ABSTRACT

Robust localization of self-reproducing autocatalytic chemistries is a key step in the realization of heritable and evolvable chemical systems. While autocatalytic chemical reaction networks already possess attributes such as heritable self-reproduction and evolvability, localizing functional multispecies networks within complex primitive phases, such as coacervates, has remained unexplored. Here, we show the self-reproduction of the Azoarcus ribozyme system within charge-rich coacervates where catalytic ribozymes are produced by the autocatalytic assembly of constituent smaller RNA fragments. We systematically demonstrate the catalytic assembly of active ribozymes within phase-separated coacervates-both in micron-sized droplets as well as in a coalesced macrophase, underscoring the facility of the complex, charge-rich phase to support these reactions in multiple configurations. By constructing multispecies reaction networks, we show that these newly assembled molecules are active, participating both in self- and cross-catalysis within the coacervates. Finally, due to differential molecular transport, these phase-separated compartments endow robustness to the composition of the collectively autocatalytic networks against external perturbations. Altogether, our results establish the formation of multispecies self-reproducing reaction networks in phase-separated compartments which in turn render transient robustness to the network composition.

ABSTRACT

Cleft lip and/or cleft palate (CL/P) is one of the most common congenital anomalies of the human face with a complex etiology involving multiple genetic and environmental factors. Several studies have shown the association of the paired box 7 ( PAX7 ) gene with CL/P in different populations worldwide. However, the current literature reveals no reported case-parent trio studies to evaluate the association between the PAX7 gene and the risk of nonsyndromic cleft lip and/or palate (NSCL/P) in the Indian population. Hence, the purpose of this study was to assess the PAX7 gene single nucleotide polymorphisms (SNPs) in the etiology of NSCL/P among the Indian cleft trios. Forty Indian case-parent trios of NSCL/P were included. The cases and their parents' genomic DNA were extracted. The SNPs rs9439714, rs1339062, rs6695765, rs742071, and rs618941of the PAX7 gene were genotyped using the Agena Bio MassARRAY analysis. The allelic transmission disequilibrium test was performed using PLINK software while pair-wise linkage disequilibrium by the Haploview program. The SNP rs9439714 showed evidence of association ( p -value = 0.02, odds ratio = 3) with NSCL/P. Considering the parent-of-origin effects, the SNPs rs9439714 and rs618941 showed an excess maternal transmission of allele C at rs9439714 ( p -value = 0.05) and G allele at rs618941 ( p -value = 0.04). The results of the present study suggested that the SNPs rs9439714 and rs618941 showed an excess maternal transmission of alleles suggestive of the possible role of the PAX7 gene involvement in the etiology of NSCL/P in the Indian population.

ABSTRACT

Graphene oxide (GO) was synthesized and characterized by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), atomic force microscopy (AFM), X-ray diffraction (XRD), Fourier transform-infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA). GO was then electro-chemically reduced and used for electrochemical study of mycophenolate mofetil (MMF). The electro-chemically reduced graphene oxide (ERGO) film on glassy carbon electrode (GCE) showed enhanced peak current for electrooxidation of MMF. MMF exhibited two irreversible oxidation peaks at 0.84 V (peak a1) and 1.1 V (peak a2). Effects of accumulation time, pH and scan rate were studied and various electro-chemical parameters were calculated. A differential pulse voltammetric method was developed for the determination of MMF in bulk samples and pharmaceutical formulations. Linear relationship was ob-served between the peak current and concentration of MMF in the range of 40 nM―15μM with a limit of detection of 11.3 nM. The proposed method is simple, sensitive and inexpensive and, hence, could be readily adopted in clinical and quality control laboratories.

ABSTRACT

STUDY DESIGN: A translation, cross-cultural adaptation, and validation study. OBJECTIVE: The aim of this study was to translate, adapt cross-culturally, and validate the Kannada version of the Oswestry Disability Index (ODI). SUMMARY OF BACKGROUND DATA: Low back pain is recognized as an important public health problem. Self-administered condition-specific questionnaires are important tools for assessing a patient. For low backache, the ODI is used widely. Preferred language of a region can have an effect on interpretation of questions and thus scoring. A search of literature showed no previously validated Kannada version of the ODI. METHODS: Cross-cultural adaptation and translation was carried out according to previously set guidelines. Patients were recruited from the orthopedic outpatient department. They filled out a booklet containing the Kannada version of the ODI, Kannada version of the Roland Morris Disability Questionnaire (RMDQ), and a 10-point visual analog scale for pain (VASpain). RESULTS: The Kannada ODI was answered by 91 patients and retested in 35 patients. After removing questionnaires with stray or ambiguous markings causing difficulty in computation of scores, 76 test questionnaires and 32 retest questionnaires were available for statistical analysis. The Kannada version showed an excellent internal consistency (Cronbach's alpha = 0.92). The Kannada version of the ODI showed good correlation with the RMDQ (r = 0.72) and moderate correlation with VASpain (r = 0.58). It also showed an excellent test-retest reliability (ICC = 0.96). Standard error of measurement (SEM) was also low (4.08) and a difference of 11 points is the "Minimum Detectable Change (MDC)." CONCLUSION: The Kannada version of the ODI that was developed showed consistency and reliability. It can be used for assessment of low back pain and treatment outcomes in Kannada-speaking populations. However, in view of a smaller sample size, it will benefit from verification at multiple centers and with more patients. LEVEL OF EVIDENCE: 3.

ABSTRACT

The mammalian vomeronasal organ detects social information about gender, status, and individuality. The molecular cues carrying this information remain largely unknown. Here, we show that small peptides that serve as ligands for major histocompatibility complex (MHC) class I molecules function also as sensory stimuli for a subset of vomeronasal sensory neurons located in the basal Gao- and V2R receptor-expressing zone of the vomeronasal epithelium. In behaving mice, the same peptides function as individuality signals underlying mate recognition in the context of pregnancy block. MHC peptides constitute a previously unknown family of chemosensory stimuli by which MHC genotypic diversity can influence social behavior.
