ABSTRACT

Marine organisms have evolved to survive against predators in complex marine ecosystems via the production of chemical compounds. Soft corals (Cnidaria, Anthozoa, Octocorallia) are an important source of chemically diverse metabolites with a broad spectrum of biological activities. Herein, we perform a comparative study between high-resolution proton nuclear magnetic resonance (1H-NMR) and pure shift yielded by chirp excitation (PSYCHE) experiments to analyze the metabolic profile of 24 soft corals from the Colombian Caribbean to correlate chemical fingerprints with their cytotoxic activity against three cancer cell lines (human cervical carcinoma (SiHa), human prostatic carcinoma (PC3) and human lung adenocarcinoma (A549)). All data obtained were explored using multivariate analysis using principal components analysis (PCA) and orthogonal partial least squares (OPLS) analysis. The results did not show a significant correlation between clusters using 1H-NMR data in the PCA and OPLS-DA models and therefore did not provide conclusive evidence; on the other hand, a metabolomic analysis of PSYCHE data obtained under the same parameters revealed that when a decoupled experiment is performed, it was possible to establish a statistically valid correlation between the chemical composition of soft corals and their cytotoxic activity against the PC3 cancer cell line, where the asperdiol and plexaurolone markers were putatively identified and related to the cytotoxic activity presented by extracts of Plexaurella sp. and Plexaura kukenthali, respectively. These results increase the speed, effectiveness and reliability of analyses for the study of this type of complex matrices.

ABSTRACT

La participación del Fonoaudiólogo en la rehabilitación neuromuscular orofacial y musculatura implicada en la mecánica deglutoria, se ha complementado con el uso de la terapia eléctrica funcional; esta carece de evidencia científica que respalde su efectividad en los procesos de recuperación funcional. Objetivo: Realizar una revisión documental sobre el uso de la terapia eléctrica funcional en el manejo de la disfagia, con el fin de crear un soporte teórico que respalde los procesos de intervención en fonoaudiología. Metodología: La revisión de la literatura se realizó en las bases de datos de Proquest, EBSCO, Scielo, Redalyc y Medic Latina. Fueron revisados artículos entre el 2008 y 2018. Para la selección de los artículos se utilizó el diagrama de flujo PRISMA (Preferred Reporting Hems for Systematic Reviews and Meta analyses). Resultados: Se revisaron 2980 artículos para exclusión por título, quedando 66 artículos en la primera depuración, de los cuales 21 fueron descartados por no cumplir con ningún criterio propuesto, 10 por repetición y 12 por no permitir el acceso al documento. Finalmente fueron considerados 23 artículos y 5 fuentes de consulta física para realizar el respectivo análisis. Conclusiones: Existe escasa evidencia científica que respalde la intervención fonoaudiológica en la disfagia con electroterapia.


The participation of the speech therapist within the orofacial neuromuscular rehabilitation and the musculature involved in swallowing mechanics, has been complemented with the use of functional electrical therapy, which has lacked scientific evidence that supports its effectiveness in faster recovery processes in terms of functionality. Objective: To carry out a documentary review of information related to the use of functional electric therapy on the management of dysphagia, to create a theoretical support that supports the processes of speech therapy intervention. Methodology: The review of the literature was carried out in the databases of Proquest, EBSCO, Scielo, Redalyc and Medic Latina. Articles were reviewed between 2008 and 2018. For the selection of the articles the PRISMA flow chart (Preferred Reporting Hems for Systematic Reviews and Meta analyzes) was used. Results: 2980 articles were reviewed for exclusion by title, leaving 66 articles in the first debugging, of which 21 were discarded for not complying with any proposed criteria, 10 for repetition and 12 for not allowing access to the document. Finally, 23 articles and 5 sources of physical consultation were considered to perform the respective analysis. Conclusions: There is little scientific evidence to support the speech therapy intervention in dysphagia with electrotherapy.

ABSTRACT

Soft corals (Cnidaria, Anthozoa, Octocorallia) are a diverse group of marine invertebrates that inhabit various marine environments in tropical and subtropical areas. Several species are recognized as prolific sources of compounds with a wide array of biological activities. Recent advances in analytical techniques, supported by robust statistical analyses, have allowed the analysis and characterization of the metabolome present in a single living organism. In this study, a liquid chromatography-high resolution mass spectrometry metabolomic approach was applied to analyze the metabolite composition of 28 soft corals present in the Caribbean coast of Colombia. Multivariate data analysis was used to correlate the chemical fingerprints of soft corals with their cytotoxic activity against tumor cell lines for anticancer purpose. Some diterpenoids were identified as specific markers to discriminate between cytotoxic and non-cytotoxic crude extracts of soft corals against tumor cell lines. In the models generated from the comparative analysis of PLS-DA for tumor lines, A549 and SiHa, the diterpene 13-keto-1,11-dolabell-3(E),7(E),12(18)-triene yielded a high score in the variable importance in projection. These results highlight the potential of metabolomic approaches towards the identification of cytotoxic agents against cancer of marine origin. This workflow can be useful in several studies, mainly those that are time consuming, such as traditional bioprospecting of marine natural products.

ABSTRACT

The qualitative and quantitative anthocyanin composition of four wild tropical fruits from Colombia was studied. Compounds of "mora pequeña" ( Rubus megalococcus Focke.), "uva de árbol" ( Myrciaria aff. cauliflora O. Berg), coral, and motilón ( Hyeronima macrocarpa Mull. Arg.) fruits were separately extracted with methanol-acetic acid (95:5, v/v). The anthocyanin-rich extracts (AREs) were obtained by selective adsorption on Amberlite XAD-7. Each extract was analyzed by HPLC-PDA and HPLC-HRESI-MS(n) with LCMS-IT-TOF equipment in order to characterize the anthocyanin pigments and the coinjection in HPLC using standards allowed identifying the major constituents in each extract. The antioxidant activity was measured by electron paramagnetic resonance (EPR) and UV-vis spectroscopy, using ABTS and DPPH free radicals. The ARE of motilón ( H. macrocarpa Müll. Arg) exhibited the highest radical scavenging activity in comparison to the other extracts. A second-order kinetic model was followed in all of the cases. These results suggested that the studied fruits are promising not only as source of natural pigments but also as antioxidant materials for food industry.
