ABSTRACT

The enlargement of the prostate gland in the reproductive system of males is considered a form of prostate cancer (PrC). The survival rate is considerably improved with earlier diagnosis of cancer; thus, timely intervention should be administered. In this study, a new automatic approach combining several deep learning (DL) techniques was introduced to detect PrC from MRI and ultrasound (US) images. Furthermore, the presented method describes why a certain decision was made given the input MRI or US images. Many pretrained custom-developed layers were added to the pretrained model and employed in the dataset. The study presents an Equilibrium Optimization Algorithm with Deep Learning-based Prostate Cancer Detection and Classification (EOADL-PCDC) technique on MRIs. The main goal of the EOADL-PCDC method lies in the detection and classification of PrC. To achieve this, the EOADL-PCDC technique applies image preprocessing to improve the image quality. In addition, the EOADL-PCDC technique follows the CapsNet (capsule network) model for the feature extraction model. The EOA is based on hyperparameter tuning used to increase the efficiency of CapsNet. The EOADL-PCDC algorithm makes use of the stacked bidirectional long short-term memory (SBiLSTM) model for prostate cancer classification. A comprehensive set of simulations of the EOADL-PCDC algorithm was tested on the benchmark MRI dataset. The experimental outcome revealed the superior performance of the EOADL-PCDC approach over existing methods in terms of different metrics.

ABSTRACT

Object detection in pedestrian walkways is a crucial area of research that is widely used to improve the safety of pedestrians. It is not only challenging but also a tedious process to manually examine the labeling of abnormal actions, owing to its broad applications in video surveillance systems and the larger number of videos captured. Thus, an automatic surveillance system that identifies the anomalies has become indispensable for computer vision (CV) researcher workers. The recent advancements in deep learning (DL) algorithms have attracted wide attention for CV processes such as object detection and object classification based on supervised learning that requires labels. The current research study designs the bioinspired Garra rufa optimization-assisted deep learning model for object classification (BGRODL-OC) technique on pedestrian walkways. The objective of the BGRODL-OC technique is to recognize the presence of pedestrians and objects in the surveillance video. To achieve this goal, the BGRODL-OC technique primarily applies the GhostNet feature extractors to produce a set of feature vectors. In addition to this, the BGRODL-OC technique makes use of the GRO algorithm for hyperparameter tuning process. Finally, the object classification is performed via the attention-based long short-term memory (ALSTM) network. A wide range of experimental analysis was conducted to validate the superior performance of the BGRODL-OC technique. The experimental values established the superior performance of the BGRODL-OC algorithm over other existing approaches.

ABSTRACT

The Al/Au alloy was investigated to improve the ohmic characteristic and light efficiency of reflective infrared light-emitting diodes (IR-LEDs). The Al/Au alloy, which was fabricated by combining 10% aluminum and 90% gold, led to considerably improved conductivity on the top layer of p-AlGaAs of the reflective IR-LEDs. In the wafer bond process required for fabricating the reflective IR-LED, the Al/Au alloy, which has filled the hole patterns in Si3N4 film, was used for improving the reflectivity of the Ag reflector and was bonded directly to the top layer of p-AlGaAs on the epitaxial wafer. Based on current-voltage measurements, it was found that the Al/Au alloyed material has a distinct ohmic characteristic pertaining to the p-AlGaAs layer compared with those of the Au/Be alloy material. Therefore, the Al/Au alloy may constitute one of the favored approaches for overcoming the insulative reflective structures of reflective IR-LEDs. For a current density of 200 mA, a lower forward voltage (1.56 V) was observed from the wafer bond IR-LED chip made with the Al/Au alloy; this voltage was remarkably lower in value than that of the conventional chip made with the Au/Be metal (2.29 V). A higher output power (182 mW) was observed from the reflective IR-LEDs made with the Al/Au alloy, thus displaying an increase of 64% compared with those made with the Au/Be alloy (111 mW).

Background: Mammalian target of rapamycin (mTOR) inhibitors, such as everolimus and sirolimus, may be efficacious in preserving renal function in liver transplantation (LT) recipients while preventing hepatocellular carcinoma (HCC) recurrence. Materials and Methods: In this study, we retrospectively evaluated the safety, efficacy, and renoprotective effects of mTOR inhibitors in LT recipients. Among the 84 patients enrolled, mTOR inhibitor was commenced during the first year after LT. Renal function was measured by estimated glomerular filtration rate (eGFR) using the Modification of Diet in Renal Disease equation. Results: Regarding the type of mTOR inhibitor, everolimus was used in 71 patients and sirolimus in 13 patients. Concomitant tacrolimus was used in 63 patients (75.0%). For total enrolled patients, kidney function did not significantly change during 12 months after initiation of mTOR inhibitors, although tacrolimus-withdrawn patients (n = 21) showed better kidney function compared to tacrolimus-minimized patients (n = 63) after conversion. However, a significant improvement in kidney function was observed in the eGFR <60 ml/min/1.73 m2 group (n = 19) 12 months after initiation of mTOR inhibitors, for both patient groups with early + mid starters (n = 7, stating within 1 year after LT) and late starters (n = 12, starting over 1 year after LT). mTOR inhibitors were safely administered without serious adverse events that led to drug discontinuation. Conclusion: We demonstrated that patients with renal impairment showed significant improvement in renal function regardless of the timing of mTOR inhibitor start, suggesting that switch to mTOR inhibitors may be beneficial when renal function declines.

ABSTRACT

A predictive biomarker of immune checkpoint inhibitor (ICI)-based treatments in hepatocellular carcinoma (HCC) has not been clearly demonstrated. In this study, we focused on the infiltration and programmed death ligand 1 (PD-L1) expression of tumor-associated macrophages (TAMs) in the tumor microenvironment of HCC. Immunohistochemistry demonstrated that PD-L1 was preferentially expressed on CD68+ macrophages in the tumor microenvironment of HCC, suggestive of its expression in TAMs rather than in T cells or tumor cells (P < 0.05). A co-culture experiment using activated T cells and M2 macrophages confirmed a significant increase in T cell functionality after the pretreatment of M2 macrophages with anti-PD-L1. Syngeneic mouse model experiments demonstrated that TAMs expressed PD-L1 and tumors treated with anti-PD-L1 showed smaller diameters than those treated with IgG. In these mice, anti-PD-L1 treatment increased activation markers in intratumoral CD8+ T cells and reduced the size of the TAM population. Regarding nivolumab-treated patients, three of eight patients responded to the anti-PD-1 treatment. The percentage of Ki-67-positive CD4+ and CD8+ T cells was higher in responders than non-responders after nivolumab. Overall, PD-L1 expression on TAMs may be targeted by immune-based HCC treatment, and ICI treatment results in the reinvigoration of exhausted CD8+ T cells in HCC.

ABSTRACT

The Internet of things (IoT), the Internet of vehicles, and blockchain technology have become very popular these days because of their versatility. Road traffic, which is increasing day by day, is causing more and more deaths worldwide. The world needs a product that would reduce the number of road accidents. This paper suggests combining IoT and blockchain technology to mitigate road hazards. The new intelligent transportation system technologies and the subsequent emergence of 5G technologies will be a blessing, delivering the necessary speed to ensure both safety and quality of service (QoS). Hashgraph technology, a distributed ledger technology is used to create communication networks between the different vehicles and other relevant parameters. Scheduling the requests according to the priorities for ensuring better QoS quotient can be effectively done using hashgraph. We demonstrated how the hashgraph outstrips other equivalents platforms. The proposed model was simulated using OMNeT++ with proper design and network description files. A hardware implementation of the proposed model was also done. Messages were transferred between the vehicles and prioritized using a hashgraph. This paper proposes an effective model in reducing the accidents in terms of parameters like speed, security, stability, and fairness.

The rapid increase in the emergence of antifungal-resistant Candida albicans strains is becoming a serious health concern. Because antimicrobial peptides (AMPs) may provide a potential alternative to conventional antifungal agents, we have synthesized a series of peptides with a varying number of lysine and tryptophan repeats (KWn-NH2). The antifungal activity of these peptides increased with peptide length, but only the longest KW5 peptide displayed cytotoxicity towards a human keratinocyte cell line. The KW4 and KW5 peptides exhibited strong antifungal activity against C. albicans, even under conditions of high-salt and acidic pH, or the addition of fungal cell wall components. Moreover, KW4 inhibited biofilm formation by a fluconazole-resistant C. albicans strain. Circular dichroism and fluorescence spectroscopy indicated that fungal liposomes could interact with the longer peptides but that they did not release the fluorescent dye calcein. Subsequently, fluorescence assays with different dyes revealed that KW4 did not disrupt the membrane integrity of intact fungal cells. Scanning electron microscopy showed no changes in fungal morphology, while laser-scanning confocal microscopy indicated that KW4 can localize into the cytosol of C. albicans. Gel retardation assays revealed that KW4 can bind to fungal RNA as a potential intracellular target. Taken together, our data indicate that KW4 can inhibit cellular functions by binding to RNA and DNA after it has been translocated into the cell, resulting in the eradication of C. albicans.

This work reports the design of and experimentation with a topographically patterned cell culture substrate of variable local density and anisotropy as a facile and efficient platform to guide the organization and migration of cells in spatially desirable patterns. Using UV-assisted capillary force lithography, an optically transparent microstructured layer of a UV curable poly(urethane acrylate) resin is fabricated and employed as a cell-culture substrate after coating with fibronectin. With variable local pattern density and anisotropy present in a single cell-culture substrate, the differential polarization of cell morphology and movement in a single experiment is quantitatively characterized. It is found that cell shape and velocity are exquisitely sensitive to variation in the local anisotropy of the two-dimensional rectangular lattice arrays, with cell elongation and speed decreasing on symmetric lattice patterns. It is also found that cells could integrate orthogonal spatial cues when determining the direction of cell orientation and movement. Furthermore, cells preferentially migrate toward the topographically denser areas from sparser ones. Consistent with these results, it is demonstrated that systematic variation of local densities of rectangular lattice arrays enable a planar assembly of cells into a specified location. It is envisioned that lithographically defined substrates of variable local density and anisotropy not only provide a new route to tailoring the cell-material interface but could serve as a template for advanced tissue engineering.
