ABSTRACT

Direct synthesis and cytotoxicity activity of new series of pyrido[2,3-d]pyrimidine was described. Nicotinamide 2 was synthesized via cyclization of N-cyclohexyl derivative with cyanoacetamide. The o-aminonicotinonitrile 2 was subjected to acylation or thio acylation process followed by intramolecular heterocyclization to afford the desired pyrido[2,3-d]pyrimidine (3-10) and pyrido triazine 11. Compounds 4 and 11 exhibited remarkable cytotoxicity against MCF-7 cells with IC50 values of 0.57 µM and 1.31 µM and IC50 values of 1.13 µM and 0.99 µM against HepG2 cells. Interestingly, compounds 4 and 10 had potent PIM-1 kinase inhibition with IC50 values of 11.4 and 17.2 nM, respectively, with inhibition of 97.8% and 94.6% compared to staurosporine (IC50 = 16.7 nM, with 95.6% inhibition). Moreover, compound 4 significantly activated apoptosis in MCF-7 cells, increasing the cell apoptosis by 58.29-fold by having 36.14% total apoptosis in treated cells compared to 0.62% for control. Moreover, it arrested the cell cycle at the G1 phase. PIM-1 kinase inhibition was virtually elucidated by the molecular docking study, highlighting binding interactions of the lead compound 4 towards the PIM-1 protein. Accordingly, compound 4 was validated as a promising PIM-1 targeted chemotherapeutic agent to treat breast cancer.

ABSTRACT

Ulcerative colitis is a common type of inflammatory bowel disease that affects millions of individuals around the world. Traditional UC treatment has focused on suppressing immune responses rather than treating the underlying causes of UC, which include oxidative stress, inflammation, and microbiota dysbiosis. Diosmin (DIO), a naturally occurring flavonoid, possesses antioxidant and anti-inflammatory properties. This study aimed to assess the efficacy of DIO in treating dextran-sulfate sodium (DSS)-induced colitis, and to investigate some of its underlying mechanisms, with an emphasis on Akkermansia muciniphila abundance, inflammatory markers, and intestinal barrier function. C57BL/6 mice were given 4% (w/v) DSS to induce colitis. DSS-induced mice were administered DIO (100 and 200 mg/kg) or sulfasalazine orally for 7 days. Every day, the disease activity index (DAI) was determined by recording body weight, diarrhea, and bloody stool. Changes in fecal A. muciniphila abundance, colonic MUC1 and MUC2 expression, as well as oxidative stress and inflammatory markers were all assessed. Histopathological changes, colonic PIK3PR3 and ZO-1 levels, and immunohistochemical examinations of occludin and claudin-1, were investigated. DIO administration resulted in a dose-dependent decrease in DAI, as well as increase in A. muciniphila abundance and MUC2 expression while decreasing MUC1 expression. DIO also dramatically reduced colonic oxidative stress and inflammation by regulating the NF-κB and Nrf2 cascades, restored intestinal barrier integrity by inhibiting PIK3R3 and inducing ZO-1, and improved occludin/claudin-1 gene expression and immunostaining. This study provides the first evidence that DIO preserves intestinal barrier integrity and increases A. muciniphila abundance in DSS-induced colitis. However, more research is required to explore the impact of DIO on the overall composition and diversity of the gut microbiota. Likewise, it will be important to fully understand the molecular mechanisms by which A. muciniphila maintains intestinal barrier function and its potential use as an adjuvant in the treatment of UC.

ABSTRACT

INTRODUCTION: The aim of this study was to compare two CRISPR/Cas9-based orthogonal strategies, paired-Cas9 nickase (paired-Cas9n) and RNA-guided FokI (RFN), in targeting 18S rDNA locus in Chinese hamster ovary (CHO) cells and precisely integrating a bicistronic anti-CD52 monoclonal antibody (mAb) expression cassette into this locus. METHODS: T7E1 and high-resolution melt (HRM) assays were used to compare the ability of mentioned systems in inducing double-strand break (DSB) at the target site. Moreover, 5'- and 3'-junction polymerase chain reactions (PCR) were used to verify the accuracy of the targeted integration of the mAb expression cassette into the 18S rDNA locus. Finally, anti-CD52 mAb gene copy number was measured and, its expression was analyzed using ELISA and western blot assays. RESULTS: Our results indicated that both paired-Cas9n and RFN induced DSB at the target site albeit RFN performance was slightly more efficient in HRM analysis. We also confirmed that the anti-CD52 mAb cassette was accurately integrated at the 18S rDNA locus and the mAb was expressed successfully in CHO cells. CONCLUSION: Taken together, our findings elucidated that both paired-Cas9n and RFN genome editing tools are promising in targeting the 18S rDNA locus. Site specific integration of the bicistronic anti-CD52 mAb expression cassette at this locus in the CHO-K1 cells was obtained, using RFN. Moreover, proper expression of the anti-CD52 mAb at the 18S rDNA target site can be achieved using the bicistronic internal ribosome entry site (IRES)-based vector system.

ABSTRACT

The use of annular one-dimensional (1D) photonic crystals (PCs) for salinity sensing is studied in this research. Annular 1D-PCs provide small and integrated structures that facilitate the creation of portable and miniaturized sensor equipment appropriate for field use. In order to generate annular 1D-PCs, the research explores the finite element method (FEM) simulation technique utilizing the COMSOL Multiphysics approach, highlighting the significance of exact control over layer thickness and uniformity. Furthermore, we construct a 1D annular PCs structure in the form [Formula: see text], where A is silicon ([Formula: see text]) and B is silicon dioxide ([Formula: see text]) of 40 nm and 70 nm, respectively, with a number of periods equal to 9. By incorporating a central defect layer of saline water (220 nm thickness), the sensor achieves optimum performance at normal incidence with a sensitivity (S) of [Formula: see text], a quality factor (Q) of 10.22, and a figure of merit (FOM) of [Formula: see text]. The design that is suggested has several advantages over past work on planners and annular 1D-PCs, including ease of implementation, performance at normal incidence, and high sensitivity.

ABSTRACT

Excessive usage of chemical fertilizers has detrimental effects on the environment and the safety of food. Conversely, utilizing organic fertilizers such as sage offers several advantages, including cost-effectiveness, soil enhancement, and promotion of root development. A two-year field experiment was conducted to investigate the impact of different potassium fertilizer sources and biofertilizers (specifically Bacillus cereus (MBc)) on potato plants. The experiment employed a split-plot design with three replicates, where the main plot factor was MBc (with and without), and the subplot factor was the sources of potassium fertilizer (control without K fertilizer, 100% Feldspar (FD), 100% Filter cake (FC), 75% FD + 25% FC, 25% FD + 75% FC, and 50% FD + 50% FC). The purpose was to examine the growth response of potato plants to these treatments. The results indicated that all treatments increased plant height, stem count, and tuber dry matter compared to the control. Furthermore, all treatments exhibited a higher uptake of macronutrients (N, P, and K) compared to the control. Notably, the plants treated with 100FC combined with MBc showed a significant 104.74% increase in total tuber weight compared to the control treatment. Additionally, the addition of 100FC with MBc significantly enhanced the availability of N, P, and K by 73.13%, 110.33%, and 51.88% respectively, compared to the control treatment. Apart from the biofertilizers, the individual application of FC and its combination with FD also demonstrated positive effects on soil fertility, potato growth, and yield.

ABSTRACT

This paper introduces a novel design of a thin-film solar cell based on CZTS and ZnO composite materials with the help of ITO as the front contact layer. This study primarily focuses on how the cells' optical absorbance at visible wavelengths can be improved. COMSOL Multiphysics is employed as a powerful tool for the investigation of the numerical simulation. The numerical findings showed that the optimum thicknesses of the ITO and ZnO are 80 and 350 nm, respectively. In this regard, with a normal incidence, a wide range of incoming light wavelengths from 450 nm to 800 nm might result in optical absorption of the examined cell of above 0.9. However, this value decreased significantly to reach less than 0.75 when the angle of incidence increased to 50°. To minimize this reduction, on the top surface of the cell, a texture-designed anti-reflective coating designed from a single period of well-known one-dimensional photonic crystals is deposited. The findings demonstrated that the cell's absorption at normal incidence could reach over 0.96 through the overall incident wavelengths. Therefore, CZTS/ZnO thin-film solar cells with an anti-reflecting coating of texturing configuration showed enormous potential for manufacturing effective solar cells.

SUMMARY: The main cause of mortality and disability globally is myocardial infarction (MI). Isoproterenol (ISO), a β-adrenoceptor agonist, has been used to induce rat myocardial necrosis. Whereas interleukin-37 (IL-37) has anti-inflammatory and cytoprotective properties. The study aimed to investigate the potential protective effects of IL-37 administration on cardiac architecture, oxidative stress, and inflammatory markers during ISO-induced MI in rats. Three groups of adult male rats were used in this study, the normal control group (n=8), ISO-induced MI group (n=8) that received isoproterenol hydrochloride (ISO) (100 mg/kg/day, SC, for the first 2 consecutive days), and IL-37-treated group (ISO+IL-37) (n=8) that received recombinant human IL-37 (40 µg/kg /day, intraperitoneally, for 2 weeks during and after ISO injections. Heart rate (HR.) and ECG changes were monitored. Some oxidative stress markers such as superoxide dismutase (SOD), nitric oxide (NOx), malondialdehyde (MDA), and glutathione (GSH) tissue levels in the tissue homogenate were assayed. Interleukin- 6 (IL-6), tumor necrosis factor- α (TNF-α), caspase-8, P53, and C- reactive protein (CRP) were among the inflammatory markers examined. In addition, serum levels of creatinine kinase (CK-MB) and lactate dehydrogenase (LDH) were analyzed to evaluate the myocardial injury. For histological analysis, tissues were sectioned, fixed in paraffin, and stained with hematoxylin and eosin (H&E), Masson Trichrome and, immunohistochemical against NF-kB, TNF-α, and Caspase-9. IL-37 improved ECG changes, cardiac enzyme markers, and some inflammatory markers of oxidative stress in ISO-induced MI. It also improved the histopathological and immunohistochemical changes in MI. In conclusion: IL-37 might be a promising therapeutic modality in myocardial infarction.


La principal causa de mortalidad y discapacidad a nivel mundial es el infarto de miocardio (IM). El isoproterenol (ISO), un agonista de los receptores adrenérgicos β, se ha utilizado para inducir necrosis miocárdica en ratas. Mientras que la interleucina-37 (IL-37) tiene propiedades antiinflamatorias y citoprotectoras. El estudio tuvo como objetivo investigar los posibles efectos protectores de la administración de IL-37 en la arquitectura cardíaca, el estrés oxidativo y los marcadores inflamatorios durante el infarto de miocardio inducido por ISO en ratas. En este estudio se utilizaron tres grupos de ratas macho adultas, el grupo control normal (n=8), el grupo con IM inducido por ISO (n=8) que recibió clorhidrato de isoproterenol (ISO) (100 mg/kg/día, SC, durante los primeros 2 días consecutivos) y el grupo tratado con IL-37 (ISO+IL- 37) (n=8) que recibió IL-37 humana recombinante (40 µg/kg/día, por vía intraperitoneal, durante 2 semanas durante y después de las inyecciones de ISO. Se monitorearon la frecuencia cardíaca (FC) y los cambios en el ECG. Se analizaron algunos marcadores de estrés oxidativo como la superóxido dismutasa (SOD), el óxido nítrico (NOx), el malondialdehído (MDA) y los niveles tisulares de glutatión (GSH) en el homogeneizado de tejido. La interleucina-6 (IL-6), el factor de necrosis tumoral-α (TNF-α), la caspasa-8, la P53 y la proteína C reactiva (CRP) se encontraban entre los marcadores inflamatorios examinados. Se analizaron los niveles de creatinoquinasa (CK-MB) y lactato deshidrogenasa (LDH) para evaluar la lesión miocárdica; para el análisis histológico se seccionaron los tejidos, se fijaron en parafina y se tiñeron con hematoxilina y eosina (H&E), Tricromo de Masson e inmunohistoquímica contra NF-kB, TNF-α y Caspasa-9. IL-37 mejoró los cambios de ECG, los marcadores de enzimas cardíacas y algunos marcadores inflamatorios de estrés oxidativo en el IM inducido por ISO. Además mejoró los cambios histopatológicos e inmunohistoquímicos en MI. En conclusión: la IL-37 podría ser una modalidad terapéutica prometedora en el infarto de miocardio.

INTRODUCTION: Female Pattern Hair Loss (FPHL) is considered the most common type of hair loss in females. Women with FPHL may suffer from psychological distress and defective social functioning. Patients with psychiatric and neurodegenerative disorders almost have a deficient brain and blood brain-derived neurotrophic factor levels (BDNF). This serum BDNF level may act as a diagnostic marker for negative psychological impact in FPHL patients. OBJECTIVES: Evaluate the levels of serum BDNF in patients with FPHL and correlate its level to the severity of alopecia and the degree of psychological impact. METHODS: Forty-six female patients with FPHL and 41 healthy age-matched female volunteers as a control were included in the study. Patients filled out a Dermatology Life Quality Index questionnaire. Both patients and controls filled Beck Depression Inventory, Beck Anxiety Inventory, and Perceived Stress Scale (PSS) questionnaires. Serum levels of BDNF were measured for all the participants using the ELISA technique. RESULTS: Patients with FPHL had significantly lower levels of BDNF and significantly higher Beck depression inventory score and PSS questionnaire scores. There is a significant negative correlation between serum levels of BDNF and Beck Depression Inventory, Beck Anxiety Inventory, and PSS questionnaire scores. CONCLUSIONS: Patients with FPHL are at a high risk to develop chronic stress and depression. The serum level of BDNF is a good predictor for the assessment of chronic stress and depression in FPHL patients.

ABSTRACT

During the past decades, tremendous advances have occurred in manufacturing recombinant therapeutic proteins in Chinese hamster ovary (CHO) cells. Nevertheless, the production of stable high-producing cell lines has remained a major obstacle in the development process of the CHO cell line. It has been shown that genomic regulatory elements can promote cell line development efficiency by improving transgenes' productivity and stability. Such elements include insulators, ubiquitous chromatin opening elements, scaffold/matrix attachment regions, and antirepressors. In addition, tDNA elements are shown to act as insulators in mammalian cells. This study examines the effect of the tDNA insulator on stable expression of a vascular endothelial growth factor receptor-Fc fusion protein.

ABSTRACT

BACKGROUND: Chinese hamster ovary (CHO) cells are the most predominantly utilized host for the production of monoclonal antibodies (mAbs) and other complex glycoproteins. A major challenge in the process of CHO cell culture is the occurrence of cell death following different stressful conditions, which hinders the production yield. Engineering genes involved in pathways related to cell death is a remarkable strategy to delay apoptosis, improve cell viability and enhance productivity. SIRT6 is a stress-responsive protein that regulates DNA repair, maintains genome integrity, and is critical for longevity and cell survival in organisms. METHODS AND RESULTS: In this study, SIRT6 was stably overexpressed in CHO-K1 cells and the impact of its expression on apoptosis related gene expression profile, viability, apoptosis, and mAb productivity was investigated. While a significant increase was observed in Bcl-2 mRNA level, caspase-3 and Bax mRNA levels were decreased in the SIRT6 engineered cells compared to the parental CHO-K1 cells. Moreover, improved cell viability and decreased rate of apoptotic progression was observed in a SIRT6-derived clone in comparision to the CHO-K1 cells during 5 days of batch culture. anti-CD52 IgG1 mAb titers were improved up to 1.7- and 2.8-fold in SIRT6-derived clone during transient and stable expression, respectively. CONCLUSIONS: This study indicates the positive effects of SIRT6 overexpression on cell viability and anti-CD52 IgG1 mAb expression in CHO-K1 cells. Further studies are needed to examine the potential of SIRT6-engineered host cells for the production of recombinant biotherapeutics in industrial settings.

ABSTRACT

In this study, the biosensing capabilities of conventional and hybrid multilayer structures were theoretically examined based on surface plasmon resonance (SPR). The transfer matrix method is adopted to obtain the reflectance spectra of the hybrid multilayer structure in the visible region. In this regard, the considered SPR sensor is configured as, [prism (CaF2)/Al2O3/Ag/Al2O3/2D material/Al2O3/Sensing medium]. Interestingly, many optimization steps were conducted to obtain the highest sensitivity of the new SPR biosensor from the hybrid structure. Firstly, the thickness of an Al2O3 layer with a 2D material (Blue P/WS2) is optimized to obtain an upgraded sensitivity of 360° RIU-1. Secondly, the method to find the most appropriate 2D material for the proposed design is investigated to obtain an ultra-high sensitivity. Meanwhile, the inclusion of black phosphorus (BP) increases the sensor's sensitivity to 466° RIU-1. Thus, black phosphorus (BP) was obtained as the most suitable 2D material for the proposed design. In this regard, the proposed hybrid SPR biosensing design may pave the way for further opportunities for the development of various SPR sensors to be utilized in chemical and biomedical engineering fields.

ABSTRACT

Natural feed additives and their potential benefits in production of safe and highly nutritious food have gained the attention of many researchers the last decades. Cordia myxa is a nutrient-dense food with various health benefits. Despite this fact, very limited studied investigated the physicochemical and sensory impacts of incorporation of fermented camel milk with Cordia myxa and its biological effects. The current study aimed to assess the physical, chemical, and sensory characteristics of fermented camel milk (FCM) fortified with 5, 10, and 15% Cordia myxa pulp. The study demonstrated that fortification of camel milk efficiently enhanced protein, total solids, ash, fiber, phenolic substance, and antioxidant activity. When compared to other treatments, FCM supplemented with 10% Cordia myxa pulp had the best sensory features. In addition, FCM fortified with 10% Cordia myxa pulp was investigated as a potential inhibitor of hypercholesterolemia agents in obese rats. Thirty-two male Wistar rats were split into two main groups including normal pellet group (n = 8) served as negative control group (G1) and a group of hyperlipidemic animals (n = 24) were feed on a high-fat diet (HFD). Hyperlipidemic rats group (n = 24) were then divided into three subgroups (8 per each); second group or positive control (G2) which include hyperlipidemic rats received distilled water (1 mL/day), the third group (G3) involved hyperlipidemic rats feed on FCM (10 g/day) and the fourth group (G4) included hyperlipidemic animals feed on 10 g/day FCM fortified with 10% of Cordia myxa pulp by oral treatment via an intestinal tube for another 4 weeks. In contrast to the positive control group, G4 treated with Cordia myxa showed a substantial decrease in malondialdehyde, LDL, cholesterol, triglycerides, AST, ALT, creatinine, and urea levels, while a significant increase in HDL, albumin, and total protein concentrations. The number of large adipocytes decreased while the number of small adipocytes increased after consumption of fortified FCM. The results indicated that fermented milk fortified with Cordia myxa pulp improved the functions of the liver and kidney in hyperlipidemic rats. These results demonstrated the protective effects of camel milk and Cordia myxa pulp against hyperlipidemia in rats.

This investigation reports the synthesis and characterization of (1-x)Ni0.5Zn0.5Fe2O4/(x)Zn0.95Co0.05O nanocomposites, with 0.0 ≤ × ≤ 0.5. Fourier transform infrared (FTIR) and Raman spectroscopies confirmed the purity of the samples and the presence of bands corresponding to octahedral and tetrahedral iron occupancies for Ni0.5Zn0.5Fe2O4 nanoparticles. A shift in peak positions of these bands was detected upon the addition of Zn0.95Co0.05O nanoparticles. The magnetic properties of the nanocomposites were examined using Mössbauer spectrometry at both room temperature and 77 K. Room temperature analysis showed the existence of both ferromagnetic and superparamagnetic behaviors, while at 77 K, all nanocomposites showed ferromagnetic behavior. The adsorption performance of the nanocomposite on the removal of malachite green (MG) dye solution was investigated by varying the contact time, adsorbent concentration, and reaction temperature. The adsorption reaction followed the second-order kinetics and the sample with x = 0.3 showed the highest adsorption rate. The adsorption rate showed an increase with the increase in the reaction temperature. The adsorption isotherm was determined by applying different adsorption isotherms (Langmuir, Freundlich, and Temkin isotherms), and the results are well-fitted with the Langmuir theoretical model.

ABSTRACT

For sustainable water desalination, there is a worldwide push towards solar thermal desalination with the objective to limit the amount of consumed energy in other desalination technologies and maximize the resulting freshwater from saline water. Here, we demonstrate a photonic crystals solar umbrella that covers the saline water surface, demanding to absorb all the incident electromagnetic wave and remit it as greater wavelengths in the range of mid-infrared (MIR) to be highly absorbed and localized close to the water surface. The temperature of the saline water with a refractive index of 1.3326 is reached to [Formula: see text] after one hour of illumination with the incident power intensity equal 680 [Formula: see text]. Hence, by adding one-dimensional PCs the surface temperature is reached [Formula: see text]. Also, by adding 2D PCs to allow the vapor to flow up through the pores of the structure with the diameter of the pore equal to 500 nm, the surface temperature is reached [Formula: see text] after three hour of illumination. Thus, the effective use of electromagnetic waves and warmth localization at the surface of saline water is accomplished by radiative coupling with the effect of 2D PCs. We design the considered structure by using COMSOL multiphysics which based on the finite element method (FEM).

ABSTRACT

We have designed a unique structure for a liquid sensor based on two-dimensional PCs with a triangular lattice constant in the periodicity by drilling a hexagonal cylinder in a dielectric host material. Using the COMSOL multiphysics approach, we investigated the given structure and sensing performance based on the finite element method. We will optimize two-dimensional hexagonal photonic crystals to localize the photonic band gap region in the mid and far infra-red frequency range, as water is a good absorber for this range of frequencies. Then, we inject the central hexagonal cylinder with saline water and calculate the sensor parameters for different values of the refractive index of saline water at different frequencies related to photonic band gaps. We could reach the optimum conditions of the salinity sensor as the half diagonal of the hexagonal shape (R) = 500 nm, the perpendicular distance between the two diagonal hexagonal (D) = 250 nm, and the number of periods (N) = 5, which gives a high efficiency with sensitivity (S) = 525 nm/RIU, figure of merit (FOM) = 80.7 RIU-1, and quality factor (Q) = 375. The effects of structural characteristics on sensing performance are investigated, with new approaches for improving salinity sensors proposed. Furthermore, traditional salinity sensors may be replaced by the proposed method in the photo-sensing application, which is simple and practical for use in the thermal desalination techniques.

BACKGROUND: Prevalence, morphology, and molecular characteristics of isopodiosis in the European seabass, Dicentrarchus labrax, in Egypt were assessed using light and electron microscopy and polymerase chain reaction targeting the mitochondrial COI (cytochrome oxidase c) gene. RESULTS: Adult parasites were found mainly in the branchial cavity between gill arches and to a lesser extent in the buccal cavity. They were morphologically identified as the Cymothoidae Livoneca redmanii Leach, 1818 (Crustacea: Isopoda). Obviously, a 23% prevalence rate of isopods was reported in D. labrax from Egyptian Mediterranean waters. Destructive and degenerative necrotic alterations with complete sloughing of gill lamellae of the infested fish were observed. DNA sequencing of the mitochondrial COI gene confirmed the identification of the parasite which was deposited in the GenBank under accession numbers MW600099, MZ208984, and MZ208985. Furthermore, the phylogenetic analysis demonstrated that parasites emerged from a monophyletic clade closely affiliated with L. redmanii and were clearly distinguished from other isopod genospecies. CONCLUSION: The present investigation addresses L. redmanii infestation in D. labrax in Egypt and affirmed morphological properties via the scanning electron microscopy (SEM) and molecular characteristics of this isopod species. The drastic effects of this parasite on the infected fish were proven both clinically and histopathologically.

ABSTRACT

Objectives: This study aimed to prove the effects of Escherichia coli isolates isolated from diseased broilers to form biofilms, describe their antimicrobial sensetivity, and determine the effect of allicin and cinnamon essential oils on the expression of some genes (fimH, int1, and luxS) through quantitative polymerase chain reaction (q-PCR). Materials and Methods: 140 samples were obtained from diseased broilers in Beni-Suef Governorate, Egypt. These samples were examined by conventional bacteriology methods to detect the causative agent. The antimicrobial susceptibility of the isolated bacteria was assessed using the disc diffusion method, The ability of yeast extract-casamino acids Congo Red Agar to generate phenotypic biofilms was next tested. The presence of resistance and virulence genes in some multidrug resistant isolates was genotypically investigated. The antibacterial effects of allicin and cinnamon oil were evaluated against the growth of multidrug-resistant E. coli. Finally, q-PCR was utilized to assess changes in some genes' expression. Results: Escherichia coli was isolated from 61 samples (43.6%). An antimicrobial susceptibility test revealed that multidrug-resistance (MDR) (could resist more than three antimicrobial classes) E. coli prevalence was 100%. 40.8% of isolates phenotypically produce biofilms. The detection of resistance and virulence genes by PCR showed that all tested isolates carry aadB, fimH, int1, qnrS, and luxS genes, while only 40% harbor iss genes. q-PCR showed that after treatment with allicin and cinnamon oils, gene expression went down. Conclusion: This investigation highlights that E. coli showed resistance against most of the tested antimicrobials; all isolates were MDR. The study showed wide dissemination of virulence and resistance genes among E. coli. Allicin and cinnamon oils have antimicrobial activities and could be used as alternatives to synthetic antimicrobial agents.

BACKGROUND: The purpose of this study was to compare the functional results of latissimus dorsi (LD) tendon transfer with those of subscapularis (SS) muscle release versus sliding. METHODS: Fifty-six patients with internal rotation contracture and external rotation (ER) weakness as sequelae of Erb's palsy were included in the study. Of the patients, 24 were included in group 1 (11 boys and 13 girls), with a mean age of 2 years 8 months (range 1.5-5 years) and a follow-up period of 62 months (range 38-68 months). The patients in group 1 underwent LD tendon transfer, with internal rotation contracture and SS release procedures. Thirty-two patients were included in group 2 (18 boys and 14 girls), with a mean age of 2 years 6 months (range 1.5-4.8 years) and a follow-up period of 58 months (range 38-68 months). The patients in group 2 underwent LD tendon transfer with SS sliding. RESULTS: A significant improvement in preoperative passive ER from - 3.6° to 67.3° after operation was observed in group 1. In group 2, preoperative passive ER in adduction improved from 0° to 72.3°. We found no significant difference (P = 0.1) in postoperative improvement in active ER in both groups (group 1 vs. group 2: 75° vs. 77.3°). Similarly, no significant difference (P = 0.7) in postoperative improvement in passive ER was found between the groups (group 1 vs. group 2: 71° vs. 72.3°). CONCLUSIONS: LD tendon transfer with SS release or sliding is an effective procedure to improve shoulder ER in patients with OBPP, with no inferiority of SS muscle release or sliding for internal rotation contractures and increased passive range of shoulder motion. BACKGROUND: Neonatal sepsis is a serious condition. Recent clinical studies have indicated that microRNAs (miRNAs) are key players in the pathogenesis of sepsis, which could be used as biomarkers for this condition. PATIENTS AND METHODS: A total of 90 neonates with sepsis and 90 healthy neonates were enrolled in this study. qRT-PCR was performed to measure the expression levels of serum miR-34a-5p and miR-199a-3p. RESULTS: miR-34a-5p and miR-199a-3p serum levels were significantly reduced in neonates with sepsis compared with those in healthy neonates (P = 0.006 and P = 0.001, respectively). Significant correlations of miR-34a-5p and miR-199a-3p with each of TLC, RDW, RBS, and C-reactive protein (CRP) as well as SNAPII were observed, indicating their associations with the severity of neonatal sepsis. CONCLUSION: miR-34a-5p and miR-199a-3p may be useful as novel biomarkers in neonatal sepsis and may provide a new direction for its treatment.
