ABSTRACT

A core challenge in perception is recognizing objects across the highly variable retinal input that occurs when objects are viewed from different directions (e.g. front versus side views). It has long been known that certain views are of particular importance, but it remains unclear why. We reasoned that characterizing the computations underlying visual comparisons between objects could explain the privileged status of certain qualitatively special views. We measured pose discrimination for a wide range of objects, finding large variations in performance depending on the object and the viewing angle, with front and back views yielding particularly good discrimination. Strikingly, a simple and biologically plausible computational model based on measuring the projected three-dimensional optical flow between views of objects accurately predicted both successes and failures of discrimination performance. This provides a computational account of why certain views have a privileged status.

ABSTRACT

Measurement tools from which valid interpretations can be made are critical for assessing early emerging depressive symptoms, as depressive symptoms in childhood are associated with increased risk for early-onset depressive disorder, recurrence, suicidality, and other psychopathology. The Depression Self-Rating Scale for Children (DSRS) is a widely used self-report scale assessing youth depressive symptoms. The relatively few studies investigating the DSRS' latent structure have yielded mixed results, and measurement invariance (MI) based on sex and age has not been examined. We examined the factor structure and MI of the DSRS across sex and age in a community sample of 6-9-year-olds (N = 352; Mage = 7.57 years, SD = .70). Consistent with the largest prior structural study of the DSRS, a two-factor structure, with factors reflecting elevated negative affect (NA) and low positive affect (PA), showed strong model fit. Although this structure was consistent across sex and age (i.e., configural invariance), loadings of DSRS items varied across sex and age (i.e., metric noninvariance). Allowing the loadings of items contributing to noninvariance to vary across groups improved model fit. ABSTRACT

Over the past decade, thousands of bacteriophage genomes have been sequenced and annotated. A striking observation from this work is that known structural features and functions cannot be assigned for >65% of the encoded proteins. One approach to begin experimentally elucidating the function of these uncharacterized gene products is genome-wide screening to identify phage genes that confer phenotypes of interest like inhibition of host growth. This study describes the results of a screen evaluating the effects of overexpressing each gene encoded by the temperate Cluster F1 mycobacteriophage Girr on the growth of the host bacterium Mycobacterium smegmatis. Overexpression of 29 of the 102 Girr genes (~28% of the genome) resulted in mild to severe cytotoxicity. Of the 29 toxic genes described, 12 have no known function and are predominately small proteins of <125 amino acids. Overexpression of the majority of these 12 cytotoxic no known functions proteins resulted in moderate to severe growth reduction and represent novel antimicrobial products. The remaining 17 toxic genes have predicted functions, encoding products involved in phage structure, DNA replication/modification, DNA binding/gene regulation, or other enzymatic activity. Comparison of this dataset with prior genome-wide cytotoxicity screens of mycobacteriophages Waterfoul and Hammy reveals some common functional themes, though several of the predicted Girr functions associated with cytotoxicity in our report, including genes involved in lysogeny, have not been described previously. This study, completed as part of the HHMI-supported SEA-GENES project, highlights the power of parallel, genome-wide overexpression screens to identify novel interactions between phages and their hosts.

ABSTRACT

BACKGROUND: MUPPITS-2 was a randomized, placebo-controlled clinical trial that demonstrated mepolizumab (anti-IL-5) reduced exacerbations and blood and airway eosinophils in urban children with severe eosinophilic asthma. Despite this reduction in eosinophilia, exacerbation risk persisted in certain patients treated with mepolizumab. This raises the possibility that subpopulations of airway eosinophils exist that contribute to breakthrough exacerbations. OBJECTIVE: We aimed to determine the effect of mepolizumab on airway eosinophils in childhood asthma. METHODS: Sputum samples were obtained from 53 MUPPITS-2 participants. Airway eosinophils were characterized using mass cytometry and grouped into subpopulations using unsupervised clustering analyses of 38 surface and intracellular markers. Differences in frequency and immunophenotype of sputum eosinophil subpopulations were assessed based on treatment arm and frequency of exacerbations. RESULTS: Median sputum eosinophils were significantly lower among participants treated with mepolizumab compared with placebo (58% lower, 0.35% difference [95% CI 0.01, 0.74], P = .04). Clustering analysis identified 3 subpopulations of sputum eosinophils with varied expression of CD62L. CD62Lint and CD62Lhi eosinophils exhibited significantly elevated activation marker and eosinophil peroxidase expression, respectively. In mepolizumab-treated participants, CD62Lint and CD62Lhi eosinophils were more abundant in participants who experienced exacerbations than in those who did not (100% higher for CD62Lint, 0.04% difference [95% CI 0.0, 0.13], P = .04; 93% higher for CD62Lhi, 0.21% difference [95% CI 0.0, 0.77], P = .04). CONCLUSIONS: Children with eosinophilic asthma treated with mepolizumab had significantly lower sputum eosinophils. However, CD62Lint and CD62Lhi eosinophils were significantly elevated in children on mepolizumab who had exacerbations, suggesting that eosinophil subpopulations exist that contribute to exacerbations despite anti-IL-5 treatment.

ABSTRACT

Imagine staring into a clear river, starving, desperately searching for a fish to spear and cook. You see a dark shape lurking beneath the surface. It doesn't resemble any sort of fish you've encountered before - but you're hungry. To catch it, you need to anticipate which way it will move when you lunge for it, to compensate for your own sensory and motor processing delays1,2,3. Yet you know nothing about the behaviour of this creature, and do not know in which direction it will try to escape. What cues do you then use to drive such anticipatory responses? Fortunately, many species4, including humans, have the remarkable ability to predict the directionality of objects based on their shape - even if they are unfamiliar and so we cannot rely on semantic knowledge about their movements5. While it is known that such directional inferences can guide attention5, we do not yet fully understand how such causal inferences are made, or the extent to which they enable anticipatory behaviours. Does the oculomotor system, which moves our eyes to optimise visual input, use directional inferences from shape to anticipate upcoming motion direction? Such anticipation is necessary to stabilise the moving object on the high-resolution fovea of the retina while tracking the shape, a primary goal of the oculomotor system6, and to guide any future interactions7,8. Here, we leveraged a well-known behaviour of the oculomotor system: anticipatory smooth eye movements (ASEM), where an increase in eye velocity is observed in the direction of a stimulus' expected motion, before the stimulus actually moves3, to show that the oculomotor system extracts directional information from shape, and uses this inference to predict and anticipate upcoming motion.

ABSTRACT

Reducing health inequalities by addressing the social circumstances in which children are conceived and raised is a societal priority. Early interventions are key to improving outcomes in childhood and long-term into adulthood. Across the UK nations, there is strong political commitment to invest in the early years. National policy interventions aim to tackle health inequalities and deliver health equity for all children. Evidence to determine the effectiveness of socio-structural policies on child health outcomes is especially pressing given the current social and economic challenges facing policy-makers and families with children. As an alternative to clinical trials or evaluating local interventions, we propose a research framework that supports evaluating the impact of whole country policies on child health outcomes. Three key research challenges must be addressed to enable such evaluations and improve policy for child health: (1) policy prioritisation, (2) identification of comparable data and (3) application of robust methods.

ABSTRACT

This paper explores the impacts that the New Zealand government's lockdown measures to contain the COVID-19 virus during 2020 had on the activity and experiences of outdoor recreationists in New Zealand. Concepts related to coping strategies such as rationalisation, displacement and substitution, have been used frequently to explain the behavioural changes and processes involved in outdoor recreation when disruptions arise such as crowding and recreational conflict. However, such concepts have rarely been applied to rapid on-set disruptors such as pandemics. This paper adopts coping strategy theory to help document the strategies adopted by outdoor recreationists in response to the national lockdown in 2020. Based on a qualitative analysis of twenty interviews with outdoor recreationists in New Zealand, various coping mechanisms such as temporal, activity, and spatial displacement are identified. These include increased appreciation for outdoor settings, discovery of local activities and microadventures, and increased walking activity across the restricted, reaction and reset periods. Outdoor recreation coping strategies may contribute to increased resilience to disruptive and rapid on-set events and enhance understanding of how recreationists respond and adapt to disruption. This research presents a unique insight of coping strategies adopted in response to the national lockdown that may have implications for participation and management of outdoor recreation in New Zealand in the coming years. This paper also offers a new perspective on the behaviourist tradition in the field of outdoor recreation which may be fruitful for future research examining rapid on-set disruptions and crises.

ABSTRACT

Although medical masks have played a key role in decreasing the transmission of communicable disease, they simultaneously reduce the availability of nonverbal cues fundamental to social interaction. In the present study, we determined the collective impact of medical masks on emotional expression recognition and perceived intensity as a function of actor race. Participants completed an emotional expression recognition task involving stimuli with or without medical masks. Across six basic emotional facial expressions, medical masks were associated with significantly more emotional expression recognition errors. Overall, the effects associated with race varied depending on the emotion and appearance of masks. Whereas recognition accuracy was higher for White relative to Black actors for anger and sadness, the opposite pattern was observed for disgust. Medical mask-wearing exacerbated actor-race related recognition differences for anger and surprise, but attenuated these differences for fear. Emotional expression intensity ratings were significantly reduced for all emotions except fear, where masks were associated with increased perceived intensity. Masks further increased already higher intensity ratings for anger in Black versus White actors. In contrast, masks eliminated the tendency to give higher intensity ratings for Black versus White sad and happy facial expressions. Overall, our results suggest that the interaction between actor race and mask wearing status with respect to emotional expression judgements is complex, varying by emotion in both direction and degree. We consider the implications of these results particularly in the context of emotionally charged social contexts, such as in conflict, healthcare, and policing.

Parenting and child impulsivity are consistent predictors of children's externalizing symptoms; however, the role of the range of parenting (i.e., variation in parenting across contexts), and its interactions with child impulsivity, are poorly understood. We examined whether characteristic parenting practices and parenting range predicted the course of externalizing symptoms in 409 children (Mage = 3.43 years at baseline, 208 girls) across ages 3, 5, 8, and 11. We assessed parent positive affectivity (PPA), hostility, and parenting structure at child age 3 using three behavioral tasks that varied in context, examining range by modeling a latent difference score for each parenting dimension. Greater PPA range, mean structure, and parenting structure range all predicted fewer symptoms at age 3 for children with higher impulsivity. Lower mean hostility predicted fewer symptoms at age 3 for children with lower impulsivity. Greater PPA, and smaller PPA range, predicted a decrease in symptoms for children higher in impulsivity. Lower hostility range predicted a decrease in symptoms for children with lower impulsivity but predicted maintaining symptoms for children with higher impulsivity. Results demonstrate the differential roles average parenting practices and parenting range play in the development of child externalizing psychopathology, especially in the context of child impulsivity.

ABSTRACT

OBJECTIVE: Interventions to tackle the social determinants of health can improve outcomes during pregnancy and early childhood, leading to better health across the life course. Variation in content, timing and implementation of policies across the 4 UK nations allows for evaluation. We conducted a policy-mapping review (1981-2021) to identify relevant UK early years policies across the social determinants of health framework, and determine suitable candidates for evaluation using administrative data. METHODS: We used open keyword and category searches of UK and devolved Government websites, and hand searched policy reviews. Policies were rated and included using five criteria: (1) Potential for policy to affect maternal and child health outcomes; (2) Implementation variation across the UK; (3) Population reach and expected effect size; (4) Ability to identify exposed/eligible group in administrative data; (5) Potential to affect health inequalities. An expert consensus workshop determined a final shortlist. RESULTS: 336 policies and 306 strategy documents were identified. Policies were mainly excluded due to criteria 2-4, leaving 88. The consensus workshop identified three policy areas as suitable candidates for natural experiment evaluation using administrative data: pregnancy grants, early years education and childcare, and Universal Credit. CONCLUSION: Our comprehensive policy review identifies valuable opportunities to evaluate sociostructural impacts on mother and child outcomes. However, many potentially impactful policies were excluded. This may lead to the inverse evidence law, where there is least evidence for policies believed to be most effective. This could be ameliorated by better access to administrative data, staged implementation of future policies or alternative evaluation methods.

BACKGROUND: According to the Elaboration Likelihood Model, persuasion can occur via two different routes (the central route and peripheral route), with the route utilized dependent on factors associated with motivation and ability. This study aimed to explore the moderating role of need for cognition (NFC) and perceived relevance on the processing of physical activity messages designed to persuade via either the central route or the peripheral route. METHOD: Participants (N = 50) were randomized to receive messages optimized for central route processing or messages optimized for peripheral route processing. Eye-tracking devices were used to assess attention, which was the primary outcome. Message perceptions and the extent of persuasion (changes in physical activity determinants) were also assessed via self-report as secondary outcomes. Moderator effects were examined using interaction terms within mixed effects models and linear regression models. RESULTS: There were no detected interactions between condition and NFC for any of the study outcomes (all ps > .05). Main effects of personal relevance were observed for some self-report outcomes, with increased relevance associated with better processing outcomes. An interaction between need for cognition and personal relevance was observed for perceived behavioral control (p = 0.002); greater relevance was associated with greater perceived behavioral control for those with a higher need for cognition. CONCLUSION: Matching physical activity messages based on NFC may not increase intervention efficacy. Relevance of materials is associated with greater change in physical activity determinants and may be more so among those with a higher NFC.

OBJECTIVE: To investigate the effect of bidirectional fecal microbial transplant (FMT) between male and female rats on methamphetamine (MA)-induced hyperthermia. METHODS: FMT was performed between male and female rats prior to MA (10 mg/kg, sc) treatment. Core body temperature, plasma drug and norepinephrine (NE) levels were measured and compared between treatment groups. 16S rRNA gene sequencing of bacterial communities between male and female rats was performed. RESULTS: MA treatment resulted in significantly higher core body temperatures in male groups (control and FMT-treated) compared to MA-treated female groups (control and FMT-treated). Plasma concentrations of MA and amphetamine were higher in females than males. Whereas, plasma norepinephrine (NE) levels were not different between male and female rats 90 minutes after MA treatment. At the phyla level, the microbiome of male and female control rats were dominated by Firmicutes and Bacteroidetes. Males had a higher relative abundance of Firmicutes and lower relative abundances of Bacteroidetes than females. The FMT procedure changed the recipient group towards their donor with males getting closer to their donors than females. In the control groups following MA treatment, Firmicutes increased and Bacteroides decreased in females and males. Conversely, in the FMT treatment groups following MA treatment, Firmicutes decreased while Bacteroidetes increased in females and males. CONCLUSIONS: Although definite differences in the structure and diversity of the gut microbiome were observed using 16S rRNA gene sequencing of bacterial communities between male and female rats, these differences do not seem to contribute to the sex-based differences in MA-induced hyperthermia.

Emotional stimuli can disrupt or enhance task performance according to factors that are presently poorly understood. One potentially important determinant is the sensory modality involved. In unimodal visual paradigms (visual task-irrelevant stimuli during a visual task) emotional stimuli frequently produce distraction effects; however, the effects across modalities appear more complex and may also depend on factors related to stimulus timing. It is entirely unclear how task-irrelevant visual stimuli impact auditory task performance in cross-modal paradigms. This project explored task performance as a function of sensory modality, emotional valence, and stimulus timing. In Study 1, participants (N = 50) completed a visual stimulus detection task in the presence of task-irrelevant negative and neutral images and sounds. Accuracy was disrupted in the presence of visual but not auditory emotional stimuli, particularly when the target and task-irrelevant stimulus appeared simultaneously. In Study 2, participants (N = 38) completed an equivalent auditory stimulus detection task. In sharp contrast to the effects observed with visual targets, response times and accuracy were enhanced in the presence of auditory emotional stimuli at the first timepoint but disrupted at later timepoints. However, there was no effect of task-irrelevant visual stimuli on auditory task performance. These findings demonstrate the importance of both sensory modality and timing in determining how emotional stimuli affect task performance and lay the groundwork for future studies examining the interaction between emotional and attentional processes. ABSTRACT

The discovery of mental rotation was one of the most significant landmarks in experimental psychology, leading to the ongoing assumption that to visually compare objects from different three-dimensional viewpoints, we use explicit internal simulations of object rotations, to 'mentally adjust' one object until it matches the other1. These rotations are thought to be performed on three-dimensional representations of the object, by literal analogy to physical rotations. In particular, it is thought that an imagined object is continuously adjusted at a constant three-dimensional angular rotation rate from its initial orientation to the final orientation through all intervening viewpoints2. While qualitative theories have tried to account for this phenomenon3, to date there has been no explicit, image-computable model of the underlying processes. As a result, there is no quantitative account of why some object viewpoints appear more similar to one another than others when the three-dimensional angular difference between them is the same4,5. We reasoned that the specific pattern of non-uniformities in the perception of viewpoints can reveal the visual computations underlying mental rotation. We therefore compared human viewpoint perception with a model based on the kind of two-dimensional 'optical flow' computations that are thought to underlie motion perception in biological vision6, finding that the model reproduces the specific errors that participants make. This suggests that mental rotation involves simulating the two-dimensional retinal image change that would occur when rotating objects. When we compare objects, we do not do so in a distal three-dimensional representation as previously assumed, but by measuring how much the proximal stimulus would change if we watched the object rotate, capturing perspectival appearance changes7.

Clinical manifestations of severe COVID-19 include coagulopathies that are exacerbated by the formation of neutrophil extracellular traps (NETs). Here, we report that pulmonary lymphatic vessels, which traffic neutrophils and other immune cells to the lung-draining lymph node (LDLN), can also be blocked by fibrin clots in severe COVID-19. Immunostained tissue sections from COVID-19 decedents revealed widespread lymphatic clotting not only in the lung but also in the LDLN, where the extent of clotting correlated with the presence of abnormal, regressed, or missing germinal centers (GCs). It strongly correlated with the presence of intralymphatic NETs. In mice, tumor necrosis factor α induced intralymphatic fibrin clots; this could be inhibited by DNase I, which degrades NETs. In vitro, TNF-α induced lymphatic endothelial cell upregulation of ICAM-1 and CXCL8, among other neutrophil-recruiting factors, as well as thrombomodulin downregulation; in decedents, lymphatic clotting in LDLNs. In a separate cohort of hospitalized patients, serum levels of Myeloperoxidase-DNA (MPO-DNA, a NET marker) inversely correlated with antiviral antibody titers, but D-dimer levels, indicative of blood thrombosis, did not correlate with either. Patients with high MPO-DNA but low D-dimer levels generated poor antiviral antibody titers. This study introduces lymphatic coagulation in lungs and LDLNs as a clinical manifestation of severe COVID-19 and suggests the involvement of NETosis of lymphatic-trafficking neutrophils. It further suggests that lymphatic clotting may correlate with impaired formation or maintenance of GCs necessary for robust antiviral antibody responses, although further studies are needed to determine whether and how lymphatic coagulation affects adaptive immune responses.

ABSTRACT

The rare genetic neurodevelopmental disease Angelman syndrome (AS) is caused by the loss of function of UBE3A, a ubiquitin ligase. The disease results in a lifetime of severe symptoms, including intellectual disability and motor impairments for which there are no effective treatments. One avenue of treatment for AS is the use of gene therapy to reintroduce a functional copy of the UBE3A gene. Our group had previously shown that recombinant adeno-associated virus (rAAV) expressing mouse Ube3a could rescue deficits in a mouse model of AS. Here, we expand on this work and show that this approach could be successfully replicated in a second AS model using the human UBE3A gene. Furthermore, we address the challenge of limited vector distribution in the brain by developing a novel modified form of UBE3A. This modified protein, termed STUB, was designed with a secretion signal and a cell-penetrating peptide. This allowed transduced cells to act as factories for the production of UBE3A protein that could be taken up by neighboring non-transduced cells, thus increasing the number of neurons receiving the therapeutic protein. Combining this construct with intracerebroventricular injections to maximize rAAV distribution within the brain, we demonstrate that this novel approach improves the recovery of behavioral and electrophysiological deficits in the AS rat model. More importantly, a comparison of rAAV-STUB to a rAAV expressing the normal human UBE3A gene showed that STUB was a more effective therapeutic. These data suggest that rAAV-STUB is a new potential approach for the treatment of AS.

To create an accurate percept of the world, the visual system relies on past experience and prior assumptions.1 For example, although the retinal projection of an object moving in depth changes drastically, we still perceive the object at a constant size and velocity.2,3 Consequently, if we see the same object with a constant retinal size at two different depth levels, the perceived size differs (illustrated by the Ponzo illusion). Past experience also directly influences perceptual judgments, an effect known as serial dependence.4,5 Such sequential effects have also been reported for oculomotor behavior, even on the trial-by-trial level.6-10 An integration of past experiences seems like a smart and sophisticated mechanism to reduce uncertainty and improve behavior in a world full of statistical regularities. By leveraging the Ponzo illusion to dissociate perceived size and speed from retinal signals, we show that serial-dependence effects for oculomotor control are mediated by retinal error signals. These sequential effects likely take place in early sensory processing because they transfer to different visual stimuli. In contrast to recently reported history effects for perceptual decisions,11 sequential effects for oculomotor control deviate from perceptual mechanisms by not integrating spatial context and by ignoring size and velocity constancy. Although this dissociation might appear suboptimal, we argue that this effect reveals the different goals of the oculomotor and perceptual systems. The oculomotor system tries to reduce retinal error signals to bring and keep the target close to the fovea, whereas the visual system interprets retinal input to achieve an accurate representation of the world.12.

ABSTRACT

Our environment contains an abundance of objects which humans interact with daily, gathering visual information using sequences of eye-movements to choose which object is best-suited for a particular task. This process is not trivial, and requires a complex strategy where task affordance defines the search strategy, and the estimated precision of the visual information gathered from each object may be used to track perceptual confidence for object selection. This study addresses the fundamental problem of how such visual information is metacognitively represented and used for subsequent behaviour, and reveals a complex interplay between task affordance, visual information gathering, and metacogntive decision making. People fixate higher-utility objects, and most importantly retain metaknowledge about how much information they have gathered about these objects, which is used to guide perceptual report choices. These findings suggest that such metacognitive knowledge is important in situations where decisions are based on information acquired in a temporal sequence.

ABSTRACT

BACKGROUND: The effectiveness of hand rubbing with alcohol-based handrub (ABHR) is impacted by several factors. To investigate these, World Health Organization (WHO) commissioned a systematic review. AIM: To evaluate the impact of ABHR volume, application time, rubbing friction and hand size on microbiological load reduction, hand surface coverage or drying time. METHODS: Medline, CINAHL, Web of Science and ScienceDirect databases were searched for healthcare or laboratory-based primary studies, published in English, (1980- February 2021), investigating the impact of ABHR volume, application time, rubbing friction or hand size on bacterial load reduction, hand coverage or drying time. Two reviewers independently performed data extraction and quality assessment. The results are presented narratively. FINDINGS: Twenty studies were included in the review. Categories included: ABHR volume, application time and rubbing friction. Sub-categories: bacterial load reduction, hand size, drying time or hand surface coverage. All used experimental or quasi-experimental designs. Findings showed as ABHR volume increased, bacterial load reduced, and drying times increased. Furthermore, one study showed that the application of sprayed ABHR without hand rubbing resulted in significantly lower bacterial load reduction than poured or sprayed ABHR with hand rubbing (- 0.70; 95%CI: - 1.13 to - 0.28). Evidence was heterogeneous in application time, volume, technique, and product. All studies were assessed as high risk of bias. CONCLUSIONS: There is insufficient evidence to change WHO recommendation of a palmful of ABHR in a cupped hand applied for 20-30 s or manufacturer-recommended volume applied for about 20 s (Centers for Disease Control and Prevention). Future hand hygiene research should standardise volume, application time, and consider hand size.

VanLee is a singleton phage that was isolated from soil in Florida using Gordonia rubripertincta NRRL B-16540 as the host. The genome is 84,560 bp and has a GC content of 67.8%. VanLee has 164 predicted protein-coding genes and one tRNA. VanLee can infect Gordonia terrae with the same efficiency as G. rubripertincta.
