ABSTRACT

Yunnan Province is one of the rich biodiversity hotspots with abundant resources of lignicolous freshwater fungi. A total of 281 species of lignicolous freshwater fungi from 1986 to the present in Yunnan Province. They are mostly distributed in the classes Dothideomycetes and Sordariomycetes, a few species in the Eurotiomycetes and Leotiomycetes, and rarely reported in Orbiliomycetes and Pezizomycetes. Lignicolous freshwater fungi can decompose lignocellulose substrates and release energy and nutrients, and thus playing an important role in freshwater environment. This study briefly reviewed the biodiversity and taxonomic status of lignicolous freshwater fungi in Yunnan, the ecological functions of lignicolous freshwater fungi, factors affecting community distribution, application status, and research difficulties.

ABSTRACT

BACKGROUND: In this study, a new species Neomonodictysaquatica was collected from submerged decaying wood in Erhai Lake, Yunnan Province, China. NEW INFORMATION: Neomonodictysaquatica is characterised by acrogenous, solitary, oval, dictyospores (most are transverse septum, occasionally vertical septum, in immaturity the septum is clear, but when mature, the conidia becomes darker so the septum is not clear), smooth-walled conidia. The immature conidia are usually hyaline to olivaceous and mature conidia are usually darkened to black, sometimes with one pale basal cell. Phylogenetic analyses of combined ITS and LSU sequence data showed that the new collection is distinct from other Neomonodictys species. Description and illustration are provided as well.

ABSTRACT

BACKGROUND: Pleosporales is the largest order of Dothideomycetes. In recent years, systematics of Pleosporales have undergone considerable revisions. Dictyosporiaceae is one of the newly established families within this order proposed to accommodate holomorphic saprobic Dothideomycetes. Currently 18 genera are recognised in Dictyosporiaceae. NEW INFORMATION: The new species, Jalapriyaaquaticum sp. nov. and J.apicalivaginatum sp. nov. were collected from freshwater habitats in Gansu and Yunnan Provinces, China, respectively and are introduced, based on morphology and molecular analysis of combined ITS, LSU, SSU and TEF1-α sequence data. We also recovered one fresh collection of Dictyocheirosporavinaya D'souza, Bhat & K.D. Hyde, which is a new record for China. Jalapriyaaquaticum differs from extant species of Jalapriya in rows converging at the apex and apical cells with spherical-like appendages. Jalapriyaapicalivaginatum differs from extant species of Jalapriya in having the rows of conidia mostly arranged in a plane. The phylogenetic analysis place the new collections within Dictyosporiaceae (Pleosporales). Descriptions and illustrations of Jalapriyaaquaticum, J.apicalivaginatum and Dictyocheirosporavinaya are provided. A synopsis of characters of species of Jalapriya is also provided.

ABSTRACT

During investigations into freshwater fungi from the Great Mekong Subregion, four Distoseptispora taxa were collected from China and Thailand. Based on morphological characteristics, and phylogenetic analyses of combined LSU, ITS, SSU, TEF1-α, and RPB2 sequence data, two new species Distoseptisporabangkokensis and D.lancangjiangensis are introduced, and two known species D.clematidis and D.thysanolaenae were first reported in freshwater habitat. Illustrations and descriptions of these taxa are provided, along with comparisons with extant taxa in the genus.

ABSTRACT

Ganodermadianzhongense sp. nov. and G.esculentum sp. nov. are proposed as two new species based on both phenotypic and genotypic evidences. Ganodermadianzhongense is characterized by the stipitate basidiomata, laccate and oxblood red pileus, gray white pore surface, duplex context and broadly ellipsoid basidiospores (9.0-12.5 × 6.5-9.0 µm) with coarse interwall pillars. Ganodermaesculentum is characterized by its basidiomata with slender stipe, white pore surface, homogeneous pileus context, and slightly truncate, narrow basidiospores (8.0-12.5 × 5.0-8.0 µm). Phylogenetic analyses were carried out based on the internal transcribed spacer (ITS), translation elongation factor 1-α (TEF1-α) and the second subunit of RNA polymerase II (RPB2) sequence data. The illustrations and descriptions for the new taxa are provided.

ABSTRACT

Freshwater hyphomycetes are a highly diverse group of fungi with a worldwide distribution and have been mostly reported from tropical and subtropical regions. During investigations of freshwater fungi from the Greater Mekong subregion in China and Thailand, sixteen freshwater hyphomycetes (three of them belong to the class Dothideomycetes while thirteen belong to the class Sordariomycetes) were collected. Based on morphology and multi-gene phylogenetic analyses, Neospadicoides thailandica, Pseudodactylaria aquatica, Sporidesmium nujiangense, Tetraploa thailandica, Vamsapriyaaquatica and Wongia fusiformis are described as new species; Aquapteridospora bambusinum is proposed as a new combination; Acrodictys liputii, Chloridium gonytrichii, Pseudoberkleasmium chiangmaiense, Pleomonodictys capensis, Sporidesmium aturbinatum and Vamsapriya indica are reported as new country records; and Sporidesmium tropicale, Sporoschisma chiangraiense and Sporoschisma longicatenatum are introduced as three new collections. In addition, a checklist of freshwater fungi from China over the last five years is also provided.

ABSTRACT

BACKGROUND: During the survey of pathogenic fungi on medicinal plant leaves in Yunnan Province, China, two Cladosporium-like taxa were isolated from leaf spots of Parispolyphylla. Based on morphological characteristics and phylogenetic analysis of combined ITS, TEF1-α and ACT genes, two new species were discovered. NEW INFORMATION: Two new species Cladosporiumyunnanensis and C.paris are introduced, the detailed descriptions and illustrations are provided. Morphology of the two new species is compared with other related Cladosporium species. This study widens the host diversity of the genus Cladosporium.

ABSTRACT

During a study of diversity and taxonomy of lignicolous freshwater fungi in China, nine species of Acrogenospora were collected. Seven of these were new species and they are described and illustrated. With morphology, additional evidence to support establishment of new species is provided by phylogeny derived from DNA sequence analyses of a combined LSU, SSU, TEF1α, and RPB2 sequence dataset. Acrogenospora subprolata and A. verrucispora were re-collected and sequenced for the first time. The genus Acrogenospora is far more species rich than originally thought, with nine species found in a small area of Yunnan Province, China.

ABSTRACT

Lignicolous freshwater fungi represent one of the largest groups of Ascomycota. This taxonomically highly diverse group plays an important role in nutrient and carbon cycling, biological diversity and ecosystem functioning. The diversity of lignicolous freshwater fungi along a north-south latitudinal gradient is currently being studied in Asia. In this paper, we introduce two novel freshwater taxa viz. Tingoldiago hydei sp. nov. and T. clavata sp. nov. which were collected from freshwater substrates in Eastern Thailand. Morphological comparison based on the size of ascomata, asci and ascospores, as well as multi-gene phylogenetic analyses based on LSU, SSU, ITS and TEF1-α DNA sequences, supports their placement in Tingoldiago (Lentitheciaceae). Descriptions and illustrations of these two new species are provided.

ABSTRACT

Mycobacterium tuberculosis (Mtb) serine/threonine protein phosphatase PstP plays an important role in regulating Mtb cell division and growth by reversible phosphorylation signaling. However, the substrates of Mtb with which the PstP interacts, and the underlying molecular mechanisms are still largely unknown. In this study, we performed an Mtb proteome microarray to globally identify the PstP bindings. In this way, we discovered 78 interactors between PstP and Mtb proteins, and found a novel connections with EthR. The interaction between PstP and EthR has been validated by Bio-Layer interferometry and Yeast-two-hybrid. And functional studies showed that PstP significantly enhances the binding between EthR and related DNA domain through its interaction with EthR. Phenotypically, overexpression of PstP promoted the resistance of Mycobacterium smegmatis with the antibiotic of ethionamide. Overall, we hopefully wish that the PstP interactors identified in this study will serve as a useful resource for further systematic studies of the roles that PstP plays in the regulation of Mtb dephosphorylation. SIGNIFICANCE: Mycobacterium tuberculosis (Mtb) is the causative agent of tuberculosis, which is responsible of ~1.5 million death per year. Understanding the knowledge about the basic biological regulation pathways in Mtb is an effective approach to discover the novel drug targets for cure TB. PstP is a serine/threonine protein phosphatase in Mtb, and plays important roles in regulating Mtb cell division and growth by reversible phosphorylation signaling. In this study, we identified 78 PstP interacting Mtb proteins using Mtb proteome microarray, which could preliminarily explain the roles of PstP played in Mtb. Moreover, functional analysis showed that a novel transcription factor EthR had been found regulated by PstP through binding, which could enhance the resistance to the antibiotic ETH. Overall, this network constructed with PstP-Mtb proteins could serve as a valuable resource for studying Mtb growth.

ABSTRACT

NHL (NDR1/HIN1-like) genes play crucial roles in pathogen induced plant responses to biotic stress. Here, we report the possible function of NHL6 in plant response to abscisic acid (ABA) and abiotic stress. NHL6 was highly expressed in non-germinated seeds, and its expression was strongly induced by ABA and multiple abiotic stress signals. Loss-of-function of NHL6 decreased sensitivity to ABA in the early developmental stages including seed germination and post-germination seedling growth of the nhl6 mutants. However, overexpression of NHL6 increased sensitivity to ABA, salt and osmotic stress of the transgenic plants. Further studies indicated that the increased sensitivity in the 35S::NHL6 overexpressing plants could be a result of both ABA hypersensitivity and increased endogenous ABA accumulation under the stress conditions. It was also seen that the ABA-responsive element binding factors AREB1, AREB2 and ABF3 could regulate NHL6 expression at transcriptional level. Our results indicate that NHL6 plays an important role in the abiotic stresses-induced ABA signaling and biosynthesis, particularly during seed germination and early seedling development in Arabidopsis.

ABSTRACT

We are studying the freshwater lignicolous fungi along a north-south latitudinal gradient in Asia. In this paper, fresh collections of Minimelanolocus from submerged wood in streams in Yunnan Province, China are characterised based on morphology and molecular phylogeny based on three rDNA regions: 18S (SSU), ITS1-5.8S-ITS2 (ITS) and 28S nuclear rDNA (LSU). The phylogenetic analysis of combined LSU and SSU sequence data and a separate analysis of ITS placed the isolates within the family Herpotrichiellaceae, order Chaetothyriales. An updated phylogenetic backbone tree for Chaetothyriales is provided with available ex-type and additional isolates. One of the isolates collected was identified as Minimelanolocus obscurus based on morphology and molecular data. Minimelanolocus aquaticus, M. asiaticus, M. curvatus and M. melanicus are described as new species considering the interspecific ITS variability and morphology. The phylogenetic placement of Minimelanolocus in Chaetothyriales is novel and provides new sequence data for the genus as a distinct lineage in Chaetothyriales. The conidial characters of all the known species in the genus are summarized. Descriptions and illustrations are provided for the five species of Minimelanolocus with notes on their taxonomy and phylogeny.

ABSTRACT

A MADS box gene HoMADS2 was cloned from Hyacinthus orientalis L. in this study. Sequence comparison revealed that HoMADS2 was highly homologous to the class B MADS box genes. Furthermore, phylogenetic analysis showed that HoMADS2 was closely related to PI gene family and this was also supported by the presence of specific diagnostic sites of PI homologs in K box domain and C terminal region,suggesting that HoMADS2 might be a PI-like gene. HoMADS2 mRNA was accumulated in all floral organs,different from the expression patterns of PI homologs in dicots. During in vitro flower development, HoMADS2 expression was constitutively expressed and not affected by the presence of cytokinin and auxin in the regenerated flowers. Our results indicated that the expression of HoMADS2 is different from those of both HAG1 and HoMADS1 in responding to plant hormones during in vitro flower development.
