ABSTRACT

Mucor circinelloides is a dimorphic fungus that is a non-pathogen strain belonging to zygomycetes. In this research, a part of hypothetical mechanism on yeast-like cell induction of M. circinelloides in CO2 atmosphere was reported from the viewpoint of gene expression. To explain the relation between the change and the expressions of some genes involved in morphological changes of the strain, these were analyzed on the filamentous and yeast cell by real-time qPCR. The compared genes were Nce103, Ras3, Cyr1, Pde, and Efg1 encoding carbonic anhydrase, GTPase, adenylate cyclase, phosphodiesterase, and elongation factor G1, respectively. In anaerobic grown yeast cell with 70%N2 + 30%CO2, the Nce103 and Ras3 gene expressions decreased to 24 h whereas that of the filamentous cell increased. However, a downstream gene of Cyr1 expression level in the yeast cell was higher than that of filamentous cell. A lower level of Pde in the yeast cell than that of the filamentous cell indicated intracellular cAMP accumulation. The actual cAMP in the yeast cell remained whereas that of the filamentous cell decreased with cultivation. The Efg1 expression level controlling hyphal elongation was suppressed in the yeast cell. The intracellular cAMP accumulation and Efg1 expression regulate hyphal elongation or yeast forming.

ABSTRACT

Spiculisporic acid (SA) is a fatty acid-type biosurfactant with one lactone ring and two carboxyl groups. It has been used in metal removers and cosmetics, because of its low propensity to cause irritation to the skin, its anti-bacterial properties, and high surface activity. In the present study, we report an effective method for producing SA by selecting a high-producing strain and investigating the effective medium components, conditions, and environments for its culture. Among the 11 kinds of Talaromyces species, T. trachyspermus NBRC 32238 showed the highest production of a crystalline substance, which was determined to be SA using NMR. The strain was able to produce SA under acidic conditions from hexoses, pentoses, and disaccharides, with glucose and sucrose serving as the most appropriate substrates. Investigation of nitrogen sources and trace metal ions revealed meat extract and FeCl3 as components that promoted SA production. Upon comparing the two types of cultures with glucose in a baffle flask or aeration bioreactor, SA production was found to be slightly higher in the flask than in the reactor. In the bioreactor culture, sucrose was found to be an appropriate substrate for SA production, as compared to glucose, because with sucrose, the lag time until the start of SA production was shortened. Finally, fed-batch culture with sucrose resulted in 60 g/L of SA, with a total yield of 0.22 g SA/g sucrose and a productivity of 6.6 g/L/day.

ABSTRACT

Stereoscopic depth perception is supported by a combination of correlation-based and match-based representations of binocular disparity. It also relies on both transient and sustained temporal channels of the visual system. Previous studies suggest that the relative contribution of the correlation-based representation (over the match-based representation) and the transient channel (over the sustained channel) to depth perception increases with the disparity magnitude. The mechanisms of the correlation-based and match-based representations may receive preferential inputs from the transient and sustained channels, respectively. We examined near/far discrimination by observers using random-dot stereograms refreshed at various rates. The relative contribution of the two representations was inferred by changing the fraction of dots that were contrast reversed between the two eyes. Both representations contributed to depth discrimination over the tested range of refresh rates. As the rate increased, the correlation-based representation increased its contribution to near/far discrimination. Another experiment revealed that the match-based representation was constructed by exploiting the variability in correlation-based disparity signals. Thus, the relative weight of the transient over sustained channel differs between the two representations. The correlation-based representation dominates depth perception with dynamic inputs. The match-based representation, which may be a nonlinear refinement of the correlation-based representation, exerts more influence on depth perception with slower inputs.

ABSTRACT

Two ß-glucosidases (BGLs 1 and 2) were purified to homogeneity from the extracellular enzyme preparations of the ethanol-fermenting Mucor circinelloides NBRC 4572 statically grown on rice straw. BGLs 1 and 2 are monomeric glycoproteins whose apparent molecular masses (Ms) are around 78 kDa, which decreased by approximately 10 kDa upon enzymatic deglycosylation. Both BGLs showed similar enzyme characteristics in optimal temperature and pH, stability, and inhibitors. They were active against a wide range of aryl-ß-glucosides and ß-linked glucose oligosaccharides. Their amino acid sequences shared 81% identity and exhibited less than 60% identity with the known family-3 BGLs. Considering properties such as reduced inhibition by ethanol, glucose, and cellobiose, low transglucosylation activity, wider substrate range, less binding affinity to lignocellulosic materials, and abundant expression, BGL1 is likely to be more suitable for bioethanol production than BGL2 via simultaneous saccharification and fermentation of rice straw with M. circinelloides.

ABSTRACT

A newly developed quantitative near-infrared spectroscopy (NIRS) system was used to measure changes in cortical hemoglobin oxygenation during the Verbal Fluency Task in 32 healthy controls, 15 subjects with mild cognitive impairment (MCI), and 15 patients with Alzheimer's disease (AD). The amplitude of changes in the waveform, which was quantitatively calculated by a signal processing method, was significantly lower in the frontal, and the bilateral parietal areas in the AD group, whereas that in the MCI group was significantly lower only in the right parietal area. The NIRS system may be a potential tool for the primary screening of AD.
