ABSTRACT

Beer is among the most popular beverages in the world, with the production distributed uniformly between the biggest continents, so the utilization of brewing by-products is essential on a global scale. Among their potential recipients, the plastics industry offers extensive range of potential products. Herein, the presented study investigated the application of currently underutilized solid brewing by-products (brewers' spent grain, spent hops, spent yeast) as fillers for highly-filled poly(ε-caprolactone)-based composites, providing the first direct connection between spent hops or spent yeast and the polymer composites. Comprehensive by-product characterization revealed differences in chemical composition. The elemental C:O ratio, protein content, and Trolox equivalent antioxidant capacity varied from 1.40 to 1.89, 12.9 to 32.4 wt%, and 2.41 to 10.24 mg/g, respectively, which was mirrored in the composites' structure and performance. Morphological analysis pointed to the composition-driven hydrophilicity gap limiting interfacial adhesion for high shares of brewers' spent grain and spent hops, due to high hydrophilicity induced by carbohydrate content. Phytochemicals and other components of applied by-products stimulated composites' oxidative resistance, shifting oxidation onset temperature from 261 °C for matrix over 360 °C for high spent yeast shares. Simultaneously, spent yeast also provided compatibilizing effects for poly(ε-caprolactone)-based composites, reducing complex viscosity compared to other fillers and indicating its highest affinity to poly(ε-caprolactone)due to the lowest hydrophilicity gap. The presented results indicate that the proper selection of brewing by-products and adjustment of their shares creates an exciting possibility of engineering composites' structure and performance, which can be transferred to other polymers differing with hydrophilicity.

ABSTRACT

The tyrosinase pathway takes part in the enzymatic process of food browning and is primarily responsible for food spoilage - manifesting itself from a decrease in its nutritional value to a deterioration of taste, which consequently leads to a gradual loss of shelf life. Finding safe and bio-based tyrosinase inhibitors and anti-browning agents may be of great importance in agriculture and food industries. Herein, we showed that Cyrene™ exhibits tyrosinase inhibitory activity (IC50: 268.2 µM), the 1.44 times higher than ascorbic acid (IC50: 386.5 µM). Binding mode studies demonstrated that the carbonyl oxygen of Cyrene™ coordinates with both copper ions. Surprisingly, both hydroxyl groups of Cyrene gem-diol perform a monodentate binding mode with both copper ions, at similar distances. This fact suggests that both compounds could have a similar binding mode and, as consequence, similar biological activities in tyrosinase inhibition assays and anti-browning activities.

ABSTRACT

In aqueous solutions, cetyltrimethylammonium cations bind to carrageenan polyanions, and the resulting ionic associates form macroscopic aggregates due to hydrophobic interaction. At certain ratios of cetyltrimethylammonium to carrageenan, the resulting colloidal particles auto-flocculate. According to visual observations, the ratio ranges from 1 to 3 mmol/g; otherwise the suspensions are stable. By measuring the sedimentation rate and particle size distribution, the most extensive flocculation was found to be from 1.7 to 2.3 mmol/g. The ratio corresponding to the fastest auto-flocculation was precisely determined by titrating the reagents with small increments and recording the turbidity. The turbidimetric titration plots contain distinct break points corresponding to the most extensive flocculation. These break points occur at the same ratio of carrageenan to cetyltrimethylammonium over a wide range of reagent concentrations. The precise values of the critical ratio were found to be 1.78 and 1.53 mmol/g, respectively, during the titration of cetyltrimethylammonium with carrageenan and vice versa. The number of anionic sulfate groups in carrageenan was measured by ICP OES and found to be 1.35 mmol/g. This value is consistent with the critical ratio of the auto-flocculation.

ABSTRACT

The purpose of this study was to modify plasticised PVC to obtain a material with antimicrobial properties and selected mechanical properties. Natural cassia oil (CO) was used to modify plasticised PVC materials. The modified material was produced by extrusion. The introduced modifier had a maximum concentration of 20 phr. Rheological and mechanical properties were evaluated, and the glass transition temperature was determined. The antioxidant and antimicrobial activity of the agar diffusion method was investigated by analysing the growth inhibition zones against Enterococcus faecalis and Listeria monocytogenes. A favourable effect of the cassia oil content on the increase in antioxidant activity of the developed polymeric materials was found with an increase in the modifier content and the duration of action (30 days). The largest growth restriction zones were observed for L. monocytogenes, i.e., they showed the highest sensitivity to the modified material. The simultaneous decrease in modulus of elasticity, increase in elongation at break, and decrease in Tg indicate that the modifier has a plasticising effect on PVC. The developed material may find application as an active and/or functional material, especially as an emitter of antimicrobial agents, in the packaging used to store minimally processed food.

ABSTRACT

The aim of this study was to develop a dressing with bioactive lavender in a new form of nanoemulsion, and to verify its biosafety and effectiveness in burn wound healing. As part of this research, the composition of the bioactive carrier of lavender oil in the form of a nanoemulsion obtained using ultrasound was optimised. The mean particle size of the internal phase and polydispersity were determined using the dynamic light scattering method using a Zestasizer NanoZS by Malvern and using cryo-transmission electron microscopy (TEM). These studies confirmed that the selected formulation had a particle size of approximately 180 nm and remained stable over time. The preparation was also subjected to rheological analysis (viscosity approximately 480 mPa·s) and a pH test (approximately 6). A macroemulsion (ME) with the same qualitative composition was developed as a reference. Nanoformulations and MEs were tested for skin penetration using Raman spectroscopy in an in vitro model. Research has shown that both formulations deliver oil to living layers of the skin. Subsequently, studies were conducted to confirm the effect of lavender oil in emulsion systems on the mitigation of the inflammatory reaction and its pro-regenerative effect on the wound healing process in an in vitro cell culture model. The safe concentration of the oil in the emulsion preparation was also determined based on preliminary in vivo tests of skin sensitisation and irritation as well as an hemocompatibility test of the preparation.

ABSTRACT

This article presents the results of research on obtaining new polyurethane (PUR) foams modified with thermally expanded vermiculite. The filler was added in amount of 3 wt.% up to 15 wt.%. The additionally applied procedure of immersion the non-organic filler in H2O2 was performed to increase the exfoliation effect of thermally treated mineral and additional oxidation the surfaces. The effect of fillers on foaming process, cell structure, thermal insulation, apparent density, compressive strength, thermal properties, and flammability are assessed. The foaming process of PUR foams modified with vermiculite was comparable for all systems, regardless of the content of the filler. A slight increase in reactivity was observed, confirmed by a faster decrease in dielectric polarization for the system with modified vermiculite by H2O2. The modification of the reference system with the vermiculite increased the content of closed cells from 76% to 91% for the foams with the highest vermiculite content. Coefficient of thermal conductivity of reference foam and foams modified with vermiculite was in the range 24-26 mW/mK. The use of vermiculite up to 15 wt.% did not influence significantly on mechanical properties and flammability, which from an economic point of view is important because it is possible to reduce the cost of materials by introducing a cheap filler without deteriorating their properties.

ABSTRACT

The paper presents the characteristics of unplasticized PVC composites modified with biofiller obtained from the waste eggshells of hen eggs. The composites obtained by extrusion contained from 10 phr to 40 phr of biofiller. The filler was characterized using the SEM, TG, and sieve analysis methods. The influence of the filler on the processing properties was determined using plastographometric and MFR tests. Fundamental analysis of mechanical properties was also performed, i.e., Charpy impact strength and determination of tensile properties. The mechanical properties were supported with dynamical mechanical thermal analysis, time of thermal stability, and thermogravimetric analysis. Structure analysis was also performed using SEM and X-ray microcomputed tomography (micro-CT). The processing properties of the tested composites do not give grounds for disqualifying such material from traditional processing PVC mixtures. Notably, the biofiller significantly improves thermal stability. Ground eggshells (ES) work as scavengers for the Cl radicals released in the first stage, which delays the PVC chain's decay. Additionally, a significant increase in the value of the modulus of elasticity and softening point (VST) of the composites concerning PVC was found. Ground hen eggshells can be used as an effective filler for PVC composites.

ABSTRACT

Rotational molding is a technology in which polymeric thin-walled products can be made. The newest descriptions of this technology concern the possibility of obtaining polymer composite materials. There are two main methods of incorporating fillers into a polymer matrix. Dry blending is based on mixing fillers with polymer powders before rotational molding by hand or using automatic mixers. In the melt compounding method, fillers are mixed with the polymer in the preliminary step by melt processing and then grinding or pulverization to obtain polymer powders for rotational molding. This work aimed to investigate the impact of the processing procedure on the structure and properties of biobased composites with expanded vermiculite. Produced rotomolded parts were examined using mechanical tests to assess changes in tensile, flexural, and impact properties. The most significant difference in mechanical properties was noted for samples with 10 wt% expanded vermiculite (EV). The elasticity modulus increases by almost 2 fold when the sample is prepared in a two-step process, the tensile strength is 4-fold higher, the flexural modulus is 3-fold higher, and the flexural strength is 5-fold higher. We also investigated thermomechanical properties in DMA measurement. The void volume content was also measured to control the quality of obtained parts. The porosity of dry blended samples containing more than 2 wt% EV is almost 2-fold higher. Other methods to control quality and structure were optical and scanning electron microscopy used for rotomolded parts and polymer powders. The investigations of rotomolded parts were supplemented with a complete description of used materials, including the particle size distributions of polymer powders and filler. Analysis of the thermal properties and chemical structure was also performed despite all the mechanical tests. The emerging conclusions from the research clearly show that the two-step process allows for achieving a more beneficial mechanical performance of the composites made of the biobased polymer in rotational molding technology.

ABSTRACT

In this work, thermally expanded vermiculite (TE-VMT) was surface modified and used as a filler for composites with a polylactide (PLA) matrix. Modification of vermiculite was realized by simultaneous ball milling with the presence of two PLA chain extenders, aromatic carbodiimide (KI), and 4,4'-methylenebis(phenyl isocyanate) (MDI). In addition to analyzing the particle size of the filler subjected to processing, the efficiency of mechanochemical modification was evaluated by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The composites of PLA with three vermiculite types were prepared by melt mixing and subjected to mechanical, thermomechanical, thermal, and structural evaluation. The structure of composites containing a constant amount of the filler (20 wt%) was assessed using FTIR spectroscopy and SEM analysis supplemented by evaluating the final injection-molded samples' physicochemical properties. Mechanical behavior of the composites was assessed by static tensile test and impact strength hardness measurements. Heat deflection temperature (HDT) test and dynamic thermomechanical analysis (DMTA) were applied to evaluate the influence of the filler addition and its functionalization on thermomechanical properties of PLA-based composites. Thermal properties were assessed by differential scanning calorimetry (DSC), pyrolysis combustion flow calorimetry (PCFC), and thermogravimetric analysis (TGA). The use of filler-reactive chain extenders (CE) made it possible to change the vermiculite structure and obtain an improvement in interfacial adhesion and more favorable filler dispersions in the matrix. This translated into an improvement in impact strength and an increase in thermo-mechanical stability and heat release capacity of composites containing modified vermiculites.

ABSTRACT

Difficulties with interpersonal communication experienced by individuals with autism spectrum disorders (ASD) significantly contribute to their underrepresentation in the workforce as well as problems experienced while in employment. Consistently, it is vital to understand how communication within the employment cycle of this group can be improved. This study aims to identify and analyze the possibilities of modifying the communication processes around recruitment, selection, onboarding, and job retention to address the specific characteristics and needs of the representatives of this group. This qualitative study is based on 15 in-depth interviews conducted with 21 field experts, i.e.,: therapists, job trainers, and entrepreneurs employing people with ASD. The findings of this research informed the creation of an inclusive communication model supporting the employment cycle of individuals with ASD. The most important recommendations within the model that was created include the modification of job advertisements, use of less structured job interviews, providing opportunities for mentorship, and supportive and non-direct, electronically mediated communication. To apply the above-mentioned solutions and take full advantage of the talents of people with ASD, it is also necessary to provide tailored sensitivity and awareness training programs for their direct addressees as well as their neurotypical colleagues, including managerial staff.

ABSTRACT

The Covid-19 global pandemic is a crisis like no other, forcing governments to implement prolonged national lockdowns in an effort to limit the spread of the disease. As organizations aim to adapt and remain operational, employers can suspend or reduce work activity for events related to Covid-19 and claim government support to subsidize employee wages. In this way, some employees are placed on furlough (i.e., temporary unemployment) as opposed to being made redundant. While the impact of such schemes on global economy attracted much attention, their micro-level impact on individual employees is still unknown. Building on the ability-motivation-opportunity (AMO) framework, this pilot study explores how employees' perceptions of abilities, motivation, and opportunities are affected as a result of furlough. Rapid ethnography including interviews, observations, and document analysis in a British organization provided insights into the perceptions and experiences of employees put on furlough and highlighted that all three elements of AMO are affected by the current situation, either positively or negatively. We identify theoretical contributions and suggest a number of AMO enhancing practices in the context of furlough.

ABSTRACT

This paper presents the effects of accelerated weathering on the properties of polylactide (PLA) composites filled with linseed cake. The particle-shaped waste filler with different linseed oil content (0.9-39.8 wt %) was incorporated with constant amount of 10 wt % to a polymeric matrix and subjected to accelerated weathering tests with different exposition times. The structure of the composites, their mechanical, thermal, and thermo-mechanical properties were evaluated by means of scanning electron microscopy, tensile test, dynamic mechanical thermal analysis, and differential scanning calorimetry prior to and after weathering. The results of the measurements were analyzed in reference to the amount of crude oil contained in the filler. The behavior of the multiphase composite during weathering was described. It was found that the oil-rich samples during the first stage of the process showed increased resistance to hydrolytic degradation due to their relatively high crystallinity. The presence of water and elevated temperatures caused swelling of the filler and cracking of the polymeric matrix. Those discontinuities enabled the plasticizing oil to be rinsed out of the composite and thus water penetrated into the samples. As a result, the PLA-based composites containing oil-rich linseed cake were found to be more vulnerable to hydrolytic degradation in a longer time.

Bacterial cellulose modified with chitosan (MBC) is an innovative biomaterial used in regenerative medicine which may potentially improve treatment outcomes mesh for hernia repair surgery by facilitating better absorption in native tissue with less risk of mesh-related infections. The aim of the present study was to evaluate the biocompatibility of mesh based on MBC, and determine whether immunological reactions occur due to hypersensitivity to the implants. Forty five Imp:WIST rats were randomly assigned to be implanted with one of three mesh types: simple polypropylene mesh (n = 15), mesh modified by bacterial cellulose only (n = 15) and MBC mesh (n = 15) and evaluated after one and three months following intramuscular implantation. For MBC mesh, basic toxicological studies, i.e. Acute Dermal Irritation, Intradermal Reactivity and Acute Sensitization (GPMT), were also carried out on 9 Imp:BN albino rabbits and 15 Imp:D-H guinea pigs. The lowest immune response and the highest degree of fibroplasia were observed for MBC mesh both after one and three months after implantation. Toxicological studies classified the tested MBC mesh as a barely perceptible irritant with no signs of sensitization or allergic reactions observed during the studies. The findings indicate that MBC mesh does not irritate, does not sensitize and does not cause hypersensitivity in the implant site, and therefore presents a low risk of provoking such reactions in humans.

OBJECTIVES: The aim of this study was to investigate plasma concentrations of four pro-inflammatory cytokines (IL17a, IL6, TNFα and IL12p70) in patients with myocardial infarction and to analyse them according to presence of depression observed during first 6 months after myocardial infarction. METHODS: In 44 patients with the first acute STEMI (ST segment elevation myocardial infarction) plasma levels of IL17a, IL6, TNFα and IL12p70 were measured on the 3rd and 5th day after the MI. Cytokine concentrations were analyzed according to the presence of depression during 6 months of observation. RESULTS: Two groups of patients distinguished according to presence of depression during 6 months of observation differed in their inflammatory reaction to MI. In the depression group all four cytokines on the 3rd day after the MI were elevated compared to control and on the 5th day two of them: IL17a and IL6 were still elevated. In the group without depression on the 3rd day only two of four investigated cytokines were elevated and on the 5th day only IL6 concentration remained higher. CONCLUSIONS: It can be assumed that more pronounced inflammatory response as an element of stress reaction after MI can predispose to depression. IL17a increase can play particularly important role in this process.

ABSTRACT

Network models have been used to capture, represent and analyse characteristics of living organisms and general properties of complex systems. The use of network representations in the characterization of time series complexity is a relatively new but quickly developing branch of time series analysis. In particular, beat-to-beat heart rate variability can be mapped out in a network of RR-increments, which is a directed and weighted graph with vertices representing RR-increments and the edges of which correspond to subsequent increments. We evaluate entropy measures selected from these network representations in records of healthy subjects and heart transplant patients, and provide an interpretation of the results.

ABSTRACT

This study describes the egg membrane structures of angelfish (Pterophyllum scalare), morpho-physiological changes during angelfish embryogenesis from activation to hatching under optimal conditions (28°C; pH 6.8), the developing larvae and fry, the effect of alkaline pH on the early developmental stages of the species, the relationship between food item size and fry survival. Egg membranes (thin, transparent, 1.67-2.18 µm thick) are covered by a sticky substance. The amber-coloured angelfish eggs were oval in shape, with average diameters of 1.436 and 1.171 mm, i.e., a mean volume of 1.033 ± 0.095 mm³. The survival rate of embryos and larvae kept in water with an elevated, slightly alkaline pH was very low: as few as 2% of the embryos survived, while in the batch kept in optimal water conditions very few eggs died. The first larvae hatched after 1288 h of embryonic development. The newly hatched larvae measured on average 2.60 ± 0.093 mm and had large (0.64 ± 0.077 mm³) yolk sacs. They attached themselves to the substrate with a secretion of thin, viscous threads, which was released from glands situated on the top of the head. The glands vanished on day 5. The 1-day-old larvae showed the first pigment cells on the body and the eyes of the 2-day-olds were already fully pigmented. Between day 4 and 5 of larval life, the larvae began feeding on live food. The 23-day-old fry looked like a miniature versions of the adults. Mortality of the angelfish larvae during their first days after hatching was higher in those fed brine shrimp (Artemia salina) nauplii than those fed protozoans and rotifers.


En este trabajo se ha descrito la estructura de las túnicas ovulares del escalar o pez ángel (Pterophyllum scalare), las modificaciones morfo-fisiológicas que transcurren durante la embriogénesis del escalar desde el momento de activización para el desove en condiciones ambientales óptimas (28ºC y pH 6,8) y, se han caracterizado las larvas y los alevines desarrollándose. Adicionalmente, se ha estudiado el efecto del pH básico del agua sobre los primeros estadios de desarrollo y la dependencia entre la cantidad de alimento y la supervivencia de los alevines. Las túnicas ovulares del escalar son finas (1,67-2,18 mm), transparentes, cubiertas de una sustancia viscosa. Los huevos de color ámbar tienen forma ovalada de diámetros medios 1,436 y 1,171 mm y de un volumen medio de 1,033 ± 0,095mm³. El porcentaje de supervivencia de embriones y de larvas en agua con pH aumentado, ligeramente básico, fue muy bajo, ya que sólo el 2% de los embriones sobrevivió, mientras que en agua de parámetros óptimos sólo algunos huevos palidecieron. Las larvas recién salidas del huevo medían 2,60 ± 0,093 mm por término medio y poseían grandes (0,64 ± 0,077 mm³) sacos vitelinos. Se han adherido al substrato mediante una secreción en forma de filamentos finos y viscosos. Esta secreción se ha desprendido de las glándulas ubicadas en la cumbre de la cabeza. Las glándulas desaparecieron al 5º día de vida de las larvas. Las larvas de un día poseían ya las primeras células pigmentarias en el cuerpo, los ojos de las larvas de dos días estaban pigmentadas plenamente. Entre el 4º y 5º día de vida las larvas empezaron a tomar alimento. Las larvas de 23 días se parecían a una versión miniaturizada de los adultos. La mortalidad de las larvas del escalar alimentadas durante los primeros días a partir del desove con larvas de artemia salina (Artemia salina) fue mayor que la de las alimentadas con protozoarios y rotíferos.

The heart rate responds dynamically to various intrinsic and environmental stimuli. The autonomic nervous system is said to play a major role in this response. Multifractal analysis offers a novel method to assess the response of cardiac interbeat intervals. Twenty-four hour ECG recordings of RR interbeat intervals (of 48 elderly volunteers (age 65-94), 40 middle-aged persons (age 45-53) and 36 young adults (age 18-26)) were investigated to study the effect of aging on autonomic regulation during normal activity in healthy adults. Heart RR-interval variability in the very low frequency (VLF) band (32-420 RR intervals) was evaluated by multifractal tools. The nocturnal and diurnal signals of 6 h duration were studied separately. For each signal, the analysis was performed twice: for a given signal and for the integrated signal. A multifractal spectrum was quantified by the h(max) value at which a multifractal spectrum attained its maximum, width of a spectrum, Hurst exponent, extreme events h(left) and distance between the maxima of a signal and its integrated counterpart. The following seven characteristics are suggested as quantifying the age-related decrease in the autonomic function ('int' refers to the integrated signal): (a) h(sleep)(max) - h(max)(wake) > 0.05 for a signal; (b) h(int)(max) > 1.15 for wake; (c) h(int)(max) - h(max) > 0.85 for sleep; (d) Hurst(wake) - Hurst(sleep) < 0.01; (e) width(wake) > 0.07; (f) width(int) < 0.30 for sleep; (g) h(int)(left) > 0.75. Eighty-one percent of elderly people had at least four of these properties, and ninety-two percent of young people had three or less. This shows that the multifractal approach offers a concise and reliable index of healthy aging for each individual. Additionally, the applied method yielded insights into dynamical changes in the autonomic regulation due to the circadian cycle and aging. Our observations support the hypothesis that imbalance in the autonomic control due to healthy aging could be related to changes emerging from the vagal function (Struzik et al 2006 IEEE Trans. Biomed. Eng. 53 89-94).

ABSTRACT

BACKGROUND: In recent years the WTMM (wavelet transform modulus maxima) and MDFA (multifractal detrended fluctuation analysis) methods have become widely used techniques for the determination of nonlinear, multifractal heart rate (HR) dynamics. The purpose of our study was to compare multifractal parameters of heart rate calculated using both methods in a group of 90 patients with reduced left ventricular systolic function (rlvs group) and in a group of 39 healthy persons (nsr group). METHODS: For each subject from the rlvs group (LVEF < or =40%) and the nsr group, a 24-hour ECG Holter monitoring was performed. The width of the multifractal spectrum and global Hurst exponent were calculated by means of WTMM and MDFA methods for 5-hour daytime and nighttime subsets. RESULTS: The width of the multifractal spectrum was significantly lower and the Hurst exponent was significantly higher in rlvs group in comparison to nsr group both during diurnal activity and nocturnal rest according to MDFA and only during diurnal activity according to WTMM method. In both groups we observed significant differences of the multifractal spectrum width and the global Hurst exponent between the nighttime and daytime recordings. CONCLUSIONS: MDFA seems to be more sensitive as compared with WTMM method in differentiation between multifractal properties of the heart rate in healthy subjects and patients with left ventricular systolic dysfunction.

ABSTRACT

OBJECTIVE: The sympathetic nervous system is a key modulator of prognosis and outcome in cardiac ischaemia and infarction. The effects of acute cardiac ischaemia on sympathetic neural traffic in humans are unknown. We tested the hypothesis that angioplasty, and associated transient myocardial ischaemia, elicits changes in neural circulatory control, including direct intraneural measures of sympathetic traffic. METHODS: We measured muscle sympathetic nerve activity (MSNA), blood pressure and heart rate in 12 patients (11 men, one woman) undergoing clinically indicated elective coronary angioplasty of the left anterior descending (n = 7) or circumflex (n = 5) coronary artery. Baseline data were obtained for 2 min before occlusion. Each balloon inflation was designed to last up to 120 s. RESULTS: Coronary occlusion had no significant effect on blood pressure or heart rate. In contrast, occlusion resulted in a significant increase in MSNA, in 10 of the 12 patients, and in all seven of those patients undergoing angioplasty of the left anterior descending artery. The group mean increase in MSNA during occlusion was 36 +/- 11% (P = 0.008 versus preocclusion). MSNA increased within 60 s of occlusion (129 +/- 12% of baseline; n = 12; P = 0.04), and increased further during the next 60 s to 141 +/- 12% of baseline levels (n = 10; P < 0.001). Increases in MSNA were similar in patients with and without significant chest pain (39 +/- 9% versus 34 +/- 13%, respectively; P = 0.84). CONCLUSION: Acute coronary occlusion during angioplasty increases central sympathetic outflow, but there is no systematic change in heart rate. The increase in sympathetic nerve traffic cannot be explained by blood pressure changes or occlusion-related chest pain.

ABSTRACT

In the present study three factors were investigated that may affect the process of the first polar body extrusion in pig oocytes matured in vitro: IVM medium, oocyte diameter and donor genotype at the ryanodine receptor (RYR1) locus. In the first experiment, COCs were collected by the aspiration of slaughterhouse ovaries. Oocytes were matured in vitro at 39 degrees C, in humidified 5% CO(2) atmosphere for 44 h using the following media: (1) TCM199+hCG+eCG+follicular fluid (FF), (2) TCM199+hCG+17beta-estradiol and (3) NCSU23+hCG+eCG+FF. According to cytogenetic analysis, 98.1% of cells reached the second metaphase stage (MII). No significant differences were observed among IVM groups in terms of diploidy level. In the second experiment, oocytes collected by the aspiration or slicing of individual ovaries were matured in vitro in groups reflecting their origin. One ovary was considered a donor. IVM was carried out under conditions described in experiment I, with the use of TCM199+hCG+17beta-estradiol. A total of 68 ovaries/donors were included in this study. Granulosa cells collected from each ovary were used as DNA source in molecular (RFLP) analysis. Genotype frequencies at the RYR1 locus were as follows: CC, 0.46; CT, 0.48 and TT, 0.06. After maturation the diameter of each denuded oocyte was determined with the use of a computer aided system. Five size categories were distinguished: <90, 90-100, 100.1-110, 110.1-120 and >120 microm. The average diameter of haploid oocytes at MII stage was 111.7 microm, whereas that of diploid cells was 110.4 microm. According to statistical analysis, diploidy was not related to the oocyte diameter. That trait, however, was influenced by the donor genotype at the RYR1 locus. The TT genotype was associated with a higher rate of diploidy.
