ABSTRACT

The excitation of quasi-particles near the extrema of the electronic band structure is a gateway to electronic phase transitions in condensed matter. In a many-body system, quasi-particle dynamics are strongly influenced by the electronic single-particle structure and have been extensively studied in the weak optical excitation regime. Yet, under strong optical excitation, where light fields coherently drive carriers, the dynamics of many-body interactions that can lead to new quantum phases remain largely unresolved. Here, we induce such a highly non-equilibrium many-body state through strong optical excitation of charge carriers near the van Hove singularity in graphite. We investigate the system's evolution into a strongly-driven photo-excited state with attosecond soft X-ray core-level spectroscopy. We find an enhancement of the optical conductivity of nearly ten times the quantum conductivity and pinpoint it to carrier excitations in flat bands. This interaction regime is robust against carrier-carrier interaction with coherent optical phonons acting as an attractive force reminiscent of superconductivity. The strongly-driven non-equilibrium state is markedly different from the single-particle structure and macroscopic conductivity and is a consequence of the non-adiabatic many-body state.

ABSTRACT

The Adenoviridae family is composed by a high diversity of viruses that are extremely resistant in environment and are frequently excreted in animal reservoir feces for long periods. The knowledge of adenovirus (AdV) diversity among wild species may be important for the understanding of the epidemiology of putative emerging diseases. Cavia aperea aperea, commonly known as wild guinea pigs, wild cavies, or preas, are small herbivorous rodents widely distributed throughout South America and classified in Caviidae family, as well as domestic guinea pigs and capybaras. In order to investigate their potential role as reservoir of zoonotic agents, the present study aimed to verify the presence of AdV in fecal samples of 14 preas from Northeast Brazil. When submitted to nested PCR, two out of 14 samples (14.28%) were positive for AdV and classified as human Mastadenovirus C (HAdV-C) using DNA sequencing and phylogenetic analysis. Wild guinea pigs are synanthropic rodents that live in close contact with humans. The investigation of viral agents in rodents is important due to their potential role as reservoirs of human and animal pathogens. Moreover, the present work presents the first known evidence of HAdV in wild guinea pig stool samples, which may represent both the impact of anthropogenic pollution to wild animals and an important knowledge in terms of human health.

ABSTRACT

BACKGROUND: Ontario's human-rights legislation and institutions' support for students with disabilities have influenced these students' increased enrolment in the postsecondary environment. Yet more attention is paid to in-classroom than to out-of-classroom experiences. Students with disabilities enrolled in nursing are least likely to be accommodated. This has significant implications for access and equity upon graduation, as clinical placements are a bridge between school and employment. OBJECTIVES: To explore clinical instructors' and students who identify with disabilities' perspectives on nursing clinical placements. METHOD: A descriptive case-report was used: interactive take-home diaries were completed and semi-structured interviews were conducted with clinical instructors (n-14) and nursing students with disabilities (n-14) in Ontario. An iterative inductive analysis was undertaken. RESULTS: Three overarching themes were identified: (a) an immovable clinical environment in relation to technology and time, (b) a perceived conflict between nursing and disability identity, and (c) a need to shift responsibility upwards while listening to lived experience. The clinical environment is a highly contested space; different identity groups have distinct roles and differing degrees of power within it. CONCLUSION: Our research calls for a shift in the nursing landscape to be led by education and medical institutions that can build supportive networks, develop clear policies and listen to nursing students with disabilities.

ABSTRACT

Clinically meaningful molecular subtypes for classification of breast cancers have been established, however, initiation and progression of these subtypes remain poorly understood. The recent development of desorption electrospray ionization-mass spectrometry imaging (DESI-MSI) facilitates the convergence of analytical chemistry and traditional pathology, allowing chemical profiling with minimal tissue pretreatment in frozen samples. Here, we characterized the chemical composition of molecular subtypes of breast cancer with DESI-MSI. Regions of interest were identified, including invasive breast cancer (IBC), ductal carcinoma in situ (DCIS), and adjacent benign tissue (ABT), and metabolomic profiles at 200 µm elaborated using Biomap software and the Lasso method. Top ions identified in IBC regions included polyunsaturated fatty acids, deprotonated glycerophospholipids, and sphingolipids. Highly saturated lipids, as well as antioxidant molecules [taurine (m/z 124.0068), uric acid (m/z 167.0210), ascorbic acid (m/z 175.0241), and glutathione (m/z 306.0765)], were able to distinguish IBC from ABT. Moreover, luminal B and triple-negative subtypes showed more complex lipid profiles compared with luminal A and HER2 subtypes. DCIS and IBC were distinguished on the basis of cell signaling and apoptosis-related ions [fatty acids (341.2100 and 382.3736 m/z) and glycerophospholipids (PE (P-16:0/22:6, m/z 746.5099, and PS (38:3), m/z 812.5440)]. In summary, DESI-MSI identified distinct lipid composition between DCIS and IBC and across molecular subtypes of breast cancer, with potential implications for breast cancer pathogenesis. SIGNIFICANCE: These findings present the first in situ metabolomic findings of the four molecular subtypes of breast cancer, DCIS, and normal tissue, and add to the understanding of their pathogenesis.

ABSTRACT

In this work, lemon and onion biomasses commonly found in street markets are for the first time used to develop a facile, fast and low-cost one-step microwave-assisted carbonization method for synthesis of highly fluorescent carbon dots (CDs). The structure and optical properties of CDs were investigated by TEM, XRD, XRF, UV-Vis, FTIR, and fluorescence spectroscopy. CDs displayed satisfactory optical proprieties, a high quantum yield of 23.6%, and excellent water solubility, and the particle size was 4.23-8.22 nm with an average diameter of 6.15 nm. An efficient fluorescent resonance energy transfer (FRET) between the CDs and riboflavin was achieved with CDs acting as donor and riboflavin as acceptor. A linear relationship between FRET and the riboflavin concentration from 0.10 to 3.0 µg/mL was observed, allowing the development of an accurate and fast analytical method to determine this vitamin in multivitamin/mineral supplements. Despite the potential interferences in these supplements, CDs were selective for riboflavin under optimized conditions. A paired t-test at a 95% confidence level indicated no statistically significant difference between the proposed and the reference methods. Recovery test presented values ranged from 96.0% to 101.4%. The limit of detection and relative standard deviation were estimated at 1.0 ng/mL and <2.6% (n = 3), respectively. CDs were successfully synthesized in a domestic microwave oven (1450 W, 6 min), presenting satisfactory parameters when compared with results of other studies reported in the literature, suggesting that the proposed method is a potentially useful method for the synthesis of CDs and determination of riboflavin.

ABSTRACT

O objetivo deste estudo foi determinar a soroprevalência de lentivírus de pequenos ruminantes (LVPR) e identificar os fatores de risco para a ocorrência de caprinos e ovinos soropositivos no semiárido do Estado da Paraíba. Foram utilizados 1.733 animais, sendo 1.274 caprinos procedentes de 62 Unidades de Produção (UPs) e 459 ovinos provenientes de 32 UPs. Para o diagnóstico sorológico da infecção por lentivírus foi utilizado o teste de imunodifusão em gel de ágar (IDGA). Dos 1.274 caprinos analisados 15 (1,18%) foram soropositivos, enquanto que todos os 459 ovinos foram soronegativos. Das 62 propriedades caprinas analisadas oito (12,9%) apresentaram pelo menos um animal soropositivo. Os fatores de risco para a ocorrência de caprinos soropositivos foram área da propriedade (odds ratio = 3,28; p = 0,044), ausência de capacitação dos produtores (odds ratio = 8,29; p = 0,042) e uso de monta natural não controlada (odds ratio = 6,78; p = 0,012). Conclui-se que a infecção por lentivírus de pequenos ruminantes, demonstrada pela detecção de anticorpos, está disseminada em rebanhos caprinos do semiá­rido paraibano, e sugere-se o incentivo à capacitação contínua dos produtores, manutenção de reprodutores negativos ao LVPR e utilização de inseminação artificial com o intuito de evitar o contato físico entre macho e fêmeas.(AU)


The aim of this survey was to determine the seroprevalence of small ruminant lentivirus (SRLV) and to identify risk factors for the occurrence of seropositive goats and sheep in the semiarid region of Paraiba State. It were used 1,733 animals, being 1,274 goats from 62 Production Units (PU) and 459 sheep from 38 PU. For the serological diagnosis of lentivirus infection the agar gel immunodiffusion test (AGID) was used. Of the 1,274 goats 15 (1.18%) were seropositive, and all 459 sheep were seronegative. Of the 62 goat herds eight (12.9%) presented at least one seropositive animal. Risk factors for the occurrence of seropositive goats were area of the property ≤35 ha (odds ratio = 3.28; p=0.044), not training of producers (odds ratio = 8.29; p=0.042) and use of uncontrolled natural mating (odds ratio = 6.78; p=0.012). It is concluded that lentivirus infection detected by serology is spread in goat flocks in the semiarid of the State of Paraíba, and it is suggested to encourage the continous capacitation of owners, maintenance of reproducers negative for SRLV and use of artificial insemination aiming to avoid the physical contact among male and females.(AU)

ABSTRACT

The prebiotic ability of several rice bran fractions obtained by debranning (RBD) using human microbiota was studied in anaerobic batch cultures with agitation and pH-controlled. Fraction C (3.8-5% w/w pearling) from RBD increased the number of bifidobacteria and lactobacteria compared with the positive control, raftilose P95. RBD fermentation induced changes in the short-chain fatty acid (SCFA) profile. In addition, Fraction C revealed the highest growth of positive lactobacteria than commercial control. The present work illustrates the prebiotic capacity of RBD to modulate human microbiota and highlights that fraction C could be an economical source for use in human food as well as an interesting alternative to valorise a by-product of cereal industry.

Most of the commercialized lactic acid fermented products are dairy-based. Hence, the development of non-dairy fermented products with probiotic properties draws significant attention within the functional foods industry. The microorganisms used in such products have complex enzyme systems through which they generate diverse metabolites (volatile and non-volatile) that provide significant flavour attributes of importance for fermented foods. The correlation of the volatile flavour compounds of a malt beverage fermented with a Bifidobacterium breve strain with its unique sensory characteristics was performed. The volatile composition analysis exposed the presence of 12 components. Eight of these flavour volatiles were produced through the metabolic activity of the bifidobacteria strain. Notably acetic acid, of reported sour flavour characteristics, exhibited the greatest intensity. Four components of considerable organoleptic characteristics were identified as Maillard-derived products, namely maltol, pyranone, 2 (5H)-furanmethanol and 3-furanmethanol. The sensory evaluation exhibited that the fermented cereal beverage had a sour flavour with mild sweet and malty notes. These results indicate that the volatile compounds identified can be appointed as significant flavour markers of the novel fermented cereal beverage.

ABSTRACT

Fermentation by using Aspergillus oryzae has been reported to increase antioxidant activity of soybeans significantly. The effectiveness of the extract from fermented soybeans was studied in 3 model foods with different complexities, i.e., linoleic acid emulsion, sunflower oil emulsions and bulk sunflower oil. For the emulsion systems, oxidation at two different pH values (4.5 and 7) was also compared. A reparameterised logistic equation was used to describe and to predict the experimental data. In general, a good agreement between experimental trends and simulated data from the model was found. A crude antioxidant extract (5 mg/g) showed a comparable antioxidant activity to 0.26 mg/g of butylated hydroxytoluene (BHT) in the linoleic acid emulsions. The extract exhibited a better capability to retard primary products in the linoleic acid systems than the secondary products. The opposite effect was observed in the bulk sunflower oil and its emulsion systems.

Este trabalho teve como objetivo determinar a prevalência de rebanhos ovinos positivos (focos) e de animais soropositivos para Brucella ovis na mesorregião do Sertão, Estado da Paraíba, Nordeste do Brasil, bem como identificar fatores de risco. Foram colhidas amostras de sangue de 1.134 animais procedentes de 103 rebanhos em 17 municípios. Para o diagnóstico sorológico da infecção por B. ovis foi utilizado o teste de imunodifusão em gel de ágar (IDGA). Um rebanho foi considerado positivo quando apresentou pelo menos um animal soropositivo. Das 103 propriedades utilizadas 21 (20,39%) apresentaram pelo menos um animal soropositivo e dos 1.134 animais, 59 (5,20%) foram soropositivos. Realizar higiene nas instalações com periodicidade anual (odds ratio = 7,13; IC 95% = 1,56-32,47; p=0,011) e aquisição de animais (odds ratio = 6,06; IC 95% = 1,39-26,48; p=0,017) foram identificados como fatores de risco. Com base na análise de fatores de risco, recomenda-se a realização de diagnóstico da infecção por B. ovis previamente à aquisição de animais e realização periódica de higienização das instalações.


The aim of this investigation was to determine the seroprevalence of Brucella ovis in sheep flocks and individual sheep in the Sertão mesorregion, Paraíba state, Northeastern Brazil, as well as to identify risk factors. Blood samples were collected from 1,134 sheep from 103 flocks in 17 counties. For the serological diagnosis of B. ovis infection the agar gel immunodiffusion test (AGID) was carried out. A flock was considered positive when there was at least one seropositive animal. Of the 103 flocks used, 21 (20.39%) presented at least one seropositive sheep, and of the 1,134 sheep examined 59 (5.20%) seropositive animals were diagnosed. Cleaning of facilities (odds ratio = 7.13; 95% CI=1.56-32.47; p=0.011) and purchase of animals (odds ratio = 6.06; 95% CI=1.39-26.48; p=0.017) were identified as risk factors. Based on the risk factor analysis, it is recommended the diagnosis of B. ovis infection prior to purchase of sheep and the periodic cleaning of the facilities on the farm.

O objetivo do presente trabalho foi determinar as prevalências de propriedades positivas e animais soropositivos para Chlamydophila abortus em ovinos deslanados da região semiárida do Nordeste do Brasil, bem como identificar fatores de risco. Foram colhidas amostras de sangue de 476 ovinos procedentes de 72 propriedades em 14 municípios na mesorregião do Sertão, Estado da Paraíba. Para o diagnóstico sorológico da infecção por Chlamydophila abortus foi utilizada a reação de fixação de complemento (RFC). Uma propriedade foi considerada positiva quando apresentou pelo menos um animal soropositivo. Das 72 propriedades usadas 38 (52,8%) apresentaram pelo menos um animal soropositivo, e dos 476 animais 94 (19,7%) foram soropositivos. Participação em exposições (odds ratio =4,31; IC 95% =1,80-10,35; p=0,011) foi identificada como fator de risco. Sugere-se que a infecção por Chlamydophila abortus encontra-se disseminada em ovinos da região, e baseando-se na análise de fatores de risco, recomenda-se o controle sanitário nas exposições de animais.

Participation in animal expositions (odds ratio= 4.31; 95% CI= 1.80-10.35; p=0.011) was identified as risk factor. It is suggested that C. abortus infection is spread in sheep of the region, and based on the risk factor analysis sanitary control in animal expositions is recommended.

A cross-sectional study based on a planned sampling was carried out to determine flock-level risk factors associated to Leptospira spp. infection in dairy goat flocks in a semiarid region of Northeastern Brazil. Serum samples from 975 adult dairy goats from 110 flocks were examined for Leptospira spp. antibodies by MAT using 24 serovars. A structured questionnaire focusing on risk factors for leptospirosis was completed for each flock. Of the 110 flocks 48 (43.6%; 95% CI: 34.2-53.4%) presented at least one seropositive animal, and most frequent serovar was Autumnalis (10.9%). Ninety-eight (8.7%; 95% CI: 5.7-12.9%; design effect=4.23) of the 975 goats tested seropositive at MAT, and serovar Autumnalis was also the most frequent (1.74%). Presence of rodents (OR=2.78; P=0.015) was identified as a risk factor. There was also association between history of infertility (OR=14.74; P=0.015) and prevalence of positive flocks. We suggest that a program of rodent control should be included in the flock management practices aiming to reduce transmission of the agent and then to reduce prevalence of positive flocks and occurrence of reproductive disorders such as impaired fertility.

Few data are available on the prevalence and risk factors of Chlamydophila abortus infection in goats in Brazil. A cross-sectional study was carried out to determine the flock-level prevalence of C. abortus infection in goats from the semiarid region of the Paraíba State, Northeast region of Brazil, as well as to identify risk factors associated with the infection. Flocks were randomly selected and a pre-established number of female goats > 12 mo old were sampled in each of these flocks. A total of 975 serum samples from 110 flocks were collected, and structured questionnaire focusing on risk factors for C. abortus infection was given to each farmer at the time of blood collection. For the serological diagnosis the complement fixation test (CFT) using C. abortus S26/3 strain as antigen was performed. The flock-level factors for C. abortus prevalence were tested using multivariate logistic regression model. Fifty-five flocks out of 110 presented at least one seropositive animal with an overall prevalence of 50.0% (95%; CI: 40.3%, 59.7%). Ninety-one out of 975 dairy goats examined were seropositive with titers >32, resulting in a frequency of 9.3%. Lend buck for breeding (odds ratio = 2.35; 95% CI: 1.04-5.33) and history of abortions (odds ratio = 3.06; 95% CI: 1.37-6.80) were associated with increased flock prevalence.


São escassos os trabalhos publicados sobre a prevalência e fatores de risco associados à infecção por Chlamydophila abortus em caprinos no Brasil. Foi conduzido um estudo transversal para determinar a prevalência de rebanhos positivos para a infecção por C. abortus em caprinos do semiárido do Estado da Paraíba, Nordeste do Brasil, bem como identificar os fatores de risco associados com a infecção. Os rebanhos foram selecionados aleatoriamente e um número pré-estabelecido de cabras com idade >12 meses foi amostrado por rebanho. No total, foi colhido sangue de 975 animais procedentes de 110 rebanhos, e no momento da colheita foi aplicado um questionário epidemiológico a cada proprietário. Para o diagnóstico sorológico foi utilizado o teste de fixação de complemento (FC) usando a estirpe de C. abortus S26/3 como antígeno. Os fatores de risco para a prevalência de C. abortus em nível de rebanho foram testados com o uso de modelo de regressão logística multivariada. Cinquenta e cinco rebanhos dos 110 analisados apresentaram pelo menos um animal soropositivo, com uma prevalência de 50,0% (IC 95%: 40,3-59,7%). Noventa e um animais entre os 975 examinados foram soropositivos com título >32, resultando em uma frequência de 9,3%. Compartilhar reprodutores (odds ratio = 2,35; IC 95%: 1,04-5,33) e histórico de abortamentos (odds ratio = 3,06; IC 95%: 1,37-6,80) foram associados com o aumento da prevalência de rebanhos.

Foram colhidas amostras de sangue de 1.275 animais procedentes de 117 rebanhos em 19 municípios da mesorregião do Sertão, Estado da Paraíba. Para o diagnóstico sorológico da leptospirose foi utilizado o teste de soroaglutinação microscópica (SAM), com 24 sorovares de Leptospira spp. como antígenos. Um rebanho foi considerado positivo quando apresentou pelo menos um animal soropositivo. Das 117 propriedades utilizadas 33 (28,20%) apresentaram pelo menos um animal soropositivo, e dos 1.275 animais 69 (5,41%) foram soropositivos, com títulos variando de 100 a 3200. O sorovares reagentes foram Autumnalis (49,30%), Andamana (27,53%), Sentot (17,39%), Whitcomb (4,34%) e Australis (1,44%). Possuir mais de 48 animais no rebanho (odds ratio = 2,26; IC 95% = 1,33 -5,07; p = 0,021) e participação em exposições (odds ratio = 9,05; IC 95% = 0,96 - 85,71; p = 0,055) foram identificados como fatores de risco. Sugere-se a necessidade de estudos acerca do isolamento do agente, da caracterização de sua patogenicidade e do seu impacto econômico nos rebanhos ovinos da região, bem como recomenda-se maior controle sanitário nas aglomerações de animais.


The aim of this investigation was to determine the herd-level and animal-level prevalence of leptospirosis in sheep from the semiarid of Paraíba State, Northeast Brazil, as well as to identify risk factors. Blood samples were collected from 1,275 sheep from 117 flocks in 19 counties in the Sertão mesoregion, Paraíba. For the serological diagnosis of leptospirosis the microscopic agglutination test (MAT) using 24 Leptospira spp. serovars as antigens was carried out. Of the 117 flocks studied 33 (28.20%) presented at least one seropositive sheep, and of the 1,275 animals 69 (5.41%) were seropositive with titers ranging from 100 to 3,200. Reactant serovars were Autumnalis (49.30%), Andamana (27.53%), Sentot (17.39%), Whitcomb (4.34%) and Australis (1.44%). Herd size > 48 sheep (odds ratio =2.26; 95% CI =1.33-5.07; p=0.021) and participation in animal exhibits (odds ratio =9.05; 95% CI =0.96-85.71; p=0.055) were identified as risk factors. The need was suggested for studies on the isolation of the agent, characterization of its pathogenicity and its economic impact on sheep flocks of the region, and sanitary control in sheep agglomerations was recommended.

ABSTRACT

Este trabalho teve como objetivos determinar a prevalência de propriedades de caprinos leiteiros positivas e de animais soropositivos para leptospirose no semiárido paraibano. A amostragem foi delineada para a determinação da prevalência de propriedades positivas (focos) e de animais soropositivos para a infecção por Leptospira spp. No total, foram colhidas amostras de sangue de 975 animais procedentes de 110 propriedades leiteiras localizadas no Munícipio de Monteiro, microrregião do Cariri Ocidental, Estado da Paraíba. Para o diagnóstico da infecção por Leptospira spp., foi utilizado o teste de soroaglutinação microscópica (SAM), utilizando como antígenos 24 sorovares. Uma propriedade foi considerada foco quando apresentou pelo menos um animal soropositivo. As prevalências de propriedades positivas e de animais soropositivos foram de 43,6% (IC 95% = 34,2-53,4%) e de 8,7% (IC 95% = 5,7-12,9%), respectivamente. Nos animais, os sorovares mais frequentes foram Autumnalis (1,74%; IC 95% = 0,97-3,09%), Sentot (1,71%; IC 95% = 0,82-3,52%) e Whitcomb (1,39%; IC 95% = 0,65-2,93%), e nas propriedades, os sorovares Autumnalis (10,9%; IC 95% = 5,8-18,3%), Whitcomb (8,2%; IC 95% = 3,8-15,0%) e Sentot e Patoc (7,3%; IC 95% = 3,2-13,8%) foram os mais frequentes. Sugere-se que a infecção está distribuída em caprinos da região, e que há necessidade de implantação de medidas de controle e prevenção, com o intuito de reduzir a ocorrência da infecção e, consequentemente, diminuir perdas econômicas ocasionadas e bloquear a possível transmissão do agente aos seres humanos.


This study aimed to determine the prevalence of positive dairy goat herds and seropositive animals for leptospirosis in the semiarid region of Paraíba state, Brazil. Sampling was designed to determine the prevalence of positive herds (foci) and seropositive animals for leptospirosis. A total of 975 serum samples were collected from goats of 110 dairy herds in the Monteiro municipality, Western Cariri microregion of Paraiba. For the serological diagnosis of leptospirosis the microscopic agglutination test (MAT) was carried out using 24 Leptospira spp. serovars as antigens. A herd was considered positive when existed at least one seropositive animal. The prevalence of positive herds and seropositive animals were 43.6% (95% CI = 34.2-53.4%) and 8.7% (95% CI = 58.7-12.9%) respectively. The most frequent serovars in goats were Autumnalis (1.74%; 95% CI = 0.97-3.09%), Sentot (1.71%; 95% CI = 0.82-3.52%) and Whitcomb (1.39%; 95% CI = 0.65-2.93%), and by herd, serovars Autumnalis (10.9%; 95% CI = 5.8-18.3%), Whitcomb (8.2%; 95% CI = 3.8-15.0%) and Sentot and Patoc (7.3%; 95% CI = 3.2-13.8%) were the most frequent. It is suggested that leptospirosis is spread in goats in the region, and that there is need to implement control and preventive measures in order to reduce infection and economic losses, and to avoid possible transmission of infection to humans.

ABSTRACT

In the present work, single and mixed cereal substrates were fermented with lactic acid bacteria to study and compare the effect of the media formulation on fermentation parameters. Three cereal flours namely malt, barley and barley mixed with malt (barley-malt) were selected and fermented with two probiotic strains: Lactobacillus plantarum (NCIMB 8826) and Lactobacillus acidophilus (NCIMB 8821). The effect of the single and mixed cereal flour suspensions on the fermentation of these two strains of lactic acid bacteria (LAB) was studied at an incubation temperature of 30 °C for 28 h. It was found that the LAB growth was enhanced in media containing malt and significant amounts of lactic acid were produced (0.5-3.5 g/L). A cell concentration between 7.9 and 8.5 Log10 CFU/mL and a pH below 4.0 was achieved within 6 h of fermentation. Though the cell populations in the mixed culture fermentations of mixed substrates were similar to the ones obtained with single cereal flours, significant differences in the production of lactic acid were observed. These results suggest that the functional and organoleptic properties of these cereal-based probiotic drinks could be considerably modified through changes in the substrate or inocula composition.

ABSTRACT

This study introduces the first micro-flow-batch analyzer (µFBA). A simple, low-cost, deep urethane-acrylate photo-resist ultraviolet-lithographic technique was used in its development. Details of the microfabrication process are presented including; the use of two superimposed photo-masks to improve the micro-channel and stop chamber border definition, as well as integration of an LED/phototransistor photometric pair, while using an open nylon-thread (fishing line) micro-mixing system for solutions homogenization. The system was used for photometric determination of Fe(II) in oral solution iron supplements employing the well-known 1,10-phenanthroline method, with instantaneously prepared micro-chamber calibration solutions. All analytical processes were accomplished by simply changing the timing parameters in the control software. It must be emphasized here that there was no outside preparation of the standard calibration solutions; the mixing was all done in-chamber/in-line, with all solutions maintained flowing while being proportioned for the measurement processes. The µFBA results were acceptable when compared to the reference method, and comparable to normal flow-batch systems. It was possible both to project and build a low-cost probe with high sample throughput (about 120 h(-1)), low relative standard deviations (about 1.1%), and reduced reagent consumption (30 times less than the reference method). The µFBA system based on urethane-acrylate presented satisfactory physical and chemical properties while keeping the flexibility, versatility, robustness, and multi-task characteristics of normal flow-batch analyzers. The µFBA system contributes to the advance of micro-analytical instrumentation, while realizing the basic principles of "Green Chemistry".

ABSTRACT

Estudou-se a ocorrência de anticorpos anti-Leptospira e os fatores de risco associados à infecção em primatas do gênero Cebus mantidos em cativeiro no Nordeste do Brasil. Foram analisadas 139 amostras de soro sanguíneo de diferentes espécies de primatas de ambos os sexos e idades variadas. Para a pesquisa de anticorpos empregou-se a técnica de Soroaglutinação Microscópica (SAM) e para estudo dos fatores de risco utilizou-se análise multivariada. Foram observados anticorpos anti-Leptospira, sorovar Patoc em 6,5 por cento dos primatas (09/139). O fator de risco identificado nesse estudo foi a não higienização diária de bebedouros (Odds ratio=12,095; IC 95 por cento = 1,73-84,52; p=0,012,). Conclui-se que a população de Cebus mantidos em cativeiros no Nordeste do Brasil está exposta à infecção por anticorpos anti-Leptospira e que medidas corretivas de manejo dos animais, especificamente aquelas relacionadas à higienização diária de bebedouros devem ser implementadas para reduzir o risco de infecção por sorovares de Leptospira spp.


This study aimed to verify the occurrence of anti-Leptospira antibodies and the risk factors associated with infection in the Cebus spp. mantained in captivity in the northeast of Brazil. One hundred and thirty nine blood samples of different species of primates of both sexes and varied ages were analyzed. For the study of antibodies the Microscopic Agglutination Test (MAT) was used, and for the risk factor study a multivariate analysis was used. Antibodies anti-Leptospira, serovar Patoc were observed in 6.5 percent of primates (09/139). The risk factor identified in this study was the lack of daily hygienization of drinking fountains (Odds ratio=12.095; IC 95 percent = 1.73-84.52; p=0.012,). The conclusion of the study was that the Cebus population maintained in captivity in the northeast of Brazil is exposed to infection by antibodies anti-Leptospira and that corrective measures of animal handling, especially those related to daily hygienization of drinking fountains, must be implemented to reduce the risk of infection by serovars of Leptospira spp.

ABSTRACT

The aim of this study was to evaluate the survivability of Bifidobacterium breve NCIMB 702257 in a three malt-based media supplemented with cysteine and yeast extract, and to determine the protective effect of these growth factors. A number of parameterised mathematical models were used to predict of kinetics of viability and total acidity during storage at different temperatures. Results demonstrated a good fit to the experimental mathematical model. The Arrhenius equations showed only reasonable fits and the polynomial plots contained a large area without data between 4 and 25 degrees C. In addition, it was shown that cysteine promotes growth and acid production by bifidobacteria, but does not extend survivability. On the other hand, increasing the yeast extract content of the fermentation media enhances the survivability of B. breve. To our knowledge, this is the first study to address the modelling of the survivability of probiotic bacteria in a cereal based fermentation media at different temperatures, introducing a more quantitative approach to the study of the shelf-life of a probiotic product.

ABSTRACT

AIMS: Assessment of left and right systolic atrial reservoir function in asymptomatic mitral stenosis (MS) by strain and strain rate imaging (SRI) and their prognostic power at 3 year follow-up was the purpose of this study. There is clear indication to treat (by surgery or percutaneous valvotomy) symptomatic patients with MS, whereas for the asymptomatic ones, the question is much debated. So, we need new echocardiographic parameters helpful for the management of asymtomatic patients. Atrial reservoir function by SRI could help in evaluation of these patients. METHODS AND RESULTS: Fifty-three asymptomatic patients with MS and 53 healthy controls were evaluated by the standard echo-Doppler study [mitral valve (MV) area, mean gradient, systolic pulmonary pressure, left atrial (LA) width, LA volumes, LA compliance index] and by Doppler myocardial imaging (velocity, strain, and SR of both atria). The endpoint at 3 year follow-up was symptoms, hospitalization for cardiac cause, atrial fibrillation, thrombo-embolic events, valvular surgery, or percutaneous commissurotomy. LA width, volumes, and systolic pulmonary pressure were significantly increased in MS patients (P < 0.001). Atrial myocardial velocities and deformation indices were significantly compromised in MS patients (P < 0.0001). Significant correlation was found between atrial myocardial velocity and MV area (by pressure half-time method: P = 0.019, R = 0.41; by planimetric method: P = 0.016, R = 0.43). Peak systolic LA myocardial strain and SR were significantly correlated with atrial volumes (strain: P = 0.03, R = -0.28; SR: P = 0.0008, R = -0.42), with atrial compliance index (strain: P = 0.04, R = 0.26; SR: P = 0.04, R = 0.16), with atrial ejection fraction (strain: P < 0.0001, R = 0.56; SR: P = 0.03, R = 0.43). At 3 year follow-up, 22 (41%) patients had events. Comparing the MS patients who had events during the 3 year follow-up with those who did not, the former had bigger LA volumes, although these parameters did not reached a significant value, whereas atrial myocardial systolic SR was significantly impaired in patients with events. In multivariate analysis, the best predictor of adverse events was LA peak systolic SR average (P = 0.04; coefficient: 0.113; SE: 0.055; cut-off value of 1.69 s(-1) for LA peak systolic SR average) with a sensitivity of 88%, specificity of 80.6%, area under the receiver operating characteristic curve of 0.852 (SE: 0.048; 95% CI: 0.74-0.93, P = 0.0001). CONCLUSION: Atrial myocardial deformation properties, assessed by SRI, are abnormal in asymptomatic patients with rheumatic MS. The degree of this impairment is predictor of events in a 3 year follow-up. SRI could be helpful in decision-making of asymtomatic patients with MS.
