ABSTRACT

Interactions between different types of immune cells and organically-modified silica nanoparticles were studied. The silica particles functionalized with amine groups were prepared by sol-gel technique. Sheep immunoglobulin labeled with fluoresceine isothiocyanate was immobilized by adsorption onto the nanoparticles. The presence of the functional groups was confirmed by infrared absorption measurements. The level of immunocompetent cells interacting with the silica nanoparticles was estimated as the amount of fluorescence-bright cells by flow cytometry method. A low level of interaction of the peripheral blood lymphocytes with the silica nanoparticles was found. On the contrary, the macrophages are actively involved in interaction with the silica nanoparticles. The influence of different size of the silica nanoparticles and incubation time on viability and functional activity of peripheral blood lymphocytes and peritoneal macrophages were investigated.

ABSTRACT

In children (aged 3-8 years old) with minimal cerebral dysfunction the immune status was studied before and after cerebrolysin administration in the dosage of 1 ml per 10 kg of child's weight, intramuscularly, within one month. The cerebrolysin administration resulted in an increase in the level of CD19(+) cells with a simultaneous normalization of serum IgG and IgA levels. The count of CD4(+) lymphocytes has risen. Normalization of a relative count of CD16(+) cells (NK) was noted after cerebrolysin therapy. Expression of activation markers (CD25 and HLA DR) in total population of lymphocytes parallelly changed, achieving the parameters of the control group without any changes in expression of CD95 molecule. Under the cerebrolysin influence the activation mainly of T helpers could be observed in vitro.

ABSTRACT

The work was outlined to elucidate contribution of soluble factors produced by peripheral neutrophils from healthy pregnant women and those with EPH-gestosis to regulation of expression of surface activation markers by donor lymphocytes. It was found out that in healthy pregnant women neutrophil factors stimulated expression of HLA-DR and CD25 antigens by T lymphocytes. In women with EPH-gestosis neutrophil factors, which comprised soluble antigen of leukocytes 2 (SAL-2), enhanced expression of CD25 antigen in B subset. Presumably, the effects were caused by activation of SAL-2 neutrophil secretion during EPH-gestosis. Impaired neutrophil secretion in women with EPH-gestosis may lead to activation of B lymphocytes through Th2 recruitment and triggering of the autoimmune reactions.
