ABSTRACT

Selenium (Se) plays a crucial role in protecting biological materials from oxidative damage through the action of the selenoprotein glutathione peroxidase (GSH-Px), and the effectiveness of this protection is often dependent upon Se supply. Recent evidence has indicated that GSH-Px mRNA expression can be upregulated in response to potential oxidative damage risk, and that this upregulation is independent of Se supply. The current study aimed to determine the effect of Se supplementation, stocking rate and tissue fatty acid profile on GSH-Px activity in breast and thigh tissue of commercial broilers. A total of 168 Ross 308 broiler chicks were enrolled onto the study. Prior to enrolment, birds were brooded as a single group and received a starter diet containing no additional Se. The study was a 2 × 2 factorial design comprising of two levels of dietary Se (high Se, 0.5 mg/kg total Se, low Se background Se only), and two stocking rates (high, 30 kg/m2, and low, 15 kg/m2). At 15 days of age, birds were blocked by live weight and randomly allocated to one of the four treatments, with six pen replicates per treatment. At 42 days of age, one bird was randomly selected from each pen replicate, euthanased and breast and thigh tissue harvested. GSH-Px activity, thiobarbituric acid reactive substances (TBARS), and fatty acid (FA) content of these tissues were determined. There was no effect (P > 0.05) of stocking rate on GSH-Px activity or TBARS. GSH-Px activity did not differ between tissue types but was greater in high Se birds (P < 0.001) compared to low Se. TBARS concentrations were greater in thigh tissue (P < 0.001), and these thigh concentrations were greater in high Se birds (P < 0.05). There were marked differences between breast and thigh tissue in most FAs (P < 0.001), with breast generally containing greater proportions of polyunsaturated FA, so that breast tissue had a higher (P < 0.001) peroxidisability index (PI) than thigh. A positive correlation between GSH-Px activity and PI in the thigh tissue of high Se birds (Pearson Correlation 0.668; P = 0.025) may indicate that increasing susceptibility to peroxidisation in lipid-rich tissues may also upregulate GSH-Px activity in Se-replete birds. This study suggests that ensuring adequate dietary selenium could be a useful tool to mitigate adverse effects on meat quality caused by oxidation, particularly in lipid-rich meat.

ABSTRACT

Antimicrobial resistance (AMR) is a major threat to humans and animals globally. Antimicrobial stewardship has been acknowledged as a primary strategy to tackle AMR. An important first step for antimicrobial stewardship is to quantify antimicrobial use (AMU). In Fiji, there are currently no data on AMU in livestock farms. This study aimed to quantify AMU in different livestock enterprises (beef, dairy, broiler, and layer) and farming systems (backyard, semi-commercial and commercial) in Central and Western divisions of Viti Levu, Fiji. A survey with 210 livestock farmers and 26 managers representing 276 enterprises was conducted between May and September 2019. The difference in AMU between different livestock enterprises and farming systems was investigated using ANOVA. In Fiji, the estimated annual antibiotic use in livestock was lower than the global average (44 compared with 118 mg/PCU). However, this use was concentrated in 56% of participant farms (the remaining 44% did not use antimicrobials). Total estimated quarterly anthelmintic use (20,797 mg) was not affected by farming systems but was highest (P < 0.001) in dairy enterprises (24,120 mg) and lowest in broiler enterprises (4 mg). Quarterly antibiotic use was different between the enterprises regardless of the metrics used to quantify the use (P < 0.05). Total estimated quarterly mg/PCU of antibiotic use was highest (P < 0.001) in broiler enterprises (12.4 mg/PCU) and lowest in beef enterprises (0.2 mg/PCU). For all other ESVAC metrics, total estimated antibiotic use was higher in poultry and lower in cattle enterprises. Backyard systems used less antibiotics (total mg) than commercial systems, but for other metrics, the trend was reversed. The use of both antibiotics and anthelmintics (rather than antibiotics or anthelmintics alone, or no AMU) was associated with dairy enterprises (Χ2 = 123, P < 0.001). Further studies should be conducted to quantify and evaluate the drivers of AMU in Fijian livestock farms. In addition, differences in AMU between different enterprises and farming systems suggest that strategies to reduce AMU should be tailored to specific settings.

ABSTRACT

This study was undertaken to determine the effect of a yeast (Candida famata) and a bacterium (Lactobacillus plantarum), administered alone or in combination in the drinking water, on the population of yeast, Lactobacillus sp. and coliforms, and the prevalence of antimicrobial resistance (AMR) and virulence genes in Escherichia coli (E. coli) isolated from digesta samples taken throughout the life of broiler chickens. Male (Ross 308) day-old chicks (220) were used. C. famata (isolated from a chicken) and L. plantarum (isolated from a pig) were administered via the drinking water. Water was provided either untreated or with C. famata (CF; 108/ml), L. plantarum (LP; 105-108/ml), or a combination of CF and LP (106-108/ml) in water hoppers on 2 days each week for 35 days. Administering probiotics did not affect the growth performance in broiler chickens. No significant interactions were observed between main effects, and neither CF nor LP had any effect on the population size of Lactobacillus sp. or coliforms. The administration of C. famata increased the population density of yeasts in the small intestine at these ages. The population density of coliforms, Lactobacillus sp. and yeast decreased with age (P < 0.001). There was no significant effect of probiotics on the prevalence of phenotypic AMR and virulence genes in these studies. The prevalence of E. coli that was resistant to ampicillin and tetracycline, as well as carrying ≥3 virulence-associated genes, was greatest at the end of the starter phase (around 8 days old), before declining through the grower and finisher phases. There was only limited evidence that administering either CF or LP affected either the AMR or the virulence of E. coli in the bird. However, tetracycline resistance in E. coli was associated (P < 0.001, P < 0.01, P < 0.05, and P < 0.05) with the carriage of the iron uptake systems of E. coli D, iron-repressible protein, increased serum survival and temperature-sensitive haemagglutinin genes respectively, suggesting that the accumulation of iron and the genetic element conferring tetracycline resistance may be intertwined.

ABSTRACT

The study aims to research the effects of varied dietary sources of n-3 polyunsaturated fatty acids (PUFA) on the immune response in broiler chickens with stress on natural killer (NK) cell activity. Diets supplemented with one of the four sources of n-3 PUFA: linseed oil-, echium oil-, fish oil (FO) or algal biomass-enriched diets at levels of 18, 18, 50 and 15 g/kg fresh weight, were provided for one-d-old male Ross 308 broilers, totaling 340 in number, until they were slaughtered. The analyses included total lipid profile using gas chromatography (GC) for plasma, spleen, thymus, and blood. Additionally, NK cell activity and cell proliferation were investigated for thymocytes and splenocytes. The results indicated that the source of n-3 PUFA had a strong influence on fatty acid composition across all tissues. NK activity was highest in splenocytes and PBMCs from broilers fed linseed oil, followed by those fed algal biomass or echium oil, and lowest for those from broilers fed FO. The proliferative response of lymphocytes from algal biomass-fed chickens tended to be the highest, followed by those fed linseed oil in most cases. Lymphocytes from chickens fed fish oil showed the lowest proliferative response. These results could mean that a docosahexaenoic acid (DHA)-rich algal product might enrich chicken meat with n-3 PUFA without significant damaging effects on chicken immunity.

ABSTRACT

Condensed tannins (CTs), which extracted from yew leaves, tilia flower and black locust leaves, were examined for their antimicrobial in vitro activity against avian pathogenic Escherichia coli (APEC). Past research demonstrated that CTs which contain procyanidins and prodelphinidins that could inhibit the growth of a wide range of bacteria. However, there is no information on how these affect pathogenic bacteria from chickens such as APEC. The high concentration of extracts, 10, 5, 2·5 mg ml-1 , affected the growth curves of APEC, which gave different inhibition values for the three CT extracts. Furthermore, these CTs had significant effects (P ≤ 0·05) on APEC biofilm and motility depending on each CT concentration and composition. However, at low concentration (0·6 mg ml-1 ), the tilia flowers, a high molar percentage of procyanidins, enhanced bacterial cell attachment and improved the swimming motility of APEC. In contrast, yew, an equal molar percentage of procyanidins/prodelphinidins, and black locust, a high molar percentage of prodelphinidins, interrupted and blocked swarming and swimming motility. The data suggested that the antimicrobial activity of the CT extracts was elicited by a positive relationship between anti-biofilm formation and anti-motility capacities. SIGNIFICANCE AND IMPACT OF THE STUDY: This study showed that condensed tannins (CTs), which were a group of secondary metabolites of many plants and rich in prodelphinidins (PD), had greater antibacterial activity against avian pathogenic Escherichia coli (APEC) than CTs that were rich in procyanidins (PC). The mode of action of the CTs was to inhibit the swimming and swarming motility of APEC, and its ability to form biofilms. The significance of this finding is that the use of PD-rich CTs to control APEC should not encourage the development of antibiotic resistance by APEC because a different mechanism is used. If confirmed in vivo, this could provide the poultry industry with a valuable and novel means of controlling the antibiotic resistance.

ABSTRACT

This study aimed to determine the effects of supplementing pregnant heifers with the organic selenium (Se) source 2-hydroxy-4-methylselenobutanoic acid (HMSeBA) during the last 8 wk of pregnancy on dam and calf Se status. A total of 42 in-calf heifers were recruited to the study and randomly allocated to 1 of 3 treatments; a negative control (Con), sodium selenite (NaSe), or HMSeBA. Animals were blocked by body weight, body condition score, and expected calving date before treatment allocation. Following enrollment, all animals underwent a 7-wk wash-out period, after which they received their respective supplements, top-dressed daily onto a basal diet for the last 8 wk of pregnancy. Heifer blood samples were taken at weekly intervals from enrollment until 2 wk before expected calving date and as soon as possible after calving for determination of whole-blood glutathione peroxidase activity (GSH-Px) and plasma Se and malondialdehyde (MDA) concentrations. Selenized AA were determined in plasma samples taken at 3 wk precalving. A colostrum sample was taken as close to parturition as possible for determination of colostrum total Se, selenized AA, and IgG concentration. Calves were blood sampled as close to birth as possible for determination of whole-blood GSH-Px activity and plasma Se and MDA concentrations. Differences in whole-blood GSH-Px activity did not become apparent until calving; GSH-Px activity was lowest in Con heifers but similar between NaSe and HMSeBA heifers. Plasma Se was lowest in unsupplemented heifers and greatest in those supplemented with HMSeBA; this was attributable to greater selenomethionine concentrations in the plasma of HMSeBA heifers. Colostrum Se was lowest in Con heifers and greatest in HMSeBA heifers. The greater Se concentration of HMSeBA heifers was attributable to a greater proportion of total Se comprising selenocysteine; the reason for this is not known. There was no effect of supplementation on colostrum IgG concentration. Plasma Se was lowest in calves born to Con heifers and greatest in those born to HMSeBA heifers. There were no effects of treatment on calf whole-blood GSH-Px activity or plasma MDA concentration. The enhanced Se status associated with HMSeBA supplementation is likely a consequence of selenomethionine supply and may confer benefits to both the dam and her calf postpartum.

ABSTRACT

In the global South, dairying is often promoted as a means of poverty alleviation. Yet, under conditions of climate warming, little is known regarding the ability of small-scale dairy producers to maintain production and/or the robustness of possible adaptation options in meeting the challenges presented, particularly heat stress. The authors created a simple, deterministic model to explore the influence of breed and heat stress relief options on smallholder dairy farmers in Odisha, India. Breeds included indigenous Indian (non-descript), low-grade Jersey crossbreed and high-grade Jersey crossbreed. Relief strategies included providing shade, fanning and bathing. The impact of predicted critical global climate parameters, a 2°C and 4°C temperature rise were explored. A feed price scenario was modelled to illustrate the importance of feed in impact estimation. Feed costs were increased by 10% to 30%. Across the simulations, high-grade Jersey crossbreeds maintained higher milk yields, despite being the most sensitive to the negative effects of temperature. Low-capital relief strategies were the most effective at reducing heat stress impacts on household income. However, as feed costs increased the lower-grade Jersey crossbreed became the most profitable breed. The high-grade Jersey crossbreed was only marginally (4.64%) more profitable than the indigenous breed. The results demonstrate the importance of understanding the factors and practical trade-offs that underpin adaptation. The model also highlights the need for hot-climate dairying projects and programmes to consider animal genetic resources alongside environmentally sustainable adaptation measures for greatest poverty impact.

ABSTRACT

There is interest in the enrichment of poultry meat with long-chain n-3 polyunsaturated fatty acids in order to increase the consumption of these fatty acids by humans. However, there is concern that high levels of n-3 polyunsaturated fatty acids may have detrimental effects on immune function in chickens. The effect of feeding increasing levels of fish oil (FO) on immune function was investigated in broiler chickens. Three-week-old broilers were fed 1 of 4 wheat-soybean basal diets that contained 0, 30, 50, or 60 g/kg of FO until slaughter. At slaughter, samples of blood, bursa of Fabricius, spleen, and thymus were collected from each bird. A range of immune parameters, including immune tissue weight, immuno-phenotyping, phagocytosis, and cell proliferation, were assessed. The pattern of fatty acid incorporation reflected the fatty acid composition of the diet. The FO did not affect the weight of the spleen, but it did increase thymus weight when fed at 50 g/kg (P < 0.001). Fish oil also lowered bursal weights when fed at 50 or 60 g/kg (P < 0.001). There was no significant effect of FO on immune cell phenotypes in the spleen, thymus, bursa, or blood. Feeding 60 g/kg of FO significantly decreased the percentage of monocytes engaged in phagocytosis, but it increased their mean fluorescence intensity relative to that of broilers fed 50 g/kg of FO. Lymphocyte proliferation was significantly decreased after feeding broiler chickens diets rich in FO when expressed as division index or proliferation index, although there was no significant effect of FO on the percentage of divided cells. In conclusion, dietary n-3 polyunsaturated fatty acids decrease phagocytosis and lymphocyte proliferation in broiler chickens, highlighting the need for the poultry industry to consider the health status of poultry when poultry meat is being enriched with FO.

ABSTRACT

Supplementing broiler diets with conventional vegetable oils has little effect on the long-chain n-3 PUFA (LC n-3 PUFA) content of the meat. The present study investigated the effect on fatty acid composition and sensory characteristics of chicken meat when broilers were fed oil extracted from soyabeans (SDASOY) that had been genetically engineered to produce C18 : 4n-3 (stearidonic acid (SDA), 240 mg/g oil). Three diets were fed to 120 birds (eight replicate pens of five birds) from 15 d to slaughter (41-50 d). Diets were identical apart from the oil added to them (45 and 50 g/kg as fed in the grower and finisher phases, respectively), which was either SDASOY, near-isogenic soya (CON) or fish oil (FISH). The LC n-3 PUFA content of the meat increased in the order CON, SDASOY and FISH. In breast meat with skin, the SDA concentration was 522, 13 and 37 (sem 14·4) mg/100 g meat for SDASOY, CON and FISH, respectively. Equivalent values for C20 : 5n-3 (EPA) were 53, 13 and 140 (sem 8·4); for C22 : 5n-3 (docosapentaenoic acid (DPA)) 65, 15 and 101 (sem 3·5); for C22 : 6n-3 (DHA) 19, 9 and 181 (sem 4·4). Leg meat (with skin) values for SDA were 861, 23 and 68 (sem 30·1); for EPA 87, 9 and 258 (sem 7·5); for DPA 95, 20 and 165 (sem 5·0); for DHA 29, 10 and 278 (sem 8·4). Aroma, taste and aftertaste of freshly cooked breast meat were not affected. Fishy aromas, tastes and aftertastes were associated with LC n-3 PUFA content of the meat, being most noticeable in the FISH leg meat (both freshly cooked and reheated) and in the reheated SDASOY leg meat.

ABSTRACT

Human consumption of long-chain n-3 polyunsaturated fatty acids (LC n-3 PUFA) is below recommendations, and enriching chicken meat (by incorporating LC n-3 PUFA into broiler diets) is a viable means of increasing consumption. Fish oil is the most common LC n-3 PUFA supplement used but is unsustainable and reduces the oxidative stability of the mat. The objective of this experiment was to compare fresh fish oil (FFO) with fish oil encapsulated (EFO) in a gelatin matrix (to maintain its oxidative stability) and algal biomass at a low (LAG, 11), medium (MAG, 22), or high (HAG, 33 g/kg of diet) level of inclusion. The C22:6n-3 contents of the FFO, EFO, and MAG diets were equal. A control (CON) diet using blended vegetable oil was also made. As-hatched 1-d-old Ross 308 broilers (144) were reared (21 d) on a common starter diet then allocated to treatment pens (4 pens per treatment, 6 birds per pen) and fed treatment diets for 21 d before being slaughtered. Breast and leg meat was analyzed (per pen) for fatty acids, and cooked samples (2 pens per treatment) were analyzed for volatile aldehydes. Concentrations (mg/100 g of meat) of C20:5n-3, C22:5n-3, and C22:6n-3 were (respectively) CON: 4, 15, 24; FFO: 31, 46, 129; EFO: 18, 27, 122; LAG: 9, 19, 111; MAG: 6, 16, 147; and HAG: 9, 14, 187 (SEM: 2.4, 3.6, 13.1) in breast meat and CON: 4, 12, 9; FFO: 58, 56, 132; EFO: 63, 49, 153; LAG: 13, 14, 101; MAG: 11, 15, 102; HAG: 37, 37, 203 (SEM: 7.8, 6.7, 14.4) in leg meat. Cooked EFO and HAG leg meat was more oxidized (5.2 mg of hexanal/kg of meat) than the other meats (mean 2.2 mg/kg, SEM 0.63). It is concluded that algal biomass is as effective as fish oil at enriching broiler diets with C22:6 LC n-3 PUFA, and at equal C22:6n-3 contents, there is no significant difference between these 2 supplements on the oxidative stability of the meat that is produced.

ABSTRACT

The effect of poultry species (broiler or turkey) and genotype (Wrolstad or BUT T8 turkeys and Ross 308 or Cobb 500 broilers) on the efficiency with which dietary long-chain n-3 PUFA were incorporated into poultry meat was determined. Broilers and turkeys of both genotypes were fed one of six diets varying in FA composition (two replicates per genotype x diet interaction). Diets contained 50 g/kg added oil, which was either blended vegetable oil (control), or partially replaced with linseed oil (20 or 40 g/kg diet), fish oil (20 or 40 g/kg diet), or a mixture of the two (20 g linseed oil and 20 g fish oil/kg diet). Feeds and samples of skinless breast and thigh meat were analyzed for FA. Wrolstad dark meat was slightly more responsive than BUT T8 (P = 0.046) to increased dietary 18:3 concentrations (slopes of 0.570 and 0.465, respectively). The Ross 308 was also slightly more responsive than the Cobb 500 (P = 0.002) in this parameter (slopes of 0.557 and 0.449). There were no other significant differences between the genotypes. There was some evidence (based on the estimates of the slopes and their associated standard errors) that white turkey meat was more responsive than white chicken meat to 20:5 (slopes of 0.504 and 0.289 for turkeys and broilers, respectively). There was no relationship between dietary 18:3 n-3 content and meat 20:5 and 22:6 contents. If birds do convert 18:3 to higher FA, these acids are not then deposited in the edible tissues.

ABSTRACT

There is clear evidence of the nutritional benefits of consuming long-chain n-3 PUFA, which are found predominantly in oily fish. However, oily fish consumption, particularly in the United Kingdom, is declining, as is the consumption of all meats with the exception of poultry, which has increased in consumption by 73% in the last 30 yr. This pattern, if less marked, is reflected throughout Europe, and therefore one means of increasing long-chain n-3 PUFA consumption would be to increase the long-chain n-3 PUFA content in the edible tissues of poultry. This review considers the feasibility of doing this, concentrating particularly on chickens and turkeys. It begins by summarizing the benefits to human health of consuming greater quantities of n-3 FA and the sources of n-3 PUFA in the human diet. The literature on altering the FA composition of poultry meat is then reviewed, and the factors affecting the incorporation of n-3 PUFA into edible tissues of poultry are investigated. The concentration of alpha-linolenic acid (ALA) in the edible tissues of poultry is readily increased by increasing the concentration of ALA in the birds' diet (particularly meat with skin, and dark meat to a greater extent than white meat). The concentration of EPA in both white and dark meat is also increased when the birds' diet is supplemented with EPA, although supplementing the diet with the precursor (ALA) does not result in a noticeable increase in EPA content in the edible tissues. Although supplementing the birds' diets with relatively high concentrations of DHA does result in an increased concentration of DHA in the tissues, the relationship between dietary and tissue concentrations of DHA is much weaker than that observed with ALA and EPA. The impact that altering the FA composition of edible poultry tissue may have on the organoleptic and storage qualities of poultry products is also considered.
