ABSTRACT

Oil exploration's devastation on health and the environment may far outweigh its economic benefits. An oil spill occurred at Egbokodo River in Delta State, Nigeria, thereby polluting the land and water bodies. The study was therefore aimed at evaluating the impacts of iron, lead, cadmium, and chromium on the zooplankton community structure of Egbokodo River and the potential health risks. Zooplankton and surface water samples were collected to investigate the concentrations of trace metals and zooplankton abundance. The associated carcinogenic and non-carcinogenic effects of the metals in the water were analyzed. Trace metal concentrations in the surface water were determined using atomic absorption spectroscopy (Philips model PU 9100) and zooplankton samples were collected using a hydrobios plankton net (mesh size 25 µm). Total petroleum hydrocarbons (TPH) and oil and grease (OG) were determined using Agilent 7890B gas chromatography coupled to flame ionization detector (GC-FID) and volumetric analysis respectively. The trend of the abundance of zooplanktons cross the river was 18 individuals (Station A) < 100 individuals (Station B) < 155 individuals (Station C). Cyclopoida proved to be the most resilient to the impacts of the oil spill. On a taxa basis, the order of abundance among Calanoida, Cyclopoida, Cladoceran, and Harpacticoida was Station C > Station B > Station A, except in Amphipoda where Station B > Station C > Station A was observed. Iron and lead posed significant carcinogenic risks that are liable to be inflicted by the ingestion of the water. The cumulative non-carcinogenic health risk in the male was the only significant (> 1) among the age groups. Total petroleum hydrocarbons (TPH), oil and grease (OG), iron, and lead had notable impacts on the general abundance of zooplankton in the aquatic habitat. The dominance of the Cyclopoida in the river buttressed the impact of the oil spill which warrants a prompt remediation measure. The pollution had notable ecological impacts on the zooplankton community structure of the aquatic habitat. The adults in the nearby human populations are liable to elicit carcinogenic health challenges associated with lead and iron ingestion. The males are at risk of non-carcinogenic illnesses which are associated with the combined toxicity effects of all the metals. The study suggests that the pollution in Egbokodo River was validated by the dominance of the Cyclopoida in the aquatic habitat. The study confers bioindicator reputation on the Cyclopoida for future biomonitoring studies.

ABSTRACT

BACKGROUND: Polygalacturonase (EC 3.2.1.15) enzyme aids in microbial spoilage of fruits and vegetables. It is very important to find economical ways to producing the enzyme so as to achieve maximum yield in industries due to its use at different areas of production process. METHODS: Isolation of polygalacturonase-producing bacterial strain from tomatoes (Lycopersicon esculentum Mill.) was studied. Polygalacturonase-producing bacterial strains were isolated and screened from tomatoes stored at normal laboratory temperature (25 ± 2°C). They were identified based on their morphological, biochemical, and molecular characteristics. The enzyme produced was partially purified by the ammonium sulphate precipitation method. Molecular weights and optimum conditions for best enzyme activity were obtained by SDS PAGE technique. RESULTS: Five bacterial isolates resulted after screening. Bacterial strain code B5 showed highest polygalacturonase activity. Optimum conditions for polygalacturonase PEC B5 were maintained at pH 4.5; temperature 35°C; substrate concentration 0.3 mg/ml, and best activity at less than 5 min of heating. The enzyme PEC B5 was found to weigh 65 kDa and 50 kDa for crude and partially purified aliquots, respectively. The result of 16S rRNA gene sequencing revealed bacterial strain code B5 as Enterobacter tabaci NR146667 having 79% similarity with the NCBI GenBank. CONCLUSION: Microorganisms should be developed for large-scale production of enzymes in developing countries.

ABSTRACT

PURPOSE: Increasing rates of clonal spread of fecal blaTEM bacilli remains a huge concern to the community health with resultant high morbidity. The fecal carriage and clonal diversity of bla TEM within the communities in Southwest Nigeria were surveyed. MATERIALS AND METHODS: Enteric bacilli obtained from fresh fecal samples randomly collected from community residents were biotyped and profiled for antibiotic susceptibility. Resistant strains were typed for beta-lactamase, extended-spectrum beta-lactamases (ESBL), AmpC and carbapenemase production while the R-plasmid carriage was detected and mating activities were examined. The presence of bla TEM gene was assayed by PCR and its phylodiversity determined with 16sRNA genomic profiling. RESULTS: Escherichia coli have the highest (28.6%) occurrence rate and Pseudomonas aeruginosa (20.5%) showing significant resistance to beta-lactamase inhibitors (ampicillin, cefuroxime and cefotaxime), and high-level multidrug resistance of more than 15.2% rate to ampicillin, cefuroxime, ceftazidime, tetracycline and imipenem. E. coli and Klebsiella oxytoca, are the highest beta-lactamase, ESBL and AmpC producers encoded with high molecular weight R-plasmid (>11.0 kbp) and significant rate of conjugation and transformational activities. Only 2/14, 1/13 and 1/6 ESBL-type of E. coli, K. oxytoca and Enterobacter cloaca, expressed bla TEM gene, clustering into five different phylodiverse groups with close genomic relatedness with other bacilli. CONCLUSION: This is an indication of clonal dissemination of ESBL bla TEM encoded enteric bacilli having high phylodiverse characteristics through fecal carriage in the Nigerian community which requires public health education, food and environmental hygiene for its prevention.

ABSTRACT

Chrysophyllum albidum G. is a tropical plant and commonly found in Nigeria. It belongs to the sapotaceae family and used in folklore in the treatment of yellow fever, malaria, diarrhea, vaginal and dermatological infections. The study was aimed at investigating the antioxidant properties of this plant by employing the in vitro and in vivo experimental models. The effect of DPPH free radical scavenging activity on the fractions of petroleum ether, ethanol, butanol, ethylacetate, and water of C. albidum was determined. The ethyl acetate fraction was purified in column chromatography to obtain myricetin rhamnoside. Structure elucidation was done by NMR and mass spectroscopic techniques. Furthermore, ethanol extract was administered to five groups of eight rats per group. The animals in the normal group were administered with vehicle alone for 7 days. The positive control animals were given vehicle on the first four days, and with the vehicle and hepatotoxin (CCl(4)) on the fifth, sixth and seventh day. The animals in the treatment category were respectively administered with 500, 1000 and 1500 mg/kg b.w. of extract & distilled water for the first four days, and with distilled water, extract and CCl(4) on the last three days. Animals were subsequently anaesthetized and blood samples were collected for catalase (CAT), malondialdehyde (MDA), reduced gluthathione (GSH) and superoxide dismutase (SOD) assays. The petroleum ether fraction showed the least antiradical activity (4057.5 ± 809.6 g/kg) while ethyl ether exhibited the highest activity (414.4 ± 92.0 g/kg). Myricetin rhamnoside also exhibited an excellent radical scavenging activity (314.1 ± 60.2) which was comparable to the positive control. Result from animal study showed that C. albidum exhibited significant (p < 0.05) differences on the activity of CAT, MDA and GSH. The plant could therefore be employed as sources of natural antioxidant boosters and for the treatment of some oxidative stress disorders in which free radicals are implicated.
