ABSTRACT

Planarians are freshwater flatworms commonly used as environmental bioindicator due to their sensitivity of response and their ease of culturing in lab. Nevertheless, to date, very few studies describing their behavior have been led. This work aims to fill the literature gap by providing preliminary results through six behavioral challenges (locomotion, exploration, light stress, planarian light/dark test, shoaling and foraging) conducted with three different species Dugesia tigrina, Schmidtea mediterranea and Schmidtea polychroa. The behavioral responses of every species in each of these six assays were recorded and differences between species were highlighted, depending on the assays and conditions. Schmidtea polychroa is less active than the two others and had the highest light aversion. Reactions observed in response to diverse and realistic stimuli helped us to select the most suitable tests and choose the species that seem the most appropriate for future ecotoxicological and neurophysiological tests. Four tests - out of the six tested- seem reliable in order to standardize planarian behavioral tests.

ABSTRACT

Catastrophic events can have profound effects on the demography of a population and consequently on genetic diversity. The dynamics of postcatastrophic recovery and the role of sexual versus asexual reproduction in buffering the effects of massive perturbations remain poorly understood, in part because the opportunity to document genetic diversity before and after such events is rare. Six natural (purely sexual) and seven cultivated (mainly clonal due to farming practices) populations of the red alga Agarophyton chilense were surveyed along the Chilean coast before, in the days after and 2 years after the 8.8 magnitude earthquake in 2010. The genetic diversity of sexual populations appeared sensitive to this massive perturbation, notably through the loss of rare alleles immediately after the earthquake. By 2012, the levels of diversity returned to those observed before the catastrophe, probably due to migration. In contrast, enhanced rates of clonality in cultivated populations conferred a surprising ability to buffer the instantaneous loss of diversity. After the earthquake, farmers increased the already high rate of clonality to maintain the few surviving beds, but most of them collapsed rapidly. Contrasting fates between sexual and clonal populations suggest that betting on strict clonality to sustain production is risky, probably because this extreme strategy hampered adaptation to the brutal environmental perturbation induced by the catastrophe.

ABSTRACT

In the context of the abnormal mass mortality of mussels in France since 2014, Flow CytoMetry (FCM) was used in 2015 and 2016 to study the DNA content and cell cycle characteristics of hemic circulating cells collected from 2000 mussels. The mussels were sampled from 12 wild and cultivated blue mussels stocks distributed along the French Atlantic coast from the south Brittany to Pertuis Charentais areas. During these surveys, various genetic abnormalities were frequently detected, and ploidy characteristics revealed contrasting profiles that corresponded to respective contrasting sanitary status, i.e. healthy mussels with high cytogenetic quality (HCQ) versus diseased mussels with low cytogenetic quality (LCQ). In the present work, FCM and hemocytology cell monolayer techniques were combined in order to determine the putative causes of the observed genetic abnormalities that were significantly associated with mortality levels. FCM and cell monolayer approaches permitted the definition of new threshold values delimiting HCQ mussels from LCQ ones. FCM histograms of mussels from the HCQ group showed one single or a largely dominant population of diploid (2n) nuclei and a large majority of normal hemocytes. Hemolymph cell-monolayer analyses showed predominantly acidophil granulocytes characterized by nuclei of normal size and a large cytoplasm with numerous granulations. In contrast, FCM histograms for the LCQ group showed, in addition to the normal diploid (2n) nuclei, populations of nuclei that displayed aneuploidy patterns in a broad ploidy range, including diploid-triploid (2-3n), tetraploid-pentaploid (4-5n) and heptaploid-octaploid levels (7-8n). The corresponding hemolymph cell-monolayer showed cellular features characteristic of disseminated neoplasia disease with frequent abnormal anaplastic cells that exhibited noticeable numbers of mitotic figures with both normal and aberrant chromosomes segregation patterns. These neoplastic cells were a rounded shape with a reduced, granulation-free cytoplasm and large (11-12 µm) to very large (up to 21 µm) round or ovoid nuclei that correspond to the 4-5n and 7-8n nuclei previously detected by FCM analyses. These characteristics suggest that the genetic abnormalities detected by means of FCM were related to an ongoing neoplastic process that is affecting blue mussels in France, at least since the onset in 2014 of the mortality that heavily impacted French blue mussels stocks.

ABSTRACT

Anthropogenic CO2 emissions are rapidly changing seawater temperature, pH and carbonate chemistry. This study compares the embryonic development under high pCO2 conditions across the south-north distribution range of the marine clam Limecola balthica in NW Europe. The combined effects of elevated temperature and reduced pH on hatching success and size varied strongly between the three studied populations, with the Gulf of Finland population appearing most endangered under the conditions predicted to occur by 2100. These results demonstrate that the assessment of marine faunal population persistence to future climatic conditions needs to consider the interactive effects of co-occurring physico-chemical alterations in seawater within the local context that determines population fitness, adaptation potential and the system resilience to environmental change.

ABSTRACT

BACKGROUND: Allopatric divergence across lineages can lead to post-zygotic reproductive isolation upon secondary contact and disrupt coevolution between mitochondrial and nuclear genomes, promoting emergence of genetic incompatibilities. A previous F ST scan on the transcriptome of the Baltic clam Macoma balthica highlighted several genes potentially involved in mito-nuclear incompatibilities (MNIs). As proteins involved in the mitochondrial oxidative phosphorylation (OXPHO) chain are prone to MNIs and can contribute to the maintenance of genetic barriers, the mitochondrial genomes of six Ma. balthica individuals spanning two secondary contact zones were sequenced using the Illumina MiSeq plateform. RESULTS: The mitogenome has an approximate length of 16,806 bp and encodes 13 protein-coding genes, 2 rRNAs and 22 tRNAs, all located on the same strand. atp8, a gene long reported as rare in bivalves, was detected. It encodes 42 amino acids and is putatively expressed and functional. A large unassigned region was identified between rrnS and tRNA (Met) and could likely correspond to the Control Region. Replacement and synonymous mutations were mapped on the inferred secondary structure of all protein-coding genes of the OXPHO chain. The atp6 and atp8 genes were characterized by background levels of replacement mutations, relative to synonymous mutations. However, most nad genes (notably nad2 and nad5) were characterized by an elevated proportion of replacement mutations. CONCLUSIONS: Six nearly complete mitochondrial genomes were successfully assembled and annotated, providing the necessary roadmap to study MNIs at OXPHO loci. Few replacement mutations were mapped on mitochondrial-encoded ATP synthase subunits, which is in contrast with previous data on nuclear-encoded subunits. Conversely, the high population divergence and the prevalence of non-synonymous mutations at nad genes are congruent with previous observations from the nuclear transcriptome. This further suggest that MNIs between subunits of Complex I of the OXPHO chain, coding for NADH dehydrogenase, may play a role in maintaining barriers to gene flow in Ma. balthica.

ABSTRACT

The introduced oriental shrimp Palaemon macrodactylus has recently become widespread in temperate estuaries worldwide. However, this recent worldwide spread outside of its native range arises after a previous introduction to the US Pacific coast, where it was restricted for more than 30 years. Using a phylogeographic approach, the present work investigates the genetic history of the invasion of this decapod worldwide. Japan acted as the main native source area for worldwide introduced populations, but other native areas (likely South Korea and China) may act as source populations as well. The recently introduced European and NW Atlantic populations result from colonization from both Japan and an unknown area of the native range, although colonization from the NE Pacific could not be ruled out. Most introduced populations had higher haplotypic diversity than most native populations. P. macrodactylus has a strong potential to become one of the most widespread introduced species and may become the dominant estuarine shrimp in Europe. The ecological and economic consequences of this invasion remain to be thoroughly evaluated.
