ABSTRACT

In 2007, sweet corn plants (Zea mays cv. Challenger) with symptoms of mosaic resembling those caused by Maize dwarf mosaic virus (MDMV) were observed in production fields in the Centre Region of France. In 10 sweet corn fields surveyed, yield production was decreased by 10 to 20%. Potyvirus-like particles were observed by electron microscopy. Twenty samples from affected plants (cv. Challenger) showing severe mosaic symptoms were tested by ELISA with rabbit polyclonal antibodies to MDMV (Montpellier SupAgro, France) and SCMV (Sugarcane mosaic virus) (Bioreba, Reinach, Switzerland). For all leaves tested, slight and strong positive reactions were observed for MDMV and SCMV, respectively, indicating that the plants were infected by SCMV. Filtered phosphate buffer extracts of infected leaves of field plants (cv. Challenger) were used to mechanically inoculate leaves of different corn plants at the three-leaf stage. Sweet corn cultivars (Atugan, Challenger, Golda, Max, and Rana) and seed corn cultivars (DK315, DK3152, DKC3420, Marcello, and PR38H20) were tested. Two weeks after inoculation, mosaic symptoms were observed on leaves. The sweet corn cultivars (with the exception of cv. Max) were considerably more sensitive to the virus. Using two lots of 1,000 seeds each (cv. Challenger), a rate of seed transmission was estimated to be 0.5 to 0.6%. Total RNA was extracted from infected leaves of cv. Challenger 3 weeks after mechanical inoculation. To confirm the potyvirus species involved, a short conserved fragment of the coat protein gene was reverse transcribed, PCR amplified and sequenced (3). The sequence was deposited in GenBank (Accession No. HM014060). Multiple sequence alignment and phylogenetic analysis clearly grouped this isolate called SCMV-MFc (Maize France centre) in the 'Maize' cluster of SCMV, which consists exclusively of isolates from maize. Nucleotide sequence comparison showed that SCMV-MXVrP, a Mexican isolate (2), and SCMV-P3, a Polish isolate (4), have the two closest sequences (285 identical nucleotides of 286). SCMV-MFc is more distantly related with the described SCMV German isolates (1). This is in contrast to SCMV isolates from the traditional Saccharum officinarum cultivars that can be clustered by geographical origin (1). This result could be a consequence of the possible seed transmission of the maize isolates of SCMV. ABSTRACT

A potyvirus (proposed name of Zea mosaic virus [ZeMV]) isolated from maize in Israel was analyzed by serology, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of capsid proteins, symptomatology, and sequencing. Parts of the nuclear inclusion b, coat protein, and 3' regions were sequenced; the amino acid sequence of ZeMV capsid was determined by time-of-flight mass spectrometry (TOFMS). The results of these analyses were compared with those of similar analyses of the following potyviruses: Maize dwarf mosaic virus (MDMV), Sugarcane mosaic virus strain MDB (SCMV-MDB), Johnsongrass mosaic virus(JGMV), Sorghum mosaic virus (SrMV), and an isolate of MDMV from Israel. Indirect enzyme-linked immunosorbent assay using ZeMV antiserum detected only ZeMV, and reciprocal tests using MDMV, JGMV, or SrMV antisera failed to detect ZeMV. ZeMV cross-reacted weakly when SCMV-MDB antiserum was used. The mass of ZeMV capsid was determined to be 36,810 Da by SDS-PAGE and 34,216 Da by TOFMS. The ZeMV systemically infected johnsongrass (Sorghum halepense), but did not infect oat (Avena sativa), pearl millet (Pennisetum glaucum), barley (Hordeum vulgare), or rye (Secale cereale). Necrosis was caused in 19 sorghum lines by SrMV, in 15 by ZeMV, in 14 by MDMV, and in 5 by JGMV and SCMV-MDB. The nucleic acid and amino acid sequences of ZeMV clearly showed that it is not a strain of JGMV, MDMV, SCMV, or SrMV.

ABSTRACT

By using slow thermal transients of reduced amplitude (+/- 3 degrees C (thermoneutrality in humans sleeping nude) during only 1 night (experimental, EX), we have advanced the minimum of rectal temperature (Tre) and the peak of their paradoxical sleep propensity (PPSP) of sleeping subjects. During this EX night Tre minimum was significantly (P = 0.0001) advanced by 143 min versus that observed during baseline night spent at thermoneutrality. The advance of PPSP was objectivated by the more rapid cumulation of paradoxical sleep (P = 0.02) during the second half of EX night, i.e. strictly after the occurrence (around 0330 h) of the new Tre minimum, and by the earlier occurrence of its barycentric point (P = 0.04) between 0330 and 0700 h. The involvement of the central thermoregulatory system on phase-shifting mechanisms is discussed.

ABSTRACT

cDNA complementary to the 3'-terminal half of RNA 1 of wheat spindle streak mosaic virus (WSSMV) from Southern France has been cloned and sequenced. One large open reading frame (ORF) of 4410 nucleotides and a nontranslated region (NTR) of 213 nucleotides at the 3'-end excluding the poly(A)-tail were found. Because of the amino acid sequence homology to the polyprotein of barley yellow mosaic virus (BaYMV) RNA 1, the encoded polyprotein of the sequenced region of WSSMV is supposed to comprise the C-terminal part of the putative cytoplasmic inclusion (CI) protein, the nuclear inclusion a (NIa) proteinase, the (NIb) RNA-polymerase and the capsid protein. The first 19 N-terminal amino acids of the capsid protein were determined by direct sequencing of proteins of purified WSSMV particles and confirmed this hypothesis. The deduced capsid protein has 294 amino acids and shows 74% identity with the BaYMV capsid protein sequence. This high sequence homology with BaYMV, in addition to the significant identities with barley mild mosaic virus (BaMMV, 35%) and its marginal homology to capsid protein sequences of aphid and mite-borne potyviruses (22-24%), supports the classification of WSSMV as a distinct member of the genus Baymovirus, family Potyviridae.

ABSTRACT

A principal components analysis was carried out on subjective data obtained from the sleep questionnaire systematically used in our laboratory on the morning following a polysomnographic recording. This technique, based on 70 healthy subjects, revealed three principal axes which explore the questionnaire. The first axis reflects the subjective evaluation of the quality of sleep. The second underlines the importance of dreams and of the brief waking periods which immediately follow. Finally, the third axis brings out the subjective assessment of sleep efficiency. The third axis was the most ambiguous and the only one to show a gender difference. Women tend to better estimate general sleep events, while men tend to evaluate better the more circumstantial events. Finally, given the results, it would be interesting to apply a similar analysis to populations presenting sleep disorders in order to establish the presence of an eventual pattern specific to the disorder.

ABSTRACT

The echocardiographic appearances of a patient with a rare congenital malformation, interatrial septal aneurysm (IASA) associated with a hemodynamically significant left-to-right shunt, are described. A 52 year old man with Noonan's syndrome was admitted to hospital for atrial flutter with right heart failure, which, on investigation, led to the diagnosis of a rare type of atrial septal defect; 2D echocardiography showed an isolated aneurysm of the interatrial septum; cardiac catheterisation demonstrated a significant left-to-right (5 volumes of oxygen per 100 ml). The patient underwent surgery which confirmed the presence of a multiperforated aneurysm of the interatrial septum in the region of the fossa ovalis. This abnormality was resected and the interatrial defect closed; there were no postoperative complications and the cardiomegaly regressed significantly. The authors underline the M mode and 2D appearances of this condition; although it may be difficult on M mode examination to distinguish an IASA from other conditions giving rise to mobile intra-right atrial echos, such as Chiari's malformation, tricuspid valve vegetations, intraatrial tumour or thrombus or Eustachian valve, 2D examination using different incidences and contrast injections generally confirms the diagnosis. Surgery is often necessary after the detection of this echocardiographic abnormality: the surgical indications are described.
