ABSTRACT

The development of geopolymer building composites at a lower cost with a smaller carbon footprint may lessen the growing concerns about global warming brought on by emissions of a critical greenhouse gas (CO2) paired with the high production costs in the cement sector. Diatomaceous earth, commonly used as an admixture or partial replacement of cement owing to its most effective pozzolanic properties, has been investigated as a precursor in geopolymer concrete development. Several studies have been examined to develop a greater understanding of its characterization, inclusion status, and impacts on the performance aspects of concrete. The literature review showed that using diatomaceous earth is one of the effective ways to create sustainable, insulating, lightweight building materials while minimizing the harmful economic and environmental effects of industrial solid wastes. However, since most studies have focused on its integration as a partial cement substitute or a replacement for fine aggregate, further research on diatomaceous earth-based clinker-free concrete is required. A lack of research on geopolymer concrete's reinforcement with either natural or synthetic fibers, or a combination of the two, was also discovered. This review also showed that there has been remarkably little effort made towards theoretical property correlation modeling for predicting concrete performance. It is anticipated that the detailed overview presented herein will guide potential researchers in defining their future paths in the study area.

ABSTRACT

Abstract The first report of the genus and species Zefevazia peruana (Boucomont, 1902) for Ecuador is presented. The species is previously known only from a few localities in northern Peru, and is now also recorded from Santa Elena, southern Ecuador. This report represents the northernmost record of the genus and species.


Resumen Se presenta el primer reporte del género y la especie Zefevazia peruana (Boucomont, 1902) para Ecuador. La especie es conocida previamente de unas pocas localidades en el norte de Perú, y ahora también es registrada en Santa Elena, sur de Ecuador. Este reporte representa el registro más al norte del género y la especie.

ABSTRACT

Identifying dietary nutrient gaps and interpreting their public health significance are essential for improving poor diets and reducing malnutrition. Evidence indicative of the burden of nutrient deficiencies and inadequate nutrient intake or availability exists in many countries yet is often misinterpreted or underused in decision-making. Clear guidance is lacking on how to synthesize and interpret the relevant evidence, which comes in many forms. To fill this methodological gap, an approach called Comprehensive Nutrient Gap Assessment was created to enable use of existing evidence to assess the public health significance of nutrient gaps and identify evidence gaps. Comprehensive Nutrient Gap Assessment requires ≥ 2 experts in nutritional assessment but does not require primary data collection or secondary quantitative data analysis. It can be implemented relatively quickly with low costs, for specific countries and subnational regions, and updated on the basis of new data with minimal effort. The findings from a Comprehensive Nutrient Gap Assessment are easily interpretable by nontechnical decision makers yet include clear justification for technical audiences.

ABSTRACT

Clearance of the airway is dependent on directional mucus flow across the mucociliary epithelium, and deficient flow is implicated in a range of human disorders. Efficient flow relies on proper polarization of the multiciliated cells and sufficient ciliary beat frequency. We show that NO, produced by nNOS in the multiciliated cells of the mouse trachea, controls both the planar polarity and the ciliary beat frequency and is thereby necessary for the generation of the robust flow. The effect of nNOS on the polarity of ciliated cells relies on its interactions with the apical networks of actin and microtubules and involves RhoA activation. The action of nNOS on the beat frequency is mediated by guanylate cyclase; both NO donors and cGMP can augment fluid flow in the trachea and rescue the deficient flow in nNOS mutants. Our results link insufficient availability of NO in ciliated cells to defects in flow and ciliary activity and may thereby explain the low levels of exhaled NO in ciliopathies.

ABSTRACT

The objective of this study was to estimate the prevalence of canine visceral leishmaniasis (CVL) and to identify the differences in associated factors to its occurrence in urban area and countrysides in the municipality of Santa Luzia located in the semi-arid region of Paraíba. In the years 2015 and 2016, 779 blood samples from dogs were collected. The prevalence was determined by three serological techniques, ELISA-S7® Kit, DPP® Rapid Test and EIE-LVC® Kit, considering positive the samples that reacted in at least two assays. Associated factors were determined by univariate and multivariate analyzes of the guardians' responses to the epidemiological questionnaire. The prevalence of anti-Leishmania infantum antibodies in the studied municipality was 15.00% (117/779), being higher in the urban area (15.20%) than in the countryside (13.60%). The neighborhood with the highest prevalence was Frei Damião with 26.40% (33/125), being considered a hotspot (OR 1.245, p=0.007). Other associated factors were the semi-domiciliary breeding (OR 1.798, p=0.025), in the urban area, and hunting dog (OR 18.505, p=0.016), contact with cattle (OR 17.298, p=0.022) and environment where the dog is raised (OR 4.802, p=0.024) in the countryside. In the municipality of Santa Luzia, the prevalence of canine visceral leishmaniasis is high and the disease is widely distributed. Epidemiological differences between urban area and the countryside could be observed demonstrating the need for more adequate control measures for each locality and proving the urbanization process.(AU)


O objetivo desta pesquisa foi estimar a prevalência da leishmaniose visceral canina (LVC) e identificar as diferenças nos fatores relacionados à sua ocorrência nas zonas urbana e rural do município de Santa Luzia, localizado no semiárido paraibano. ABSTRACT

A qualitative 3 min one-step assay for detecting beta-lactam, sulfonamide, and tetracycline antibiotics was validated following milk screening test guidelines developed by FDA-CVM, AOAC-RI, and IDF. The validated 90% detection levels with 95% confidence were: penicillin G 2 part per billion (ppb); amoxicillin 4 ppb; ampicillin 9 ppb; ceftiofur plus metabolites 50 ppb; cloxacillin 9 ppb; cephapirin 15 ppb; sulfadimethoxine 8 ppb; sulfamethazine 9 ppb; chlortetracycline 34 ppb; oxytetracycline 53 ppb; and tetracycline 42 ppb. Detection levels were lower than U.S. and Canadian allowable limits for milk and were consistent with most European Maximum Residue Limits. Tests of raw commingled cows' milk indicated a low positive error rate of <0.3% with no interferences demonstrated by 1.08 MM/mL somatic cells, Gram-positive or Gram-negative bacteria < 300 K/mL, freeze/thawing, or non-targeted drugs. Detection of incurred residues were similar to, or more sensitive to, fortified samples. Some cross reactivity across drug families occurred in interference studies and therefore initial positive samples should be confirmed with drug family specific screening methods. The National Conference of Interstate Milk Shipments approval as a bulk tank/tanker screening test was completed in three stages for each drug family, including a tetracycline confirmation procedure to target U.S. tolerance levels. Detection and robustness were found to be appropriate for multiple countries' regulatory requirements for screening tests. The method development, validation, and approval was intended to diversify and increase the verification tools for the control of the major antibiotic drug families used in managing cows' health and welfare.

ABSTRACT

BACKGROUND: Adolescence presents an opportunity to influence diet, which impacts present and future health outcomes, yet adolescent diets globally are poorly understood. OBJECTIVE: We generate evidence on adolescent diets globally and explore patterns and trends by subpopulation. METHODS: We estimated mean frequency of consumption and prevalence of less-than-daily fruit and vegetable consumption, at-least-daily carbonated beverage consumption, and at-least-weekly fast-food consumption among school-going adolescents aged primarily 12 to 17 years from the Global School-based Student Health Surveys in Africa, Asia, Oceania, and Latin America between 2008 and 2015. Random-effects meta-analysis was used to pool estimates globally and by subgroup. RESULTS: On average, adolescents consumed fruit 1.43 (95% confidence interval [CI] 1.26-1.60) times per day, vegetables 1.75 (1.58-1.92) times per day, carbonated soft drinks 0.99 (0.77-1.22) times per day, and fast food 1.05 (0.78-1.32) times per week. Overall, 34.5% (95% CI 29.4-39.7) consumed fruit less than once per day, 20.6% (15.8-25.9) consumed vegetables less than once per day, 42.8% (35.2-50.7) drank carbonated soft drinks at least once per day, and 46.1% (38.6-53.7) consumed fast food at least once per week. Mean daily frequency of fruit consumption was particularly low in South and East Asia (1.30 [1.02-1.58]); carbonated soft drink consumption high in Latin America (1.54 [1.31-1.78]), high-income countries (1.66 [1.29-2.03]), and modern food system typologies (1.44 [0.75-2.12]); and mean weekly fast food consumption high in mixed food system typologies (1.29 [0.88-1.71]). CONCLUSIONS: School-going adolescents infrequently consume fruits and vegetables and frequently consume carbonated soft drinks, but there is wide variability by subpopulation.

ABSTRACT

Eggs are a highly nutritious food but have been shown to be infrequently consumed in many low-income countries, especially by women and children. We collate country-level data on egg production, availability, consumption, prices, industry structure, and contextual trends and use these to estimate current patterns and likely future outcomes under four alternative scenarios. These scenarios are as follows: incremental change based on expected economic growth and urbanisation (the base scenario); enhanced productivity of independent small producers; aggregated production in egg hubs; and the accelerated spread of large-scale intensive production. All scenarios are modelled out to 2030 using a mix of regression and deterministic models. We find that children's consumption of eggs is highly correlated with national availability, and both are a function of egg prices. Eggs are unavailable, expensive, and infrequently consumed by children in much of South Asia and sub-Saharan Africa. The base scenario results in modest increases in production in low-income regions. Focusing efforts on independent small producers can only boost rural consumption in a handful of countries where poultry ownership is unusually high and would be expensive and logistically challenging to scale. Aggregation of production, with minimum flock sizes of 5,000 layers per farm, is a more promising pathway to increasing availability in rural areas. To meet the needs of urban populations, large-scale intensive production is needed. Intensive production brings down prices significantly, allowing many more poor households to access and consume eggs. Recent experience in countries such as Thailand confirms that this is both feasible and impactful.

ABSTRACT

Multiciliated cells of the airways, brain ventricles, and female reproductive tract provide the motive force for mucociliary clearance, cerebrospinal fluid circulation, and ovum transport. Despite their clear importance to human biology and health, the molecular mechanisms underlying multiciliated cell differentiation are poorly understood. Prior studies implicate the distal appendage/transition fiber protein CEP164 as a central regulator of primary ciliogenesis; however, its role in multiciliogenesis remains unknown. In this study, we have generated a novel conditional mouse model that lacks CEP164 in multiciliated tissues and the testis. These mice show a profound loss of airway, ependymal, and oviduct multicilia and develop hydrocephalus and male infertility. Using primary cultures of tracheal multiciliated cells as a model system, we found that CEP164 is critical for multiciliogenesis, at least in part, via its regulation of small vesicle recruitment, ciliary vesicle formation, and basal body docking. In addition, CEP164 is necessary for the proper recruitment of another distal appendage/transition fiber protein Chibby1 (Cby1) and its binding partners FAM92A and FAM92B to the ciliary base in multiciliated cells. In contrast to primary ciliogenesis, CEP164 is dispensable for the recruitment of intraflagellar transport (IFT) components to multicilia. Finally, we provide evidence that CEP164 differentially controls the ciliary targeting of membrane-associated proteins, including the small GTPases Rab8, Rab11, and Arl13b, in multiciliated cells. Altogether, our studies unravel unique requirements for CEP164 in primary versus multiciliogenesis and suggest that CEP164 modulates the selective transport of membrane vesicles and their cargoes into the ciliary compartment in multiciliated cells. Furthermore, our mouse model provides a useful tool to gain physiological insight into diseases associated with defective multicilia.

ABSTRACT

Chibby1 (Cby1) is a small, conserved coiled-coil protein that localizes to centrioles/basal bodies and plays a crucial role in the formation and function of cilia. During early stages of ciliogenesis, Cby1 is required for the efficient recruitment of small vesicles at the distal end of centrioles to facilitate basal body docking to the plasma membrane. Here, we identified family with sequence similarity 92, member A (FAM92A) and FAM92B, which harbor predicted lipid-binding BAR domains, as novel Cby1-interacting partners using tandem affinity purification and mass spectrometry. We found that in cultured cell lines, FAM92A colocalizes with Cby1 at the centrioles/basal bodies of primary cilia, while FAM92B is undetectable. In airway multiciliated cells, both FAM92A and -92B colocalize with Cby1 at the base of cilia. Notably, the centriolar localization of FAM92A and -92B depends largely on Cby1. Knockdown of FAM92A in RPE1 cells impairs ciliogenesis. Consistent with the membrane-remodeling properties of BAR domains, FAM92A and -92B in cooperation with Cby1 induce deformed membrane-like structures containing the small GTPase Rab8 in cultured cells. Our results therefore suggest that FAM92 proteins interact with Cby1 to promote ciliogenesis via regulation of membrane-remodeling processes.

ABSTRACT

Motile multiciliated cells (MCCs) have critical roles in respiratory health and disease and are essential for cleaning inhaled pollutants and pathogens from airways. Despite their significance for human disease, the transcriptional control that governs multiciliogenesis remains poorly understood. Here we identify TP73, a p53 homolog, as governing the program for airway multiciliogenesis. Mice with TP73 deficiency suffer from chronic respiratory tract infections due to profound defects in ciliogenesis and complete loss of mucociliary clearance. Organotypic airway cultures pinpoint TAp73 as necessary and sufficient for basal body docking, axonemal extension, and motility during the differentiation of MCC progenitors. Mechanistically, cross-species genomic analyses and complete ciliary rescue of knockout MCCs identify TAp73 as the conserved central transcriptional integrator of multiciliogenesis. TAp73 directly activates the key regulators FoxJ1, Rfx2, Rfx3, and miR34bc plus nearly 50 structural and functional ciliary genes, some of which are associated with human ciliopathies. Our results position TAp73 as a novel central regulator of MCC differentiation.

ABSTRACT

Anaerobic digestion is used to treat effluents with a lot of organics, such as molasses distillery wastewater (MDW) which is the effluent of bioethanol production from molasses. The raw MDW requires a lot of dilution water before biodigestion, while the digested MDW has high level of recalcitrants which are problematic for its discharge. This study investigated ferric coagulation, Fenton, Fenton-like (with ferric ions as catalyst) processes and their combinations on the biodegradability of digested MDW. The Fenton and Fenton-like processes after coagulation increased the MDW biodegradability defined by (BOD5/COD) from 0.07 to (0.4-0.6) and saved 50% of H2O2 consumed in the classic Fenton process. The effluent from coagulation coupled to a Fenton-like process was used as dilution water for the raw MDW before the anaerobic digestion. The process was stable with volumetric loading of approx. 2.7 g COD/L/d. It resulted in increased overall biogas recovery and significantly decreased the demand for the dilution water.

ABSTRACT

Airway cilia provide the coordinated motive force for mucociliary transport, which prevents the accumulation of mucus, debris, pollutants, and bacteria in our respiratory tracts. As airway cilia are constantly exposed to the environment and, hence, are an integral component of the pathogenesis of several congenital and chronic pulmonary disorders, it is necessary to understand the molecular mechanisms that control ciliated cell differentiation and ciliogenesis. We have previously reported that loss of the basal body protein Chibby (Cby) results in chronic upper airway infection in mice due to a significant reduction in the number of airway cilia. In the present work, we demonstrate that Cby is required for normal ciliary structure and proper distribution of proteins involved in the bidirectional intraflagellar transport (IFT) system, which consists of 2 distinct sub-complexes, IFT-A and IFT-B, and is essential for ciliary biogenesis and maintenance. In fully differentiated ciliated cells, abnormal paddle-like cilia with dilated ciliary tips are observed in Cby-/- airways and primary cultures of mouse tracheal epithelial cells (MTECs). In addition, IFT88, an IFT-B sub-complex protein, robustly accumulates within the dilated tips of both multicilia in Cby-/- MTECs and primary cilia in Cby-/- mouse embryonic fibroblasts (MEFs). Furthermore, we show that only IFT-B components, including IFT20 and IFT57, but not IFT-A and Bardet-Biedl syndrome (BBS) proteins, amass with IFT88 in these distended tips in Cby-/- ciliated cells. Taken together, our findings suggest that Cby plays a role in the proper distribution of IFT particles to preserve normal ciliary morphology in airway ciliated cells.

ABSTRACT

Com este estudo objetivou-se descrever a topografia do cone medular do macaco-prego (Sapajus libidinosus) a fim de fornecer suporte para que a realização de procedimentos anestésicos, bem como exames de mielografia e coleta de líquor, dentre outros procedimentos que utilizam a via epidural. Para tanto foram dissecados oito animais, sendo seis machos e duas fêmeas, de diferentes faixas etárias. Rebateu-se a pele para retirada da musculatura da região dorsal, exposição de toda a coluna vertebral e identificação das vértebras lombares e sacrais. Para estabelecer o final da medula espinhal e medir o comprimento do cone medular, foi aberto todo o canal vertebral lombossacro, seccionando-se lateralmente os arcos vertebrais. Em seguida a duramáter foi seccionada para visualização do cone medular e observação da relação topográfica deste com as vértebras. Todos os animais apresentaram cinco vértebras lombares e três vértebras sacrais. As vértebras se apresentaram, de forma geral, muito próximas e com os processos espinhosos bem desenvolvidos e direcionados em sentido cranial. O cone medular dos macacos-prego situou-se entre as vértebras L2 e L5, com a base localizando-se com maior frequência na altura da vértebra L3, enquanto o ápice em L4. O comprimento corporal (espaço interarcual occiptoatlântico até o espaço interarcual sacrocaudal) variou de 22,9 a 31,8cm, com média de 27,44 ±3,1cm enquanto que comprimento do cone medular variou de 1,70 a 3,51cm, com média de 2,47 ±0,57cm. Não houve correlação entre o tamanho do corpo e o comprimento do cone medular (r = 0,212). Conclui-se que apesar das variações do comprimento e posicionamento do cone medular, o seu ápice não ultrapassa a articulação lombossacral, tornando seguro o acesso ao espaço epidural por esta via.(AU)


This study aimed to describe the topography of the conus of capuchin monkey (Sapajus libidinosus) to provide support for anesthetic procedures, as well as examinations of myelography and CSF collection, among other procedures using the epidural route. Eight animals were dissected, six males and two females, of different ages. The skin was countered for removal of the dorsal musculature for exposure of the entire spine and identification of the lumbar and sacral vertebrae. To establish the end of the spinal cord and to measure the length of the conus medullaris, we opened the lumbosacral spinal canal through side section of the vertebral arches. The dura mater was sectioned to visualize the conus and to observe the topographical relation with the vertebrae. All animals showed five lumbar vertebrae and three sacral vertebrae. The vertebrae were in general very closed with the spinous processes well developed and directed cranially. The conus of capuchin monkeys was located between the L2 and L5 vertebrae, with the base mostly in the L3 vertebra, and the peak in L4. The body length (interarcual space occiptoatlântico until sacrocaudal interarcual space) ranged from 22.9 to 31.8cm, with a mean of 27.44±3.1cm while the medullary cone length ranged from 1.70 to 3.51cm, with a mean of 2.47±0.57cm. There was no correlation between body size and length of the medullary cone (r=0.212). It is concluded that despite the variations in length and positioning of the medullary cone, its height does not exceed the lumbosacral joint, making safe access to the epidural space in this way.(AU)

ABSTRACT

Airway multiciliated epithelial cells play crucial roles in the mucosal defense system, but their differentiation process remains poorly understood. Mice lacking the basal body component Chibby (Cby) exhibit impaired mucociliary transport caused by defective ciliogenesis, resulting in chronic airway infection. In this paper, using primary cultures of mouse tracheal epithelial cells, we show that Cby facilitates basal body docking to the apical cell membrane through proper formation of ciliary vesicles at the distal appendage during the early stages of ciliogenesis. Cby is recruited to the distal appendages of centrioles via physical interaction with the distal appendage protein CEP164. Cby then associates with the membrane trafficking machinery component Rabin8, a guanine nucleotide exchange factor for the small guanosine triphosphatase Rab8, to promote recruitment of Rab8 and efficient assembly of ciliary vesicles. Thus, our study identifies Cby as a key regulator of ciliary vesicle formation and basal body docking during the differentiation of airway ciliated cells.
