ABSTRACT

Anisotropic colloidal particles have the ability to self-assemble into cholesteric structures. We used molecular dynamics to simulate the self-assembly of ellipsoidal particles with the objective to establish a general framework to reveal the primary factors driving chiral interactions. To characterize these interactions, we introduce a characteristic parameter following the crowding factor (CF) theory. Our simulations and statistical analysis showed good agreement with the CF theory; at the early stages of the assembly process, the ellipsoidal particles go through a critical aggregation point followed by further clustering toward nematic order. Furthermore, we demonstrate that under high CF conditions, small initial clusters may induce a chiral twist, which subsequently forms a cholesteric structure with no directional preference in higher organization states.

ABSTRACT

The SREB (Super-conserved Receptors Expressed in Brain) family of G protein-coupled receptors is highly conserved across vertebrates and consists of three members: SREB1 (orphan receptor GPR27), SREB2 (GPR85), and SREB3 (GPR173). Ligands for these receptors are largely unknown or only recently identified, and functions for all three are still beginning to be understood, including roles in glucose homeostasis, neurogenesis, and hypothalamic control of reproduction. In addition to the brain, all three are expressed in gonads, but relatively few studies have focused on this, especially in non-mammalian models or in an integrated approach across the entire receptor family. The purpose of this study was to more fully characterize sreb genes in fish, using comparative genomics and gonadal expression analyses in five diverse ray-finned (Actinopterygii) species across evolution. Several unique characteristics were identified in fish, including: (1) a novel, fourth euteleost-specific gene (sreb3b or gpr173b) that likely emerged from a copy of sreb3 in a separate event after the teleost whole genome duplication, (2) sreb3a gene loss in Order Cyprinodontiformes, and (3) expression differences between a gar species and teleosts. Overall, gonadal patterns suggested an important role for all sreb genes in teleost testicular development, while gar were characterized by greater ovarian expression that may reflect similar roles to mammals. The novel sreb3b gene was also characterized by several unique features, including divergent but highly conserved amino acid positions, and elevated brain expression in puffer (Dichotomyctere nigroviridis) that more closely matched sreb2, not sreb3a. These results demonstrate that SREBs may differ among vertebrates in genomic structure and function, and more research is needed to better understand these roles in fish.

ABSTRACT

This article describes a process of fabricating highly porous paper from cellulosic fibers and carbon black (CB) with tunable conductivity. By embossing such paper, its porosity decreases while its conductivity increases. Tuning the porosity of composite paper alters the magnitude and trend of conductivity over a spectrum of concentrations of conductive particles. The largest increase in conductivity from 8.38 × 10-6 to 2.5 × 10-3 S/m by a factor of ∼300 occurred at a percolation threshold of 3.8 wt % (or 0.36 vol %) with the composite paper plastically compressed by 410 MPa, which caused a decrease of porosity from 88% to 42% on average. Our composite paper showed stable piezoresistive responses within a broad pressure range from 1 kPa up to 5.5 MPa for 800 cycles. The piezoresistive sensitivities of the composite paper were dependent on concentration and decreased with pressure. Composite paper with 7.5 wt % CB had sensitivities of -0.514 kPa-1 over applied pressures ranging from 1 to 50 kPa and -0.215 kPa-1 from 1 to 250 kPa. This piezoresistive paper with embossed patterns enabled touch sensing and detection of damage from darts and punches. Understanding the percolation behavior of three-phase composites (cellulosic fibers/conductive particles/air) and their response to damage, pressure, and processing conditions has the potential to enable scalable applications in prosthetics and robotics, haptic feedback, or structural health monitoring on expansive surfaces of buildings and vehicles.

ABSTRACT

Abstraction allows us to discern regularities beyond the specific instances we encounter. It also promotes creative problem-solving by enabling us to consider unconventional problem solutions. However, the mechanisms by which this occurs are not well understood. Because it is often difficult to isolate human high-level cognitive processes, we utilized a nonhuman primate model, in which rhesus monkeys appear to use similar processes to consider an unconventional solution to the difficult reverse-reward problem: i.e., given the choice between a better and worse food option they must select the worse one to receive the better one. After solving this problem with only one specific example-one vs. four half-peanuts-three of four monkeys immediately transferred to novel cases: novel quantities, food items, non-food items, and to the choice between a larger, but inferior vegetable and a smaller, but superior food item (either grape or marshmallow), in which they selected the inferior vegetable to receive the superior option. Thus, we show that nonhuman animals have the capacity to comprehend abstract non-perceptual features, to infer them from one specific case, and to use them to override the natural preference to select the superior option. Critically, we also found that three monkeys had a large learning and performance advantage over the fourth monkey who showed less generalization from the original one and four half-peanuts. This difference suggests that abstraction promoted problem-solving via cascading activation from the two food item options to the relation between them, thus providing access to an initially nonapparent problem solution.

ABSTRACT

We elucidate the effects of incorporating surfactants into electrospun poly (ɛ-caprolactone) (PCL) scaffolds on network homogeneity, cellular adherence and osteogenic differentiation. Lecithin was added with a range of concentrations to PCL solutions, which were electrospun to yield functionalized scaffolds. Addition of lecithin yielded a dose-dependent reduction in scaffold hydrophobicity, whilst reducing fiber width and hence increasing specific surface area. These changes in scaffold morphology were associated with increased cellular attachment of Saos-2 osteoblasts 3-h postseeding. Furthermore, cells on scaffolds showed comparable proliferation over 14 days of incubation to TCP controls. Through model-based interpretation of image analysis combined with gravimetric estimates of porosity, lecithin is shown to reduce scaffold porosity and mean pore size. Additionally, lecithin incorporation is found to reduce fiber curvature, resulting in increased scaffold specific elastic modulus. Low concentrations of lecithin were found to induce upregulation of several genes associated with osteogenesis in primary mesenchymal stem cells. The results demonstrate that functionalization of electrospun PCL scaffolds with lecithin can increase the biocompatibility and regenerative potential of these networks for bone tissue engineering applications. © 2017 The Authors Journal of Biomedical Materials Research Part A Published by Wiley Periodicals, Inc. ABSTRACT

Falls and diminished walking capacity are impairments common in persons with transfemoral amputation (TFA). Reducing falls and optimizing walking capacity through such means as achieving a more normal gait speed and community ambulation should be considered when formulating the prosthetic prescription. Because walking capacity and balance confidence are compromised with TFA, these outcomes should be considered when evaluating interfaces for transfemoral prosthetic users. The purpose of this study was to compare the effect of TFA interface design on walking capacity and balance confidence A retrospective cohort design was utilized involving unilateral TFA patients who used ischial ramus containment (IRC) and High-Fidelity (HiFi) interfaces (independent variables). Dependent variables included the Activity-specific Balance Scale (ABC) and the two-minute walk test (2MWT). Complete records were available for 13 patients (n = 13). The age range was 26 to 58 years. Three patients functioned at the K4 activity level, whereas all others functioned at the K3 level. Mean ABC scores were significantly different (p ≤ 0.05) at 77.2 (±16.8; 35.6 to 96.9) for IRC and 90.7 (±5.7; 77.5 to 98.7) for HiFi. The mean distance walked on the 2MWT was 91.8 m (±22.0, 58.3 to 124.7) for IRC compared to 110.4 m (±28.7; 64.7 to 171.1) for the HiFi socket (p ≤ 0.05). Alternative transfemoral interface design, such as the HiFi socket, can improve walking capacity and balance confidence in higher-functioning TFA patients.

ABSTRACT

The deformation micromechanics of bacterial cellulose (BC) and microfibrillated cellulose (MFC) networks have been investigated using Raman spectroscopy. The Raman spectra of both BC and MFC networks exhibit a band initially located at ≈ 1095 cm(-1). We have used the intensity of this band as a function of rotation angle of the specimens to study the cellulose fibril orientation in BC and MFC networks. We have also used the change in this peak's wavenumber position with applied tensile deformation to probe the stress-transfer behavior of these cellulosic materials. The intensity of this Raman band did not change significantly with rotation angle, indicating an in-plane 2D network of fibrils with uniform random orientation; conversely, a highly oriented flax fiber exhibited a marked change in intensity with rotation angle. Experimental data and theoretical analysis shows that the Raman band shift rate arising from deformation of networks under tension is dependent on the angles between the axis of fibrils, the strain axis, the incident laser polarization direction, and the back scattered polarization configurations. From this analysis, the effective moduli of single fibrils of BC and MFC in the networks were estimated to be in the ranges of 79-88 and 29-36 GPa, respectively. It is shown also that for the model to fit the data it is necessary to use a negative Poisson's ratio for MFC networks and BC networks. Discussion of this in-plane "auxetic" behavior is given.

ABSTRACT

Decision making is one of the principal cognitive processes underlying goal-directed behaviour and thus there is justifiably strong interest in modeling it. However, many of these models have yet to be tested outside of the laboratory. At the same time, field work would benefit from the use of experimental methods developed in the laboratory to determine the causal relationships between environmental variables and behaviour. We therefore adapted a laboratory-derived experimental paradigm to test decision making in the wild. The experiment used an indifference-point procedure to determine the influence of both the amount and distance of food on choice behaviour. Free-ranging rhesus monkeys were given the choice between a smaller amount of food at a closer distance and a larger amount farther away. In four conditions, we held the closer amount constant across trials and varied the farther amount to determine the point at which the monkeys were indifferent to the choice alternatives. For example, in condition one, we used one piece of food at the closer location, and determined how many pieces would be equivalent in the farther location. Four different closer amounts were tested to obtain an indifference point curve, with the indifference amounts at the farther location plotted against the closer amounts. The slope of the obtained linear indifference curve was surprisingly high, suggesting that rhesus monkeys significantly discount food that is farther away. Possible reasons for this steep spatial discounting are discussed.

ABSTRACT

This study shows that it is possible to affect the morphology and spreading of fibroblast cells on amino acid-modified cellulose-based fibrous networks. Hydrophilic (Gly, Ser), aliphatic (Ala, Val, Leu, Ile) and aromatic amino acids (Phe, Tyr, Trp), coupled to the cellulose via esterification, give rise to different cell morphologies. Modified cellulosic substrates are analysed using time of flight secondary ion mass spectrometry (TOF-SIMS), demonstrating that amino acids are coupled and uniformly distributed over the surface of the samples. Remarkably, it is shown that it is the aromatic amino acids, and in particular Trp, that give rise to significantly enhanced cell spreading. This enhanced effect is shown to be linked to an increase in the adsorption of fibronectin in the presence of aromatic bound amino acids.

ABSTRACT

We have utilized reversible cyclic peptides (RCPs)-peptides containing alternating l- and d-amino acids with N- and C-termini derivatized with thiol-containing groups allowing reversible peptide cyclization-to solubilize and noncovalently functionalize carbon single-walled nanotubes (SWNTs) in aqueous solution. Solubilization occurs through wrapping of RCPs around the circumference of a SWNT, followed by the formation of head-to-tail covalent bonds, yielding closed rings on the nanotubes. By controlling the length of the RCPs, we have demonstrated limited diameter-selective solubilization of the SWNTs as revealed by UV/vis/NIR and Raman spectroscopies, as well as atomic force microscopy.

ABSTRACT

The application of theoretical models to describe the structure of the types of fibrous network produced by the electrospinning of polymers for use in tissue engineering and a number of other applications is presented. Emphasis is placed on formal analyses of the pore size distribution and porosities that one would encounter with such structures and the nature of their relationships with other structural characteristics likely to be important for the performance of nanofibrous materials. The theoretical structures considered result from interactions between randomly placed straight rods that represent fibres with nanoscale dimensions. The dominant role of fibre diameter in controlling the pore diameter of the networks is shown and we discuss the perhaps counter-intuitive finding that at a given network mass per unit area and porosity, increasing fibre diameter results in an increase in mean pore radius. Larger pores may be required for ingrowth of cells to nanofibrous networks, hence this study clarifies that simply making the diameters of the fibres smaller might not be the way to improve cell proliferation on such substrates. An extensive review of structural features of the network such as the distribution of mass, inter-fibre contacts and available surface for cell attachment, fibre contact distributions for integrity of the networks and the porosity and pore size distributions is given, with emphasis placed on nanofibre dimensions for the first time.

ABSTRACT

OBJECTIVES: This study was conducted to test the effectiveness of an abbreviated cognitive-behavioral insomnia therapy (ACBT) with primary DESIGN: A single-blind, randomized group design was used in which study patients were randomized to either a brief, 2-session ACBT or a similarly brief intervention (SHC) that included only generic sleep hygiene recommendations. SETTING: A university-affiliated Department of Veterans Affairs medical center. PARTICIPANTS: Twenty (2 women) veteran patients (M(age) = 51.0 yrs., SD = 13.7 years) who met criteria for chronic primary insomnia. MEASUREMENTS AND RESULTS: Participants completed sleep logs for 2 weeks and questionnaires to measures insomnia symptoms, sleep-related self-efficacy, and dysfunctional beliefs about sleep before treatment, during a 2-week posttreatment assessment, and again at a 3-month posttreatment follow-up. Statistical analyses showed that ACBT produced significantly larger improvements across a majority of outcome measures than did SHC. Case-by-case analyses showed that only the ACBT produced consistent positive effects across study patients, and a sizeable proportion of these patients receiving this treatment achieved clinically significant improvements by their study endpoints. Approximately 52% of those receiving the ACBT reported at least a 50% reduction in their wake time after sleep onset, and 55.6% of ACBT-treated patients who entered the study with pathologic scores on an Insomnia Symptom Questionnaire (ISQ), achieved normal ISQ scores by their final outcome assessment. CONCLUSIONS: ACBT is effective for reducing subjective sleep disturbance and insomnia symptoms in primary care patients.

ABSTRACT

The current study examined service era differences in a sample of 172 Gulf and Vietnam outpatient veterans with combat-related posttraumatic stress disorder (PTSD). Participants completed the MMPI-2 and several additional self-report measures of symptom severity (PTSD, depression, anxiety, hostility, and health complaints). Results indicated that MMPI-2 profiles differed significantly according to service era with Vietnam veterans scoring higher on scales 2, 8, and 0 and lower on scale 9 than did Gulf veterans. Examination of group means derived from parametric analysis of MMPI-2 data suggested a mean two-point code type of 2-8/8-2 for Vietnam veterans and 1-8/8-1 for Gulf veterans. In contrast, when the data were examined using descriptive techniques based on frequency counts of individual MMPI-2 profiles, the most frequently occurring two-point codetype was 7-8/8-7 for Vietnam veterans, and 6-8/8-6 for Gulf veterans. In addition, Gulf veterans reported a greater number of total health complaints than Vietnam veterans, whereas Vietnam veterans reported a greater number of physician-diagnosed physical conditions. Potential advantages of incorporating descriptive approaches versus parametric methods when examining profile data are also presented.
