ABSTRACT

We report the synthesis of the triphosphate of 5-methyl 4-N-[6-(p-bromobenzamido)hex-1-yl]-2'-O-deoxycytidine 3A. We also analyzed the formation of intramolecular H-bonds of 5-methyl 4-N-[n-[6-(p-bromobenzamido) caproyl amino]alk-1-yl]-2'-deoxycytidine compounds, and confirmed their presence by 1H-NMR studies. In vitro DNA labeling with modified nucleotides is preliminarily evaluated.

ABSTRACT

Se estudiaron por medio de ecodoppler color 40 hombres impotentes, con factores de riesgo vascular como antecedentes de su disfunción eréctil. El estudio fue realizado con el pene en estado de flaccidez en escala de grises para la detección de anomalías estructurales, y postinyección de drogas vasoactivas (Alprostadil 10 microgramos) a los 5,15 y 25 minutos. En todos los casos fueron medidas las velocidades de pico sistólico, velocidades de fin de diástole y el calibre arterial. La medición fue hecha sobre la artyeria cavernosa derecha o izquierda, tomando en cuenta el mejor resultado. Observamos que en estado de flaccidez no se pueden obtener datos suficientes para establecer un diagnóstico correcto. En cambio, a los 15 minutos obtuvimos datos suficientes para determinar patrones morfológicos y hemodinámicos de la arteria cavernosa como para realizar diagnóstico de impotencia vasculogénica

ABSTRACT

A simple and rapid method for the determination of 1-octacosanol in plasma, based on an improved Folch extraction technique and capillary gas chromatography, has been developed taking into account the analytical criteria for the pharmacokinetic studies. The procedure was validated in the range of 50-2000 ng/ml. Despite the complexity of the obtained fingerprints, the efficiency and the separation power of GC allowed the determination of 1-octacosanol in plasma samples. The high recoveries (94.5-98.7%) and precision (1.8-5.8%) obtained are in accordance with the established validation criteria. The validity of this method for pharmacokinetic purposes was shown using an endovenous experiment in animals.

ABSTRACT

Artemisia absynthium L. is a commonly used medicinal plant for parasitic diseases all over the world. By means of high-performance liquid chromatography with diode-array detection and the PU6100 solvent optimization system, two sesquiterpene lactones, alpha-santonin and ketopelenolid-A, were tentatively identified in methanolic extracts of this plant. alpha-Santonin is a well known antiparasitic compound and could be one of the active principles of this plant species. Reconstructed spectra are potentially useful in scanning a complex chromatogram for pharmacologically active compounds.

ABSTRACT

A prospective study on biliary and pancreatic obstruction during gallstone migration was performed in patients without acute pancreatitis. From January to October 1986, 125 patients with upper abdominal pain due to cholelithiasis were admitted to the hospital. Ultrasonography performed in all patients at admission demonstrated a distal bile duct measuring 7 mm or more in 39 patients, who were monitored for diameter changes of the biliary and pancreatic duct every 24 h and their stools screened for gallstones. Patients underwent surgery at least 8 days after admission. Gallstone migration was found preoperatively in 10 patients, of whom 6 had total serum bilirubin values lower than 2 mg/100 ml. Migration time was accurately determined by the sudden decrease in bile duct caliber. Simultaneous dilatation of biliary and pancreatic duct was found in 4 out of 10 patients with migrating gallstones and in 7 out of 23 patients without gallstone migration, though differences proved non-significant. Acute pancreatitis developed in 2 patients with lithiasis of the distal bile duct who ingested a fatty meal against medical advice. Gallstone migration, even of small stones, was preceded by a period of biliary obstruction. Pain and jaundice before migration were not as frequent as expected.

Estos dos alcaloides indólicos se encuentran en el material vegetal en muy pequeña proporción y su purificación es difícil, lo que los hace muy costosos y justifica así estudios de cultivo de células y tejido in vitro. Se describe la iniciación de callos y suspensiones celulares de esta planta, la obtención de líneas productoras de alcaloides indólicos y la identificación de siete de ellos: ajmalicina, vindolinina, 19-epivindolinina, tetrahidroalstonina, yohimbina, horhammericine y pleiocarpamina, así como se reportan medios de cultivo útiles para crecimiento y producción

ABSTRACT

En la presente comunicación se informa el estudio de 5 alcaloides extraídos del Strempeliopsis strempelioides, realizado mediante un tamizaje farmacológico integrado por 4 técnicas dirigidas hacia la detección de una posible acción antiarrítmica, hipotensora arterial, cardiotónica o broncodilatora. Los alcaloides fueron la pleiocarpamina (1), la aspidospermina (II), la desacetilaspidospermina (III), la desmetilaspidospermina (IV) y la vallesina (V). Como posible antiarrítmico se evaluaron los alcaloides I, II y III; como posible hipotensor, los I, II, III y IV y los 5 en las dos técnicas restantes. Esto dependió de la cantidad disponible de producto para los ensayos. Los resultados fueron: 1, Arritmia inducida por el cloroformo en el ratón. Sólo la aspidospermina protegió de la arritmia, en dosis que no evidenciaron toxicidad aguda(AU)

ABSTRACT

The effects of propranolol (100 mg/kg), atenolol (200 mg/kg) and acebutolol (1000 mg/kg) administered daily by gavage to spontaneously hypertensive rats (SHRs) from their 6th to 20th weeks of age were investigated on genetic hypertension development (GHD) and at regular intervals on heart rate (HR), cardiac output (CO), stroke volume (SV), peripheral resistence (PR), plasma renin concentration (PRC) and heart weight/body weight ratio (HW/BW). Treatment with propranolol and especially atenolol markedly inhibited GHD while acebutolol was ineffective. No correlation was found between GHD prevention and (1) the degree of beta-adrenergic blockade (2) the reductions in HR and CO and (3) the decrease in PRC induced by the three treatments. Although none of the three drugs prevented the progressive increase in PR which develops in SHRs during their growth, propranolol and atenolol opposed GHD mainly by reducing CO, this effect being however partly counterbalanced for atenolol by a secondary potentiation of PR increase. With acebutolol, a similar reinforcement of PR increase occurred which completely neutralized the reduction in CO, resulting in the drug's ineffectiveness against GHD. These differential effects of the three drugs probably reflect different early induced structural modifications at cardiac and/or vascular levels.

In spontaneously hypertensive rats (SHRs) chronically treated during their growth with beta-adrenoceptor blocking drugs, no correlation was found between the reduction in heart rate and the prevention of genetic hypertension development.
