ABSTRACT

Results of the analysis of a number of parameters which determine the efficiency of using the photodynamic action for treating rheumatoid arthritis are reported. The investigations are based on determining the character of sensitizer stabilization in the joint tissue and evaluating its stability. As the sensitizer chlorin e6 was chosen. We have established the fact of contrast accumulation of chlorin e6 in the synovial membrane and cartilage, developed the system of intrajoint introduction of the pigment, studied the kinetics of sensitizer destruction under irradiation.

ABSTRACT

The influence of the chemical structure of porphyrin pigments on their accumulation and localization in HeLa cells has been examined by the scanning fluorescence microphotometry. It has been found that the replacement of carboxyl groups of chlorine e6 for methyl and amino groups has no influence on the pigment distributions in cells. All the pigments are bound by cell membrane structures. The chemical modification of chlorine e6 structure is essential for the ability of pigment to be accumulated by cells that can be used to increase the efficiency of cancer phototherapy. The charge and hydrophobic properties of pigment molecules are of great importance for accumulating porphyrin sensitizers by cells.

ABSTRACT

Kinetics of hemolysis of human erythrocytes photosensitized by porphyrin and chlorin derivatives was investigated by small angle light scattering method. The compounds used were arranged in the following order of photosensitizing activity decrease: ethylendiamide of chlorine e6, chlorine e6 dimethylester chlorine e6 haematoporphyrin diacetate tetracarboxyphenylporphyn. The substances having a greater number of binding sites with liposomes (chlorine e6 dimethylester) or with albumin (ethylendiamide of chlorine e6) showed the greatest photohemolytic activity. The photohemolytic activity of porphyrins was suggested to depend on the number of the pigment molecules bound by erythrocytes membranes.

ABSTRACT

Hemolysis of human erythrocytes photosensitized by chlorophyll derivative--chlorin e6 has been studied. This process was found to be oxygen-dependent. High photodynamic activity of chlorin e6 is explained both by its specific binding with cells and by its photophysics parameters.

ABSTRACT

The experimental investigation of the influence of HCl and Zn, Cd and Cu acetates on association of pheophytin alpha and rodin g7 was carried out in water-ethanolic (4 : 1) media. It is shown that during acidation of a binary solvent mixtures containing pheophytin alpha associate, the formation of product with the absorption maximum 702nm takes place, which is the pigment associated form. The formation of the same product is found under the action of metal acetates on pigment associated complexes. It is assumed that under treatment of water-ethanolic solutions of associates of pheophytin and its derivatives by hydrochloric acid and metal salts. at first introduction of these compound molecules into the aggregate sphere takes place and then the incorporation of the H+ and Me2+ positive ions into the centre of an associate dimer cell occurs along with the transformation of the structure of the pigment associated complexes.

ABSTRACT

Protochlorophyll with a high yield is isolated under oxidation of chlorophyll a by tetrachloro-o-quinone. A pigment, designated as protochlorophyll b, is isolated by the same way from chlorophyll b. A method of purification of the pigments obtained is worked out, their properties are studied and their structures are identified. The proposed method can be recommended for isolation of protochlorophyll a and b.
