ABSTRACT

The objective of this study was to investigate the protective effects and the underlying mechanisms of resveratrol (RES) against hydrogen peroxide (H2O2)-induced oxidative stress in bovine skeletal muscle cells (BMCs). Pretreatment of BMCs with RES prior to H2O2 exposure increased cell viability, attenuated reactive oxygen species, and stabilized the redox state. H2O2 exposure activated sirtuin type 1 (SIRT1) and nuclear factor E2-related factor 2 (NRF2)-mediated signaling pathways. Pretreatment with RES did not alter SIRT1-regulated genes but inhibited the upregulation of NRF2, whereas enhanced heme oxygenase 1 (HO-1) expression. Pretreatment with RES prior to H2O2 exposure failed to suppress NRF2 expression when NRF2 was knocked down by RNA interference. However, HO-1 expression still could be induced by RES. These results suggest that RES has benifical effects against oxidative stress. NRF2-mediated pathway play an important role, and HO-1 upregulation is the key process in RES regulation. RES may be used as a therapeutic agent for meat quality improvement in beef cattle.

ABSTRACT

To evaluate the effects of Spirulina platensis and probiotics on growth, immunity indexes, fecal flora, and fecal odor in mice, 40 mice were randomly allotted to four groups, and each was administrated with nothing, S. platensis, probiotics, or both for 28 days, respectively. Then, many indexes were measured. The results showed that S. platensis was more effective (P < 0.001) than probiotics in improving mice's feed conversion ration (FCR). In immunity, probiotics administration increased (P < 0.042) serum IgE, IgM, IFN-γ, colonic AHR, TLR4, and NF-κB protein expression and decreased (P < 0.039) serum IL-1α, IL-21, IL-22, and colonic ARNT gene expression. However, the S. platensis showed weaker effect, which increased (P < 0.025) the serum IgE, IgM, TNF-α, and the colonic AHR and NF-κB protein expression, and decreased (P < 0.01) serum IL-21. Probiotics consumption decreased the fecal odor by decreasing (P < 0.02) fecal Escherichia coli, indole-3-acetic acid (IAA), and skatole contents, and the S. platensis decreased (P = 0.04) the IAA. These results indicated that oral administration of probiotics, S. platensis, or both of them in mice probably benefited body's immunity and reduced fecal odor. However, their mechanisms were still unclear and need further study.

ABSTRACT

MicroRNAs are short non-coding RNAs that regulate gene expression. Several microRNAs, useful for coronary artery disease assessment, have previously been identified. MicroRNA-33 is located within SREBP introns and controls cholesterol homeostasis. In order to find the possibility of microRNA-33 as a potential biomarker in high cholesterol disease, we developed a mouse model for coronary heart disease by feeding mice with a high-fat diet. The expression differences of microRNA-33, SREBP and ABCA1 genes in the liver, muscle, and lipid tissues were compared between a high-cholesterol group and control group in mice. The results showed that ABCA1 was up-regulated by high cholesterol conditions in liver, muscle and lipid tissues. SREBP1C was up-regulated by high cholesterol conditions in the liver and lipid tissues and down-regulated by high cholesterol conditions in the muscle tissue. MicroRNA-33 and SREBP2 were down-regulated by high cholesterol conditions in the liver and muscle tissues and up-regulated by high cholesterol conditions in the lipid tissue. Our study suggests that antisense therapeutic targeting of microRNA-33 may be a potential biomarker for cardiovascular disease.

ABSTRACT

Conservationists strive for practical, cost-effective management solutions to forest-based species conservation and climate change mitigation. However, this is compromised by insufficient information about the effectiveness of protected areas in increasing carbon storage, and the co-benefits of species and carbon conservation remain poorly understood. Here, we present the first rigorous quantitative assessment of the roles of giant panda nature reserves (NRs) in carbon sequestration, and explore the co-benefits of habitat conservation and climate change mitigation. Results show that more than 90% of the studied panda NRs are effective in increasing carbon storage, with the mean biomass carbon density of the whole NRs exhibiting a 4.2% higher growth rate compared with lands not declared as NRs over the period 1988-2012, while this effectiveness in carbon storage masks important patterns of spatial heterogeneity across the giant panda habitats. Moreover, the significant associations have been identified between biomass carbon density and panda's habitat suitability in ~85% NRs and at the NR level. These findings suggest that the planning for carbon and species conservation co-benefits would enhance the greatest return on limited conservation investments, which is a critical need for the giant panda after its conservation status has been downgraded from "endangered" to "vulnerable".

ABSTRACT

Spatiotemporal patterns of forest carbon (C) sinks and accurate estimation of such patterns are crucial to sustainable forest management. We combined individual tree biomass equations and a Random Forest algorithm to assess the spatiotemporal changes in biomass C sequestration and to further quantify the relative contributions of forest areal expansion and growth to biomass C sinks in Sichuan Province, China, over the past 25 yr. Forest area and average biomass C density increased from 10.5 million ha and 45.7 Mg C ha in 1988 to 14.2 million ha and 52.3 Mg C ha in 2012. Average C density was generally larger in the north and west of Sichuan Province compared with other regions. The expanded forest area and enhanced C density have jointly led to a rise in total C storage by 54.9% over this period in Sichuan Province. It was estimated that the forest areal expansion has been a larger contributor to C sinks than forest growth in Sichuan Province (69 vs. 31%), especially in the regions of the northwestern high mountains and the hilly country of the Sichuan basin. However, the relative contributions of areal expansion exhibited different trends in five subregions and 15 forest species groups in this province. Our study suggests that it is necessary to develop a new forestry management mode to maintain the long-term health of forest ecosystems in Sichuan Province, which should attach more importance to improving forest quality and selecting tree species in different subregions while increasing forested area in the future.

Hypoglycemic polypeptide (PA) was extracted from Momordica charantia seeds with organic acid and ethanol and purified with Sephadex G-50 gel filtration and RP-HPLC. PA was judged as plant insulin on the base of the analysis of its SDS-PAGE electrophoresis and amino acid composition.
