ABSTRACT

【Objective】 To explore the expression of USP9X in platelets and its effect on platelet function. 【Methods】 The expression of USP9X in human and mouse was evaluated by PCR and Western blot. Platelets from young and old mice were separated and prepared, and the expression of USP9X was detected. USP9X inhibitos were used to assess the regulation of USP9X in platelet function, including aggregation, ATP release and spreading. Platelet lysates were collected in different time points to evaluate the change of phosphorylation of Akt in USP9X inhibitors treated platelets. 【Results】 Both human and mouse platelets expressed USP9X. Compared to the young mice, the old mice showed significantly enhanced expression of USP9X(P<0.05). To assess the effect of USP9X on platelet function, USP9X inhibitor was used to pre-incubate platelets for 30 min and platelet function were examined later. Results showed that USP9X inhibitor significantly decreased platelet activation including aggregation, ATP release and spreading(P<0.05). Compared to the control group, the inhibitor treated group showed a significant decrease in the spreading area after 45 minutes. The Western blot results showed a significant decrease in Akt phosphorylation levels of platelets in the USP9X inhibitor treated group. 【Conclusion】 Both human and mouse platelet express USP9X, and inhibition of USP9X decreased platelet function including aggregation, ATP release and spreading. USP9X can also influence the phosphorylation of Akt. The inhibitor of USP9X may become a potential therapeutic target for thrombosis intervention.

ABSTRACT

【Objective】 To investigate the influence of knowledge popularization concerning blood donation and blood use on the knowledge, attitudes and practice(KAP) of voluntary blood donation of patients′ families. 【Methods】 Knowledge popularization on voluntary blood donation and clinical blood use was conducted to family members of tumor patients. A questionnaire was designed according to KAP theory to capture the general situation, blood donation history and demographic characteristics of patients′ family members, their knowledge on blood donation and clinical blood use, as well as their intention, attitude and behavior changes on voluntary blood donation before and after popularization. 【Results】 13.49%(104/771) of the family members of tumor patients donated, and most of them aged 28~37 years old(19.7%, 26/132). The awareness rate of patients′ family members on voluntary blood donation and clinical blood use after popularizing was significantly increased as compared with before(P<0.05). Especially, the awareness rate of "blood transfusion significance and the risk of infectious diseases", and " patients were given priority to use blood if their family members donated blood" increased to 61.35%(473/771) and 68.74%(530/771), respectively.Their intention, attitude and behavior of blood donation also changed significantly(P<0.05). The intention of supporting voluntary blood donation increased to 78.21%(603/771), and non-supporting decreased to 4.15%(32/771). Such three behaviors led to a donation rate higher than 70% as donating for charity and sense of responsibility(74.06%, 571/771), donating blood after reassuring(70.69%, 545/771), and promoting their family members′ priority in blood use(71.47%, 551/771). 【Conclusion】 The popularization of knowledge concerning blood donation and blood use can change the intention and attitude of patients′ family members towards voluntary blood donation and further effectively promote their donation behavior.

ABSTRACT

Objective: To observe the effects of music electro-acupuncture and pulsed electro-acupuncture on locomotor avtivity and hippocampal astrocytes in fast-aging (SAMP8) mice, and to compare the anti-dementia mechanism.Methods: Thirty 8-month-old male SAMP8 mice were randomly divided into model group (group M), music electroacupuncture group (group MA), pulsed electro-acupuncture group (group EA) (n=10), with homologous normal aging SAMR1 mice as control group (group C) (n=10) . Acupuncture stimulation was applied to"Baihui" (GV20) 、"Yintang" (GV29) and"Renzhong" (DU26) for 20 min per day for 15 days. The Morris water maze was used to assess learningmemorize ability. Immunohistochemical DAB staining was used to observe GFAP exptession in hippocampus. Detect GFAP protein levels by Western blot in hippocampus. Results: The Morris water maze test showed: Compared with group C, the escape latency were increased (P < 0.01), swimming distance were increased in space exploration experiment in group M (P < 0.01) . Compared with group M, the escape latency were reduced in group MA and EA (P < 0.01, P < 0.05) and the swimming distance were shortened (all P < 0.05) . The immunohistochemical DAB staining showed: Compared with group C, the average optical density of GFAP in the hippocampal CA1 area of group M increased significantly (P <0.01); the optical density of GFAP in CA1 area of hippocampus in both groups MA and EA was significantly decreased (all P < 0.01), while the optical density of GFAP in MA group was lower than that of EA group (P < 0.05) . Western blot test showed: Compared with group C, the expression of GFAP protein obviously increased (P < 0.01) . Compared with group M, the expression of GFAP protein obviously decreased and had a downregulation tendency in group MA and EA (all P < 0.05), while the expression of GFAP protein decreased in MA group compare with EA group (P < 0.05) .Conclusion: Both music electro-acupuncture and pulsed electro-acupuncture can improve the learning and memory abilities of SAMP8 mice and decrease the expression of GFAP, which may be related to its mechanism of reducing neuroinflammatory reaction, improving apoptosis and eventually protecting neurons, and music electro-acupuncture has a better tendency than pulsed electro-acupuncture.

ABSTRACT

OBJECTIVE To prevent the nosocomial infection event in the clinical HIV laboratory. METHODS There were risk factors of hospital infection existing in clinical HIV laboratory.To improve the management,enhancing rules and regulations,and correspond controlling measures were necessary. RESULTS By the occurrence of hospital infection in the clinical HIV laboratory could effectively prevented. CONCLUSIONS The clinical HIV laboratory can effectively prevent the occurrence of laboratory hospital infection,through improving the management,enhancing the necessary rules and regulations,improving the organization and realizing the corresponding controlling measures.
