ABSTRACT

River water quality is influenced by natural processes and human activities. Multi-scale landscape patterns can affect river water quality by altering the generation and transport processes of pollutants at different spatial scales. Taking Taizi River Basin in Northeast China as an example, we analyzed the relationship between landscape patterns and non-point source pollution in rivers based on water quality monitoring data and land use data by using correlation analysis and redundancy analysis methods. We aimed to determine the key spatial scales for the responses of landscape patterns to non-point source pollution and identify the key landscape indices influencing river non-point source pollution. The results showed that water quality of Taizi River Basin had seasonal differences, with better water quality during the flood season than non-flood season. Spatially, total nitrogen (TN) and total phosphorus (TP) were higher at the confluence points of tributaries and downstream areas. The impact of landscape patterns on non-point source pollution was stronger during the non-flood season than the flood season, while the influence on TN was stronger than on TP. At the spatial scale of within 500 m buffer zone during the flood season and at the sub-watershed scale during the non-flood season, landscape patterns showed the highest explanatory power for the variations of TN and TP. At the type level, built-up land, cropland, and bare land were positively correlated with TN and TP, while forest was negatively correlated with TN and TP, which were the key types influencing non-point source pollution. At the landscape level, patch density, percentage of like adjacencies, and contagion index were key indicators affecting watershed water quality. Lower patch density was associated with better connectivity and aggregation of "sink" landscapes, leading to better purification effects on TN, but more pronounced retention effects on TP. Conversely, higher landscape diversity and denser pattern of multiple types would cause the deterioration of water quality. Our results suggested that rational allocation of landscape types within the watershed and riparian buffer zones, appropriately enriching landscape diversity, and optimizing landscape aggregation and connectivity would be effective measures for improving water quality and achieving sustainable ecological management.

ABSTRACT

Long noncoding RNAs (lncRNAs) have been highlighted to be involved in the pathological process of ischemic stroke (IS). The purpose of the present study was to investigate the expression profile of lncRNAs in peripheral blood mononuclear cells (PBMCs) of acute IS patients and to explore their utility as biomarkers of IS. Distinctive expression patterns of PBMC lncRNAs were identified by an lncRNA microarray and individual quantitative real-time PCR (qRT-PCR) in four independent sets for 206 IS, 179 healthy controls (HCs), and 55 patients with transient ischemic attack (TIA). A biomarker panel (lncRNA-based combination index) was established using logistic regression. LncRNA microarray analysis showed 70 up-regulated and 128 down-regulated lncRNAs in IS patients. Individual qRT-PCR validation demonstrated that three lncRNAs (linc-DHFRL1-4, SNHG15, and linc-FAM98A-3) were significantly up-regulated in IS patients compared with HCs and TIA patients. Longitudinal analysis of lncRNA expression up to 90 days after IS showed that linc-FAM98A-3 normalized to control levels by day 7, while SNHG15 remained increased, indicating the ability of lncRNAs to monitor IS dynamics. Receiver-operating characteristic (ROC) curve analysis showed that the lncRNA-based combination index outperformed serum brain-derived neurotrophic factor (BDNF) and neurone-specific enolase (NSE) in distinguishing IS patients from TIA patients and HCs with areas under ROC curve of more than 0.84. Furthermore, the combination index increased significantly after treatment and was correlated with neurological deficit severity of IS. The panel of these altered lncRNAs was associated with acute IS and could serve as a novel diagnostic method.

ABSTRACT

BACKGROUND: Dobutamine, a commonly used vasoactive drug, has been reported to reduce pulmonary edema and protect against acute lung injury (ALI) by up-regulating aquaporin 5 (AQP5) expressions. However, the underlying mechanism is still elusive. METHODS: ALI was induced by intravenous injection of LPS. Seventy male New Zealand white rabbits were randomly divided into seven groups: sham group, ALI group, dobutamine low-dose group [group ALI + Dob (L)], dobutamine medium-dose group [group ALI + Dob (M)], dobutamine high-dose group [group ALI + Dob (H)], ALI + Dob (H) + ICI group and sham + ICI group. ICI 118,551, a potent and specific beta-2 antagonist, could block the effect of dobutamine. The animals were sacrificed at 3 h after endotoxic shock and lungs were removed. The arterial blood gas was analyzed. The lung wet to dry (W/D) ratio was determined. The level of cyclic AMP (cAMP) in lung tissue was assessed by ELISA. The expression of AQP5 protein was determined by western blotting and immunohistochemistry. The pathological alteration in lung tissue was evaluated by optical microscopy and electron microscope, and lung injury score was assessed. RESULTS: Dobutamine increased AQP5 protein expression and cAMP level in a dose-dependent manner. Meanwhile, the degree of lung pathological and ultrastructural lesion was ameliorated and arterial blood gas was improved obviously. Additionally, W/D ratio and histological scores decreased significantly. However, the AQP5 protein expression and cAMP level were significantly decreased in group ALI + Dob (H) + ICI than that in group ALI + Dob (H), the degree of lung pathological and ultrastructural lesion was more serious in group ALI + Dob (H) + ICI than that in group ALI + Dob (H) and the arterial blood gas was not obviously improved. CONCLUSIONS: These results suggested that protective effect of dobutamine against endotoxin shock-induced ALI may be due to its ability of up-regulating AQP5 protein expression via increasing intracellular cAMP concentration.

ABSTRACT

In order to correctly evaluate wetland ecological benefit and restoration in the natural reserves of western Jilin Province, an index system of wetland ecological benefit assessment for nature reserves was established based on ecosystem service theory and catastrophe theory. Considering the diversity characteristics of ecological benefit, and consulting the research results of ecological benefit in study area in 2001, the analytic hierarchy process and entropy method were introduced to analyze the ecological benefit and restoration in 2005 and 2012. The results showed that the ecological benefit level was very high in Momoge Nature Reserve since 2005, high in Chaganhu and Xianghai nature reserves, and moderate in Dabusu Nature Reserve. This study showed that the assessment combining the catastrophe progression method with the analytic hierarchy process and entropy method could reveal the dynamics and the integrality of wetland ecological benefit, and accurately reflect the reality of the ecological restoration in study area. In recent years, some achievements in ecological protection and environmental remediation had been obtained in the west of Jilin Province, and the whole ecological benefit of the wetlands had been improved and enhanced by means of man-made effective interference.
