoma cells was extremely low, or even undetectable, and expression levels of smooth muscle cell differentiation markers were also minimal, and cells were in the de-differentiated state. Expression of exogenous myocardin can improve the expression of smooth muscle cell differentiation markers to induce cell re-differentiation. LPS stimulation can activate NF-κB to inhibit myocardin expression, thereby inducing cell dedifferentiation. CONCLUSIONS: NF-κB can inhibit the differentiation of hysteromyoma cells by decreasing the expression level of myocardin.

ABSTRACT

AIM: To compare diagnostic value of two-dimensional (2D) ultrasonography and elastosonography for suspected axillary lymph node metastasis of breast cancer. MATERIALS AND METHODS: Elastosonography and 2D ultrasonography were performed on 78 axillary lymph nodes of 78 patients with suspected breast cancer. Scores of shape, long- to short-axis ratio, cortical thickness, and lymph node hilum were summed as the score of each lymph node at 2D ultrasonography, while a four-point scale was adopted for elasticity scoring. The combined score of each lymph node was obtained by summing the score at 2D ultrasonography and that at elasticity scoring. The strain ratio was calculated by comparison of the average strain of the lymph node with that of the subcutaneous tissue. Diagnostic efficacies of 2D ultrasonography, elasticity scoring, and the combined method were compared. RESULTS: There were 78 axillary lymph nodes, including 34 non-metastatic and 44 metastatic nodes. The elasticity scores of non-metastatic and metastatic axillary lymph nodes were 1.44±0.82 and 3.11±0.75, respectively (p<0.05). The difference in area under the operating characteristic curve (AUC) was statistically significant between 2D ultrasonography and the combined method (p<0.05). The sensitivity, specificity, and accuracy of 2D ultrasonography and elasticity scoring were 77.3% versus 86.4%, 76.5% versus 85.3%, and 76.9% versus 85.9%, respectively (all p>0.05), and those of the combined method were 93.2%, 73.5%, and 84.6%, respectively. There was a significant difference in sensitivity between 2D ultrasonography and the combined method (p<0.05). CONCLUSIONS: Combined application of 2D ultrasonography with elastosonography can improve the diagnostic capability for metastatic axillary lymph node characterisation in breast cancer.

ABSTRACT

Breast cancer, which derives from the epithelium of the mammary glands, is one of the most common cancers diagnosed in women globally. To date, the authors of many studies have reported that the deregulation of microRNAs (miRNAs) plays a crucial role in the occurrence, development, and metastasis of tumors. Here, we discovered that miR-660-5p was upregulated in the breast cancer cell lines MCF7 and MDA-MB-231 compared with that in the normal control cell line CCD-1095Sk. We then inhibited the expression of miR-660-5p to investigate its biological function in cancer development, progression, and metastasis. We determined the changes in the levels of expression of transcription factor CP2 (TFCP2) and CDKN1A to further clarify the specific mechanism involved. The results showed that downregulation of miR-660-5p significantly suppressed the proliferation, migration, and invasion of MCF7 breast cancer cell. Moreover, inhibition of miR-660-5p promoted cell cycle G1 arrest and reduced apoptosis in breast cancer cells. The specific mechanism studies confirmed that TFCP2 was a direct downstream target of miR-660-5p. Aberrant expression of miR-660-5p repressed TFCP2 expression, whereas inhibition of miR-660-5p decreased TFCP2 protein expression, which is a vital factor in the downstream signaling pathway. In conclusion, miR-660-5p can regulate the proliferation, migration, and invasion of human breast cancer cells, and is a novel potential therapeutic target for the clinical treatment of breast cancer.

In this study, anti-hepatitis B virus (HBV) immunity was evaluated in mice using several regimens of the HBV recombinant protein vaccine HBSS1 that expressed in CHO cells containing S (1-223 aa) and preS1 (21-47 aa) and recombinant adenovirus rAdSS1 vaccine. Further, the protective efficacy of these vaccine regimens was studied in a mouse model. High titres of antigen-specific antibodies and neutralizing activity were elicited in mice after vaccination. However, robust multi-antigen (preS1 and S)-specific cell-mediated immunity (CMI) was only detected in mice primed with HBSS1 and boosted with rAdSS1. Moreover, functional T-cell responses with high levels of cytokines and antigen-specific cytotoxic T-cell responses (CD107a+ CD8+ ) were also detected in the mice. Rapid clearance of hepatitis B surface antigen and HBV DNA in blood and significantly decreased hepatitis B envelope antigen levels were observed in mice immunized with the heterogeneous prime-boost vaccine after hepatitis B virus challenge by hydrodynamic injection (HI) of pCS-HBV1.3. The clearance of HBV correlated well with antigen-specific CMI (Th1 and CTL responses) and cytokine profiles (IFN-γ, TNF-α, IL-2) elicited by vaccination. Taken together, our results might contribute to the development of new human HBV vaccines and a better understanding of the mechanisms underlying immune protection and clearance of hepatitis B virus infection.

ABSTRACT

In order to understand the effect of grain moisture of inbred lines at the silking and physiological maturity stages on kernel dehydration rate, 59 maize inbred lines from six subgroups were selected. Grain moisture was measured and QTLs associated with kernel dehydration were mapped. A rapid dehydration evaluation and association analysis revealed eight inbred lines with faster dehydration rate, including Yuanwu 02, K36, Zhonger/O2, Lo1125, Han 49, Qi 319, Hua 160, and PH4CV. A single sequence repeat analysis using 85 pairs detected five QTLs with phenotypic variation contribution ≥10% in the permanent F2 generation populations Zheng 58 x S1776 and Chang 7-2 x K1131, which had LOD threshold values ≥ 3 in both 2013 and 2014. The chromosome region of qFkdr7b had not previously been reported and is preliminarily identified as a new major QTL. A false positive field verification of grain dehydration rate of 53 inbred lines indicated that the screening result of the rapid dehydration inbred lines by specific amplification with marker Phi114 was most similar to the field assessment result, followed by markers Phi127 and Phi029. The rapid dehydration lines selected based on primer Phi114 amplification were also similar to the field dehydration rate and can thus be used for molecular marker-assisted selection. A significant effort is needed to improve stress resistance and shorten the growth period via fast kernel dehydration in intermediate materials of the inbred lines K36, Zhonger/ O2, Lo1125, Han 49, Hua 160, and PH4CV, and further using the selected lines for new combinations.

ABSTRACT

OBJECTIVE: To evaluate the influence of overweight/obesity on the incidence of hypertension among adults in China. METHODS: The subjects of this prospective study were 13 739 Chinese adults aged 35-74 years recruited at the baseline surveys of China Multicenter Collaborative Study of Cardiovascular Disease Epidemiology and International Collaborative Study of Cardiovascular Disease in Asian. Baseline surveys were conducted in 1998 and during 2000-2001, respectively, and the follow-up was conducted during 2007-2008. According to the body mass index, the subjects were divided into four groups: underweight group(<18.5 kg/m(2)), normal weight group(18.5-23.9 kg/m(2)), overweight group(24.0-27.9 kg/m(2))and obesity group(≥28.0 kg/m(2)). Age-standardized cumulative incidence of hypertension was calculated for each group, respectively. The relative risks(RRs)and 95% confidence intervals(CIs)for the incidence of hypertension of underweight, overweight and obesity groups were estimated by using generalized linear regression model with normal weight group as reference. RESULTS: During 8.1 years of follow-up, 4 271 hypertension cases were detected(2 012 in men and 2 259 in women). Age-standardized cumulative incidence of hypertension for the underweight, normal weight, overweight and obesity groups were 20.3%, 30.9%, 43.6% and 50.8% in men, respectively; and 22.9%, 30.4%, 41.1% and 50.8% in women, respectively. Compared with the normal weight group, multivariate-adjusted RR(95% CI)for the incidence of hypertension in underweight, overweight and obesity groups were 0.78(0.64-0.95), 1.22(1.13-1.30)and 1.28(1.16-1.42)in men, respectively; and 0.89(0.77-1.03), 1.16(1.09-1.23)and 1.28(1.18-1.38)in women, respectively. The overweight and obese subjects had higher risk for the incidence of hypertension, with the population attributable risk proportion of 7.4% in men and 8.8% in women, respectively. CONCLUSION: Overweight or obese people are at an increased risk of developing hypertension, thus prevention and control of overweight/obesity are needed to reduce hypertension incidence among adults in China.

ABSTRACT

Compared with traditional de-ionized pressurized-water cooled magnet of ECRIS, evaporative cooling magnet has some special characteristics, such as high cooling efficiency, simple maintenance, and operation. The analysis is carried out according to the design and operation of LECR4 (Lanzhou Electron Cyclotron Resonance ion source No. 4, since July 2013), whose magnet is cooled by evaporative cooling technology. The insulation coolant replaces the de-ionized pressurized-water to absorb the heat of coils, and the physical and chemical properties of coolant remain stable for a long time with no need for purification or filtration. The coils of magnet are immersed in the liquid coolant. For the higher cooling efficiency of coolant, the current density of coils can be greatly improved. The heat transfer process executes under atmospheric pressure, and the temperature of coils is lower than 70 °C when the current density of coils is 12 A/mm(2). On the other hand, the heat transfer temperature of coolant is about 50 °C, and the heat can be transferred to fresh air which can save cost of water cooling system. Two years of LECR4 stable operation show that evaporative cooling technology can be used on magnet of ECRIS, and the application advantages are very obvious.

ABSTRACT

LECR4 (Lanzhou ECR ion source No. 4) is a room temperature electron cyclotron resonance ion source, designed to produce high current, high charge state ion beams for the SSC-LINAC injector (a new injector for sector separated cyclotron) at the Institute of Modern Physics. LECR4 also serves as a PoP machine for the application of evaporative cooling technology in accelerator field. To achieve those goals, LECR4 ECR ion source has been optimized for the operation at 18 GHz. During 2014, LECR4 ion source was commissioned at 18 GHz microwave of 1.6 kW. To further study the influence of injection stage to the production of medium and high charge state ion beams, in March 2015, the injection stage with pumping system was installed, and some optimum results were produced, such as 560 eµA of O(7+), 620 eµA of Ar(11+), 430 eµA of Ar(12+), 430 eµA of Xe(20+), and so on. The comparison will be discussed in the paper.
