ABSTRACT

Objectives: The identification of molecular biomarkers for bipolar disorder is anticipated to greatly improve the diagnosis and treatment of this disease. The objective of this case-control study is to determine whether the blood thyroid hormone levels in bipolar disorder patients are associated with different types of first onset. Methods: From August 1, 2020 to July 31, 2021 a total of 120 female patients diagnosed with bipolar disorder and hospitalized at Qingdao Mental Health Center were recruited as the case group, including 60 patients with depression as their first onset (depression first-episode group, DF) and 60 with mania/hypomania as their first onset (mania/hypomania first-episode group, M/HF). A group of 60 healthy adult females matching general demographic data, such as race and age, were selected as the control group. Blood samples were taken from both groups to measure serum triiodothyronine (T3), thyroxine (T4), free triiodothyronine (FT3), free thyroxine (FT4), and thyroid stimulating hormone (TSH) concentrations. Results: The duration of current onset in the M/HF group was significantly less than that in the DF group (23.1 ± 20.2 vs. 125.2 ± 41.0 days). About 27% of patients in the M/HF group had thyroid abnormalities, in contrast to 60% in the DF group. The blood T3 and T4 levels in both the M/HF group and the DF group, as well as the TF3 levels in the DF group, were significantly lower as compared to control. The M/HF group had significantly higher T3 and FT3 levels than the DF group. The blood T3 levels were inversely correlated with the Young's Mania Scale score and the Hamilton Depression Scale score in both the M/HF and DF groups. Conclusion: Thyroid dysfunction resulting in reduced levels of blood thyroid levels may be involved in the disease progression of bipolar disorder, and correlated with the clinical symptoms in patients with depression or mania as the first episode.

ABSTRACT

The Huangshui National Wetland Park (HNWP) is a unique national wetland park in a city on the Qinghai-Tibetan Plateau, containing three zones: Haihu, Beichuan, and Ninghu. In this study, a total of 54 soil samples (18 sampling points with depths of 0-10 cm, 10-20 cm, and 20-30 cm) were collected in these three zones, and the contents of heavy metals (Cr, Cd, Cu, Hg, Ni, Pb, Zn, and As) of each sample were determined. The ecological risk of eight kinds of heavy metals was evaluated by using the geo-accumulation index (Igeo), and the ecological risk-controlling effect of the Xining urban wetlands on heavy metals was explored by comparative analysis, and the possible sources of heavy metals in the soil were analyzed via correlation analysis and principal component analysis (PCA). The results revealed that the total heavy metal concentration order was Haihu > Beichuan > Ninghu zone. As and Cu presented vertical accumulation characteristics in the surface and lower horizon, respectively. Cr, Cd, Hg, Ni, Pb, and Zn accumulated downwards along the depth. On the spatial scale, the enrichments of Cd and Hg brought non-negligible ecological risks in plateau urban wetlands. The results of PCA indicated that soil heavy metals mainly came from compound sources of domestic and atmospheric influences, traffic pollution sources, and industrial pollution sources. The study has revealed that human activities have inevitable negative impacts on wetland ecosystems, while the HNWP provides a significant weakening effect on heavy metal pollution.

ABSTRACT

Although the ecological risks of antibiotics have been extensively researched globally, fewer studies have been conducted in sensitive and fragile plateau wetland ecosystems. To evaluate the ecological risk of antibiotics in plateau urban wetlands, 18 water samples, 10 plant samples, and 8 sediment samples were collected in March 2022 in the Xining urban wetlands on the Qinghai-Tibet Plateau. The liquid chromatography-electrospray ionization tandem mass spectrometry method was utilized to measure the concentrations of 15 antibiotics in three categories in three types of environmental media. Risk quotients were adopted to assess the ecological risk of antibiotics, and the principal component analysis-multiple linear regression model was used to analyze the source of antibiotics. The results showed that (1) the maximum concentrations of antibiotics in water samples, plants, and sediments reached 1220.86 ng/L, 78.30 ng/g, and 5.64 ng/g, respectively; (2) Tylosin (TYL), norfloxacin (NFX), ofloxacin (OFX), and ciprofloxacin (CFX) in water were at medium and high-risk levels, and OFX had the highest risk value, of 108.04; and (3) the results of source apportionment indicate that 58.94% of the antibiotics came from the Huangshui river and wastewater treatment plant (WWTP) near the wetlands. The current study may provide a reference for the risks and management of antibiotics in plateau urban wetlands.

ABSTRACT

BACKGROUND: Hypereosinophilia (HE) is defined as a peripheral blood eosinophil count of > 1.5 × 109/L and may be associated with tissue damage. The clinical presentations of HE vary; however, myocardial fibrosis and thrombosis can threaten the lives of patients with sustained eosinophilia. Cerebral venous sinus thrombosis (CVST) in the setting of eosinophil-related diseases has seldom been reported. Here, we review the literature on HE with CVST to increase knowledge and encourage early diagnosis. CASE SUMMARY: A previously healthy 41-year-old man was admitted to hospital with diarrhea and abdominal pain. He was treated with antibiotics for suspected acute colitis. Three days later, he experienced headache and vomiting. Brain computed tomography (CT) revealed thrombosis of the left jugular vein to the left transverse sinus vein. Platelet (PLT) count decreased to 60 × 1012/L, and absolute eosinophil count (AEC) increased to 2.41 × 109/L. He was treated with low-molecular-weight heparin. PLT count progressively decreased to 14 × 109/L, and we terminated anticoagulation and performed PLT transfusion. Six days after admission, he complained of a worsening headache. Brain CT revealed right temporal lobe and left centrum semiovale intracerebral hemorrhage, and AEC increased to 7.65 × 109/L. We used prednisolone for HE. The level of consciousness decreased, so emergency hematoma removal and decompressive craniectomy for right cerebral hemorrhage were performed. The patient was alert 2 d after surgery. He was treated with anticoagulation again 2 wk after surgery. Corticosteroids were gradually tapered without any symptomatic recurrence or abnormal laboratory findings. CONCLUSION: HE can induce CVST, and we need to focus on eosinophil counts in patients with CVST.

Background: A rapid and reliable method to predict significant early hematoma growth in the acute setting is of great important to better inform clinicians and researchers in their efforts to improve outcomes for patients. Methods: We established a 10-point score system to predict hematoma growth including four parameters: baseline intracerebral hemorrhage (ICH) volume > 30 mL, time to initial CT scan ≤ 3 h, island sign and black hole sign. Then, we reviewed our ICH database and assessed the predict value of the score system. Results: A total of 216 ICH patients were included. Patients with hematoma growth at 24 h had higher score than those without hematoma growth (7.6 ± 3.0 vs. 2.0 ± 2.4, p < 0.001). The optimal cut-off value of the score for predicting hematoma growth was 3 (area under curve, 0.937; 95% CI, 0.899-0.975, p < 0.001), with 95% CI of 0.896-0.965 in bootstrapping analysis. The sensitivity, specificity, positive predictive and negative predictive value of the score ≥ 3 for predicting hematoma growth were 97.8, 92.7, 90.9, and 98.3%. Conclusion: The 10-point score system could predict hematoma growth with high accuracy.

ABSTRACT

Charcot-Marie-Tooth (CMT) disease is the most common inherited peripheral neuropathy characterized by progressive distal muscle weakness and atrophy with decreased or absent tendon reflexes. Mutations in LRSAM1 have been identified to cause CMT disease type 2P. We report a novel LRSAM1 mutation c.2021-2024del (p.E674VfsX11) in a Chinese autosomal dominant CMT disease type 2 family. The phenotype was characterized by late onset and mild sensory impairment. Electrophysiological findings showed normal or mildly to moderately reduced motor and sensory nerve conduction velocities in lower and upper limb nerves.

ABSTRACT

This study examined adaptive changes in protein kinase A (PKA) and kappa opioid receptor (KOR) in selected addiction-related brain regions before and after conditioned place aversion (CPA). Seventy-two male SD rats were randomly assigned to an experimental group (morphine + naloxone, "MN") and 2 control groups: MS (morphine + saline) and SN (saline + naloxone). MN rats were intraperitoneally injected with morphine twice per day for 6.5 days and naloxone once and trained to establish CPA model. MS and SN rats were injected with equivalent volumes of morphine plus saline and saline plus naloxone, respectively. PKA and KOR in AcbSH, CeA and VTA were measured by immunohistochemistry. Before CPA, there were no significant differences in PKA and KOR expression levels in the AcbSH, CeA and VTA between MN and 2 control groups. After CPA, significant differences in PKA expression were detected in the AcbSH (P<0.001) and VTA (P=0.018) between MN and 2 control groups. The average gray intensity of MN group (109.50±4.66) in AcbSH was significantly higher than that of MS (126.50±3.70, P<0.001) and MN (133.50±6.364, P<0.001) groups. Significant differences in KOR expression were also detected between MN and 2 control groups in CeA (P<0.001). In MN group, PKA and KOR expression levels showed adaptive changes at different points of CPA. These findings demonstrated that neuroadaptation mediated by PKA and KOR may be an important molecular neurobiology basis for CPA. The upregulation of AC-cAMP-PKA-CREB signaling pathway in AcbSH and VTA has some role in the neurobiological mechanism of CPA.

ETHNOPHARMACOLOGICAL RELEVANCE: Wuling Capsule is a single herbal formula from mycelia of precious Xylaria nigripes (Kl.) Sacc and its pharmacological function have a tranquilizing effect on the central nervous system. The aim of the study to evaluate the efficacy and safety of Wuling capsule in treatment of insomnia. MATERIALS AND METHODS: We performed a multicenter, randomized, double-blind, placebo-controlled study. The participants received either placebo (n=92) or Wuling capsule (n=94) for 4 weeks and a follow-up period for 2 weeks. RESULTS: Compared between pre-treatment and post-treatment, the global Pittsburgh sleep quality index (PSQI) scores in both Wuling capsule group and placebo group improved significantly (P<0.01). However, there was no significant difference between Wuling capsule group and placebo group (P>0.05). Scores of clinical global impressions scale (CGI-I) at each week in Wuling capsule group was similar to those in placebo group (P>0.05). Compared between pre-treatment and post-treatment, scores of the four components of world health organization on quality of life brief scale (WHOQOL-BREF) in both Wuling capsule group and placebo group improved significantly (P<0.01). However, there were no difference between the two groups (P>0.05). The rate of adverse events was 10.10% in Wuling group, and 6.73% in placebo group (P>0.05). CONCLUSIONS: Wuling capsule can improve insomnia when compared with pre-treatment for 4 weeks and be a well tolerated by all the patients at the 6 weeks of study period. However, there are no significant in the results of the variables tested when compared with placebo control. Further additional rigorous randomized clinical trials are still required.

ABSTRACT

OBJECTIVE: To explore neurobiological mechanisms of the withdrawal-induced aversion. The changes of protein kinase A were measured in central amygdaloid nucleic (CeA) of conditioned place aversion (CPA) model rats. METHODS: (1) All 72 male SD rats were divided into three groups, model group (MN group), and control group (MS group and SN group). MN group was injected with morphine,6.5 days, 10 mg/kg, intraperitoneally (ip), twice per day, naloxone injection, 0.3 mg/kg, ip, along with conditioned place aversion training, to develop the CPA model. The MS group was administrated equivalent volume of morphine and saline. Also the SN group was injected with equivalent volume of saline and naloxone. (2) During the process of morphine-induced CPA, the expression of protein kinase A was assayed with immunohistochemistry in the CeA. RESULTS: In the MN group, protein kinase A expressions in the CeA occurred adaptive changes at different points of CPA (P < 0.05). Protein kinase A expressions after establishment(Day7,134.43 +/- 4.481, P < 0.05), and after extinction (Day 13, 141.01 +/- 3.360, P < 0.01), and after reinstatement (Day 14,137.18 +/- 40.330, P < 0.05) were also lower than those before the establishment of the CPA (Day 5, 124.48 +/- 6.722). However, PKA expressions were not significantly different both in MS group (P > 0.05)and SN group (P > 0.05). CONCLUSION: (1) Protein kinase A expression, in turn regulating the aversion expression, in the CeA probably is a key pathway contributing to the development of CPA. (2) The neuroadaptation mediated by protein kinase A may be one of the important molecular underpinnings of CPA.

ABSTRACT

OBJECTIVE: To appraise the clinical efficacy, safety and compliance of the intervention of spleen-restoring decoction combined with dormancy hygiene education and the intervention of spleen-restoring decoction alone on sub-healthy insomnia of deficiency of both the heart and spleen pattern. METHOD: Study design took multi-centers, blind and randomized control trial, 107 participants with sub-healthy insomnia of deficiency of both the heart and spleen pattern were assigned to A group (52 cases) which was treated with the intervention of spleen-restoring decoction combined with dormancy hygiene education and B group (55 cases) which was treated with the intervention of spleen-restoring decoction single, compared by efficacy, PSQI score, CGI score, WHOQOL-BREF score and security. RESULT: The efficacy of two group was 79.58%. There was no significant different between them. The PSQI scores before treatment was (11.80 +/- 2.08) and which afer treatment was (6.78 +/- 2.84) of A group. The PSQI scores before treatment was (11.61 +/- 2.00) and which afer treatment was (6.73 +/- 2.27) of B group. There was significant difference in PSQI scores both A group and B group after treatment (P < 0.01); the results of CGI score and WHOQOL-BREF score before and after measurement showed the same as PSQI. There were no significant difference between two groups in all scores after treatment and there was no interaction between time pots and treatment factors . Withdrawal reaction analysis: comparing CGI after withdraw 2 weeks and at the end of treatment course, there was no significant difference between two groups. The same result was in comparison among groups. CONCLUSION: Both the intervention of spleen-restoring decoction integrating with dormancy hygiene education and spleen-restoring decoction had obvious clinical efficacy on treating subhealthy insomnia of deficiency of both the heart and spleen pattern, and had high compliance and safety. The intervention of spleen-restoring decoction integrating with dormancy hygiene education showed no better clinical efficacy than spleen-restoring decoction did.

There is evidence that adenosine acting at A(2A) receptors (A(2A)R) can influence striatal plasticity and cognitive functions. We examined spatial working memory in wild-type (WT) and A(2A) receptor knock-out (KO) mice using two assessments: the eight arm radial maze and a repeated trial Morris water maze (MWM) paradigm. Compared to WT littermates, A(2A)R KO mice displayed enhanced working memory as evidenced by a decrease in escape latency in trial 2 compared to trial 1 in the repeated trial MWM, and by a reduction in working memory errors in the radial arm maze. Both MWM and radial maze results indicated that this enhancement of working memory in A(2A)R KO mice was selective for this specific short-term memory. The decrease in escape latency in MWM was detected with an inter-trial interval of 15 s but not with intervals of 10 or 60 min. In the radial maze, spatial reference memory and memory retention after prolonged training (15 days but not 6 days) were not affected by the A(2A)R KO. These results demonstrate preferential improvement in spatial working memory by genetic inactivation of the A(2A)R and support a modulatory role of the A(2A)R in spatial working memory in mice.

ABSTRACT

In the present paper a method was proposed for the determination of Fe, Cr, Ni, Si, Al and Ba in Bi-based superconductor powder (BSCCO, Bi(2-x), Pb(x)Sr2Ca2Cu3O8) by inductively coupled plasma atomic emission spectrometry(ICP-AES). The operation parameters for ICP-AES were optimized, and the influences of acid kind and concentration were examined. The interferences of matrix elements Bi, Sr, Pb, Ca and Cu in Bi-based superconductor powder on the determination of impurities were studied systematically. The interference coefficients of Bi, Sr, Pb, Ca and Cu were obtained for each target elements respectively, and the mutual interferences matrix was constituted with the above coefficients. The equated concentrations of background from matrix elements were calculated by interferences coefficients matrix and concentrations of Bi, Sr, Pb, Ca and Cu using Gaussian elimination with full pivoting method. Then the concentrations of impurities were obtained by calculation of differences. By means of the mutual interference coefficients matrix, the artificial sample was determined for the target elements with the recovery coefficient of 99.5%-100.5%. The superconductor powers were analysed with satisfactory results also, and the results were identical with the reference values from ICP-MS.
