ABSTRACT

Rapid urbanization and industrialization have greatly contributed to boosting regional economic growth and mitigating the problem of poverty, but blind expansion of cities and towns has not only caused the inefficient use of urban land resources but also caused the deterioration in the urban ecological environment. Within the current context of emphasizing high-quality development, achieving synergy between the efficient use of urban land and ecological environmental protection is an urgent task for promoting new urbanization construction. In this study, cities in the upper reaches of the Yangtze River (URYR) were adopted as the research object, a theoretical analysis framework for the urban land use efficiency (ULUE) and ecological environment quality (EEQ) was established, the ULUE was measured by using the Slacks-Based Measure (SBM) model, the coupling coordination and interactive corresponding response relationship between the ULUE and EEQ were analyzed, and the influencing factors of the coupling coordination between these two systems were explored by using the random forest model. The following conclusions can be obtained: in 2020, compared with those in 2006, both the ULUE and EEQ were improved, and the two systems exhibited interactions and significant spatiotemporal heterogeneity. The coupling coordination degree (CCD) between the ULUE and EEQ could facilitate maintaining the original state, and the transfer of the CCD exhibited a significant spatial correlation with the state of neighbouring cities. The effect of the ULUE on the EEQ indicated nonlinear characteristics, while the effect of the EEQ on the ULUE was manifested as inhibition initially and then promotion. The random forest regression results showed that the population density, landscape agglomeration and connectivity, market conditions, government intervention, and industrial institutions are the key influencing factors of the CCD. Finally, this study provides policy implications for innovative urban land use modelling, environmental regulation, and industrial transformation and upgrading.

ABSTRACT

Background: Neuroinflammation is an important factor causing numerous neurodegenerative pathologies. Inflammation can lead to abnormal neuronal structure and function and even death, followed by cognitive dysfunction. There is growing evidence that chlorogenic acid has anti-inflammatory effects and immunomodulatory activity. Purpose: The aim of this study was to elucidate the potential targets and molecular mechanisms of chlorogenic acid in the treatment of neuroinflammation. Methods: We used the lipopolysaccharide-induced neuroinflammation mouse model and the lipopolysaccharide-stimulated BV-2 cells in vitro model. Behavioral scores and experiments were used to assess cognitive dysfunction in mice. HE staining and immunohistochemistry were used to assess neuronal damage in the mouse brain. Immunofluorescence detected microglia polarization in mouse brain. Western blot and flow cytometry detected the polarization of BV-2 cells. The migration of BV-2 cells was detected by wound healing assay and transwell assay. Potential targets for chlorogenic acid to exert protective effects were predicted by network pharmacology. These targets were then validated using molecular docking and experiments. Results: The results of in vivo experiments showed that chlorogenic acid had an obvious ameliorating effect on neuroinflammation-induced cognitive dysfunction. We found that chlorogenic acid was able to inhibit BV-2 cells M1 polarization and promote BV-2 cells M2 polarization in vitro while also inhibiting the abnormal migration of BV-2 cells. Based on the network pharmacology results, we identified the TNF signaling pathway as a key signaling pathway in which chlorogenic acid exerts anti-neuroinflammatory effects. Among them, Akt1, TNF, MMP9, PTGS2, MAPK1, MAPK14, and RELA are the core targets for chlorogenic acid to function. Conclusion: Chlorogenic acid can inhibit microglial polarization toward the M1 phenotype and improve neuroinflammation-induced cognitive dysfunction in mice by modulating these key targets in the TNF signaling pathway.

ABSTRACT

Objectives: Assess the validity of the Chinese version of the Dominic Interactive (DI), a 91-item, video-based diagnostic screening instrument for children that assesses four internalized disorders (phobias, separation anxiety disorder, generalized anxiety disorder, and major depressive disorder) and three externalized disorders (attention-deficit/hyperactivity disorder, conduct disorder, oppositional defiant disorder). Methods: (1) Compare DI-generated "probable" or "possible" diagnoses to diagnoses based on the Development and Well-Being Assessment (DAWBA) instrument in 113 psychiatric outpatients and 20 community controls. (2) Administer DI to 1,479 children from elementary schools in Tianjin. Results: In the validation sample, DI with DAWBA concordance was much greater for internalized disorders (mean Kappa = 0.56) than for externalized disorders (mean kappa = 0.11). The positive predictive value of DI diagnoses ranged from 0.96 (generalized anxiety disorder) to 25% (oppositional defiant disorder) and negative from 0.81 to 0.96. Using "probable" cuts provides better results. In the survey, prevalence of probable DI disorders ranged from 1.0% (conduct disorder) to 13.1% (phobias). Internal consistency of all DI items was excellent (Cronbach alpha = 0.93) and that of the seven subscales ranged from 0.64 (phobias) to 0.87 (major depressive disorder). In multilevel SEM analyses, SRMR (Standardized root mean square residual) or each of the seven diagnoses was below 0.08 and each coefficient of determination was below 0.60. Conclusion: The Chinese DI is a convenient method of screening common mental disorders in Chinese children mainly for internalized disorders, which are the most prevalent diagnoses in that population. However its high negative predictive values for externalized could be used for screening.

BACKGROUND: Ulcerative colitis (UC) is a common type of inflammatory bowel disease. Due to the elusive pathogenesis, safe and effective treatment strategies are still lacking. Fraxini Cortex (FC) has been widely used as a medicinal herb to treat some diseases. However, the pharmacological mechanisms of FC for UC treatment are still unclear. METHODS: An integrated platform combining network pharmacology and experimental studies was introduced to decipher the mechanism of FC against UC. The active compounds, therapeutic targets, and the molecular mechanism of action were acquired by network pharmacology, and the interaction between the compounds and target proteins were verified by molecular docking. Dextran sulfate sodium (DSS)-induced colitis model was employed to assess the therapeutic effect of FC on UC, and validate the molecular mechanisms of action predicted by network pharmacology. RESULTS: A total of 20 bioactive compounds were retrieved, and 115 targets were predicted by using the online databases. Ursolic acid, fraxetin, beta-sitosterol, and esculetin were identified as the main active compounds of FC against UC. PPI network analysis identified 28 FC-UC hub genes that were mainly enriched in the IL-17 signaling pathway, the TNF signaling pathway, and pathways in cancer. Molecular docking confirmed that the active compounds had high binding affinities to the predicted target proteins. GEO dataset analysis showed that these target genes were highly expressed in the UC clinical samples compared with that in the healthy controls. Experimental studies showed that FC alleviated DSS-induced colitis symptoms, reduced inflammatory cytokines release, and suppressed the expression levels of IL1ß, COX2, MMP3, IL-17 and RORγt in colon tissues. CONCLUSION: FC exhibits anti-UC properties through regulating multi-targets and multi-pathways with multi-components. In vivo results demonstrated that FC alleviated DSS-induced colitis.

BACKGROUND: Suicidal ideation in children has received less attention than in adolescents. This study aimed to explore the self-reported prevalence of suicidal ideation among children aged 6-12 and to identify the relationship between self-reported suicidal ideation and children's mental health reported by different informants in Chinese setting. METHOD: The study sample was 1479 children aged 6-12 from three elementary schools in Tianjin. Children completed the Dominic Interactive to report their mental health and suicidal ideation. Parents and teachers completed the Socio-Demographic Questionnaire and the Strengths and Difficulties Questionnaire (SDQ). RESULTS: The prevalence of suicidal thoughts and death thoughts was 18.05 % and 16.90 %, respectively. Parent reported emotional symptoms, ADHD, and externalized problems were associated with death thoughts, and ADHD was associated with suicidal thoughts. For teacher's reports, emotional symptoms, and impact were associated with death thoughts, and ADHD, peer problems, internalized problems, and internalized and externalized comorbidity were associated with suicidal thoughts. All of the children's self-reported mental health problems were associated with suicidal thoughts and death thoughts. LIMITATIONS: Causality cannot be inferred in a cross-sectional study. CONCLUSION: Suicidal ideation is not uncommon in Chinese children. The relationships between mental health problems and suicidal ideation varied in different informants. Suicide prevention in young children should be enhanced, and screening for suicidal ideation is recommended at the onset of different informants who reported specific mental health problems.

ABSTRACT

Ulcerative colitis (UC) is a chronic inflammatory gastrointestinal disease mainly associated with immune dysfunction and microbiota disturbance. Cinnamaldehyde (CIN) is an active ingredient of Cinnamomum cassia with immunomodulatory and anti-inflammatory properties. However, the therapeutic effect and detailed mechanism of CIN on UC remains unclear, and warrant further dissection. In this study, network pharmacology and molecular docking analyses were introduced to predict the potential targets and mechanism of CIN against UC. The therapeutic effect and the predicted targets of CIN on UC were further validated by in vivo and in vitro experiments. Seven intersection targets shared by CIN and UC were obtained, and four hub targets, i. e., toll-like receptor 4 (TLR4), transcription factor p65 (NF-κB), NF-kappa-B inhibitor alpha (IκBα), prostaglandin G/H synthase 2 (COX2) were acquired, which were mainly involved in NF-κB, tumor necrosis factor (TNF), Toll-like receptor and NOD-like receptor signaling pathways. CIN alleviated the symptoms of dextran sulfate sodium (DSS)-induced colitis by decreasing the disease active index (DAI), restoring colon length, and relieving colonic pathology. CIN attenuated systemic inflammation by reducing serum myeloperoxidase (MPO), TNF-α, interleukin-6 (IL-6), and interleukin-1ß (IL-1ß), down-regulating TLR4, phosphorylated-NF-κB (p-NF-κB), phosphorylated-IκBα (p-IκBα), and COX2 expression in colonic tissues, and decreasing NOD-like receptor protein 3 (NLRP3), Caspase-1, and IL-1ß protein expression in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. These results indicate that CIN alleviates DSS-induced colitis inflammation by modulating TLR4/NF-κB signaling pathway and NLRP3 inflammasome activation.

Background: Although evidence-based and effective treatments are available for people with major depressive disorder (MDD), a substantial number do not seek or receive help. Therefore, this study aimed to (1) investigate the total help-seeking rate and first-time help-seeking choices; (2) explore the perceived helpfulness of 23 potential sources; and (3) evaluate the factors related to help-seeking behaviors among patients with MDD. Materials and Methods: Data came from the Tianjin Mental Health Survey (TJMHS), which included a representative sample of adult community residents (n = 11,748) in the Chinese municipality of Tianjin. Of these, 439 individuals were diagnosed with lifetime MDD according to the Diagnostic and Statistical Manual-fourth edition (DSM-IV) and administered a help-seeking questionnaire. Results: In a survey, 28.2% of patients with MDD living community reported that they had ever sought any help during their entire lifetime before the interview, with 8.2% seeking help in mental healthcare settings, 8.0% only in other healthcare settings, and 12.0% only in non-healthcare sources (e.g., family, friends, and spiritual advisor). Among help-seekers, the first help mainly was sought in non-healthcare sources (61.3%), followed by healthcare settings (25.8%) and mental healthcare settings (12.9%). The majority of MDD individuals thought the non-healthcare sources were not helpful and mental healthcare settings were helpful or possibly helpful to solve mental problems. Female, having 10-12 or higher education years, comorbid anxiety disorders were associated with increased help-seeking. Conclusion: A small percentage of individuals with MDD living in community of Tianjin sought help. They preferred non-healthcare sources to healthcare settings. Demographic and clinical features were associated with help-seeking behaviors.

The land development by human beings has changed from the initial small-scale and low-level transformation to the comprehensive utilization of large-scale and high-intensity implementations. The contradiction between production-living-ecology space (PLES) has become increasingly prominent while drawing land dividends. As one of the important birthplaces of Chinese civilization and the ecological barrier in the northern region, the rapid urbanization and industrialization of the Yellow River Basin (YRB) make the ecological environment very fragile, and the imbalance of land and space development is extremely serious. Therefore, according to the multifunctional characteristics of land use, this paper establishes a classification system of production space (PS), living space (LS) and ecology space (ES), and discusses the spatiotemporal evolution and conflict distribution characteristics of the PLES with the help of the transfer matrix and spatial conflict index (SCI). The results are as follows. In 1990-2020, agricultural production space (APS), grassland ecology space (GES) and other ecology space (OES) yielded the largest proportion of PLES in the YRB. However, compared with 1990, the area of these spatial types decreased in 2020, while the urban living space (ULS) expanded rapidly. The distribution pattern of PLES was generally consistent, and the transformation between PLES in Ningxia, central Inner Mongolia, Loess Plateau and downstream areas was relatively intense. The conflict index of PLES showed an upward trend, but it was generally in a controllable range. The stable and controllable areas were concentrated in the upstream of the urban agglomeration, and the midstream and downstream were basic out of control and seriously out of control, respectively.

ABSTRACT

The outbreak of COVID-19 has prompted consideration of the importance of urban resilience. Based on a multidimensional perspective, the authors of this paper established a comprehensive evaluation indicator system for evaluating urban resilience in the Yellow River basin (YRB), and various methods such as the entropy value method, Theil index, exploratory spatial data analysis (ESDA) model, and geographical detector model were used to measure the spatiotemporal characteristics and influencing factors of urban resilience in the YRB from 2011 to 2018. The results are as follows. (1) From 2011 to 2018, the urban resilience index (URI) of the YRB showed a "V"-shaped dynamic evolution in the time series, and the URI increased by 13.4% overall. The resilience of each subsystem showed the following hierarchical structure: economic resilience > social resilience > ecological resilience > infrastructure resilience. (2) The URI of the three major regions-upstream, midstream, and downstream-increased, and the resilience of each subsystem in the region showed obvious regional characteristics. The comprehensive difference in URI values within the basin was found to be shrinking, and intraregional differences have contributed most to the comprehensive difference. (3) There were obvious zonal differences in the URI from 2011 to 2018. Shandong Peninsula and Hohhot-Baotou-Ordos showed a "High-High" agglomeration, while the southern and southwestern regions showed a "Low-Low" agglomeration. (4) Among the humanist and social factors, economic, fiscal, market, urbanization, openness, and innovation were found to be the factors that exert a high impact on the URI, while the impacts of natural factors were found to be low. The impact of the interaction of each factor is greater than that of a single factor.
