ABSTRACT

Physical activity (PA), sedentary behavior (SB), and sleep duration are known to have an individual effect on clinic blood pressure (BP) of older adults. However, whether different patterns of these so-called movement behaviors over the 24h-cycle on BP remains poorly investigated. The study aimed to identify movement behavior patterns associated with clinic BP among older adults with chronic diseases. Cross-sectional study with 238 older adults (80.3% female; mean age 68.8 ± 6.6) with at least one chronic disease. PA, SB, and sleep duration were measured by a triaxial accelerometer. Clinic systolic BP (SBP) and diastolic BP (DBP) were obtained through an automated method following standard procedures. Non-hierarchical K-means cluster and linear regression modeling were employed to identify the clusters of movement behaviors and to examine the associations. Two clusters were identified [active and non-sedentary, n = 103 (i.e., sufficient sleep duration, higher LPA and MVPA, and lower SB) and sedentary and inactive, n = 135 (i.e., sufficient sleep duration, lower LPA and MVPA, and higher SB). Active and non-sedentary older adults presented lower systolic BP compared to sedentary and inactive ones, even after adjustments for sociodemographic and clinical characteristics (ß = 6.356; CI 95% from 0.932 to 11.779; P = 0.022). No associations were found for diastolic BP. In conclusion, higher PA and lower SB were associated with lower systolic BP in older adults with chronic diseases. However, sleep duration did not modify this association. Therefore, interventions focusing on concomitantly increasing PA levels and reducing SB should be the priority for controlling blood pressure.

ABSTRACT

Menopause is characterized by a reduction in sex hormones in women and is associated with metabolic changes, including fatty liver and insulin resistance. Lifestyle changes, including a balanced diet and physical exercise, are necessary to prevent these undesirable changes. Strength training (ST) has been widely used because of the muscle and metabolic benefits it provides. Our study aims to evaluate the effects of ST on hepatic steatosis and insulin resistance in ovariectomized mice fed a high-fat diet (HFD) divided into four groups as follows: simulated sedentary surgery (SHAM-SED), trained simulated surgery (SHAM-EXE), sedentary ovariectomy (OVX-SED), and trained ovariectomy (OVX-EXE). They were fed an HFD for 9 weeks. ST was performed thrice a week. ST efficiently reduced body weight and fat percentage and increased lean mass in OVX mice. Furthermore, ST reduced the accumulation of ectopic hepatic lipids, increased AMPK phosphorylation, and inhibited the de novo lipogenesis pathway. OVX-EXE mice also showed a better glycemic profile, associated with greater insulin sensitivity identified by the euglycemic-hyperinsulinemic clamp, and reduced markers of hepatic oxidative stress compared with sedentary animals. Our data support the idea that ST can be indicated as a non-pharmacological treatment approach to mitigate metabolic changes resulting from menopause.

ABSTRACT

CONTEXT: Vascular changes can be a risk factor for recurrent and new events of stroke. However, few information is known regarding the variables related to aortic pulse wave morphology in stroke individuals. OBJECTIVE: To analyze aortic pulse wave morphology (arterial stiffness indices, hemodynamics and vascular variables) and to compare the paretic and non-paretic sides in individuals after chronic stroke. DESIGN: In this cross-sectional study stroke individuals had arterial stiffness indices, hemodynamics and vascular variables assessed with brachial artery oscillometry. T-test (CI95%) was used in order to compare the variables between the paretic and non-paretic sides. RESULTS: Twenty individuals were included, 65% men (60.3 SD 16.7 years). The following variables: (mean difference [CI95%]): coefficient of reflection (-2.33 [-4.60 to -0.07]), peak of ejection wave, P1 (5.32 [2.75 to 7.90] and peak of ejection wave, P2 (6.17 [2.55 to 9.78]), central diastolic blood pressure (mean difference [IC95%]): (-3.75 [-6.09 to -1.40]), central systolic blood pressure (-6.17 [-9.74 to -2.59]), mean arterial pressure (-4.46 [-7.08 to -1.84]), peripheral diastolic blood pressure (-3.48 [-5.94 to -1.02]) and peripheral systolic blood pressure (-5.53 [-9.54 to -1.52]) were higher in paretic than non-paretic side. Hemodynamics parameters were similar in both sides. CONCLUSIONS: In this study we demonstrated, for the first time, that many parameters from aortic pulse wave were higher in paretic compared with non-paretic side in individuals after chronic stroke, suggesting that peripheral vascular changes affect heart-vascular coupling in these individuals.

ABSTRACT

In the search for new strains of edible mushrooms in the Brazilian Amazon Forest, we found Lentinula specimens different from Lentinula raphanica. These were described morphologically and evaluated phylogenetically within the Lentinula clade. The mating system was determined, and interbreeding compatibility with L. raphanica was verified. The basidiomata have a cinnamon or deep orange to fulvous brown, moist to dry pileus, occasionally with whitish scales; crowded whitish cream lamellae; and an eccentric to lateral stipe. The typical, predominant basidiospores are 4.4-7.2 µm in length. Endogenous, elongate (7.8-14 µm) basidiospores were also found in some specimens. The long spores seem rare and occasional, but nonetheless a novelty for the group. Basidia are homogeneous in size, cheilocystidia are pyriform or bulboid, and caulocystidia are long and spheropedunculate. The hyphae of the pileipellis are pigment-encrusted. The characteristics match those of Agaricus ixodes originally described from Guyana, currently a synonym of Lentinula boryana. In the phylogenetic trees, such taxon appears distinct from L. boryana and sister to L. raphanica with strong support. This unique lineage was confirmed to be reproductively isolated from sympatric L. raphanica strains. Lentinula ixodes comb. nov. is the second species of the genus reported in the Amazon Forest.

ABSTRACT

Paullinia cupana var. sorbilis, known in Brazil as guarana plant, is an important plant and a major traditional crop in the State of Amazonas. It is a native Brazilian species of great economic and social importance, particularly in the Amazon region. Anthracnose caused by Colletotrichum spp. is the main challenge for this crop. Therefore, the present study verified whether C. siamense, an endophytic fungus infected with a mycovirus, could protect the seedlings and reduce or eliminate the characteristic symptoms. Total proteins and enzymatic activities of pathogenesis-related proteins (PRPs), including peroxidase (POX), chitinase (CHI), and phenylalanine ammonia lyase (PAL), were quantified. Guarana seedlings of cultivar Maués were sprayed with a C. siamense conidia suspension (5.0 × 103 conidia/mL). After ten days, the seedlings were sprayed with a suspension of the phytopathogen's conidia (1.0 × 106 conidia/mL). One group of these seedlings received the fungicide indicated for this crop. The fungicide was applied twice with an interval of 15 days between applications. Negative control seedlings did not receive any treatment (except water and fertilization), and positive control seedlings were treated only with the phytopathogen. The experiment was conducted between December 2019 and February 2020 in a greenhouse. The treatments were applied at an average temperature of 25°C and 85% relative humidity. Leaflets were randomly collected from each treatment group at 0, 48, 72, and 96 hours after pathogen inoculation and analyzed for total protein and enzyme production (POX, PAL, and CHI). After 28 days, the percentage of leaf lesions on the seedlings was evaluated. C. siamense inoculation reduced lesions. There were differences in total proteins and PRPs at different timepoints after inoculation, except for CHI activity, among treatments. To the best of our knowledge, this is the first record of resistance induction in guarana plants.

ABSTRACT

Vascular disorders have a direct link to mortality in the acute phase of Trypanosoma cruzi infection. However, the underlying mechanisms of vascular dysfunction in this phase are largely unknown. We hypothesize that T. cruzi invades endothelial cells causing dysfunction in contractility and relaxation of the mouse aorta. Immunodetection of T. cruzi antigen TcRBP28 was observed in endothelial cells. There was a decreased endothelial nitric oxide synthase (eNOS)-derived NO-dependent vascular relaxation, and increased vascular contractility accompanied by augmented superoxide anions production. Endothelial removal, inhibition of cyclooxygenase 2 (COX-2), blockade of thromboxane A2 (TXA2) TP receptors, and scavenger of superoxide normalized the contractile response. COX-2, thromboxane synthase, inducible nitric oxide synthase (iNOS), p65 NFκB subunit and p22(phox) of NAD(P)H oxidase (NOX) subunit expressions were increased in vessels of chagasic animals. Serum TNF-α was augmented. Basal NO production, and nitrotyrosine residue expression were increased. It is concluded that T. cruzi invades mice aorta endothelial cells and increases TXA2/TP receptor/NOX-derived superoxide formation. Alongside, T. cruzi promotes systemic TNF-α increase, which stimulates iNOS expression in vessels and nitrosative stress. In light of the heart failure that develops in the chronic phase of the disease, to understand the mechanism involved in the increased contractility of the aorta is crucial.

ABSTRACT

We present a detailed structural analysis for small Tin (n = 2-15) clusters based on ab initio quantum mechanical calculations of their binding energies, frontier orbital gaps, and second energy derivatives. Local density approximation calculations revealed that while the smaller clusters (n < or = 8) prefer hexagonal atomic arrays with bulklike crystal symmetry, the bigger clusters prefer pentagonal atomic arrays. From the stability criteria of the magic number clusters we could identify three magic number clusters Ti7, Ti13, and Ti15. While the most stable configuration of Ti7 is a decahedral bipyramid induced by tetrahedral atomic arrays, the most stable configuration of Ti13 is an icosahedron. The other stable cluster Ti15 takes a closed-shell icosahedron-like configuration with both pentagonal and hexagonal symmetries. The stability of the Tin clusters strongly depends on their geometries and charge states. The HOMO-LUMO gap of the Tin clusters approaches its bulk value for n > 8. While there is not much difference between the HOMO and LUMO isosurface charge distributions for the Ti7 and Ti13 clusters in their most stable configurations, they are very different in the case of Ti15. Such a distinct charge distribution in Ti15 indicates its singular chemical selectivity over the other two magic number clusters.
