ABSTRACT

"External magnetic-field (EMF)" has been proved as an additional process parameter like voltage and current affecting the weld arc form, molten metal-flow, microstructure, and characteristics of the weld joint. This article analyzed the research work that has been done to promote EMF application in welding and discussed the recent development trends and research in the design and fabrication of EMF setup to the controlled arc welding process. It is found that even after the successful application of EMF in welding. Still, there is no mass level initiation to integrate EMF with welding machines that hinder researchers and manufacturers to accept it as a regular process parameter to control weld quality.

ABSTRACT

This study attempted to analyze and optimize the effect of an external magnetic field (EMF) on the aspect ratio and heat input for AZ31B weld joints that were welded using the gas metal arc welding (GMAW) process. The response surface methodology (RSM) was adopted for the critical analysis, and subsequently, mathematical models were developed based on the experimental results. It was observed that the EMF and its interaction with the wire feed rate significantly affected the aspect ratio and heat input, respectively. At 119 G (magnetic field), 700 mm/min (welding speed), 5.8 m/min feed rate, and 11.5 L/min (gas flow rate), the aspect ratio was 2.26, and the corresponding heat input factor (HIf) was 0.8 with almost full weld penetration.

ABSTRACT

RNAi is an evolutionary conserved, highly efficient, and cost effective technique of gene silencing. It holds considerable promise and success has been achieved both in vitro and in vivo experiments. However, it is not devoid of undesirable side effects as dsRNA can trigger the immune response and can also cause non-specific off-target gene silencing. In the present study, silencing of myostatin gene, a negative regulator of myogenesis, was evaluated in caprine fetal fibroblasts using three different shRNA constructs. Out of these three constructs, two constructs sh1 and sh2 showed, 72% and 50% reduction (p<0.05) of myostatin mRNA, respectively. Efficient suppression (42-86%) of MSTN gene (p<0.05) was achieved even by reducing the concentration of shRNA constructs. The induction of classical interferon stimulated gene (Oligoandenylate Synthetase-1, OAS-1) was studied to analyze the immune response against shRNAs. Notably, a reduction in the potency of shRNAs to induce interferon response was observed at lower concentration for OAS1 gene. The results obtained in the study would be helpful in the abrogation of the bystander effects of RNAi for long term stable expression of anti-MSTN expression constructs in the muscle.

ABSTRACT

For protection against cholera, it is important to develop efficient vaccine capable of inducing anti-toxin as well as anti-colonizing immunity against Vibrio cholerae infections. Earlier, expression of cholera toxin B subunit (CTB) in tomato was reported by us. In the present investigation, toxin co-regulated pilus subunit A (TCPA), earlier reported to be an antigen capable of providing anti-colonization immunity, has been expressed in tomato. Further, to generate more potent combinatorial antigens, nucleotides encoding P4 or P6 epitope of TCPA were fused to cholera toxin B subunit gene (ctxB) and expressed in tomato. Presence of transgenes in the tomato genome was confirmed by PCR and expression of genes was confirmed at transcript and protein level. TCPA, chimeric CTB-P4 and CTB-P6 proteins were also expressed in E. coli. TCPA protein expressed in E. coli was purified to generate anti-TCPA antibodies in rabbit. Immunoblot and G(M1)-ELISA verified the synthesis and assembly of pentameric chimeric proteins in fruit tissue of transgenic tomato plants. The chimeric protein CTB-P4 and CTB-P6 accumulated up to 0.17 and 0.096% of total soluble protein (TSP), respectively, in tomato fruits. Whereas expression of TCPA, CTB-P4 and CTB-P6 in E. coli can be utilized for development of conventional vaccine, expression of these antigens which can provide both anti-toxin as well as anti-colonization immunity, has been demonstrated in plants, in a form which is potentially capable of inducing immune response against cholera infection.
