ABSTRACT

Japanese quail (Coturnix japonica) is an avian model used to evaluate the reproductive and developmental toxicity of chemicals. The National Institute for Environmental Studies (NIES) of Japan established a strain of Japanese quail, NIES-L, which may be a better model because of its highly inbred characteristics. To understand sexual differentiation of the reproductive organs and the value of using NIES-L quails for avian toxicity assessment, we profiled estradiol and androgen plasma levels by enzyme-linked immunosorbent assay; the mRNA levels of estrogen receptor-α (ERα), ERß, and androgen receptor (AR) in the gonads, Müllerian ducts, Wolffian ducts; and the mRNA levels of steroidogenic enzymes, cholesterol side chain cleavage enzyme (P450scc), 17α-hydroxylase/C17-20 lyase (P45017α, lyase), 3ß-hydroxysteroid dehydrogenase (3ß-HSD), 17ß-hydroxysteroid dehydrogenase (17ß-HSD), and aromatase (P450arom), anti-Müllerian hormone (AMH), and AMH receptor type 2 (AMHR2) in the gonads of NIES-L Japanese quails on embryonic days 9, 12, and 15 using a real-time quantitative PCR method. The plasma estradiol concentration was higher in females than males on these embryonic days, but no sex difference was found in the plasma androgens. The mRNA levels of all examined steroidogenic enzymes were significantly higher in female than male embryos. In particular, the P450arom mRNA levels showed a striking sex difference: P450arom was expressed in female but not male gonads. In contrast, the AMH and AMHR2 mRNA levels in the gonads were higher in males than females. The ERα, ERß, and AR mRNA levels increased in the left female gonad and peaked on embryonic day 15, but not in the left and right male gonads; therefore, there was a female-biased sex difference. The ERα, ERß, and AR mRNA levels in the left Müllerian duct, but not in the right Müllerian duct, of females increased and peaked on embryonic day 15, which resulted in asymmetric mRNA levels. The Wolffian ducts expressed ERα, ERß, and AR in both sexes, and no sex difference or asymmetry of mRNA levels was found. The information obtained from this study helps elucidate the molecular endocrinological basis of sexual dimorphism formation of reproductive organs and clarify the value of NIES-L quails for toxicity assessment.

ABSTRACT

The present study aimed to measure the levels of coagulation factors in stored whole blood of pregnant women and to determine their usefulness in treating pregnant women who developed coagulopathy. A prospective study to measure coagulation factors in stored donated whole blood from pregnant and non-pregnant women was conducted. Fibrinogen, FV, FVII, FVIII, FXIII, and von Willebrand factor were measured in blood stored at 4°C for 0, 1, 3, and 5 weeks. All coagulation factors except for factor XIII decreased during storage. Fibrinogen and factor VII in the blood collected from pregnant women gradually decreased over time and their levels were significantly higher after 5 weeks of storage than those of non-pregnant women at week 0. Whole blood donated by pregnant women for autologous blood transfusion and stored at 4°C may be expected being effectively for the prevention of coagulopathy and the treatment of circulatory blood volume loss.

ABSTRACT

Currently, no simple assays are available for routine quantitative detection of Escherichia coli-produced Shiga toxin 2e (Stx2e) that causes porcine edema disease. Here, we present a novel quantitative detection method for Stx2e based on the measurement of Stx2e binding to the specific globotetraosylceramide (Gb4) receptor by ELISA (Gb4-ELISA). No cross-reactivity was found with the other Shiga toxins Stx1 and Stx2, indicating high specificity. When the recombinant Stx2e B subunit (Stx2eB) was used, the absorbance measured by Gb4-ELISA increased linearly with Stx2eB concentration in the range of 20-2,500 ng/ml. The Gb4-ELISA method can be easily performed, suggesting that it would be a useful diagnostic tool for porcine edema disease.

ABSTRACT

In May 2006, the positive list system for residues of pesticide and veterinary medicines in food was introduced in Japan. Under this system, the number of regulated pesticides remarkably increased. The information of food items and foods/origins combinations in which pesticides were frequently detected over the detection limits and/or exceeding MRL in the foreign pesticides residue monitoring are useful to achieve the Japanese pesticides residue monitoring programme more efficiently and effectively. We investigated pesticides residue monitoring data published by food safety authorities of North America, Europe and Oceania countries in their web sites. We focused food items and foods/origins combinations which Japan imported in large quantity. Most of pesticides residues detected in twelve food items were mainly fungicides and insecticides, and some residues level of their samples were exceeding the MRLs (maximum residue limits) established in Japan. Most of pesticides residues detected in European countries were also fungicides (e.g. maneb-group and procymidone) and insecticides (e.g. pirimiphos-methyl and malathion), but herbicides were less detected. We provided the collection of Web links to information sites of MRLs and the pesticides residue monitoring reports published by food safety authorities of other countries.

ABSTRACT

Usage of pesticides in food items in export countries was studied, focusing items which Japan imports in large quantity. Japan has imported field crops such as wheat, corn and soy bean, and also grapefruit in large quantity on a weight base, mainly from United States, Australia and Canada. While, Japan has imported various kinds of vegetables in which China had the largest share. We collected usage data of pesticides for 44 food items of 17 countries of 2004. Pesticides which were used frequently (usage rank within top ten in each item/country) were dichlorvos, carbofuran, chlorpyrifos, dimethoate (insecticides), mancozeb, carbendazim, thiophanate-methyl, chlorthalonil (fungicides), glyphosate, 2,4-D, paraquat, acetochlor (herbicides). Carbendazim, thiophanate-methyl, acetochlor and dichlorvos were mainly used in China. Dithiocarbamates are used frequently in various food items in various countries, and also frequently detected in monitoring in foreign countries. Some pesticides such as bisultap, monosultap, etaboxam and triazmate were used only in certain countries, and available information on toxicity or analytical method was very limited. Some of pesticides described above have not been analyzed in the pesticide residue monitoring in Japan before 2005,however, many of them are subjects of analysis for import food after 2006 with the enforcement of positivelist system for residues of pesticide and veterinary medicines in food in Japan.

ABSTRACT

Databases for ADI (Acceptable Daily Intake) and relevant information on food additives, pesticides and veterinary drugs were developed. The databases we developed are easily accessible on the web, and contain ADIs, latest evaluation year, classification and use, as well as synonym and CAS registry number. The databases are designed to be easily updated by researchers as ADI and relevant information are updated or added without delay. The database for food additives has already provided from the homepage of NIHS, and the access log of the web site was 1325/month in December 2005 and 2179/month in March 2006.
