ABSTRACT

Background: The regulation of p21 in the pyroptosis of cartilage cells still needs to be further clarified. We aimed to explore the regulation of p21 on the pyroptosis of cartilage cells and to reveal the improvement of osteoarthritis. Methods: Chondrocytes were collected and isolated from patients with osteoarthritis (average age 58.64 ± 4.32) in Xuzhou Third People's Hospital, China in 2019, and healthy volunteers (average age 58.23 ± 3.91) were enrolled as the control group. mRNA expression levels of p21 and pyroptosis-related proteins (NLRP3, ASC, total caspase1 and cleaved-Caspase1) were detected by Western blot and real-time PCR. Cell activity, total number of cells and number of dead cells were calculated with CCK-8, MTT. And the regulation of p21 on the pyroptosis of cartilage cells was verified with overexpression and knockdown of p21 in cartilage cells. Results: In cartilage cells of patients with osteoarthritis, the transcription and translation levels of pyrolysis-related genes (NLRP3, cleaved-caspase 1, and ASC) significantly increased (P<0.01). p21 expression was up-regulated and positively correlated with the changing trend of pyrolysis-related proteins (P<0.01). Overexpressing p21 genes in normal cartilage cells significantly increased the expression of pyrolysis-related proteins (P<0.01). Conclusion: The pyroptosis of cartilage cells is causally related to the process of osteoarthritis, and can be regulated by transcription factor p21, which is a potential therapeutic target for osteoarthritis.

ABSTRACT

A circular electron positron collider (CEPC) will adopt hundreds of 650-MHz superconducting cavities with high quality factor (Q) and accelerating gradient (Eacc). Two 650-MHz single-cell cavities made of fine-grain niobium were first treated via buffered chemical polishing (BCP), which was easy and convenient. However, the vertical test results could not meet the specification of the CEPC (4 × 1010 at 22 MV/m). Therefore, electro-polishing (EP) of 650-MHz single-cell cavities was conducted, which was complicated but remarkably effective. Both 650-MHz single-cell cavities achieved state-of-the-art gradients of 35 MV/m after the EP process, which is extremely high for large elliptical cavities (frequency < 1 GHz). One cavity achieved an intrinsic quality factor (Q0) of 4.5 × 1010 at 22.0 MV/m, which was higher than the CEPC spec. The other cavity obtained a lower Q0 of 3.4 × 1010 at 22.0 MV/m, which may have resulted from the cancellation of high-temperature annealing.

ABSTRACT

This research involved the participation of 3036 Chinese students in the first and second years of senior high school. The adolescents were active users of TikTok. The mediating effect of depression, anxiety, and stress between TikTok use disorder and memory loss was investigated. A forward and backward digit span test was applied to measure memory loss. Structural equation modeling (SEM) was established, and SPSS Amos was used for analysis. The results show a partial mediation effect of depression and anxiety between TikTok use disorder and forward digit span. A partial mediation effect of depression, anxiety, and stress between TikTok use disorder and backward digit span is also shown. These results also show gender differences. Attention should be given to male students, who have more depression, anxiety, and stress than female students; they also have more memory loss.

ABSTRACT

Systematic analysis of the surface morphology, crystalline phase, chemical composition and elemental distribution along depth for nitrogen-doped niobium was carried out using different methods of characterization, including Scanning Electron Microscopy (SEM), Atomic-Force Microscopy (AFM), Grazing Incidence X-ray Diffraction (GIXRD), Rutherford Backscattering Spectrometry (RBS) and layer-by-layer X-ray Photoelectron Spectroscopy (XPS) analysis. The results showed that, after nitrogen doping, the surface was covered by densely distributed trigonal precipitates with an average crystallite size of 32 ± 8 nm, in line with the calculation result (29.9 nm) of nitrogen-enrichedß-Nb2N from GIXRD, demonstrating the phase composition of trigonal precipitates. The depth analysis through RBS and XPS indicated thatß-Nb2N was dominant in the topmost 9.7 nm and extended to a depth of 575 nm, with gradually decreased content. In addition, the successive change along depth in the naturally oxidized states of niobium after nitrogen doping, was revealed. It was interesting to find that the oxygen diffusion depth could be moderately enhanced by the nitridation process. These results established the near-surface phase composition of nitrided niobium, which is of great significance in evaluating the effect of nitrogen doping and further understanding the Q improvement of the superconducting radio frequency cavities.

ABSTRACT

OBJECTIVES: To investigate the effects of bone morphogenetic protein-2 (BMP-2) compound with fibrin on osteoporotic vertebral fracture healing in rats. METHODS: For the present study 160 Specific-Pathogen Free 32-week-old female Sprague-Dawley rats were used. 120 rats were randomly divided in three groups (experimental, model and sham operation group- n=40 per group) and were ovariectomized to establish the osteoporosis model. 40 rats served as a control group without treatment. The expression of BMP-2 in the fracture zone at the 4th, 6th, 8th, and 12th weeks was detected by qRT-PCR. The expression of BALP and CTX-I in serum at the 12th week was detected by Elisa. RESULTS: At week 8, the morphology of the sham operation group was the same and the fracture healing occurred more slowly than in the other groups. At week 12, the expression of BMP-2 in the model group was significantly higher than that in the other three groups (p<0.05). At week 12, the maximum load, maximum strain, and elastic modulus of model group were significantly lower than those of the other three groups. CONCLUSIONS: BMP-2 compound with fibrin can enhance the timing and quality of bone fracture healing in rats.

ABSTRACT

OBJECTIVE: To investigate the clinical effects of dynamic hip screw (DHS) and proximal femoral nail anti-rotation (PFNA) on senile osteoporosis patients and their effects on the expression level of bone-specific alkaline phosphatase (BALP). METHODS: 116 elderly patients with osteoporotic fracture were divided into DHS group (n=67) and PFNA group (n=49). BALP values were measured by ELISA before operation and 30 days after operation. RESULTS: The operation time, the bleeding volume, and the weight-bearing time of PFNA group was shorter than DHS group (p<0.05); the dominant blood loss and occult blood loss in PFNA group were less than those in DHS group (p<0.05); the healing time and detumescence time, the complications of PFNA group was fewer than the DHS group (p<0.05). The ten-meter walking speed and the five sitting tests in PFNA group were shorter than that in DHS group (p<0.05); the excellent and good rate and Harris score in PFNA group were higher than those in DHS group (p<0.05). The expression of BALP in PFNA group was lower than that in DHS group (p<0.05). CONCLUSION: PFNA surgery has less trauma, fewer complications, more optimistic postoperative healing and recovery degree, and is more conducive to the reduction of BALP expression level.

ABSTRACT

The quality marker(Q-marker) of traditional Chinese medicine(TCM) is a new concept of TCM quality control proposed in recent years. It is a hot issue in the research of modern Chinese medicine. The TCM efficacy is a high-level summary of the TCM therapeutic effect under the guidance of TCM theory. On this basis, it is of considerable significance to explore the TCM efficacy marker for the TCM modernization. However, the traditional research strategy based on the single herb and decoction piece in macro TCM level, or the drug research strategy based on the biological effect of the targets, is quite different from the characteristics of multiple components of TCM, as well as the weak and low-selective effect of Chinese medicine ingredients on targets. Therefore, how to select representative ingredients to characterize the TCM overall efficacy is a problematic point in establishing TCM efficacy markers. In this paper, the concept and method of Q-marker were introduced into the study of Chinese medicine efficacy. The research method for systematic TCM was used to systematically discuss the connotation of TCM efficacy markers, the principles of discovery and determination, common research ideas and techniques by taking the representative research results as an example. This study provides new ideas for the research and discovery of TCM efficacy markers.

ABSTRACT

Astragali Radix is the elixir for invigorating Qi, with the effects of invigorating Qi, promoting Yang and nourishing the body. With the deepening researches on the chemical constituents of Astragali Radix, it is used more extensively in clinical application. Based on systematic traditional Chinese medicine theory, in this paper, we characterized the effect of Astragali Radix on invigo-rating Qi from the molecular level, and explored the markers of Astragali Radix on invigorating Qi. Through TCMSP and ChEMBL databases, the active components-targets database of Astragali Radix was constructed to clarify the targets(elements) involved in Astragali Radix's Qi invigorating efficacy system. According to the relationship between the targets, the protein interaction network was constructed, and the network modules(structure) were divided according to the theoretic clustering algorithm molecular complex detection(MCODE), and the boundary of the Qi invigorating efficacy system was defined by the pharmacological function of Astragali Radix. The active components of Astragali Radix for invigorating Qi were characterized from the aspects of composition, target and efficacy. The results showed that eight key components of Astragali Radix, such as hederagenin, quercetin, calycosin, formononetin, jaranol, isorhamnetin, astragalosideⅢ, and 9,10-dimethoxypterocarpan-3-O-β-D-glucoside, could act on eight functional modules composed of 17 key targets, and participate in G-protein coupled receptor protein signaling pathway, regulation of lipid metabolic process, positive regulation of nitrogen compound metabolic process, positive regulation of programmed cell death, fatty acid metabolic process and other biological processes to produce pharmacological effects such as regulating immune function, strengthening heart, protecting myocardial cells, improving material metabolism, and antioxidation effects, thus playing the role of invigorating Qi. Based on the systematic Chinese medicine theory, this study explored the effective markers of Astragali Radix at the level of molecular network, which provided new ideas for the interpretation of the effective substance basis of systematic traditional Chinese medicine and the quality control of traditional Chinese medicine. In the future, it can focus on the compatibility research of these components, and then carry out more in-depth studies on the efficacy of Astragali Radix in invigorating Qi, and strengthen the development of the corresponding pharmacological mechanism and related preparations.

ABSTRACT

Lonicerae Japonicae Flos has a long history of heat-clearing and detoxifying effect. The description of its efficacy in Chinese Pharmacopoeia of past dynasties is relatively stable, and it is an excellent carrier for the study of efficacy markers. Guided by the theory of systematic traditional Chinese medicine, heat-clearing and detoxifying effect efficacy system of Lonicerae Japonicae Flos was taken as an example in this study to clarify the elements(active ingredients) of Lonicerae Japonicae Flos in heat-clearing and detoxifying efficacy system, determine the boundary(signal pathway), establish the structure(system dynamics model), identify the system functions corresponding to pharmacology, efficacy and effects(heat-clearing and detoxifying effect), and explore the application of system dynamics model in the discovery of efficacy markers of traditional Chinese medicine. In this paper, the dynamic models of interleukin 1(IL-1) and interleukin 6(IL-6) in vivo were established to predict the expression of related factors in IL-1 and IL-6 signaling pathways of different components and their combinations in Lonicerae Japonicae Flos by dynamic network, so as to find the effective markers of heat-clearing and detoxification of Lonicerae Japonicae Flos. The results showed that the lower the concentration of chlorogenic acid, the higher the inhibition rate of Jun N-terminal kinase(JNK) at downstream of IL-1 by the combination of chlorogenic acid and linalool; the higher the concentration of luteolin in IL-6 pathway, the higher the inhibition rate of C-reactive protein(CRP) at downstream of IL-6 by the combination of chlorogenic acid and luteolin. It revealed that the potential efficacy markers of Lonicerae Japonicae Flos in heat-clearing and detoxifying effect based on IL-1 signaling pathway were chlorogenic acid and linalool, and the potential efficacy markers of Lonicerae Japonicae Flos in heat-clearing and detoxifying effect based on IL-6 signaling pathway were chlorogenic acid and luteolin. This study provided methodological guidance for the discovery of efficacy markers of traditional Chinese medicine.

ABSTRACT

This study aimed to explore the improvement of hip replacement combined with alendronate sodium on the condition of patients with osteoporotic femoral neck fracture and factors affecting the efficacy of patients. In total, 140 patients with femoral neck fracture from July 2015 to October 2017 in the Affiliated Xuzhou Hospital of Jiangsu University were collected. Of these, 61 patients were treated with hip replacement as the control group and 79 patients were treated with alendronate sodium as the observation group on the basis of the control group. ELISA was used to detect levels of carboxy-terminal opeptide of type I collagen (CTX-I) and bone alkaline phosphatase (BALP) in serum of patients before and after treatment. Harris score was used to compare the clinical efficacy of patients after treatment. Changes in the expression of CTX-I and BALP before and after treatment were compared between the two groups, and the correlation between CTX-I and BALP levels and Harris score was analyzed. According to the clinical efficacy of patients, the two groups were divided into the significant effect group and poor effect group. Risk factors affecting the efficacy of patients were analyzed, and the ROC of subjects with risk factors was drawn. After treatment, the expression of BALP in serum increased significantly compared with that before treatment, and the expression of CTX-I decreased significantly. After treatment, the expression of BALP in serum in the observation group was significantly higher than that in the control group (P<0.05). Multivariate analysis revealed that age, time of operation, CTX-I after treatment and BALP after treatment were independent risk factors affecting the efficacy of patients. In conclusion, hip replacement combined with alendronate sodium can effectively improve the clinical efficacy of patients, and age, time of operation, CTX-I after treatment and BALP after treatment are found to be independent risk factors affecting the postoperative efficacy of patients.

INTRODUCTION: Smartphones are ubiquitous in the digital society. Although this powerful device is useful because it supports and simplifies many tasks in everyday life, a growing number of researchers is concerned that smartphones might have detrimental effects on the human brain and related psychological processes. Evidence indicates that social media platforms such as WhatsApp and Facebook are essential drivers of smartphone usage. Thus, a critical, yet unexplored issue is how excessive use of those platforms is related to Smartphone Use Disorder (SUD). Furthermore, since the roles of life satisfaction and fear of missing out (FoMo) have been demonstrated to be of particular importance for Internet Use and Internet Communication Disorder, those constructs were examined in the context of SUD. METHODS: In total, n = 2299 participants filled in questionnaires assessing SUD, WhatsApp and Facebook Use Disorder, FoMO, life satisfaction, and reported owning a WhatsApp and Facebook account. RESULTS: The study revealed that SUD was linked to WhatsApp Use Disorder, and to a lesser extent, to Facebook Use Disorder. Associations between SUD and WhatsApp Use Disorder were most strongly pronounced in females. Additionally, FoMo predicted SUD, WhatsApp, Facebook Use Disorder, and (fully) mediated the relationship between life satisfaction and those variables. CONCLUSION: The findings of the present study suggest that WhatsApp use plays an important role to understand SUD. Regarding the mediating role of FoMO, the present work might help disentangle inconsistent results on the link between life satisfaction and (excessive) social networking sites use.

In the present study, we investigated the relationship between depression and Internet Use Disorder (IUD) and between burnout and IUD among German as well as Chinese college students. Due to cultural differences and their implications for the individual's psychological health, we expected Chinese college students to have in particular higher IUD than German college students. We further expected to find positive relationships between depression and IUD and between burnout and IUD. Furthermore, we believed these relationships to reflect global effects and thus to be present in both samples. The data showed that Chinese college students had higher average burnout scores in the subscales MBI Emotional Exhaustion and MBI Cynicism and also higher IUD scores, but not higher depression scores. As expected, the correlation analysis revealed significant, positive correlations between depression and IUD as well as between burnout and IUD. The results are consistent in both samples, implying that the effect is globally valid. Furthermore, we observed that the relationship between depression and IUD is stronger than the relationship between emotional exhaustion and IUD in both samples, although this effect was not significant. We conclude that burnout and depression are related to IUD and that this relationship is valid independently of the cultural background of an individual.

ABSTRACT

Attention-deficit/hyperactivity disorder (ADHD) is a neurodevelopmental disorder that is characterized by inattention, hyperactivity, and impulsivity but also by negative emotionality. The aim of the present study was to investigate whether subclinical ADHD tendencies are associated with negative emotionality in healthy adult samples. The present study is of special interest since it investigated negative emotionality with a questionnaire anchored in Neuroscience Theory-the Affective Neuroscience Personality Scales (ANPS). Furthermore, through the investigation of samples in two countries, namely Germany and China, the study aims to replicate the results across different cultures. German (n = 377; age: M = 23.25, SD = 8.47; 117 males) and Chinese (n = 389; age: M = 20.74, SD = 2.47; 279 males) subjects completed ANPS (primary emotional traits) and ASRS (ADHD tendencies) questionnaires in an online survey. Principal component analysis of the ANPS revealed one factor for negative emotionality and one factor for positive emotionality. Partial correlations between ANPS and ASRS (controlled for age) were conducted separately for nation and gender. The same correlation patterns between ADHD tendencies and negative emotionality could be found in male and female German/Chinese participants (range r= .189 to r = .352). Higher negative emotionality was always significantly associated with more inattentive, hyperactive/impulsive, or combined tendencies. However, significant negative correlations between ADHD tendencies and positive emotionality could only be observed in Chinese males (range r = - .264 to r = - .296). The results are in line with former findings in children and show that also in healthy adults, associations between negative emotionality and ADHD tendencies are robustly visible. The results were independent of the cultural background, indicating a general association between ADHD tendencies and negative emotionality, even in healthy adults.

Research on Internet-use Disorder (IUD) has increased rapidly, indicating its clinical and global importance. Past studies suggested cultural diversity regarding the prevalence of an IUD, e.g., between Asian and European countries. Additionally, it was found that personality factors, Internet-related cognitions and specific competences seem to influence IUD tendencies, but research lacks in cultural comparative studies regarding these mechanisms. This study focuses on differences between Germany and China regarding the above-mentioned characteristics. German (n = 411; M = 20.70 years, SD = 3.34 years) and Chinese participants (n = 410; M = 20.72 years, SD = 2.65 years) answered the short Internet Addiction Test, Big Five Inventories, the Internet-use Expectancies Scale, as well as the Internet Literacy Questionnaire. The results revealed higher occurrence of IUD symptoms in China. Furthermore, Chinese participants scored significantly higher on neuroticism and agreeableness, whereas German participants scored higher on extraversion and openness. Compared to German participants, Chinese showed higher expectancies to avoid negative feelings online and to be positively reinforced. Regarding Internet literacy, German participants indicated higher skills concerning the reflection and critical analysis of online content, whereas Chinese showed higher expertise in producing and interacting online. Further, simple slope analyses indicated that certain Internet literacy domains were related differentially to IUD symptoms in Germany and China. While Chinese participants with higher reflective skills indicated highest IUD symptoms, reflective skills revealed no effect in Germany. Additionally, higher self-regulative skills correlated with lower IUD symptoms in the German, but not in the Chinese sample. The results give a hint to potential cultural differences regarding IUD, especially on the predictive and protective role of Internet literacy domains.

INTRODUCTION: Several studies suggest that Asian countries are particularly afflicted by problematic Internet use (PIU). METHODS: The present study investigates whether individual differences in the acceptance of power distance can be linked to overuse of the Internet in Germany and in China. Power distance has been discussed as an essential dimension on which Asian and Western societies differ. In the present study, we investigate two large non-clinical samples from Germany (n = 297) and China (n = 556) to address this question. RESULTS: Both in Germany and in China, high acceptance of power distance was positively associated with PIU. These effects were more pronounced in China compared with Germany. Moreover, the observed effects were stronger in males compared with females. DISCUSSION: Clearly, these findings are just a starting point and need to be replicated in the future. Clinical populations and a further important difference variable - collectivism - also merit consideration in future work.

INTRODUCTION: It has been hypothesized that two distinctive forms of Internet addiction exist. Here, generalized Internet addiction refers to the problematic use of the Internet covering a broad range of Internet-related activities. In contrast, specific forms of Internet addiction target the problematic use of distinct online activities such as excessive online video gaming or activities in social networks. METHODS: The present study investigates the relationship between generalized and specific Internet addiction in a cross-cultural study encompassing data from China, Taiwan, Sweden and Germany in n = 636 participants. In this study, we assessed - besides generalized Internet addiction - addictive behavior in the domains of online video gaming, online shopping, online social networks and online pornography. RESULTS: The results confirm the existence of distinct forms of specific Internet addiction. One exception, however, was established in five of the six samples under investigation: online social network addiction correlates in large amounts with generalized Internet addiction. DISCUSSION: In general, it is of importance to distinguish between generalized and specific Internet addiction.

ABSTRACT

The aim of this research is to find an effective cardiomyocyte-induced method derived from porcine amniotic fluid stem cells (pAFS). For cardiac differentiation, the cells were formed embryoid bodies (EBs) firstly, then cultured in induced-medium including 5-azacytidine (5-aza) and vitamin C (Vc). We detected the specific markers of cardiomyocyte by immunocytochemistry, RT-PCR and transmission electron microscope. The results showed that some embryoid bodies beat rhythmically after 10 days of induction. Furthermore, analysis of t test revealed that the percentage of beating cardiomyocyte-like cell clusters was highest (33%) when induction using 0.1 mmol/L Vc and 5 micromol/L 5-aza. Immunocytochemistry analysis demonstrated that cardiomyocyte-like cell clusters expressed alpha-actin, Tnni3. RT-PCR analysis also illustrated that TbX5, Gata4, alpha-MHC and Tnni3 were expressed positive in cardiomyocyte-like cell clusters. Especially, we observed basic structures of myocardium, such as myofilament, glycogen granule and so on by transmission electron microscope. In conclusion, 5-azacytidine and vitamin C could promote differentiation of pAFS into myocardium.
