ABSTRACT

Fractions of copper-binding protein (CBP) specifically bound copper ions were extracted from the rat liver cell cytosole. These fractions of 10-14 kDa proteins are involved in specific pattern of intracellular distribution of copper ions. The imprinting effect of specific pattern of copper ions intracellular distribution has been found. The effect was detected 30 days after sequented injections of copper sulfate into the body. It was shown, that after primary injection of copper the ability of CBP to bind copper ions could increase tenfold, regardless of schemes of copper sulfate injection.

ABSTRACT

The duration and reversibility of the liver chalone-containing fraction on DNA synthesis in regenerating liver cells have been studied. It has been shown that administration of the [14C] chalone-containing fraction results in a rapid specific binding of the label to liver cells in which the label is retained for no less than 12 hours. The specific radioactivity of DNA in regenerating liver cells decreases 3-4-fold irrespective of the time of administration of the [14C] chalone-containing fraction. After repeated injections of the fraction under study, the hepatocytes loss their sensitivity towards this fraction. It is suggested that chalones exclude some part of the hepatocytes from the cell cycle which, in turn, leads to the asynchronous involvement in the cell cycle of a new hepatocyte population which is resistant to the chalone action.

ABSTRACT

The age dynamics of albino rat kidney cell proliferation in conditions of compensatory hypertrophy and influence of endogenous inhibitors on this process have been studied. The lack of influence of repeated injection of inhibitors on kidneys cell division of 1-month old rats has been shown, while the level of kidney proliferation of 3-month old rat and 12-month old rat decreased of about 50% and of 24-month old rat decreased of about 34%.

ABSTRACT

It was demonstrated that during postnatal ontogenesis, the heterogeneity of endogenous inhibitors of DNA synthesis in albino rat liver does not change. Upon ageing, the sensitivity of liver cells to the inhibiting effect of chalone-containing extracts and the functional activity of the inhibitor are diminished.

ABSTRACT

Contant composition of histones and marked alterations in the non-histone proteins were found in condensed and diffuse chtomatins of rat liver tissue during postnatal ontogenesis both in normal state and in treatment with thyroxine. Thyroxine affected the ratio histones (non-histone proteins in condensed chromatin of animals of the all age groups studied, except the old rats, in which the ration did not differ from the control values. After thyroxine administration the ratio histones) non-histone proteins was increased in diffuse chromatin of predolescent rats; the ratio was slightly decreased in diffuse chromatin of puberal rats and it was remarkably unaltered in the chromatin of adult and t was remarkably unaltered in the chromatin of adult an old animals. Considering the fact that thyroxine increased in the condensed chromatin the content of non-histone proteins and decreased the content of histones, the data obtained suggest that the hormone caused a shift in the ratio condensed/diffuse chromatin towards the increase in content of the diffuse chromatin.

ABSTRACT

It was shown that after treatment by Ca2+- and Mg2+-dependent DNAses and subsequent dosed ultrasonication the fractions of active and relatively inactive chromatins isolated from liver cell nuclei of rats differing in age contain all main types of histones, but differ considerably in the relative amounts of individual fractions of these proteins. In all age groups studied the proteins of relatively inactive chromatin are largely histones, while the amount of non-histone proteins is higher in active chromatin. In the course of postnatal development the amount of histones in both chromatin fractions is increased and that of non-histone proteins is decreased. This is probably due to heterochromatization of the chromatin complex in liver cells with ageing. In the course of postnatal ontogenesis the spectrum of non-histone proteins in both chromatin fractions is changed.
